Abstract

Bawang dayak is a medicine plant which has been known potential as an antioxidant. To isolate potential secondary metabolites as antioxidant needs many bawang dayak plants to be extracted. Utilization of endophytic microbes as a source of secondary metabolites will make the production of natural medicine raw materials is more efficient and environmentally friendly. This research was aimed to investigate secondary metabolites of endophytic fungi from bawang dayak leaves as antioxidant substance. The method of this research was a direct seed inoculating technique on the surface of Potato Dextrose Agar directly. Identification of fungi morphology by using macroscopic (visual) and microscopic (slide culture) method. The results of endophytic isolation of this research got four fungi isolates. Isolate is fermented with shake method, then it is extracted with ethyl acetate. The extract is continued for Antioxidant activity assessment used DPPH method. Secondary metabolite ethyl acetate extract of four isolates had a potention as antioxidant. Highest antioxidant activity was showed by ethyl acetate extract from d.IEP.4.2 isolate has IC50 at 82,9750 µg/mL which has been classified as strong antioxidant.

Abstract

Rheumatoid arthritis is an autoimmune disease characterized with chronic joint inflammation. Chronic inflammation leads to liver function abnormality. Previous study showed that Physalis angulata L. had antiinflammatory, hepatoprotector, and immunosupressan effect. This research aims was to evaluate the effect of ethanol extract of P.angulata as adjuvant on liver function of RA animals model. Animals was induced with 0.1 mL complete freund adjuvant (CFA) intraplantar. The dose of P.angulata extract was 50 mg/kg bw po. The extract was given for 10 days as adjuvant of 0.1125 mg/kg bw methotrexate. Evaluation of liver function consisted of measurement of aminotransferase (AST and ALT), alanine phosphatase (ALP), organ index, and histology. The result showed that CFA could increase the level of AST and ALP and decrease the level of ALT and organ index of control group. The extract of P.angulata as adjuvant of methotrexate could decrease the level of AST and ALP and increase the level of ALT and organ index compare to control group, but could not reach its normal value. Histology showed that the extract also could improve liver regeneration. The extract of P.angulata at dose of 50 mg/kg bw used as adjuvant could improve liver function of RA animals model.

Abstract

Bacterial symbiont known to be able to produce secondary metabolites, that it?s utilization as the source of bioactive compounds is increase. Study of Mulyaningsih (2016) has reported that the bacterial symbiont of sponge Spheciospongia inconstans from Harapan Island, Kepulauan Seribu has a potentcy to produce antibacterial compounds. The aim of this study was to identify species of symbiont bacterial isolates of sponge of Spheciospongia inconstans which can produce antibacterial substance based on  16S  rRNA  gene.  Total  DNA of the five bacterial isolates from previous study were isolated using Wizard Genomic DNA Purification Kit, then amplified the 16S rRNA gene using primers 27f and 1492r. The amplification results were sequenced and aligned using BLAST program. The results of this study only succeeded in identifying isolate 6FS3 of bacterial symbiont that had an 100% similarity with bacteria Bacillus thermophilus strain Sgz-10.
