Abstract

RingkasanPenggunaan langsung fosfat alam (FA) ke dalam tanah sebagai sumber pupuk P telah dilakukan selama bertahun-tahun melalui beberapa macam cara penggunaan. Kualitas FA di Indonesia umumnya rendah dan ketersediaan bahan baku yang berkualitas untuk produksi pupuk fosfat terlarut relatif terbatas. Beberapa mikroba asal tanah yang dapat melarutkan fosfat anorganik telah banyak dilaporkan. Namun, informasi yang tersedia tentang mekanisme pelarutan P dari FA lokal asal Indonesia dan P anorganik oleh Aspergillus niger BCC F194 belum banyak diteliti. Satu seri penelitian laboratorium telah dilaksanakan untuk mengetahui kemampuan A. niger BCC F194 melarutkan P. Evaluasi agronomi FA lokal (FA Cileungsi dan Madura) di rumah kaca juga telah dilakukan. A. niger BCC F194 dapat melarutkan sumber P sukar larut, yaitu FA Cileungsi dan Madura, serta senyawa Ca3(PO4)2 dan AlPO4. Kelarutan P anorganik tersebut berhubungan dengan peningkatan aktivitas proton (H*) yang menyebabkan penurunan pH medium dan produksi asam organik. Asam organik utama yang dihasilkan oleh A. niger BCC F194 dalam medium cair Pikovskaya yang dimodifikasi adalah asam oksalat (3.75 mM), asam sitrat (2.0 mM), dan asam glukonat (0.9 mM). Kelarutan FA Cileungsi lebih besar dibandingkan dengan FA Madura, dan kelarutan Ca3(PO4)2 lebih besar dibandingkan kelarutan AlPO4. Tidak ada korelasi antara kelarutan P anorganik dengan aktivitas enzim fosfatase, walaupun aktivitas enzim fosfatase cukup tinggi terdeteksi dalam medium. Satu formula biosuperfosfat telah berhasil dirakit dengan mereaksikan FA lokal dengan supernatan kultur cair (SKC) pengganti asam sulfat. Hasil percobaan padabibit kakao, karet dan kelapa sawit di rumah kaca menunjukkan bahwa prototipe pupuk biosuperfosfat dengan bahanbaku FA Cileungsi dan Madura bentuk granul maupun serbuk, memiliki nilai efektivitas agronomi yang relatif menyamai SP-konvensional. SummaryThe direct application of rock phosphate (RP) to soils as a source of phosphorus (P) fertilizer has been employed with varying popular methods over the years. The RP in Indonesia has low quality for plant fertilization and the availability of the raw material with good quality for production of soluble phosphate fertilizers is limited. Many common soil microbes that can dissolve insoluble inorganic phosphate have been extensively studied. However, there is little information on mechanism of P-solubilization from local RP of Indonesia and inorganic P by Aspergillus niger BCC F194 isolated from tropical acid soils. A laboratory study was conducted to determine the ability of phosphate solubilization A. niger BCC F194. Agronomic evaluation of bioactivated local RP, i.e. Cileungsi and Madura phosphate rocks (CRP and MRP) for direct application in greenhouse experiment was also conducted. A. niger BCC F194 was able in solubilizing different types of hardly-soluble phosphates, i.e. CRP and MRP, Ca3(PO4)2, and AlPO4 compounds. Inorganic P solubilization was directly correlated to the organic acid production and increasing proton (H+) activities causing pH to decrease and production of organic acid. The major acidic metabolites produced by A. niger BCC F 194 in modified liquid culture Pikovskaya medium were oxalic acid (3.75 mM), citric acid (2.0 mM), and  gluconic acid (0.9 mM). The solubilization of Cileungsi RP was higher than that of Madura RP, and the solubilization of Ca3(PO4)2 was better than that of AlPO4. No correlation between solubilization of inorganic P and enzyme activities, although high activities of phosphatase enzyme were detectable in the medium. A biosuperphosphate formula had been constructed by reacting local RP with liquid culture supernatant (LCS) replacing sulfuric acid. Results of cocoa, rubber, and oil palm seedlings experiments in greenhouse indicate that both granular and powder biosuperphosphate prototypes commonly had a comparable relative agronomic effectiveness value to that of the conventional SP.

