Abstract

Torbangun (Plectranthus amboinicus) is an herbaceous plant that grows in the tropical region. Torbangun can increase breast milk and has antioxidant activity. The objective of this research was to evaluate the antioxidant and cytotoxic activities of methanol extract of torbangun leaves and to identify diff erentially expressed lactation genes upon incubation of MCF-12A human cell epithelial mammary gland with methanolic extract of torbangun leaves. The results showed that the methanolic extract of torbangun leaves had a scavenging activity of DPPH radical with EC50 value of 14.14 μg/ mL. At lower concentration of methanol extracts of torbangun leaves showed no potent cytotoxic eff ect on MCF-12A with IC50 value of 155.24 μg/mL. The methanolic extract of torbangun leaves increased genes expression of prolactin receptor and glucocorticoid receptor in human mammary gland epithelial cells MCF-12A.

Abstract

Profesionalisme adalah perilaku aktif seseorang dalam mendemonstrasikan ciri-ciri profesional. Seorang yang profesional adalah seorang yang menunjukkan ciri-ciri, diantaranya pengetahuan, keterampilan dan perilaku profesi. Penelitian ini dilakukan dengan tujuan mengetahui hubungan profesionalisme apoteker dengan praktik asuhan kefarmasian. Desain penelitian observasional yang bersifat cross sectional, dilakukan pada 78 apoteker di Apotek Kabupaten Sidoarjo. Diharapkan penelitian ini dapat digunakan sebagai sumbangan pemikiran dalam menyelesaikan permasalahan praktik dalam meningkatkan kualitas asuhan kefarmasian serta memberikan masukan bagi perkembangan ilmu pengetahuan terkait peningkatan profesionalisme dan implementasi praktik asuhan kefarmasian. Hasil penelitian menunjukan ada hubungan antara profesionalisme apoteker dan praktik asuhan kefarmasian dengan nilai p = 0,000 dan tingkat hubungan (correlation coefficient) kuat r = 0,527. Dalam proses praktik asuhan kefarmasian yang dimulai dari tahap penerimaan resep, penyiapan obat dan penyerahan obat didapatkan nilai korelasi tertinggi pada tahap penyerahan obat dengan nilai p = 0,000 dengan tingkat hubungan (correlation coefficient) moderat r = 0,499.

Abstract

According to literatures, many plants of Papilionaceae family such as red bean (Phaseolus vulgaris L_), green bean (Phaseolus aureus Roxb.) and soybean (Glycine soja Sieb. & Zucc.) contain flavonoid, isoflavonoid and vitamin E. They all have antioxidant activity and are potential to reduce free radical. This research was aimed to identify secondary metabolites of crude powders and extracts of red bean, green bean and soybean. The bioactivity of each extracts was evaluated with Brine Shrimp Lethality Test (B SLT) method for preliminary assesment of toxicity. It was also evaluated for antioxidant activity using 1,1 difenil-2-pikrilhidrazil (DPPH) method. The results showed that red bean, green bean and soybean contain saponin, flavonoid, steroid/triterpenoid, coumarin, and tannin. The result of BSLT showed that LC50 (Lethality Concentration 50) of red bean was 213.21 ppm; green bean 390.71 ppm; and soybean 252.75 ppm. The result of DPPH tests showed that IC50 (Inhibition Concentration 50) of red bean was 47.54 ppm; green bean 95.08 ppm; and soybean 90.43 ppm.

Abstract

Antioxidants are substances that may inhibit free radical reactions in the body. The purpose of this research is to identify lethal dose, in vitro antioxidant activity and effect of kemboja merah flowers decoction (Plumeria rubra L.) on MDA level. Lethal dose was measured by Weil method. Antioxidant activity assays was performed in vitro by DPPH method and in vivo by measuring MDA plasma level. The level of MDA was conducted using the thiobarbituric acid reactivity test. In the test, 30 mices were divided into 6 groups: normal, negative control, positive control and 3 group treated with variation dose of kemboja merah flowers decoction. LD50 of decoction kemboja merah flowers is > 15 g/kg of body weight. In vitro, antioxidant activity showed the IC50 value of decoction kemboja merah flowers is 36.96 μg/mL.The antioxidant activity of decoction kemboja merah flowers is smaller than vitamin C (IC50 5.89 μg/mL). In vivo antioxidant activity showed levels of MDA on normal group is 1.66 nmol/mL, negative control group is 5.67 nmol/mL, positive control group is 1.45 nmol/mL, treated group which was given by decoction kemboja merah flowers at dose 0.39 g/kg of body weight is 4.32

