Abstract

Disease infection is one of the limiting factors that affect productivity in aquaculture and has caused economic losses. Luminescent vibrios and motile aeromonas septicemia (MAS) are diseases caused by Vibrio harveyi and Aeromonas hydrophila bacteria, respectively. Certain plants have antimicrobial compounds and can potentially be used to treat the diseases, such as Centella asiatica. In the present study, the crude leaves extracts of C. asiatica were examined for its antibacterial potential using methanol solvents against V. harveyi and A. hydrophila bacteria. Different concentrations of 50 mg/mL and 100 mg/mL were checked for its antibacterial activity. The crude extract was also tested for phytochemistry content and LC50 using Brine Shrimp Lethal Assay. The crude extracts of C. asiatica showed a remarkable antibacterial activity with inhibition zone of 10.57 mm against A. hydrophila and 21.14 mm against V. harveyi. The phytochemistry test result showed that C. asiatica leaves contain alkaloid, phenol, and tannin compounds. The acute lethal concentration (LC50) of C. asiatica after 24 hours exposure to the extract mixture was 254 mg/L. The results confirmed the potential use of C. asiatica extracts as a source of antibacterial compounds.

Abstract

Interactive effects between dietary inorganic phosphorus (IP) and phytase (P) on mineral digestibility and vertebral mineral content were investigated in a 30 days feeding trial followed by three weeks digestibility trial with Japanese flounder, Paralichthys olivaceus. Eight experimental diets were formulated based on two levels of dietary Ca at 0% and 0.2% combined with either 0% or 0.25% of dietary IP and either with 0 and 2,000 fytase unit (FTU)/kg of phytase in diet, respectively. Result indicated that digestibility of total phosphorus significantly increased by three dietary compounds where the highest was observed in fish fed diet contained 0.25% IP and 2,000 FTU phytase/kg and dietary Ca also included in diet. Significant interaction was only detected between dietary IP and P on this parameter. Supplementation of IP and Ca not phytase significantly improved Ca digestibility. Ca digestibility was very poor when dietary IP and Ca were not supplemented in diet even with when phytase supplemented in diet. There was significant interaction between dietary IP and Ca on Ca digestibility. Vertebral total phosphorus, Ca, and Mg content as well as Ca:P ratio were significantly enhanced by dietary IP and phytase. Dietary Ca has significant effect only on vertebral total phosphorus. Interaction between dietary IP and Ca was significantly found on vertebral Ca content and Ca:P ratio. No significant second-order interaction was observed among the three dietary mineral on overall parameters. Based on total phosphorus and Ca digestibility as well vertebral phosphorus content found in this study, dietary IP, Ca, and phytase at rate of 0.25%, 0.2%, and 2,000 FTU phytase/kg diet, respectively are needed to supplement in diet for a better mineral absorption and bone mineralization.

Abstract

Mariculture industry has been developed progressively in Indonesia, where its impact on the surrounding aquatic environment is inevitable. The particulate wastes produced such as excess food and feces will be discharged from a marine farm and dispersed into the surrounding areas. This process could lead to organic enrichment of the receiving seabed sediment and gradually degrade water quality and disturb local benthic community as well as the aquatic ecosystem of the area. This study focused on determining the level of organic enrichment of sediment underneath four currently-active farms in Pegametan Bay, North Bali, Indonesia. The results showed that high accumulation of organic matters was evident in all sampling farms indicated by a significantly high particulate organic matter (POM) between 75.20 ± 2.57 and 92.97 ± 0.59%, and low redox values between -217.41 ± 2.74 and -343.57 ± 1.48 mV. A visual-based assessment also determined that the sediment had a silt and muddy texture with deep grey to black colorization with noticeable strong H2S odor. In this case, further investigations and monitoring efforts are needed in the near future to ensure the best management programs for sustainable farming and ecosystem both fish farmers and environmental regulators such as local government.