Abstract

SummaryHeavy metal in waste water potentiallycauses environmental pollution. Generally,heavy metal pollutions come from metalplating, textile, latex-rubber goods, and otherindustries. The process of latex-rubber goodindustries uses heavy metal in the form ofZnO as accelerator for rubber vulcanizationprocess, so that Zn 2+ ion exists in wastewatereffluents in concentration as much as 300 ppm,whereas the maximum limit allowed is 2.5 ppm.The chemical way generally used to decreaseZn 2+ concentration in wastewater effluents isby adding bases, NaOH or Ca(OH) 2 until pHreached 11, hence this metal is precipitated asits hydroxide. However, the way is done, isvery high cost and has a risk of the emergencesecondary pollution caused by excess base. Analternative way to absorb Zn 2+ consideredinexpensive is by using biosorbent in the formof Saccharomyces cerevisiae biomass frombioethanol industrial waste. The research wasconducted using artificial wastewater withZn 2+ ion concentration of 300 ppm and the pHwas adjusted to the range between 3-7.Biosorption was conducted by addition of freeS. cerevisiae biomass as well as byimmobilized cells on filter paper. Observationwas carried out for Zn 2+ concentration aftercontact time of two and five hours. The resultsof the research indicated that free andimmobilized S. cerevisiae biomass couldabsorb Zn 2+ metal and decreased itsconcentration from 250-300 ppm to 20-50 ppm.The optimum contact time was reached at onehour, while optimum sorption process occurredat pH 5. At low concentration, less than20 ppm S. cerevisiae biomass absorbed lessZn 2+ The NaOH-treated biomass showed bettersorption capabilities compared to cells treatedby formaldehyde or heat treatments. Thecontinue experiment showed the high capacityof biomass treated with NaOH to absorb Zn 2+ ,until concentration 24,02- 47,95 ppm in thefirst sampling and 1,15-10,99 ppm in thesecond sampling. Combination adsorptionprocess using charcoal and zeolite couldadsorp remain concentration of Zn 2+ , so thatcould reached the limit concentration-allowed.RingkasanLogam berat di dalam air limbahmerupakan penyebab pencemaran lingkunganyang potensial. Pencemaran logam berat padaumumnya berasal dari industri penyepuhanlogam, tekstil, barang jadi lateks, serta industrilain. Pada proses industri barang jadi lateksdigunakan logam berat dalam bentuk ZnOsebagai akselerator proses vulkanisasi karet,sehingga ion Zn 2+ terbawa dalam air limbahindustri barang jadi dengan konsentrasimencapai 300 ppm, sedangkan ambang bataskonsentrasi yang diperbolehkan maksimaladalah 2,5 ppm. Cara kimia yang umum di-gunakan untuk menurunkan kandunganZn 2+ dalam air limbah adalah dengan caramenambahkan basa, umumnya NaOH atauCa(OH) 2 , sampai pH sekitar 11, sehinggalogam berat ini diendapkan sebagai hidroksida-nya. Namun demikian, cara ini sangat mahaldan beresiko munculnya pencemaran sekunderakibat kelebihan basa. Salah satu alternatifyang murah untuk penyerapan Zn 2+ adalahmenggunakan biosorben berupa biomassaSaccharomyces cerevisiae yang berasal darilimbah pabrik bioetanol.Penelitian dilakukan dengan mengguna-kan air limbah artifisial yang mengandung ionZn 2+ dengan konsentrasi 300 ppm. Limbahartifisial diatur pHnya antara 3-7. Biosorpsidilakukan dengan menambahkan biomassaS. cerevisiae bebas maupun yang diamobilisasidengan kertas saring. Pengamatan dilakukanterhadap kandungan Zn 2+ setelah waktu kontakdua dan lima jam. Hasil penelitian menunjuk-kan bahwa biomassa S. cerevisiae bebasmaupun amobil mampu menyerap logam Zn 2+dan menurunkan konsentrasinya dari 250-300 ppm menjadi 20-50 ppm. Waktu kontakoptimum dicapai setelah satu jam, sedangkanproses sorpsi optimum terjadi pada pH 5.Biomassa S. cerevisiae kurang efektifmenyerap logam Zn 2+ pada konsentrasi rendah,di bawah 20 ppm. Perlakuan biomassa meng-gunakan NaOH menunjukkan kemampuanpenyerapan yang lebih baik jika dibandingkandengan yang diperlakukan menggunakanformaldehida dan pemanasan. Percobaan padaaliran kontinyu yang menggunakan biomassayang diperlaukan menggunakan NaOH,menunjukkan bahwa limbah artifisial Zn 2+dapat diturunkan sampai konsentrasi 24,02-47,95 ppm pada sampling pertama, dan 1,15-10,99 ppm pada sampling kedua. Kombinasipenyerapan menggunakan arang aktif danzeolit dapat menyerap sisa Zn 2+ mencapai batasyang diperbolehkan.

