Abstract

In this study a High Performance Liquid Chromatography (HPLC) has been used for determination of rubraxanton on bark extracts Garcinia spp (Garcinia mangostana, Garcinia Cowa, Garcinia griffitii, Garcinia dioica and Garcinia forbesii). HPLC system consisted of C-18 reversed phase column with a length of 250 mm, diameter 4.6 mm, 20 mL injection volume, mobile phase methanol: water (gradient system with polarity) and flow rate of 1ml/min. Rubraxanton levels obtained in this study; 9.161% for G.mangostana, 6.942% for G.cowa, 6.762% for G.dioica, 0.499% for G.forbesii and 0.229% for G.griffitii. The method has been validated for specificity, linearity, accuracy, precision, limits of detection (LOD) and limits of quantitation (LOQ). The linearity of the method can be seen from the regression coefficient r=0.9996 with a linearity range from 1.72 to 55 ug/ml. Recovery of rubraxanton in the extract of G.mangostana was between 99.61 to 101.08%. Intra and inter day precision showed relatively small level of standard deviation (lower than 2%). Limit Of Detection (LOD) and Limit Of Quantitation (LOQ) are 0.55 ug/ml and 1.82 ug/ml respectively.Keywords : rubraxanton, bark extracts, Garcinia spp.

Abstract

The used of bacto agar in microbiological studies increased tremendously, however, up till now, to fullfill its high demant the scientist were stil relying on imported bacto agar eventhough domestic production of bacto agar was as goog and reliable as those produced commercially overseas. The current study we focuseon the production of bacto agar from red algae Gracilaria verrucosa using fell press technique; followed by quality analysis of the product. Red algae samples were collected from two different locations (Bekasi and Subang). Quality of the product was tested for its content, acid insoluble as content, overall as content, pH, gel strength and its ability to be use as culture media to culture Escherichia coli and Staphylococcus aureus. Microbiological test was performed via pour plate. The results clearlydemonstrated that red algae sample from Bekasi produced bacto agar that meets the criteria of commercial bacto agar. It has 10.3% water content, 3.9% overall as content, 0.4% acid insoluble as content, pH of 7.3 and gel strength of 600.8-602.8 g/square cm.Keywords : Bacto agar, red Algae, Gracilaria verrucosa, Microbe media

Abstract

This study is aimed at optimizing the dose of cyclophosphamide in thrombocytopenia rat model. A suitable drug dosage that coulds induce thrombocytopenia rat model. A suitable drug dosage that could induce thrombocytopenia in rat was then investigated. Animal model of 25 rats were randomly divided into five groups: Normal (group A), Cyclophosphamide low dose (group B), medium dose (group C), high dose (group D), very high dose (group E) with each of 5 rats. The four dosage groups were given 25, 50, 100, 150 mg/kg BW cyclophosphamide, respectively. Cyclophosphamide was given by subcutaneous injection once a day for 3 consecutive days. All groups were under investigation for 8 days. The result suggested that a decrease in the platelet count, white blood cell, and mean corpuscular volume of all group cyclophosphamide induced at the 7th day were significantly different than that of Normal. The platelets count was reduced but fluctuated greatly, all of the rats dies in group E at 7th day and group D at 8th day. Though all 4 dosages successfully initiated thrombocytopenia as the platelets number dorpped at the 7th day, the low dose was considered to be a suitable one that was of high efficacy and low toxicity. Thus, using Wistar rats challenged by subcutaneous injection of cyclophosphamide 25 mg/kg per day for 3 consecutive days showed one simple, feasible and stable rat thrombocytopenia model that could be used for phamacodynamic test of the drugs which are supposed to have platelet increasing effect. Abstract

The aim of this study was to prepare floating tablet dosage forms using coprocessed of kappa and iota carrageenan and pregelatinized cassava starch propionate (PPSP) as excipients. In this study, tablets were made by wet granulation method and using famotidine as a model drug. Several formulations of floating tablets were prepared using varying composition of the excipients coprossed carragenan with a certain ratio. The swelling and bupyancy of the floating tablets were evaluated. Furthermore, the drug release from the floating tablets were studied and analyzed using several models of kinetic equations. The result showed that formula A with excipients coprocessed carragenan (1:1) as much as 60% with 10% HPMC produce the best formula with floating lag time 11.42±1.53 minutes and total floating time for 22 hours. The formula also revealed a profile of controlled drug release and approached to Higuchi kinetics model and the non Fickian diffusion mechanism.Keywords : carragenan, PCSP, coprocess exipients, floating tablets.

