Abstract

Split-plot mixture-mixture design has been applied for optimization of Phyllanthus niruri chromatographic fingerprint. The design was applied for unreplicated and simultaneous optimization of extraction mixture and chromatographic mobile phase mixture. The whole-plot was extraction solvent contained various combinations of methanol, ethyl acetate and dichloromethane in a simplex centroid with axial design. The sub-plot was reversed phase chromatographic mobile phase in simplex centroid design contained varying proportions of methanol, acetonitrile and acetonitrile:water (55:45 v/v). Each extract was eluted with seven mobile phases of high performance liquid chromatography and detected at 210, 225 and 254 nm. The optimum condition was determined based on the number of peaks. The correlation between extraction solvent, chromatographic mobile phase and number of peak were evaluated with Ordinary Least Square method. The root mean square error of calibration and root mean square error of prediction obtained at 210, 225 and 254 nm were 1.8634 and 4.0076; 2.2220 and 5.2839; 1.5436 and 2.2606, respectively. The optimum condition was obtained when the ethyl acetate extract was eluted with acetonitrile:water (55:45 v/v) and detected at 254 nm. Precision of retention time at optimum condition was excellent, with the relative standard deviation in a range of 0.0698% and 0.3006%.

Abstract

Ketoprofen coated tripolyphosphate modified chitosan nanoparticle was resulted from ionic gelation process by ultrasonication, centrifugation, and spray dry methods. Particle was analyzed by particle Size Analyzer. Dissolution and stability of them have conducted by Row Hansen methods at 37±0.5 °C and 150 rpm and using climatic chamber at 40±2 °C and relative humidity 75±5% for 3 months, respectively. Anti-inflammatory activities have conducted by Plate Reader methods. The result showed that B formulae have particles diameter of 563.1±157.5 nm and entrapment efficiency of 89.663%, respectively. Dissolution behavior showed that ketoprofen restrained released within acid medium and maximum released within base medium of 11.35-15.87% and 45.73-99.25%, respectively. Kinetically, dissolution of ketoprofen within intestinal and gastric pH condition was Korsmeyers-Peppas kinetic models. While, stability behavior showed that B formula has ketoprofen percentage left in nanocapsule after 3 months greater than 90% with water content and shelf life of 19.68%, and 14.19 week, respectively, and degradation kinetic model followed Avrami-Eroveef equation. Anti-inflammation test showed that B formula had the COX-2 inhibitory activity of 90.86% is higher than free ketoprofen.

Abstract

Sinensetin-like molecules have been searched virtually to find a dummy template for Molecular Imprinted Polymer (MIP) synthesis. The aim of this study is to find a novel adsorbent for Solid Phase Extraction (SPE) of sinensetin based on tailored method using dummy template. Virtual searching was done using ChemDB online chemical database containing 5 million commercially available chemicals. Sinensetin as target was converted into SMILES code then it was run with threshold of molecular similarity score of 0.5. There were 90 chemical structures given as the output mainly flavone analogue, three flavonols and several non-flavonoid compounds. The suggested results are three dummy templates that derived from luteolin (flavon) or quercetin (flavonol) with slight alteration by methoxylation process.

Abstract

Preparation and application of platinum composite microelectrode (PCM) for the routine analysis of acetaminophen in pharmaceutical products has been carried out. This electrode was prepare by Pt powder and PVC in 4 mL tetrahydrofuran (THF) solvent and swirled flatly to homogeneous followed by drying in an oven at 100 °C for 3 h. The mixture was placed in 0.5 cm diameter stainless steel mould and pressed at 10 ton/cm2. The cyclic voltammetry were performed in a three electrodes system using PCM as a working electrode, an Ag/AgCl (saturated KCl) as reference electrode and platinum wire as the counter electrode. Electroanalysis of acetaminophen was performed in 0.1 M H2SO4 as an electrolyte. The result of the study showed that the correlation of determination using PCM electrode for electroanalysis acetaminophen was R2 = 0.999. Precision, recovery, LOD and LOQ of the PCM towards acetaminophen were found to be 1.04%, 100.54%, 19.52 mg/L and 65.08 mg/L, respectively. As a conclusion, the methods can be used for routine analysis of acetaminophen in pharmaceutical product. Simplicity of sample preparation and use of low cost reagents are the additional benefit of this method.