Abstract

SummarySolid wastes of bagasse and empty fruitbunch (EFB) respectively from sugarcane andpalm oil mill in Indonesia are abundant. Nowdays, up to now these solid wastes have not yetbeen optimally utilized so that the added valueis still very low and even cause an environ-mental problem. Research on bioconversion ofbagasse and EFB with two culture of white-rotfungi (WRF) i.e., Omphalina sp. and Pleurotusostreatus to produce ligninolytic enzymes wasconducted to provide added value to thislignocellulosic waste. Production of extracellular enzymes from WRF was not onlydetermined by the type of isolate but also theculture condition. This research was aimed todetermine the optimum culture condition ofsolid state fermentation in producing lignino-lytic enzymes at laboratory scale. In thisresearch, WRF was examined for ligninolyticproducing enzymes (laccase, lignin peroxidase/ LiP and Mn-peroxidase / MnP), using mediaconsisting of bagasse and EFB separately asmain substrate with supplementation of ricebran, Cu 2+ with or without rice bran. Theobservation was based on their growth andligninolytic enzyme activities. Characteristicsof optimum pH of LiP, MnP and laccaseactivity were also determined. The resultsshowed that addition of supplement was notable to increase the Cu 2+ growth of myceliaespecially in the first and second months but inthe third month the addition of supplementenhanced the mycelia growth. The growth ofmycelia on the addition of Cu 2+ with or withoutrice bran significantly lower compared to thecontrols both of Omphalina sp. and P. ostreatusin bagasse and EFB. The optimum pH oflaccase, MnP, and LiP activities was five bothfor Omphalina sp. and P. ostreatus at EFBand bagasse. Omphalina sp. was better thanP. ostreatus in producing laccase on bagasseand EFB without any supplementations. Thehighest laccase activity showed by P. ostreatuswith bagasse and EFB media treated with Cuand Cu + rice bran. Supplementation withCu 2+ was more effective in increasing laccaseactivity than rice bran. Activities of Li-P onbagasse and EFB for the two WRF cultureswere significantly influenced by supple-mentation of both of rice bran and Cu 2+ . Li-Pactivity on EFB was slightly higher than thaton bagasse. Mn-P activity was not influencedby rice bran, Cu 2+ or the combination of both.However, these enzymes activities on EFBwere higher compared to bagasse especiallyfor P. ostreatus. Suplementation of Cu wasenhance the activity of laccase and LiP both ofP. ostreatus and Omphalina sp in baggasse andEFB though inhibited the growth of those fungiespecially in the initial growth.RingkasanLimbah padat bagas tebu dan tandankosong kelapa sawit (TKKS) masing-masingdari proses pengolahan gula tebu dan minyaksawit di Indonesia jumlahnya melimpah dansampai saat ini belum mendapat penangananyang efektif sehingga nilai tambahnya masihsangat rendah dan bahkan mengganggulingkungan. Penelitian biokonversi limbahpadat bagas tebu dan TKKS menggunakan duaisolat fungi pelapuk putih (FPP) yaituOmphalina sp. dan Pleurotus ostreatus untukproduksi enzim ligninolitik dilakukan untukmeningkatkan nilai tambah limbah ligno-selulosa tersebut. Penelitian ini, mengujiaktivitas enzim ekstraseluler dari FPP antaralain lakase, lignin peroksidase (LiP), dan Mn-peroksidase (MnP) dari dua spesies FPP yaituOmphalina sp. dan P. ostreatus. Penelitianbertujuan menetapkan kondisi optimum mediafermentasi untuk produksi enzim ligninolitikdari bagas tebu dan TKKS sebagai substrat dankarakterisasi pH optimum enzim ligninolitikdari dua FPP yaitu Omphalina sp. danP. ostreatus. Pengamatan dilakukan berdasar-kan laju pertumbuhan dan aktivitas enzimligninolitik. Enzim lakase, MnP, dan LiPdiekstraksi dan dikarakterisasi pH optimumaktivitasnya. Hasil penelitian menunjukkanbahwa pemberian suplemen menghambatpertumbuhan miselia pada satu dan dua bulanpertama inkubasi, namun laju pertumbuhanmiselium khususnya pada perlakuan pemberianCu 2+ dan Cu 2+ + dedak meningkat tajam padabulan ketiga setelah