Abstract

Utilizing cover crops like Passiflora foetida L. (rambusa) can mitigate significant environmental issues in post-coal mining terrain rehabilitation. Plants thriving in extreme environments are known for producing high levels of secondary metabolites with significant biochemical activity. This study sought to assess the antifungal and antibacterial effects of P. foetida root extracts derived from rehabilitated coal mine soil in East Kalimantan. The roots were macerated with solvents of differing polarity (ethanol, ethyl acetate, and n-hexane) for extracting specific fractions. Utilizing the Kirby-Bauer disc diffusion method, the antifungal efficacy was assessed against Candida albicans, Candida tropicalis, and Candida lipolytica at a 60% concentration. Conversely, the antibacterial efficacy was assessed against Shigella dysenteriae, Streptococcus mutans, and Cutibacterium acnes at a 10% concentration. The findings revealed that the fraction consisting of ethyl acetate demonstrated the most potent and extensive antibacterial efficacy. The ethanol extract and ethyl acetate fraction exhibited significant antifungal activity, particularly against C. tropicalis, with inhibitory zones that were similar to those of the positive control, fluconazole. The treatments exhibited significant differences, as confirmed by statistical analysis (ANOVA and Tukey's HSD test). Studies indicate that that P. foetida roots, particularly those from harsh settings, are a significant source of antimicrobial chemicals, with the semi-polar ethyl acetate fraction being the most promising for further development as a natural antibacterial and antifungal agent.

Abstract

This study aimed to know the antimicrobial activity of ethyl acetate extract of Passiflora foetida L. roots against Streptococcus mutans and Candida albicans, two dominant microorganisms in the oral microbiota. Extraction was performed via maceration, followed by phytochemical screening and disc diffusion assay. Phytochemical tests confirmed the presence of flavonoids, alkaloids, and terpenoids. The antibacterial test showed inhibition zones ranging from 11.18 mm (1.5%) to 19.92 mm (3%), with the 3% concentration showing no significant difference compared to amoxicillin (p = 0.872). Antifungal activity was observed starting at 60% concentration, with the highest inhibition zone at 12.47 mm (70%), not significantly different from nystatin (p = 0.067). Statistical analysis using ANOVA and Games-Howell indicated significant differences among several treatment groups. These findings suggest that the ethyl acetate extract of Passiflora foetida L. roots has promising potential as a natural antimicrobial agent with dose-dependent activity against oral pathogenic bacteria and fungi. Abstract

Lawsone (2-hydroxy-1,4-naphthoquinone), the principal naphthoquinone of Lawsonia inermis, has attracted interest as a natural anti-inflammatory/analgesic lead that may offer a safer profile than long-term non-steroidal anti-inflammatory drugs. This structured narrative review synthesises evidence from 14 accessible and highly relevant publications (prioritising 2020 onwards) to clarify the mechanistic basis of its analgesic potential. Direct evidence from preclinical studies indicates that lawsone produces anti-inflammatory and anti-arthritic effects comparable to reference drugs and is associated with improved hepatic and renal markers at effective doses. Mechanistically, two experimentally supported axes predominate: (i) reduced prostaglandin biosynthesis, reflected by significant lowering of serum PGE2 and consistent with COX-2 modulation, and (ii) suppression of NF-κB signalling with downstream reductions in key pro-inflammatory cytokines (TNF-α, IL-1β, IL-6). In contrast, potential modulation of the P2X7 purinergic receptor (P2X7R) remains hypothetical, supported mainly by pharmacological and computational studies of structurally related 1,4-naphthoquinone analogues that bind the P2X7R allosteric pocket and inhibit ATP-driven Ca2+ influx and macropore formation. Overall, the evidence positions lawsone as a promising multi-target preclinical scaffold while highlighting the need for direct P2X7R validation, protein-level pathway confirmation, and translational studies.

Abstract

Fungal infections are a common health problem in tropical regions, including Indonesia, since humid conditions support microbial growth. Candida tropicalis is a pathogenic species frequently identified as the cause of both superficial and systemic candidiasis. The rising resistance to synthetic antifungal agents such as fluconazole and ketoconazole highlights the need for safer and more effective alternative therapies. Propolis, a resinous substance produced by Trigona sp. bees from various plant sources, contains flavonoids, phenolics, and terpenoids with important biological activities, including antimicrobial and antifungal effects. This study aimed to evaluate the effectiveness of ethanol extract of propolis from Balikpapan against the growth of Candida tropicalis using the disk diffusion method. Phytochemical screening revealed the presence of alkaloids, flavonoids, phenolics, tannins, and terpenoids in the extract. Antifungal activity testing showed that concentrations of 80%, 90%, and 100% significantly inhibited fungal growth, with the largest inhibition zone of 27.93 mm observed at 100% concentration. Statistical analysis using ANOVA confirmed significant differences among treatments. Thus, ethanol extract of Trigona sp. propolis from Balikpapan demonstrates strong potential as a natural antifungal agent. These findings support the use of propolis as a natural-based therapeutic alternative for fungal infections and contribute to the development of herbal pharmaceutical products.