Abstract

Buah naga putih (Hylocereus undatus) mengandung flavonoid yang dapat menghambat kerja enzim xantin oksidase yang dapat mengurangi kadar asam urat. Penelitian bertujuan untuk mengetahui efektivitas ekstrak etanol buah naga putih dalam menurunkan kadar asam urat darah pada mencit yang diinduksi kafein secara oral dengan dosis 0,39 mg/25 g BB. Sampel buah naga putih diambil saat berumur 50-55 hari setelah muncul bunga. Pada uji efektifitas, hewan coba mencit dibagi menjadi lima kelompok perlakuan dan diinduksi dengan kafein yaitu kelompok kontrol negatif yang hanya diinduksi kafein, kelompok kontrol positif diberikan allopurinol 0,26 mg/25 g BB, kelompok dosis 0,455 mg/25 g BB, kelompok dosis 0,91 mg/25 g BB, kelompok dosis 1,82 mg/25 g BB ekstrak etanol buah naga putih. Pengukuran kadar asam urat darah dilakukan sebelum induksi (hari ke 1), sesudah induksi atau sebelum pemberian ekstrak (hari ke 6) dan sesudah pemberian ekstrak pada (hari ke 9, 12 dan 15). Hasil penelitian menunjukkan bahwa ekstrak etanol buah naga putih dosis 1,82 mg/25 g BB dapat menurunkan kadar asam urat darah pada mencit secara bermakna dibandingkan dengan dosis 0,455 mg dan dosis 0,91 mg/25 g BB, tetapi tidak berbeda bermakna dengan kontrol positif (allopurinol) dengan presentase penurunan sebesar 92,16%. Kata kunci: Ekstrak buah naga putih, asam urat darah, kafein, allopurinol.

Abstract

Mushroom has been accepted as human food since ages or hundred years ago. Shiitake (Lentinus edodes) is one of an edible mushrooms which has fiber, carbohydrate and protein higher than Oyster (Pleurotus ostreatus). The nutrition of the fruit body and the stem has been analyzed. Comparison of the fruit body and the stem have been conducted for protein, carbohydrate, and the fiber contents. Protein content in the fruit body is higher than in the stem. However the fiber and the carbohydrate contents are higher in the stem than in their fruit body. The amount of protein, carbohydrate and fiber in the fruit body is 31.07%, 15.9% and 7.08% dry weight consecutively. And the amount of protein, carbohydrate and fiber in the stem 12.01%, 19.46% and 10.3% dry weight successively have been observed in this study. Combination of fruit body and stem would be good for human diet in the future.

Abstract

CYP2A6 belongs to the CYP2 family of P450 cytochromes were highly polimorphy. CYP2A6*1 (wild type) was an active allele, CYP2A6*9 was agen decreased allele and the CYP2A6*4 was an inactive allele. By using the polymerase chain reaction technique (PCR), the CYP2A6 polymorphism was studied among Javanese Indonesian smokers. In the process of genotyping the CYP2A6*9 allele, some subjects who had been genotyped as CYP2A6*1/*4 in our previous studies were regenotyped as CYP2A6*9 in this study. The primer forward 2A6*9S and the primer reverse 2A6*9AS-wild type were used in these study. The Promega Go Taq Green Master Mix reagent were used to amplifl y the allele CYP2A6*1 in the positions at -395 to -28 of the CYP2A6 gene. The sample studied consisted of 20 smokers with Cigarrete per-Day (CPD) <10 and 13 smoker with CPD 11-20 from Javanese Indonesian population. In theses research, the subjects had been genotyped as CYP2A6*1/*4 in our previous studies. The allele frequencies of CYP2A6*1, CYP2A6*4, and CYP2A6*9 were 48.5%, 48.5%, and 3%, respectively. When these allele were considered simultaneously, among the subject, 63.9% were genotyped for CYP2A6*1/*4 and 6.1% were genotyped for CYP2A6*1/*4/*9. Based on the data collected, it could be concluded that the polymorphism of CYP2A6 among Javanese population sample study was not aff ected on smoking behavior.