inkubasi. Pertumbuhanmiselium Omphalina sp dan P. ostreatus padamedium yang ditambah Cu 2+ dan Cu 2+ +dedaklebih rendah dibandingkan dengan kontrol.Pada inkubasi tiga bulan, aktivitas optimumlakase, MnP dan LiP diperoleh pada pH 5, baikuntuk Omphalina sp. maupun P. ostreatusyang diekstrak dari bahan lignoselulosa bagastebu dan TKKS. Aktivitas lakase dariOmphalina sp. lebih tinggi daripadaP. ostreatus pada substrat TKKS dan bagastebu tanpa suplementasi. Pemberian suplemenberupa Cu 2+ dan dedak atau kombinasinyameningkatkan aktivitas lakase baik pada bagastebu maupun pada TKKS. Aktivitas lakasetertinggi ditunjukkan oleh isolat P. ostreatuspada medium bagas tebu dan TKKS padaperlakuan pemberian Cu 2+ dengan atau tanpadedak. Aktivitas lakase nampaknya lebihdipengaruhi oleh penambahan Cu 2+ dibanding-kan dengan pemberian dedak. Aktivitas LiPbaik pada bagas tebu maupun TKKS untukkedua FPP yang diuji pada perlakuanpenambahan dedak dan Cu nyata lebih tinggidibandingkan dengan aktivitas LiP yangdiekstrak dari medium tanpa penambahansuplemen. Aktivitas LiP pada TKKS lebihtinggi dibandingkan dengan pada bagas tebukhususnya untuk P. ostreatus. Sedangkanaktivitas MnP tidak dipengaruhi penambahandedak dan Cu 2+ demikian pula kombinasikeduanya. Aktivitas MnP yang diekstrak dariTKKS lebih tinggi dibandingkan denganaktivitas MnP yang diekstrak dari bagas tebukhususnya untuk P. ostreatus. PenambahanCu 2+ meningkatkan aktivitas lakase dan LiPP. ostreatus dan Omphalina sp yang ditum-buhkan pada bagas dan TKKS walaupun ionlogam ini menghambat pertumbuhan keduaJPP ini khususnya pada awal pertumbuhan.

Abstract

SummaryRootstocks and scions interaction causesdifferent responses within individuals of scionfrom the same clone. The objectives of thisresearch were to determine compatible anduncompatible combinations of micrograftingbetween Cinchona succirubra A, andC. succirubra B with C. ledgeriana QRC205and Cib5 clones, based on anatomy structureand SDS-PAGE protein bands pattern ofstem at union area between rootstock andscion. The research was arranged in aCompletely Randomized Design, withrootstock/scion combinations CSA/QRC205,CSA/Cib5, CSB/QRC205, CSB/Cib5 and as acontrols were the combination of CSA/CSA,CSB/CSB, QRC205/QRC205, Cib5/Cib5, succiand ledger seedlings with the same age. Effectof rootstocks on scion was studied based onanatomy structure of the union area betweenrootsocks and scions and SDS-PAGE proteinbands pattern. The results showed that theunion of stem between rootstocks and scionwas initiated by callus formation, cellsdifferentiation and the vascular vesselsformation. The anatomy of stem union area ofCSA/QRC205 as a compatible combination ofrootsotck and scion was the same asunmicrografted plantlet. At uncompatiblecombination CSB/Cib5 showed the formationof stone cells as a line along stem cyrcle atunion area and heavely callus formation atoutside of rootstock and scion stems. SDS-PAGE protein bands pattern from thecompatible combination was the same asplanlet control. On the otherhand, from theuncompatible combinations CSB/Cib5 werefound protein degradation and the formation ofnew proteins with molecular weight 21 and 30kD.RingkasanInteraksi batang bawah dengan batangatas menimbulkan berbagai keragaman responsantar individu batang atas dari klon yang sama.Tujuan penelitian ini adalah untuk me-netapkan kombinasi yang kompatibel denganyang tidak kompatibel antara kina klonCinchona succirubra A, dan C. succirubra Bdengan C. ledgeriana klon QRC205 danCib5, berdasarkan anatomi dan pola pitaSDS-PAGE protein daerah pertautan antarabatang atas dengan batang bawah. Percobaandisusun dengan Rancangan Acak Lengkapkombinasi yang diuji adalah CSA/QRC205,CSA/Cib5, CSB/QRC205, CSB/Cib5 dengankombinasi kontrol CSA/CSA, CSB/CSB,QRC205/QRC205, Cib5/Cib5, tanaman kinasucci dan ledger tanpa sambungan denganumur yang sama. Pengamatan dilakukanterhadap struktur anatomi daerah pertautanantar batang bawah dengan batang atas danpola pita protein SDS-PAGE batang atas. Hasilyang