Abstract

The objective of this study was to determine the difference in density of Alona sp in fermented rice straw and unfermented rice straw media. This research was conducted at the Aquaculture Technology Laboratory and the Fish Health, Environment and Toxicology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University, Manado, from March to June 2021.The experiment was based on four treatments, namely fermented rice straw media with Effective Microorganisms-4 (EM-4) solution, unfermented rice straw media, horse manure media and water media. The experimental design used was a completely randomized design (CRD) with 4 treatments, each treatment was repeated 3 times, so there were 12 experimental units. The results obtained are the density of Alona sp. cultured on fermented rice straw media were not significantly different from those cultured on unfermented rice straw media but significantly different from horse manure and water media. There was no significant difference between fermented straw media and unfermented straw media on the increase in density of Alona sp., apparently caused by the fermentation time of EM-4 solution of rice straw which was not long enough.

Abstract

The purpose of this study was to examine the density of life feed Alona sp. on the media of water hyacinth leaf fermented with effective microorganism-4 (EM-4) solution. The method used was an experimental, using one-way ANOVA, with four treatments, namely fermented water hyacinth leaf media, unfermented water hyacinth leaf media, horse manure media, and water, each with three replications. The parameter observed was the density of Alona sp. The result showed that the density of Alona sp. cultured using fermented water hyacinth leaf media was significantly different from those cultured on unfermented water hyacinth leaf media.  It had also been found that the density of Alona sp. high in horse manure media. However, it is recommended to use fermented water hyacinth leaf media, because it is more hygienic. In addition, the availability of horse manure is increasingly rare

Abstract

The purpose of this study was to identify the bacteria Aeromonas sp. in a tilapia (Oreochromis niloticus) culture pond in Matungkas Village, Dimembe District, North Minahasa Regency. Bacterial identification was carried out at the Laboratory of the Fish Quarantine Center for Quality and Safety Control of Fishery Products, Manado. The time of the study was carried out from February 2021 - June 2021. Ten samples of fish were selected from 3 existing ponds for this study with the criteria of fish that have clinical signs of disease infection such as pale body surface, red spots and ulcers. From 10 samples, 19 isolates were obtained to be identified. The bacterial method was carried out based on the results of gram tests and biochemical tests which included: Gram Tests, Motility Test, Oxidase Test, Oxidative Fermentative Test, Tolerance Test to incubation temperature, Tolerance Test to pH of the media, Tolerance Test to the concentration of NaCl in the media and rimler – shotts selective media test. Of the 19 isolates from 10 samples identified, there were 7 isolates that were positive for A. hydrophila, namely: N1.4.1, N2.1.1, N2.1.2, N2.2.1, N2.3.2, N3.3.1, and N3.3.2. Twelve other isolates were negative for A. hydrophila. The characters of A. hydrophila identified are creamy in colour, gram negative, oxidase positive, bacteria tolerance to temperature 4 °C positive, 37 °C positive, 50 °C negative, can move (motile), fermentative, pH tolerance test media pH 3 negative, pH 5 positive, pH 9 positive, pH 11 positive, tolerance test for NaCl concentration 0.5% positive, 3% positive, 5% negative, rimler – shotts positive. All sampling sites (ponds) in Matungkas Village were infected with bacteria.

Abstract

This study aimed to examine the effect of adding ciplukan (Physalis angulata) to feed on the growth of tilapia and to find a dose of ciplukan that was able to increase the growth of tilapia (Oreochromis niloticus). The test fish used were tilapia with an average size of 5-8 cm as many as 300 fish from the Center for Freshwater Aquaculture (BPBAT), Talelu. This research was conducted at the Aquaculture Technology Laboratory, Faculty of Fisheries and Marine Sciences, Sam Ratulangi University. This study used a completely randomized design (CRD) with 4 treatments, where each treatment had 3 replications so there were 15 experimental units, the treatment used was ciplukan flour with different concentrations consisting of: A: 0 g flour/kg feed, B : 10 g flour/ kg feed, C : 15 g flour/ kg feed, D : 20 g flour/ kg feed. The data collected consisted of fish weight to measure the fish growth. The results showed that the addition of ciplukan leaf extract in feed with different concentrations gave significantly different effects on the growth of tilapia with treatment B (10 g) showing the highest fish growth reaching 5.24 g, followed by treatment C (15 g) with the highest growth. reached 4.35 g. While the fish treated with D (20 g) only reached 3.16 g in weight where this value was not much different from the growth of fish in control A. Duncan's further test results showed that treatments B, C, D were not significantly different from each other, but these three treatments were very significantly different compared to fish that were not given ciplukan leaf extract. The study showed that ciplukan leaf extract added to feed gave significantly different effects on fish growth, with the best dose of 10 g extract/kg feed. Thus it can be concluded that ciplukan leaf extract can increase growth of Nile tilapia.