Abstract

Goldfish (Carassius sp.) is the ornamental fish which has been managed its culture by fish farmers. The objective of the research is to improve the colour quality of goldfish juveniles through enrichment of feed using shrimp head meal as source of carotenoid. The research was conducted at The Research and Development Institute for Ornamental Fish Culture, Depok, using completely randomized design with four treatments of shrimp head meal ratio, 0% (control), 5%, 10%, and 15% in feed, the feed formulation contained isoprotein (30%), and isolipid (15%) and three replications of each treatment. Ten juveniles of Carassius sp. with body weight of (1.08±0.02 g) stocked in aquarium with water volumes of 20 L. The growth and colour quality in qualitative standard using TCF (Toca Colour Finder) were examined, while total carotenoid content on feed were measured by spectrophotometry method. The colour quality of goldfish were also measured at three of fins those were dorsal, ventral, and caudal. The result showed that no significant different on growth and survival rates among the treatments. Based on the present research, the optimal colour improvement to goldfish was by addition of 10% shrimp head meal in feed.

Abstract

The most important factor to high mortality rate in larval rearing is feeding success in early larval stage related to kind and size of natural live food. Copepod basically is the main source of natural food in the open ocean having some advantages such as smaller size of nauplii, attractive movement and high nutritional value. Observation on population dynamic of harpacticoid copepod Euterpina acutifrons was carried out using 5-L plastic bucket with initial density 100 ind./L. Green algae Nannochloropsis sp. was added to culture media at density of 50,000 cells/mL as a basic feed and additional feeds given were wheat flour (group A) and chicken liver (group B) at a rate of 50 mg/bucket. The result showed that there was no difference on population pattern in both groups where the incubation time took eight days to hatch, from nauplii to the copepodite stage was three days and from copepodite to adult copepod took five-to-six days. The differences came up from population number: in group (A) the highest number of copepod-bearing-egg was only 133 ind., nauplii production up to 62,833 ind. and number of copepodites was 22,333 ind. lower compared to group (B) with the highest copepod-egg was 308 ind., nauplii was 113,333 ind. and copepodite was 51,167 ind. The conclusion pointed out that the kind of food did not influence population pattern (quality) but gave effect to population growth.

Abstract

Lymphocystis virus has been implicated as the cause of severe infection, mortality, and economic loss in farmed giant snakehead (Channa striata) or gabus fish in Mandiangin South Kalimantan. In Kalimantan, the fish is locally known as Haruan fish and considered to have health-related benefits to human due to its high albumin content. This study aimed to determine the LCDV in gabus fish through histopathological and PCR investigation. Infected LCDV fish have a cluster of warts growths in the skin or fin of a walleye. Infected fishes with an average of total length of 15 cm and weight of 150 g were collected from local net cages and earthen ponds. They were transferred into a laboratory, anesthetized, and then subsequently killed by organ dissection. The infected fish tumors were aseptically cut out for histological study and PCR detection. Primer used was forward GII F: 5 ‘TGG GAT TCC AAY GGT CAA TTA-3’ with target band of 468 bp (for genotype-IIi LCDV) and primer reverse R: 5’TTA GAT TAT TGG GCA GCG TT-3' with target band of 250 bp (for genotype-II LCDV) and GIII F: 5' AGG AAA TAA CCG CAG TA GAA TGCA. Lymphocytosis in infected fish showed multifocal to diffuse white, round, firm, papilloma or tumor-like nodules on the skin of the body, fins, eyes, and mouth. The hypertrophied cell was surrounded by a thick smooth hyaline capsule. Stress condition caused by high population density, nutrition deficiencies, decreased dissolved oxygen, suboptimal water quality, and handling may increase the appearance of LCDV symptoms. The result of the present study revealed that histopathology and PCR could be used to diagnose LCDV infection.

Abstract

Possible use of pangasiid hybrids in aquaculture might generate potential impacts on wild populations. Therefore, rapid identification tools in the field such as growth rate are urgently needed. This study examines morphological characters and growth performance of P. djambal and P. hypophthalmus and their reciprocal hybrids. A detailed morphological study analysed 32 morphometric measurements and 5 meristic counts on hybrids of Pangasius djambal and P. hypophthalmus. Morphometric analysis and meristic counts showed that the reciprocal hybrids have intermediate characters except for gill rakers number which were lower than that of parental species. In general, the hybrids have tendency to be like P. hypophthalmus rather than P. djambal. The only typical character P. djambal appearing in hybrids is teeth shape, both vomerine and palatine. It was shown that the true hybrids have seven pelvic fin rays. Eight months of growth comparison in earthen ponds showed that the hybrids have a better performance for specific growth rate than the parental stock.