diperoleh menunjukkan bahwa tahapanpemulihan daerah pertautan penyambunganbatang bawah dengan batang atas diawalidengan pembentukan kalus, diferensiasi sel danterbentuknya jaringan ikatan pembuluhgabungan. Kombinasi antar batang bawahdengan batang atas yang kompatibel yaituCSA/QRC205 memperlihatkan strukturanatomi daerah pertautan yang serupa denganstruktur anatomi batang planlet yang tidakdisambung. Pada kombinasi yang tidak kom-patibel yaitu CSB/Cib5 pada daerah pertautanterbentuk sel-sel batu berbentuk garis yangmemanjang di tengah lingkaran batang. Disamping itu pada daerah pertautan terbentukkalus yang berlebih ke arah luar baik padabatang atas maupun batang bawah. Padakombinasi yang kompatibel pola pita proteinsama dengan planlet kontrol. Pada kombinasiyang tidak kompatibel yaitu kombinasiCSB/Cib5 terjadi degradasi protein danpembentukan protein baru dengan beratmolekul sekitar 21dan 30 kD

Abstract

SummaryThe utilization of lignocellulolitic emptyfruit-bunches of oil-palm (EFBOP) material asstraw mushroom (Volvariella volvacea) mediacould produce short chain organic carbon andnutrients which may be available for growth ofplant. This research was aimed to determine theeffect of spent mushroom media amendmentderived from empty fruit bunches of oil palm(SMEB) as organic fertilizer on the growth andnutrient uptake of oil palm seedling. Theexperiment was conducted at greenhouse usingacid soil. Oil palm seedling was grown in 60 x 50cm polybags. The experiment was carried out toexamine 20 treatments i.e. combinations of fourlevel of SMEB (0, 25%, 50%, and 75% w/w) andfive dosages of conventional fertilizer (0, 25%,50%, 75%, and 100%) recommended dosages.The experiment was conducted using completerandomized design with factorial type. Theresearch showed that the amendment of SMEB atthe level of 25% could increase the height ofseedling, fresh weight of root, stem, and seedlingsas well as dry weight of oil palm stem. However,for leaf dry weight, a higher addition of SMEB upto 50% was needed. Fertilizing at 25% ofrecommended dosage could increase the seedlingheight, fresh weight of stem and leaf while forseedling fresh weight and leaf number, higheraddition of fertilizer up to 50% was needed.Significantly higher uptake of K and Mg wasobserved on the amendment of 75% of SMEB.The higher addition of SMEB (up to 75% w/w)did not decrease any growth parameters andnutrient uptake. However, the addition of 100%of recommended chemical fertilizer tended todecrease various growth parameters and uptakeof N, P, K, and Mg of oil palm seedling.RingkasanPenggunaan bahan lignoselulosa sebagaimedium jamur merang (Volvariella volvacea)diketahui dapat menghasilkan senyawa karbonrantai pendek demikian pula hara tersedia,sehingga diduga bahan ini dapat digunakansebagai pupuk organik untuk tanaman. TKKS sisa medium jamurmerang (TKSJ) sesuai dosis perlakuan dicampurdengan tanah dan selanjutnya bibit kelapa sawitditanam di polibag berukuran 60 x 50 cm.Percobaan dilakukan untuk menguji 20 perlakuanyang merupakan kombinasi empat tingkat TKSJ(0, 25%, 50% dan 75% b/b) dan lima dosis pupukkonvensional (0, 25%, 50%, 75% dan 100%)dosis rekomendasi. Percobaan dilakukan meng-gunakan rancangan acak lengkap dengan polafaktorial. Hasil percobaan menunjukkan bahwapemberian TKSJ pada tingkat 25% dapatmeningkatkan tinggi bibit, bobot basah akar,batang, dan bibit serta bobot kering batangkelapa sawit. Namun, untuk peubah bobot keringdaun diperlukan pemberian TKSJ yang lebihtinggi yaitu 50%. Pemupukan pada dosis 25%rekomendasi meningkatkan tinggi, bobot basahbatang dan daun sedangkan untuk jumlah daundan bobot basah bibit diperlukan dosis pupuk50%. Serapan hara K dan Mg nyata lebih tinggipada pemberian 75% TKSJ. Pemberian TKSJpada jumlah yang tinggi (hingga 75% b/b) tidakmenyebabkan penurunan berbagai peubah per-tumbuhan dan serapan hara, namun pemberianpupuk 100% rekomendasi cenderung menurun-kan berbagai peubah pertumbuhan dan serapanhara N, P, K, dan Mg bibit kelapa sawit.

