Abstract

Ethanolic extract of red betel leaves (Piper crocatum Ruiz & Pav) has antihyperuricemic activity. In order to be applicable to consume, this extract need to be formulated into pharmaceutical dosage form. Granules which is packed in a tea sac bag was considered to be more practical andappropriate for person suff ering hyperuricemic, because they have to drink more water. In the granulation process, the he binder plays an important role so that this study aims to fi nd the optimal red betel leaf granule formula with variation on the extract dose and the polyvinyl pyrolidone (PVP) binder and prove granule activity in vivo in mouse hyperuricemia model. The extract was prepared by macerationthe leaves in 96% ethanol and sprayed dried by hot air. The powder was then formulated to granules form in 22 factorial design. The factors were the extract dosages (38% dan 47.51%) dan the amount of PVP (Polyvinyl Pyrrolidone) as a binder (0.5% dan 2%). The physical and chemical properties of the granules were evaluated, interaction dosage, PVP concentration and each properties was analysed by making the contour plot and superimposed contour plot to get the optimum formula. The in vivo antihyperuricemic activity of the granules was determined in hyperuricemic mice. The result showed that the optimum formula was found in granules that contained 42.5% extract and 1% PVP. At the equivalent dosage of 455 mg extract/kg, this granules formula had antihyperuricemic activity decreasing 54.2 %plasma uric acid level significantly. Thus it can be concluded that the optimum granules of ethanol extract of red betelleaf with 42.5% extra ct content and 1% PVP proved to have antihiperurisemia activity in mice induced by chicken extractand potassium oxonate.

Abstract

Exploring an antibiotic from microorganism is potential to discover the new antibiotic. The endophytic microorganism is one of the minimanufacturer producing the secondary metabolite such as antibiotic. Screening on the endophytic molds isolated from Brucea javanica L. Merr has been carried out. Fermentation in still and shake culture were used to produce the antibiotic using PDY medium. Antimicrobial activity was assayed using diffusion agar method. The results revealed that shake culture gave a better antimicrobial activity than still culture. Amount of eight isolate studied showed microbial activity against Gram positive bacteria: Staphylococcus aureus and Bacillus subtilis also Gram Negative bacteria: Escherichia coli and Salmonella typhi, however no microbial activity recorded againts Candida albicans. Isolate number 1.2 gave the highest antimicrobial activity on still culture and isolate number 1.7 gave the highest antimicrobial activity on shake culture. Further research should be carried out on identification and purification of the antimicrobial agent.

Abstract

Antioxidants are compounds which have ability to inhibit free radical reactions in the human body. Objectives of this research are to evaluate the antioxidant activities of extract and fractions from Pandanus canoideus Lam. fruits, to determine total phenolic and total flavonoid contents, and to correlate the total phenolic and total flavonoid contents of those fractions with their antioxidant activities. The Pandanus conoideus Lam. fruits or locally known as “buah merah” were extracted by methanol, then evaporated and dissolved in aquadest. This solution is subsequently partitioned using chloroform and ethyl acetate to obtain chloroform, ethyl acetate, and water fractions. The antioxidant activities were determined by radical scavenging assay using DPPH (2,2-diphenyl-1-picrylhydrazyl) radical. The total phenolic and total flavonoid contents were determined by mean of spectrophotometry. The results showed that fractions of water, ethyl acetate, and chloroform revealed antioxidant activities with IC50 values of 399.06, 10.41, 1416.68 µg/mL respectively, whereas the methanolic extract of Pandanus conoideus Lam. fruits before partition has IC50 of 186.31 µg/mL. The antioxidant activities of these extract and fractions are lower than that of vitamin E (IC50 8.27 µg/ml). A linier positive relationship existed between the total phenolic contents of these extract and fractions with their antioxidant activities as shown by equation of y = 13.085x + 754.91; r2 = 0,3641, while the correlation between the total flavonoid contents and their antioxidant activities revealed a linier regression of y = -282.9x + 868.05; r2 = 0.4484.

Abstract

Transfersom merupakan vesikel nano yang bersifat deformabel sehingga mampu berpenetrasi sampai ke lapisan kulit yang lebih dalam. Kofein memiliki khasiat sebagai antiselulit. Penelitian ini bertujuan untuk formulasi dan mengetahui kemampuan daya penetrasi gel transfersom kofein dibandingkan dengan gel kofein tanpa transfersom. Empat formula transfersom dibuat dengan konsentrasi kofein yang berbeda (1; 2; 3; 5%) menggunakan metode hidrasi lapis tipis. Suspensi formula 4 dengan ukuran partikel 202,35 nm, indeks polidipersitas 0,1090 dan efisiensi jerapan 58,91% dipilih untuk sediaan gel. Uji penetrasi dilakukan secara in vitro dengan sel difusi Franz. Hasil yang diperoleh adalah jumlah kumulatif kofein yang terpenetrasi dari gel transfersom lebih tinggi yaitu 1218,34+358,71 μg cm-2, persentase jumlah kumulatif 8,53+2,55% dan fluks 360,91+86,50 μg cm-2 jam-1. Sementara gel kofein tanpa transfersom memiliki jumlah kumulatif terpenetrasi 369,29+231,57 μg cm-2, persentase jumlah kumulatif 2,61+1,37% dan fluks 70,96+73,39 μg cm-2 jam-1. Dapat disimpulkan bahwa gel transfersom kofein menghasilkan daya penetrasi yang lebih baik dibandingkan gel kofein tanpa transfersom.

