Abstract

In addition to antihypertensive regimen, medication adherence plays an important role in therapeutic management of hyperensive patients. This research is aimerd to measure adherence to antihypertensive therapy in hypertensive patients at primary health center, and to investigate patient's characteristic associated with such adherence. Hypertensive patients attending two primary health centers in Sleman Yogyakarta during January-March 2011 were recruited as respondents in this research. Adherence to natihypertensive therapy was measured using MMAS (Morisky Medication Adherence Scale) containing 8 questions, and a respondent with MMAS score ≥ 6 was categorized as adherent one. Statistical analysis using Chi-square has been done to investigate respondent characteristic associated with adherence. Out of 215 recruited respondents, 62.3% were adherent to antihypertensive therapy. Respondent characteristic associated with adherence are educational background level, monthly income, presence of other chronic disease, and antihypertensive regimen. Keywords : hypertension, medication sdherence, MMAS, primary health center

Abstract

The research aimed to investigate the free radical scavenging activity of ethyl acetate fraction of roselle calyces infusion fermented by Lactobacillus casei strain Shirota against free radical agent, DPPH (2,2-di-phenyl-2-picrylhydrazyl). In sequence, the roselle calyces were extracted by infusion method with concentration at 5% w/v. The infusion was fermented using Lactobacillus casei strain Shirota during 3x24 hours at 37⁰C. As regard the fermentation process, the result of that was extracted using ethyl acetate. Free radical scavenging activity was measured using DPPH method with unfermented-ethyl acetate fraction as control negative. As the result, both fermented and unfermented-ethyl acetate fraction exhibited free radical scavenging activity at 370.68 ppm and 685.49 ppm, respectively. These results suggest that antioxidant activity of fermented-ethyl acetate fraction could be 1.85 greater than unfermented-ethyl acetate fraction of roselle calyces infusionKeywords : Hibiscus sabdariffa L., Lactobacillus casei Shirota strain, free radical scavenging, DPPH

Abstract

The aims of this study was to determine the immunostimulant potential ethanolic extract of roselle calyx through the proliferation of lymphocytes and phagocytosis ability of macrophages Balb/c mice in vitro. Dried calyx of roselle were extracted using 96% ethanol by maceration method is further polyphenols assay are qualitatively and quantitatively. Vitro test using with immunostimulants are seeing an increase in lymphocyte proliferation using the MTT method and phagocytosis activity macrophages with latex beads. Lymphocyte proliferation is expressed by the difference in absorbance values between treatment and control groups, where as the ability of phagocytosis is expressed as an index of phagocytosis and the SFA. Concentration of extract used was 10-1000 μg/mL. Ethanolic extract of roselle calyc containing polyphenols with total polyphenol content (TPC) of 2.757%. Ethanol extract of roselle calyx enhanced immune response at low concentrations. Concentration  of 100 μg/mL is the optimal concentration that increased proliferation of lymphocytest. The highest index of phagocytosis and SFA are at concentrations of 150 μg/mL and 250 μg/mL. It can be concluded that ethanolic extract of roselle calyx has potential as immunostimulants. Keywords : roselle, immune response, macrophage phagocytosis, lymphocyte proliferation, immunostimulant.

Abstract

Preparative High Performance liquid chromatography (HPLC) today is one of the popular methods to isolate anthocyanin from extract containing anthocyanins. The working principle of preparative-HPLC can be applied to the laboratory standard tupe of HPLC, so it can be used also to isolate the main anthocyanin compounds from anthocyanin mixtures. The function of fraction collector on preparative HPLC equipment were replaced by opening the mobile phase line after the exit of the detector and hold out the solution manually on the standard type of HPLC. Resolution value between the peaks and the computation time when cuts in the chromatogram peak collection mobile phase containing the analyte is of concern in these activities. The appearace of a single peak in the chromatogram of the isolates showed that the technique can be applied for the simple and fast technique of anthocyanins isolation. Keywords : isolation, anthocyanin, Ficus padana Burm.f, Preparative-HPLC, standar type of HPLC.