SummaryHeteromeric acetyl-CoA carboxylase (ht-ACCase) is one of key enzymes in palm oilbiosynthesis. Isolation and characterization ofthe gene is an important step in metabolicengineering to increase palm oil content andquality. The objective of this research was toisolate DNA fragment of gene encoding biotincarboxyl carrier protein (BCCP) subunit of ht-ACCase from three different oil palm types(Simalungun, Hibrida and Backcross) andinvestigate the variation of its DNA sequence.Total RNA was isolated from the mesocarp ofoil palm. DNA fragment encoding BCCP wasamplified by means of Reverse TranscriptasePolymerase Chain Reaction (RT-PCR) usingspecific primers with total RNA as a template.The products of RT-PCR were then purifiedfrom the gel, cloned and sequenced. The DNAsequences were analyzed for their homologiesto BCCP gene using BlastN and aligned todetect the sequence variability using ClustalWprogram from BioEdit. The results show thatone of the two RT-PCR products at about 300bp was highly homologous with the geneencoding BCCP from Glycine max, Brassicanapus and Arabidopsis thaliana. Nucleotidesequences of that BCCP fragments from thethree types of oil palm displayed some degreesof variability. Abstract

SummaryPromoter is a regulator of geneexpression for a phenotype or trait carried bythe gene. In the structure, a promoter locatedbeyond the 5’ end of the open reading frame ofthe gene on which its expression is regulated.This research aimed to isolate the DNAfragment flanking TcLFY at the 5’ end and toanalyze whether the fragment has charac-teristics of the promoter, primarily the coremotifs of promoter. Using Genome Walkingtechnique, DNA fragments flanking the TcLFYgene at its 5’ end was isolated. Analysis of theDNA sequence was done using onlinecomputer software accessible through web sitewww.softberry.com and an entry sequence ofthe flanking DNA fragment along with the 2.5kb TcLFY sequence. The result indicated thatthe flanking fragment has core motifs for apromoter at proper positions, which are TATAbox at position –80, CAT boxes (CCAAT) at -387 and –626, and GC boxes that are known asUAS were found at the -323 and –537positions. To obtain a conclusive result, thispromoter sequence needs to be furtherexamined to confirm its function.RingkasanPromoter merupakan pengendali ekspresigen untuk memunculkan fenotipe atau karakteryang dibawa oleh gen tersebut. Di dalamstrukturnya, promoter umumnya terletak didaerah ujung 5’ gen yang dikendalikanekspresinya. Tujuan penelitian ini adalahmendapatkan fragmen DNA yang mengapitgen pengendali pembungaan kakao (TcLFY)dan menganalisisnya apakah memilikikarakteristik promoter, yaitu mengandungmotif-motif inti (core motifs) dari promoter.Dengan teknik Genome Walking, fragmenDNA pengapit gen TcLFY di ujung 5’ dapatdiisolasi. Analisis sekuen menggunakanperangkat lunak komputer online (www.softberry.com) dengan input data fragmentersebut ditambah gen TcLFY 2,5 kb dibawahnya, mengindikasikan adanya beberapamotif inti promoter pada posisi yang sesuai,yaitu kotak TATA pada lokasi –80, kotak CAT(CCAAT) di posisi -387 dan –626, dan kotakGC yang merupakan UAS dijumpai padalokasi -323 dan –537. Untuk memperoleh hasilyang bersifat konklusif, sekuen promoter inimasih perlu diuji fungsinya.