Abstract

Pathogenic Vibrio spp. have been implicated as being one of the major causes of the disease problems in penaeid shrimp culture. For many pathogens, the outcome of the interaction between host and bacterium is strongly affected by the bacterial population size. Efforts for rapid molecular detection is to isolate specific genes which was owned by the pathogenic luminescent Vibrio and used it as the marker molecular in the rapid diagnosis of the disease. Haemolysin and gyrase gene known as one of the specific genes possessed pathogenic bacteria including Vibrio. The objectives of this study were to determine the specificity of designed specific primer from local isolate. Specificity test was performed for haemolysin primer (IAVhF1 and IAVhR1), gyrase primer (IAGyrF1 and IAGyr2) and commercial detection kit. The specificity test comparing the PCR result of pathogenic Vibrio DNA template as positive control, and DNA template from non pathogenic luminescent Vibrio, as well as pathogenic non Vibrios. The result showed that at annealing temperature of 60°C, haemolysin primer was more specific in detecting pathogenic Vibrio for penaeid shrimp than that of gyrase primer and commercial detection kit.
THE SURVIVAL OF TRANSPLANTED CORAL ON PYRAMID-SHAPED FISH SHELTER ON THE COASTAL WATERS OF KELAPA AND HARAPAN ISLANDS, KEPULAUAN SERIBU, JAKARTA Ofri Johan

Abstract

Research was carried out in August 2008 at two sites within Marine Protected Areas (MPA) in Kelapa and Harapan Islands of Seribu Islands, Indonesia. The purpose of this research was to compare the mortality rate of coral transplanted on the upper and base of pyramid-shaped concrete fish shelters. Fish shelters were placed on the seabed of Kelapa Island which is characterized by sandy seabed and Harapan Island which is characterized by silt. Both sites are within Marine Protected Areas. The survey recorded mortality rate of transplanted coral and percentage of live coral coverage. The research also sampled fish populations within the MPA sites and pyramid areas so that the two sites can be compared for their fish biodiversity. The data were analyzed by using T-Test Student of SPSS 11.5 computer program to compare between two locations, and upper and base of transplanted position. After one year, the average mortality rate among transplanted coral species in Kelapa Island was 34%. However, the mortality rate of transplanted coral in Harapan Island was slightly higher than in Kelapa Island: 42%. The transplanted coral at the top of the fish shelter had higher survival rate (39%) compared to the base of the fish shelter (24%). Fish populations in Kelapa Island consisted of 1,119 individuals from 35 species. This was much higher than in Harapan Island where the research found only 655 individuals from 27 species. Fish populations found around the fish  shelter area in Kelapa Island consisted of 125 individuals from 25 species. This was lower than the fish population in Harapan Island, where 206 individuals from 23 species were recorded.

Abstract

This study was aimed to produce recombinant growth hormone (rGH) from giant grouper (Epinephelus lanceolatus), giant gouramy (Osphronemus gouramy) and common carp (Cyprinus carpio) and compare their bioactivity potential by means of inducing the growth hormone of juvenile Nile tilapia (Oreochromis niloticus) as the model. DNA fragment encoding mature GH protein of giant grouper (El-mGH), giant gouramy (Og-mGH) and common carp (Cc-mGH) was amplified by PCR method. The purified PCR products were ligated to pCold-1 to generate pCold/El-mGH, pCold/OgmGH, and pCold/Cc-mGH protein expression vector, respectively. Each of the expression vectors was transformed into the Escherichia coli BL21. E. coli BL21 was cultured using 2xYT medium and protein production was induced by cold shock at 15±1oC for overnight. The inclusion bodies of E. coli transformants containing protein expression vector were isolated by sonication method, and rGH production was analyzed by SDS-PAGE. Juvenile of Nile tilapia of average body weight of 12.41±3.28 g was intramuscularly injected once a week for 4 weeks with 1 μg inclusion body containing rGH per gram fish body weight. The result showed that rGH in molecular weight of about 25 kDa was obtained. Fish injected with rGH of El-mGH, Cc-mGH and Og-mGH grew 20.94%, 18.09%, and 16.99% faster, respectively, compared with the control. This result indicated that the three rGH produced in E. coli possessed biological activity when tested on Nile tilapia and further research is needed to find its effect on the growth of other aquaculture fish species.