Abstract

Penelitian ini ditujukan untuk menganalisis pengaruh kualitas pelayanan informasi obat berdasarkan dimensi kualitas jasa yang terdiri dari variabel-variabel reliability (kehandalan), responsiveness (cepat tanggap), assurance (jaminan), empathy (empati) dan tangible (nyata) serta kepuasan pasien. Variabel-variabel tersebut digunakan untuk menganalisis pengaruh kualitas pelayanan informasi obat terhadap kepuasan pasien di Apotek X Kota Padang. Sampel yang digunakan dalam penelitian ini adalah 100 responden, teknik pengambilan sampel ditentukan dengan purposive sampling. Metode analisis yang digunakan adalah metode regresi linier berganda dengan menggunakan uji-t, uji-F dengan taraf signifikansi α = 5%, serta uji- R2 dengan program SPSS versi 17,0. Hasil dari penelitian menunjukkan kualitas pelayanan informasi obat dimensi reliability, responsiveness, assurance , empathy dan tangible bersama-sama memberikan pengaruh signifikan terhadap kepuasan pasien Apotek X Kota Padang sebesar 16,7% (F-hitung = 3,771, p=0,004). Dimensi yang paling memuaskan pasien adalah dimensi yang mempunyai gap paling kecil, yaitu dimensi emphaty (-0.90).

Abstract

Post antibiotic effect is the period required by bacteria to physiologically recover after exposure to antibiotics at certain concentrations and exposure time. This study investigated the post antibiotic effect of ciprofloxacin, one of quinolon derivative with broad spectrum antibiotic activity and highest sensitivity towards both Gram positive and Gram negative bacteria, Staphylococcus aureus ATCC 2593 and Escherichia coli ATCC 25922. Lorian gel membrane filtration technique was used and bacteria were exposed to the antibiotic at a concentration of 10 times of the minimum inhibitionconcentration (MIC) for 2 hours. The findings demonstrated strongly that post-antibiotic effect of the two bacterial strain were different. The Staphylococcus aureus (ATCC 25923) had shorter recovery time Within 2-3 hours When compared to the Escherichia coli (ATCC 25922) which recovered from the post-antibiotic effect Within 3-4 hours.

Abstract

Sambiloto (Andrographis paniculata Nees) is one of a medicinal plants containing andrographolide as its primary bioactive component, which is able to inhibit breast cancer cell proliferation. Andrographolide is a diterpene lacton that is sparingly soluble in water. The aim of this study is to improve the solubility of andrographolide in A. paniculata herbs ethyl acetate fraction by microencapsulation method prepared using spray drying. The ethyl acetate fraction of A.paniculata herbs was microencapsulated using PVP K30 and HPMC as the coating polymer in different ratios of ethyl acetat fraction and polymer (1:5; 1:7,5 and 1:10). Microspheres containing ethyl acetate fraction of A.paniculata herbs were evaluated their saturation solubility and in vitro dissolution in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium. The result showed that the microencapsulation could increase the saturation solubility and dissolution rate of andrographolide as compared to the ethyl acetate fraction and andrographolide standard. Compared to the solubility of andrographolide from A. paniculata ethyl acetate fraction, the solubility of andrographolide on ethyl acetate fraction microspheres in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium were increased approximately 23.42-39.34, 19.92-34.83 and 24.56-38.25 folds respectively. Moreover, compared to the solubility of andrographolide standard, the solubility of andrographolide on ethyl acetate fraction microspheres in aquadest, pH 6.8 phosphate and pH 1.2 chloride medium were increased approximately 16.99-28.53, 19.76-34.54 and 17.26-26.87 folds respectively.