SummaryArbuscular mycorrhizal (AM) fungi areecologically important for most vascular plantsfor their growth and survival. AM fungi areobligate symbions, and conventionally propa-gated by pot culture with a certain host plants.This papers describes the establishment ofmonoxenic cultures of Gigaspora sp. andAcaulospora sp in association with excised RiT-DNA transformed carrot roots and tomatoin vitro plants. Spores of Gigaspora sp. andAcaulospora sp. was cultured in monoxenictomato, carrot and hairy root of carrot in vitrocultures. The objectives of these studies were toobtained dual culture ( axenic and hairy root)for germination, sporulation, and infection ofGigaspora sp. and Acaulospora sp. Theresearch consisted of (i) host plant selectionwith high compatibility for hairy rootformation, (ii) media selection for potato andcarrot hairy root culture, (iii) hairy root ofGranola potato and carrot in dual culture, and(iv) germination, sporulation, and infection ofGigaspora sp. and Acaulospora sp. in vitroculture. The results showed that hairy rootsinduction were obtained from Granola,Atlantik potato and carrot in MS, B5 andWhite media. Granola, Atlantik potato andcarrot hairy root grow well in MS and Whitemedium, respectively. In dual culture media(MM media) hairy root of carrot grow well, buthairy root of Granola potato were inhibited.Germination, sporulation of Gigaspora sp.and Acaulospora sp. and root infection byboth CMA could be maintained in dual culturewith host carrot, tomato plants and carrothairy root culture in MM mediaRingkasanCendawan Arbuskular Mikoriza (CMA)secara ekologi berperan penting untuk ke-langsungan hidup tanaman. CMA adalahsimbion obligat, dan secara konvensional di-perbanyak dengan kultur pot menggunakantanaman inang tertentu. Tulisan ini menjelas-kan kultur monoksenik Gigaspora sp. danAcaulospora sp. berasosiasi dengan kultur akarrambut tanaman wortel dan tomat yangdiinokulasi dengan Ri T-DNA. Spora dariGigaspora sp. dan Acaulospora sp. dikultur-kan secara monoksenik in vitro dengantanaman tomat, wortel dan kultur akar rambutwortel. Tujuan penelitian ini adalah untukmendapatkan kultur ganda (aksenik dan akarrambut) untuk perkecambahan, sporulasi, daninfeksi Gigaspora sp. dan Acaulospora sp.Penelitian terdiri atas (i) seleksi tanaman inangdengan tingkat kompatibilitas tinggi untukpembentukan akar rambut, (ii) seleksi mediumuntuk kultur akar rambut wortel, (iii) akarrambut kentang Granola dan kultur gandawortel, dan (iv) perkecambahan, sporulasi,serta infeksi Gigaspora sp. dan Acaulosporasp. dalam kultur in vitro. Hasil yang diperolehmenunjukkan bahwa induksi kultur akarrambut diperoleh dari kentang Granola,Atlantik dan wortel dalam medium MS, B5 danWhite. Akar rambut kentang Granola, Atlantikdan wortel tumbuh baik dalam medium MSdan White. Akar rambut kentang dapat tumbuhbaik dalam medium kultur ganda, yaitumedium MM. Sebaliknya pertumbuhan kulturakar rambut kentang dalam medium yang samamengalami hambatan. Perkecambahan, sporu-lasi Gigaspora sp. maupun Acaulospora sp..serta infeksi akar oleh kedua jenis CMA dapatdilakukan dalam kultur ganda dengan tanamaninang wortel, tanaman kentang serta dengankultur akar rambut wortel dalam medium MM.

SummaryIn temporary immersion system (TIS),plant materials are exposed to the medium fora short time, therefore they are more exposedto the air and a lack of oxygen frequentlyexperienced by a liquid culture can be avoided.This experiment was conducted to determinethe procedure for callus proliferation up tosomatic embryo germination of oil palm(Elaeis guineensis Jacq.) in TIS culture.Embryogenic calli of oil palm clone MK 638from Marihat Research Institute were culturedon solid medium in the dark culture room andthen used as materials for TIS. Immersion timefor all cultures was three minutes every sixhours. Callus proliferation was conducted inDF liquid culture with 5 mg/L 2,4-D and0.1 mg/L kinetin with transfer interval of 4, 6and 8 weeks. The treatments for somaticembryo maturation were kinetin and ABA,whereas for somatic embryo germination wasIBA, kinetin and GA 3 . The results show thatthe best transfer interval for callus proli-feration was four weeks. In this treatment