Abstract

Fast swelling hydrogels were synthesized from the mixture of acrylamide (AAM)-acrylic acid (AA)-starch (15:5:1 w/v) solution by radiation crosslinking with varying irradiation doses (7.5-12.5 kGy) at room temperature. The copolymers were characterizated by Fourier transform infra red spectroscopy (FTIR) and Differential scanning calorimetry (DSC). It was found that hydrogels showed superabsorbent properties with higher swelling capacities (~400 g/g) and reached at a very short time (3 min). There was no significance difference between the effect irradiation dose on swelling of hydrogels. The hydrogels were also found to sensitive to the ionic strength of medium. The FTIR spectra and thermogram DSC of hydrogels were confirmed the crosslinking occurs in the hydrogels.

Abstract

One new dolabellane (1) and two known diterpenoids stolonidiol (2) and clavinflol B (3) have been isolated from the ethyl acetate extract of the Indonesian soft coral Anthelia sp. A new compound 1 exhibited a moderate cytotoxicity against NBT-T2 cells at 10 µg/mL, while known compounds 2 and 3 showed cytotoxicity at 1 and 0.5 µg/mL, respectively. Structure of the new compound 1 was elucidated by interpretation of NMR spectroscopic data (1D and 2D NMR data) and mass spectrometry (ESIMS data) as well as comparison with those of related ones. This finding should be useful for anti cancer drug development of the promising dolabellane-types compound.

Abstract

Preparation of calcium oxide from Achatina fulica shell has been carried out systematically by decomposition for 3 h at various temperatures i.e. 600, 700, 800 and 900 °C. Formation of calcium oxide was characterized using XR diffractometer. The calcium oxide obtained with the optimum temperature decomposition was characterized using FTIR spectroscopy to indicate the functional group in the calcium oxide. The results showed that XRD pattern of materials obtained from decomposition of Achatina fulica shell at 700 °C is similar with XRD pattern of calcium oxide standard from Joint Committee on Powder Diffraction Standards (JCPDS). The IR spectra of calcium oxide appear at wavenumber 362 cm-1 which is characteristic of CaO vibration. Application of calcium oxide from Achatina fulica shell for synthesis of biodiesel from waste cooking oil results in biodiesel with density are in the range of ASTM standard.

Abstract

Ionic-Imprinted Chitosan/Chlorella biomass sorbent (IICCb) has been synthesized for selective adsorption of Cd(II) adsorption. IICCb was prepared by coating Cd(II)-complexed-chitosan hydrogel on the biomass surface followed by cross-linking procedure. Cd(II)-imprinting ions were then eluted using Na2EDTA solution to provide a specific template for binding sites of Cd(II). Batch adsorption was carried out as function of initial pH, contact time, and initial concentration of the Cd(II) solution. Result showed that the optimum adsorption of Cd(II) was found at pH 4-6. Study of pseudo-second order kinetic showed that the adsorption of Cd(II) on IICCb went faster than that on Ionic-Imprinted Chitosan (IIC) or Non-Imprinted Ionic Chitosan/Chlorella sorbent (NIICCb). The maximum Cd(II) adsorption capacity as obtained from Langmuir adsorption isotherm was found to be 53.76 mg/g on IICCb, that was comparatively higher than that on IIC (44.44 mg/g) or on NIICCb (51.02 mg/g) adsorbent.

Abstract

In search of bioactive anti-cancer compounds from natural products has been found isobractatin from n-hexane extract of Garcinia eugenifolia. Isolation was done through the stages of maceration and column chromatography. Elucidation of the structure was analyzed by spectroscopy of UV-Vis, FTIR, LC-MS, NMR proton and carbon one and two dimension data. The results found three compounds isolated from G. eugenifolia. The three compounds were stigmasterol, prenylated xanthone isobractatin and its epimer (in a mixture). The mixture of isobractatin and its epimer showed cytotoxic activity against breast cancer cell line T47D with IC50 = 17 µg/mL.

Abstract

Determinations of the temperature effect on glycerol nitration processes have been done with two methods: the HYSYS predictions and the laboratory experiment. The aim of this study was to compare prediction method and laboratory experiment method. The highest equilibrium conversion from HYSYS predictions was obtained in the range of equilibrium temperature of 10 to 20 °C. The laboratory experiments also described that nitration of glycerol with nitric acid should be carried out at reaction temperature of 10 to 20 °C. HYSYS that was used to predict the results of experiments in the laboratory can reduce the laboratory work with minimize the range of operating conditions studied. HYSYS exactly predict temperature of nitration of glycerol. The difference in conversion between two methods due to the equipment that was used in the experiments, procedure of experiments and the accuracy of analysis.
