Abstract

The use of high-yielding strawberry varieties such as Sweet Charlie needs to be followed by proper propagation methods to produce large amounts of strawberry seeds quickly and free from disease. In vitro propagation techniques can be done through the formation of a callus. This study aims to determine the effect of the combination of NAA and CaP treatment on the in vitro induction of the Sweet Charlie strawberry variety leaf callus. This experiment was arranged using a completely randomized design (CRD) which consisted of 9 treatments, namely concentrations of NAA (0 mg L-1, 5 mg L-1, 10 mg L-1) and CaP (0 mgL-1, 1 mgL-1, 2mg L-1) which was repeated three times. The results showed that using NAA 5 mgL-1 was the best treatment to support callus induction of the Sweet Charlie strawberry variety in vitro. The supporting with the parameter percentage of callus formation reaching 100%, callus appearance time at 13.5 days after planting (DAP), callus diameter of 9.00 mm, callus weight of 0.52 g, and the percentage of crumbly textured callus was 50%.

Abstract

The effects of greenhouse gases on daily life can cause damage to the balance of ecosystems and threaten the survival of living things. One of them is crop failure; food commodities and export products are reduced, causing food shortages and lowering the economic level of the community. The initial objectives of this study were to find or filter methanotrophic, phosphate-solubilizing, and Nitrobacter bacteria on Sorong's agricultural land. The second aim was to determine methanotrophic bacteria's potential, which can also nitrate and solubilize phosphate. The findings of the screening for methanotrophic bacteria, phosphate-solubilizing bacteria, and Nitrobacter bacteria are described in this descriptive study. In addition, isolates of methanotrophic bacteria were confirmed for their ability to decompose phosphate and fix nitrogen. The results of this study were that of the ten samples observed, five samples were detected by methanotrophic bacteria, namely MFa, MFb, MFc, MFd, and MFe samples. Furthermore, phosphate-solubilizing bacteria and Nitrobacter bacteria were found in ten isolated samples. The second conclusion, from five isolates of methanotrophic bacteria, has the potential to decompose or utilize phosphate as an energy source. Meanwhile, of the five isolates, only three isolates could utilize nitrogen as an energy source, namely isolates with MFa code, MFd isolates, and MFe isolates.

Abstract

Fast and applicable seed quality testing methods are needed to support the availability of quality seeds that can be distributed. The Tetrazolium Rapid Test Method is a rapid test method that is currently being implemented in Seed Quality Testing Laboratories. Meanwhile, the Radicle Emergence/RE Rapid Test method is currently being developed to accelerate the supply of quality seeds. This research aims to determine a more precise, efficient, and efficient method. Accurate in predicting corn seed germination in the laboratory and field. This research was carried out in the laboratory using a completely randomized design and in the field using a one-factor randomized block design, namely varieties in the form of ten varieties of corn (Sukmaraga, Srikandi Ungu, Srikandi Kuning, Pulut Uri, Lamuru, Bisi 18, Bisi 2, P32, P35, and Betras 1). The results showed that the Tetrazolium Rapid Test and the RE Rapid Test had a very close correlation in estimating the value of Seed Germination in the Laboratory and the Field. Both methods can be used as Rapid Test methods in Seed Quality Testing laboratories. However, the RE Rapid Test is more economical and practical.

Abstract

This present study investigates the combination of probiotic Lactobacillus sp. and molasses applied to promote growth performance and survival of Nile Tilapia seed. Probiotic could act as an additive agent for fish seed with incomplete digestive organ development as it produces the digestion-support enzyme. The experiment was set based on the formalized fish feed, consisting of A (no molasses and probiotic provision), B (2 r/L molasses), C (108 cfu/mL probiotic), D (108 cfu/mL probiotic + 4 r/L molasses), E (1010 cfu/mL probiotic + 2 r/L molasses) and applied in three replicates each. Fish were reared in cone bottom tanks and were fed according to the treatment (2 times a day in 3 weeks). The growth and survival rate data were collected during the experiment and were analyzed using ANOVA (analysis of variance). The results indicate significant growth and survival of Nile Tilapia seed (P<0,05) generated by the enhancement of probiotics and molasses into fish feed with formalized 1010 cfu/mL probiotic + 2 r/L molasses.

Abstract

The low quality of vannamei shrimp seeds during larval rearing causes vannamei shrimp seeds to be more susceptible to disease, thus affecting production success. This situation can be overcome by increasing the immune response of vanamei larvae through the administration of vitamin C. This study aims to evaluate the effectiveness of Artemia salina bio encapsulation with vitamin C to increase immunity against vibriosis disease of vannamei shrimp larvae. The treatment tried was the addition of vitamin C through A. salina bio encapsulation with four different doses, 20, 30, 40, and 50 ppm as well as control (without vitamin C), each with three replicates using a Completely Randomized Design (CRD). Vaname shrimp seeds were reared from Mysis-PL11 stadia in 50-liter capacity crates, 40-liter volume of media water with a density of 40 fish/liter, then checked Total hemocyte count (THC), Differential hemocyte count (DHC) before and after the challenge test was carried out by using Vibrio harveyii bacteria. The results showed that the effectiveness of the immunostimulant vitamin C through A. salina bio encapsulation to prevent vibriosis had a significant effect on the immune system of Mysis-PL11 stadia vannamei shrimp larvae with the highest THC value of 41 x 104 cells/mm, DHC (66.67% hyaline, 27 .67% semi-granular, 67.67% granular) in the treatment with the addition of 50 ppm vitamin C.