therelative growth rate of callus was0.38 g/g/week. Somatic embryo initiation fromthe callus was done in DF mediumsupplemented with 1 mg/L 2,4-D and 0.1 mg/Lkinetin. The percentage of somatic embryowas 80% based on biomass fresh weight afterthe fourth subculture. The addition of 0.5 mg/Lkinetin and 0.05 mg/L ABA improved somaticembryo maturation of oil palm; the averagenumber of somatic embryos at advanced stages(torpedo and cotyledonary) was 16.3 embryosper flask. The addition of 2 mg/L IBA and0.5 mg/L kinetin in DF medium with half-strength macro-salt enhanced significantly thegermi-nation of somatic embryos. GA 3 at0.1 mg/L increased the total number ofgerminants.RingkasanPada sistem perendaman sesaat (SPS),bahan tanam hanya terpapar sebentar dalammedium sehingga paparan dengan udara lebihlama dan kekurangan oksigen yang seringterjadi pada kultur cair dapat diatasi. Penelitianini bertujuan menetapkan prosedur untukperbanyakan kalus embriogenik sampai denganperkecambahan embrio somatik kelapa sawit(Elaeis guineensis Jacq.) dalam kultur SPS.Kalus embriogenik kelapa sawit klon MK 638yang diperoleh dari Balai Penelitian Marihatdiperbanyak pada medium padat di ruang gelapyang kemudian digunakan sebagai bahan untukkultur cair SPS. Lama perendaman semuakultur di SPS diatur tiga menit denganfrekuensi setiap enam jam. Perbanyakan kalusdalam medium cair DF dengan 2,4-D 5 mg/Ldan kinetin 0,1 mg/L dilaksanakan denganinterval subkultur 4, 6 dan 8 minggu.Perlakuan pematangan embrio somatik adalahkinetin dan ABA sedangkan perlakuan untukperkecambahan embrio somatik adalah IBA,kinetin dan GA 3 . Hasil penelitian menunjuk-kan bahwa untuk proliferasi kalus embriogenikkelapa sawit, interval subkultur terbaik adalahempat minggu. Pada perlakuan ini laju tumbuhrelatif kalus mencapai 0,38 g/g/minggu.Inisiasi embrio somatik dari kalus dilakukanpada medium DF ditambah 2,4-D 1 mg/L dankinetin 0,1 mg/L. Persentase embrio somatikmencapai 80% dari total bobot basah biomassasetelah subkultur keempat. Penambahan kinetin0,5 mg/L dan ABA 0,05 mg/L meningkatkanpematangan embrio somatik kelapa sawit; rata-rata jumlah embrio somatik fase lanjut (torpedodan kotiledon) adalah 16,3 embrio per bejana.Penambahan IBA 2 mg/L dan kinetin 0,5 mg/Lpada medium DF dengan setengah garammakro meningkatkan perkecambahan embriosomatik secara nyata. Abstract

SummaryFungal treatment by using white-rot fungito reduce a wide variety of herbicide com-pounds is a specialized bioremediation pro-cess. A laboratory experiment was conductedto determine the ability of Phanerochaetechrysosporium, Ceriporiopsis subvermispora,and Pleurocybella porrigens and seven white-rot fungi isolated from a native of tropicalenvironment to grow on yeast malt extractglucose (YMG) agar containing highconcentration of (I) 2,4-dichlorophenoxy aceticacid, (R) glyphosate, and (G) paraquat. Thedata indicated that P. chrysosporium couldgrow on YMG media containing 5000 ppm of(I) 2,4-D, whereas BPBPI 02/04 isolate onYMG 250 ppm of (R) glyphosate or (G)paraquat. Relative values of growth inhibitionof these fungi are 81.1; 27.8; and 50.0%respectively. Biodegradation capability ofherbicides by candidate inoculants in soil-sandmedia was also determined in greenhouseexperiment by using peanut, sorghum, corn,and Borreria alata as bio-indicators. Peanutand B. alata were found to be the bestresponsive seedlings as bio-indicator on thepresence of (I) 2,4-D herbicide in soil-sandmedia.RingkasanTeknologi bioremediasi dengan fungipelapuk putih (FPP) digunakan untuk me-reduksi sejumlah senyawa herbisida. Dari data yangdiperoleh, diketahui bahwa Ph. chrysosporiummemiliki kemampuan tumbuh dalam mediumpadat YMG yang mengandung 5000 ppm (I)2,4-D dan isolat BPBPI 02/04 pada 250 ppm(R) glifosat dan (G) parakuat dengan nilaihambatan pertumbuhan relatif terhadap kontrol(HPR) masing-masing 81,1; 27,8; dan 50,0%.Pengujian isolat terpilih terhadap kemampuanmendegradasi herbisida di dalam mediumtanah dan pasir juga dilakukan di rumah kacadengan menggunakan kacang tanah, sorgum,jagung, dan Boreria alata sebagai bioindikator.Kacang tanah dan B. alata memberikan responterbaik terhadap keberadaan herbisida (I) 2,4-Ddi dalam medium tanah dan pasir .