Abstract

This study aimed to determine the effect stocking density on growth and determine the effect on the survival of saline tilapia. The experiment was conducted at the Aquaculture Technology Laboratory and the time of the study was from February to August 2021. The experiment was designed according to a completely randomized design (CRD) with 4 treatments and 3 replications so that there were 12 experimental units. The factors tested were differences in stocking density of tilapia with treatment A: 10 fish/20 liters, treatment B: 15 fish/20 liters, treatment C: 20 fish/20 liters, and treatment D: 25 fish/20 liters. Growth data and survival rate (%) were analyzed using analysis of variance for RAL, then LSD test (Least Significance Different) was performed. Observations of water quality include temperature, DO, pH, and salinity. The result of the analysis of variance showed that the difference in stocking density had a significant effect on growth, but on the survival rate had an insignificant effect. The results of the BNT test on growth showed that treatment A was not significantly different from treatments B and C, but treatments A, B, and C were significantly different from treatment D. The temperature range during the experimental period was between 29 0C – 30.9 0C, DO ranged from 4.07 ppm – 6.80 ppm, pH between 6.78 – 7.95, and salinity 25 ppt

Abstract

This research was conducted to examine the effect of different substrates on the growth of Sea Cucumber Holothuria scabra reared in cages in Bahoi Coastal waters. This research was conducted from April to June 2021 in Bahoi Coastal Waters, West Likupang, North Minahasa. The method used in this research is the observation method with three treatments: A (sand), B (mud), C (seagrass), and three replications. The cages are made of nets size 100 cm x 100 cm x 50 cm as many as 9 cages. The sea cucumber size data was collected every two weeks. Samples of sea cucumber were taken randomly from each cage, left for ± 10 minutes, photographed, weighed, then recorded. Data collection for water quality is carried out every week during the day at the lowest tide, between 12:00 to 14:00. Water quality parameters were measured using a thermometer, refractometer, and litmus paper. Based on the research results, the average absolute weight growth and daily growth rate of sea cucumber reared with mud substrate showed the best results, that is 0.55 g and 0.72%, while the sand and seagrass substrates showed negative values. The results of ANOVA (analysis of variance) showed that there was no significant difference between absolute growth and daily growth rate of Sea Cucumber H. scabra reared on sand, mud, and seagrass substrates.

Abstract

This study aimed to determine the water quality in the floating net cage area in the cultivation of tilapia (Oreochromis nilocitus) and to find out how the condition of the waters in Pematang Limau could support the cultivation activities.  This research was conducted using 3 (three) stations. Water quality data collection was carried out 3 times. The data used included primary and secondary data. Parameters observed in situ included temperature, brightness, current velocity and pH. Ammonia, DO and BOD were analyzed at the Calibration and Health Laboratory of Central Kalimantan in Palangka Raya. Research resulted showed that the overall water quality in the floating net cage area can still be used to maintain tilapia culture. The water quality at each station showed a value that can still be used for cultivation activities except for BOD value that was high.

Abstract

The purpose of this study was to determine the type of ectoparasites and to determine the level of prevalence, intensity, and dominance in tilapia. This research was conducted using an experimental method. Sampling was carried out in floating net cages in Pematang Limau Village, 10 samples of tilapia were taken for each sampling and then examined in the laboratory. During fish sampling, water samples were also taken to analyze the ammonia and dissolved oxygen content. Sample organs including scales, fins, mucus and gills were examined using scraping and biopsy methods. The results showed that there were 3 types of ectoparasites that attack tilapia based on the organs they attack, namely Argulus sp., Trichodina sp. and Oodinium sp. The prevalence of parasites that occur in tilapia shows that fish are very often infected, where the average prevalence of Argulus sp. 35%, Trichodina sp. 45% and Oodinium sp. 55%. The intensity of the parasite that attacked tilapia during the study was in the mild category, namely the intensity value ranged from 1.79 to 18.33 ind/fish while the type of parasite that dominated was Oodinium sp.

Abstract

This research was conducted to examine the survival and regeneration success rate of the Brown Sandfish (Bohadchia marmorata) using the Transverse-fission method in containers placed in nature and controlled. This research was conducted  from June to July 2021, in Bahoi Village, West Likupang, North Minahasa. This research used the Transverse-fission method, in which B. marmorata was tied using a rubber band until cleavage occurred. The container used is a basket covered with a net as many as 6 units, each of which is insulated in the middle so that it becomes 12 parts. Individuals who have been tied, then observed for changes shortly after the binding is done until the individual is separated (every 1 - 2 hours), followed by observing the survival and regeneration of separate body parts (every 6 hours). The results of the study were then analyzed using SPSS 22 version with T-test.  Based on the results of the research, the survival and regeneration success rate of B. marmorata using the Transverse-fission method placed in containers in natural waters was significantly different from that of B. marmorata placed in controlled containers, where the total survival rate of B. marmorata placed in in controlled containers reached 100% while the total survival rate of B. marmorata placed in natural waters only reached 3.33%.

Abstract

This study aimed to determine the effect of different doses of EM-4 in feed on the growth and survival of tilapia. This research was conducted at the Center for Freshwater Aquaculture (BPBAT) Tatelu. The study was carried out from June to August 2021. The experimental design used was a Completely Randomized Design (CRD) with 4 treatments and each treatment had 3 replications. The treatment used was Probiotic EM-4 with different doses consisting of: A: 0 ml/kg feed, B: 15 ml/kg feed, C: 20 ml/kg feed, D: 30 ml/kg feed. The results of the study used the Analysis of Variance (ANOVA) using the SPSS application (22). Giving different doses of EM-4 in the feed gave a significantly different effect on the absolute weight growth of tilapia, but the results also showed that the administration of different doses of EM-4 in solid had no effect on absolute length growth, daily length growth rate, FCR and tilapia survival. The treatment that had an effect on the absolute weight growth of tilapia was found in the treatment with a dose of EM-4 15 mL/kg of feed with an absolute weight growth of 23.89 g.