Abstract

Benalu batu (Begonia sp.) is used as traditional medicine by Wana tribe in Central Sulawesi to treat various diseases, including cancer. In an effort to search potential anticancer drugs from natural sources, investigation was conducted on the cytotoxic activity of Begonia sp. methanol extract against cervic cancer cells (HeLa) and breast cancer cells (T47D) and identification of the chemical compound groups in methanol extract that is responsible for the anticancer activity. Investigations including the extraction of Begonia sp. dried herb by soxhletation using methanol, followed by evaporation on rotary vapor until viscous extract gained. The cytotoxic test were carried out by tetrazolium bromide (MTT) method. The concentration series of Begonia sp. methanol extract were 250, 125, 62.5 and 31.25 μg /mL. Results showed that the Begonia sp. methanol extract inhibited the growth of cervic cancer cells (HeLa) (IC50 = 70.97 μg/mL) stronger than inhibition on breast cancer cells (T47D) (IC50 = 122.21 μg/mL). Analysis of chemical compound groups in the methanol extract using thin-layer chromatography (TLC) method indicated that polyphenolic flavonoid might be the main compound responsible for the anticancer activity.

Abstract

Indonesia is the second rank for the most TB (Tuberculosis) cases in the world after India. It’s estimated there are a million cases per year in Indonesia. The objective of the research was to find out the influence of Vitamin A supplementation in the medication of pulmonary TB on the acceleration of sputum conversion time at intensive phase in the Working Area of Puskesmas in Medan Kota Subdistrict. This research applied clinical testing design with experimental approach and Experimental Quasi method. The samples were 30 newly diagnosed pulmonary TB patients consisting of 15 patients in the treatment group and 15 patients in the control group. They were ≥ 18 years old, regularly consumed category I OAT with BMI ≥ 18.5. The statistical data were analyzed by Mann-Whitney statistical testing at reliability rate 95% (p < 0.05). The results of the research on the treatment group discovered that they were mostly 18-27 years old 40.0%, male 73.3%, mostly university students 26.7%, had BTA conversion in the second week 80.0%; and the sputumconversion time was 2.4 weeks in average. The results of the research on the control group demonstrated that they were mostly 18-27 years old 33.3%, male 80.0%, mostly university students and entrepreneurs 20.0%, had BTA conversion in the second week 80%; and the sputum conversion time was 4.1 weeks in average. Vitamin A supplementation in the medication of pulmonary TB patients had some influence on the acceleration of sputum conversion at intensive phase (p = <0.001) < (alpha = 0.05). Vitamin A supplementation in the medication of pulmonary TB patients had some influence on the acceleration of sputum conversion at intensive phase in the Working Area of Puskesmas in Medan Kota Subdistrict. speed up wound healing on second degree burns. White egg gel had significantly difference compared to negative control of NaCl 0.9% and no signifi cantly difference compared to Bioplacenton as positive control.

The Guttiferae family has many species with interesting phytochemical properties. Is has been widely used in the traditional medicine and contains xanthones as constituent, exhibiting biological and pharmacological activities for example, e. g. as antimalaria, antimicrobe, antioxidant, anticancer and HIV-1 protease inhibitor.

Abstract

Som Jawa (Talinum paniculatum Gaertn.) is a kind of Indonesian herbs that has similarities morphologically and chemically to Chinese ginseng. Som Jawa known as Ginseng Jawa in traditional healing has been made into various forms. The famous form is made as wine. The research on acute toxicity and efficacy test of Som Jawa as stimulant have been carried out to make Som Jawa into Indonesian Ginseng. The methodology of toxicity test is according to Weil CS (1952) and the efficacy test is according to Turner (1965). The test result showed that the value of acute toxicity of Som Jawa is 32.22 (25.59-40.56) mg/10 g BW (ip mice). At the time of starting to sleep, there was a significant difference on Som Jawa 10 mg/40g BW dose administration to Heptamyl 1.56 mg/40 g BW (p=0.00). In the swimming time extract of Som Jawa 5 and 10 mg/40 g BW showed no significant differences to Heptamyl 1.56 mg/40 g BW (p>0.05) (mean=32.88 +11.6;39.77 +14.6; 32.33 – 15.9). Som Jawa (Talinum paniculatum Gaertn.) is safe based on acute toxicity test. Extract of Som Jawa could prolong the time of starting to sleep and improve fitness.

Abstract

Beta glucan is a polysaccharide that can be found in cereals, yeast, bacteria, and algae. Beta glucan produced by Saccharomyces cerevisiae has the ability to enhance the body’s defense system. The study aims to investigate the antioxidant activity of B glucan produced by three strains of Saccharomyces sp BR 1, BR 2,and SC. Antioxidant activity assay was carried out in vitro on t BHP induced red blood cells (REC) by observing the level of catalase activity, SOD activity and MDA levels as lipid peroxidation product. The investigation was performed on 6 groups: normal control, negative control, positive control (RBC+ t-BHP + vitamin E 100 IU), and samples (RBC+t-BHP) treated with 1.4 µg/mL β-glucan from Saccharomyces strain BR1, BR2 and SC respectively. Saccharomyces cerevisiae strain SC produced the highest β-glucan of 88.70% (glucose equivalent), while the BR1 and BR2 of 22.56% and 46.28% respectively. Antioxidant activity assay showed that the highest increase in catalase activity (581.48%) was given by the B glucan from SC strain, the highest increase of SOD activity and decrease of MDA levels were given by the β- glucan from BR1 strain (359.17% and 48.18%) as compared to negative controls. Statistical test (α = 0.05) showed antioxidant activity of β-glucan from Saccharomyces Sp BR2 and SC did not differ significantly from the activity of vitamin E (100 IU).
