Abstract

Using decomposers of fungi to increase the composting process is very necessary. The research aims to determine the potential of Trichoderma sp. Gorontalo local isolate as a decomposer of maize stover. The research was carried out from November 2022‒January 2023 at the Biological Agents Laboratory, Agricultural Plant Protection Center, Gorontalo Province Agricultural Department. This research determined the ability of six isolates of Trichoderma, namely TZ11DI1 (T. asperellum), TZ21BN2 (T. breviconvenctum), TZ21BT1 (T. virens), TZ12PO1 (T. ghanence), TZ21DU1 (T. reesei), TZ21LU1 (T. dorothopsis), and control (EM4) to produce cellulase and as a maize stover decomposer. The parameters observed were the clear zone in the isolate growing media as well as the physical and chemical characteristics of the compost produced. The results showed that Trichoderma isolates produced cellulase and potential as a decomposer for maize stover, as indicated by the ability to increase NPK nutrient levels and reduce the C/N ratio of maize stover compost. Keywords: compost, decomposer, maize stover, Trichoderma

Abstract

Witches‘ broom of peanut caused by phytoplasma is a common disease found in Indonesia. Phytoplasma is able to be detected using polymerase chain reaction (PCR) technique. One of important factor which determine the successful of phytoplasma amplification is the DNA availability from fresh tissues. The research was aimed to evaluate some preservation methods of phytoplasma from infected plant samples. The aspects to be evaluated consisted of time (1, 2, 3, and 4 weeks), temperature (-20 °C, 4 °C, and 25 °C), and preservation medium (1X PGB buffer, 3 M NaCl, CTAB buffer, 70% ethanol, non medium, and FTA-card) for storing the fresh phytoplasma infected samples. Good preservation method will optimize the phytoplasma DNA amplification using PCR standard technique followed by nested-PCR. The results showed that preservation of samples at -20 °C, 4 °C, and 25 °C in CTAB buffer was able to maintain the tissue freshness for 4 weeks and was able to provide the DNA of either quality or quantity sufficiently for PCR detection. PCR standard using a primer pair P1/P7 showed that not all of the preserved DNA of phytoplasma were amplified positively. However, standard PCR followed by nested-PCR using primer pair fU5/rU3 was able to increase the DNA detectability. Preserved samples derived from various medium and stored for 4 weeks gave positive results.  This results were in contrary with previous same samples which were detected negatively by standard PCR technique.

Abstract

The Women Farmers Group or KWT is part of the agricultural community group. KWT needs to be empowered to process household waste generated from daily activities. The use of the Trichoderma sp fungus in breaking down organic waste into organic fertilizer in the form of compost. Empowering KWT in processing household waste using Trichoderma spp into compost is one way to support sustainable and environmentally friendly agricultural activities. The aim of this activity is to provide training and practice in processing household organic waste using Trichoderma spp in Bongohulawa village. The methods include counseling, discussions with the participants and followed by a KWT demonstration in processing household organic waste to make organic fertilizer/compost. KWT Mawar participants responded very enthusiastically to the implementation of service activities because of the material and training on the use of household organic waste using the Trichoderma sp fungus is something new for this community group. Demonstration activities produced samples of wet organic waste and dry waste which were stored in sacks and tightly closed for 4 weeks. The results of processing household organic waste are characterized by changes in the texture of the waste which has been crushed and has a blackish brown color. The success of the household waste decomposing household waste into trichocompost certainly motivates every KWT member to adopt a household waste processing using Trichoderma spp to produce trichocompost.