Abstract

The morbidity and mortality of cervical cancer is high in the world. To improve the outcome of cervical cancer therapy, an appropriate approach should be given to the patients. The study purpose was to describe the pattern of treatment in cervical cancer patients in Prof. Dr. Margono Soekardjo Public Hospital. A cross sectional descriptive study was done in 150 patients data which was collected from patients medical records. The patterns of treatment in cervical cancer patients of Prof. Dr. Margono Soekardjo Public Hospital was dissection therapy 0.67% in stadium III-IIIB, radiotheraphy 59.32 % in stadium II-IVB, chemotherapy 12.0% in stadium III-IIB and chemoradiation 10.01% in stadium I-IIIB. Many of chemotherapy drugs given to cervic cancer were the combination of cisplatin-doksorubisin-siklofosfamid in 66.6% and attached diseases were anemia 66.29%. Abstract

Nanopraticles can be prepared by several methods and the ionic gelation method is the easiest one. Verapamil hydrochloride is a drug which used as antiarrhythmic, antiangina, and antihypertension therapy. Nevertheless, bioavailability of orally administered verapamil is very low, only about 10 to 23%. Therefore, verapamil hydrochloride was prepared as nanoparticles dosage form to increase its  bioavailability. The purpose of the present study was to optimize ionic gelation method of chitosan and sodium tripolyphosphate to obtain the best nanoparticles formulation. Nanoparticles were obtained from four different methods (formula A, B, C and D). Particle size distribution, zeta potential, entrapment efficiency, morphology, and fourier transform infra red spectrum of each nanoparticles formula were characterized. The chosen formula was formula D which has 62.8 nm of size, 59.15% of entrapment efficiency, ±25.46 mV of zeta potential, spherical shape, and the ionic interaction was confirmed by FT-IR spectrum. The result showed that chitosan-tripolyphosphate succesfully produce the verapamil hydrochloride nanoparticles by ionic gelation method.Keywords: chitosan, ionic gelation, nanoparticles, sodium tripolyphosphate, verapamil hydrochloride

Abstract

Oyong (Luffa acutangula L.Roxb) seed is a traditional medicinal plant containing flavonoids, saponins, polyphenols and tannins which empirically used to treat diabetes mellitus. Combination of natural product along with conventional drugs has found to be beneficial in certain diseases, such as diabetes mellitus. This research aimed to investigate hypoglycemic effect of combination dose of oyong seed infuse and glibenclamide as well as metformin. Diabetic condition on animal model was conducted by induction of Alloxan as well as by glucose tolerance method. The data obtained were analyzed by one way ANOVA (p<0.05). The result showed that lowering blood glucose effect of oyong seed infusion was higher than that of glibenclamide and metformin on  aloxan induced diabetic mice. Only oyong seed infuse in single administration gave significant lowering effect of blood glucose level on glucose tolerance test.Keywords : Luffa acutangula seeds, alloxan induced diabetic test, glucose tolerance test.

Abstract

Formulation of chewable tablets of ethanolic extracts of Carica papaya L leaves and Morinda citrifolia L fruits and investigation of the activity towards liver damage induced by tuberculosis drugs has been performed. The rats were devide into six groups. The normal control without any treatment, negatif control were given INH 10 mg/200g-RIF 10 mg/200g, positif control were given INH-RIF and methicol, and the treatment group were given INH-RIF with chewable tablets of papaya extracts 120mg/200g and morinda extracts 20mg/200g of three tablets formula with PVP 1%, 3% and 5%. Each group were treated every day for 27 days. The measuring to monitor bilirubin serum, ALT and AST levels on 0,14th, 21st and 28th day. The study showed that the group that given chewable tablets of combination of papaya extract 120mg/200g and morinda extract 20mg/200g can prevent the occurrence of liver necrosis with necrosis value of 19,92% (PVP 3%) and 19,90 (PVP1%), compared to the negative control of 41.78% as well as reducing the value of ALT, AST and bilirubin. The tablet formula that can be received on the best of taste and most effective as hepatoprotektor is the chewable tablet with PVP 3%.Keywords : Hepatoprotector, Carica papaya L., Morinda citrifolia L., Isoniazid, Rifampicin

Abstract

Sea cucumber Stichopus sp. is one of the marine life that has the potential to be developed as a source of antibacteriak agents. Research on the antibacterial activity and toxicity test of sea cucumber Stichopus sp. aims to determine the antibacterial activity and the toxicity level of sea cucumber Stichopus sp. extract against Artemia salina. The method used for antibacterial activity test was the agar diffusion method, while toxicity tests using the Brine Shrimp Lethality Test (BSLT). The result showed that the extract of sea cucumber Stichopus sp. has antibacterial activity against Staphylococcus aureus, Bacillus subtilis and Vibrio eltor. Extracts of sea cucumber Stichopus sp. are active against BSLT test marked with LC50 values of less than 1000 ppmKeywords : Sticopus sp., antibacterial activity, toxicity test.

Abstract

This study aimed to find out hte rational in the use of antibiotic and antifungal of the patients with sexually transmitted infections at community health centers at banyumas district. Type of research was observational descriptive with cross sectional study and the data was taken retrospectively from medical records of patients. From all patients  who entered the study population, 112 patiens positive vulvavaginal candidiasis, 57 patients positive cervicitis and 39 patients positive bacterial vaginosis. Average age of patients affected by STIs were 24 years, female. Based on patients complaints, symptoms, and the laboratory results, we have found that candidiasis patients were treated with nystatin, cervicitis with a combination of azithromycin and cefixime, whereas patients with bacterial vaginosis were treated with metronidazole. The use of antibiotic and antifungals as the treatment of sexually transmitted infections at community health centers at Banyumas district is based on parameters of the right medicine, right dose, proper indications, appropriate patient, and at the lowest cost to them and always available to their community are 100% rational.Keywords : rationality, antibiotic, sexually transmitted infections
