Abstract

Nosocomial infections occur in hospitals and attack patients who are in the recovery process and have low immunity. The emergence of nosocomial conditions in the hospital environment is caused by bacteria and moist objects or materials, especially in the operating room and even the treatment room for children and babies. One type of bacteria that causes the infection is Pseudomonas aeruginosa. P. aeruginosa has been recognized as an irreversible problem in hospitals due to resistance to 3 antibiotic classes. The content of sweet mango seeds has a high phytochemical content in the form of steroid compounds, terpenoids, flavonoids, alkaloids, phenolics, tannins and saponins. This study aimed to determine the potential of sweet mango seed extract against nosocomial infections. The research method is quantitative experimental, using ethanol extract of sweet fragrant mango seeds with well diffusion and dilution tests (MIC and MBC). The results of the antibacterial activity test measured the inhibition zone formed and tested MIC and MBC. MIC value of sweet fragrant mango seed extract showed at 1.56 mg/mL and 0.78, and at MBC value of 25 mg/mL and 50 mg/mL. The results of sweet fragrant mango seed extract have the potential to be developed as an antibacterial agent, especially against nosocomial infections.

Abstract

Abstract

Copyright©2023 Diabetic wounds often occur in people with diabetes mellitus as a result of impaired tissue perfusion and fungal infections, one of which is excessive dermatophytes that cause tissue death. This study aims to detect dermatophyte fungi in foot wounds of diabetics with diabetes mellitus by the Polymerase Chain Reaction (PCR) method. The type of laboratory observational study with a cross sectional study design, and the sampling technique used is total sampling by swab foot wounds in diabetes mellitus patients. The research method uses the PCR method to detect the presence or absence of dermatophyte fungi characterized by the formation of DNA fragments on electrophoresis visualization. The results showed that from 8 samples detected, 5 samples or 62.5% were positive for dermatophyte fungal infection based on amplification results marked by the formation of an amplicon DNA target band measuring around 700-1000 bp using ITS1 primers (forward: 5' GGT TGG TTT CTT TTC CT 3') and ITS2 (reverse: 5' AAG TAA AAG TCG TAA CAA GG 3'), so it can be concluded that in identifying samples using the PCR (Polymerase Chain Reaction) method has a high sensitivity in detecting fungi so as to allow more specific and more accurate results.

Abstract

A fermented drink produced locally in the Sinjai area that is well known in the local community as well as widely is Minas. Minas, which means a typical Sinjai drink, used to be known as Irex. However, over time this drink was replaced with the name Minas because it was only produced in Sinjai City and used as a typical Sinjai drink has properties and benefits to increase body stamina due to fatigue after doing daily activities or work. Minas are made from cassava tapai, young coconut, eggs, coconut water, sugar water, honey, and milk and can be added with other fruits such as durian. Curved tape is a type of food produced from the fermentation process of substrates or carbohydrate foodstuffs using yeast yeast which belongs to the group of lactic acid bacteria. To determine the type of lactic acid bacteria that play a role in the fermentation process of minas drinks by isolating and identifying morphology using the gram staining method, biochemical tests using catalase tests, gas production and carbohydrate fermentation tests. Furthermore, molecular identification uses 16S RNA. The DNA sequencing results were analyzed with MEGA 6 software with the BLASTn algorithm at NCBI. Based on the results of the identification of lactic acid bacteria, homologous nucleotides were detected with the bacteria Bacillus subtilis, Bacillus and Streptococcus sp which are a group of lactic acid bacteria that are able to ferment carbohydrates to produce lactic acid.
