Abstract

A common problem related to the production of potatoes in Indonesia is the use of poor quality seed sources and declining seeds. Plant tissue culture is one of the best methods used for the provision of quality seeds. This study aims to establish the best combination of BAP concentration and the concentration of paclobutrazol in spurring the growth of cuttings and initiation of potato tubers in vitro.This research was conducted in November 2017 until June 2018 at Agronomy Laboratory of Biotechnology Division and Plant Culture Network of Agriculture Faculty of Bengkulu University. The  research  design  used  was  Completely  Randomized  Design  (CRD)  which  consist  of  18 combination of BAP treatment and Paclobutrazol was repeated 6 times. The concentration of BAP used was 0 mg / l, 5 mg / l, 10 mg / l and the concentration of Paclobutrazol 0 mg / l; 2.5 mg / l; 5 mg / l; 7.5 mg / l; 10 mg / l; 12.5 mg / l. The data of the research were analyzed by F test of 5% level and if there were different between treatments followed by Duncan's Multiple Range Test (DMRT) test of5% level. The results showed that the combination of BAP and  Paclobutrazol concentration had significant effect on all observed variables. Treatment of 5 mg / l BAP and 7.5 mg / l Paclobutrazol is the best medium in inducing potato micro tube as indicated by the fastest tuber formation time, highest percentage of productive crops, largest number of tubers per bottle, number of tubers per productive plant, tuber diameter the largest and highest wet weight of tuber per bottle. Keywords : in vitro, BAP, paclobutrazol, Potato

Abstract

Pineapple plants are usually propagated vegetatively by utilizing the base of the fruit (ratoone), stem buds (sucker), fruit buds (slip) and crown (crown). Propagation of plants by utilizing parts of the plant is less effective in meeting the needs because the planting material produced is small and requires a long time. An alternative to the propagation of pineapple to increase the amount of planting material is by tissue culture techniques. This study aims to obtain the best types of complex organic compounds and the right concentration for the growth of pineapple shoots in vitro. The research was conducted in June 2017 until October 2017 at the Laboratory of Agronomy at the Faculty of Agriculture, University of Bengkulu. Complete randomized design with 3 replications was used to allocate 9 treatments of complex organic compounds. The basic media used are the composition of Murashige and Skoog (MS). Treatment consists of 1) Coconut water 100 ml / l, 2) Coconut water 150 ml / l, 3) Coconut water 200 ml / l, 4) Banana porridge 100 gl / l, 5) Banana pulp 150 g / l, 6) Banana slurry 200 g / l, 7) Bean sprout extract 100 g / l, 8) Bean sprout extract 150 g / l, 9) Bean sprout extract 200 g / l. The results showed that at 16 weeks after MS media planting the addition of 100 g / l banana pulp was able to produce the highest plant height with an average of 3.45 cm. MS media with the addition of 200 ml coconut water resulted in the highest shoot height of 1.5 cm and followed by MS with the addition of 100 g / l media of banana pulp with an average of 0.9 cm. MS media with the addition of 100 ml / l of coconut water is suitable for the growth of pineapple roots with the average value of the number of roots and the highest length of roots. MS media with the addition of bean sprout extract with various concentrations is not able to stimulate shoot growth and has a growth response that tends to be slow. Keywords: Pineapple, In Vitro, Complex Organic Compounds.

Abstract

A successful rice production on swampland would require a planting material from high yielding varieties adaptated  to the swampy ecosystem. This study was carried out to compare the growth and yield characteristics of five rice lines and a check variety as grown on non-tidal swampland. The lines were F4 generation of bulk selection from the crosses involving Bengkulu swamp rice landraces (Hanafi Putih, Tigo-tigo, Harum Curup, and Lubuk Durian) and high yielding varieties for the irrigated field (Sidenuk and Bestari). The trial was conducted on a shallow non-tidal swampland with stagnant inundation no more than 50 cm in depth often occurred during the plant life cycle.  The lines and the check variety (Inpara 4) were arranged in a randomized complete block design with three replications. Observations were made for the agronomic performances of the plant, including plant height, total tiller number clump-1, effective tiller number clump-1, heading date, maturity date, panicle length, grain number panicle-1, 100-grain weight, and grain yield clump-1. Significant variation among the genotypes was found for all observed traits. On average, the evaluated lines showed comparable growth and yield performances to the check variety.  Tigo-tigo × Bestari was the tallest line and potential for medium depth swampland.  This line showed good overall agronomic performances and yielded relatively higher than the check variety, but delayed in attaining maturity. For shallow swampland, Hanafi Putih x Sidenuk exhibited the most potent line by having good overall agronomic and yield performances, except late in maturity.  For early maturing line, Lubuk Durian x Hanafi Putih showed its potential for shallow swampland. Abstract

Acclimatization is a process of an environmental adaptation from heterotrophic conditions to autotrophic conditions. In order to be successful in acclimatization, it requires proper planting media and enough nutrition supply. The objective of this experiment were to determine the best planting media and the best spraying frequency of leaf fertilizer on Pencil orchid growth during acclimatization period.  The experimental design used was Randomized Complete Block Design, arranged in factorial (2 factors, 3 replications). The first factor was type of planting media: Coconut Coil, Rockwool, Wood Shavings, and Fern Roots. The second factor tested was fertilizing frequencies: every 2 days, every 3 days, and every 4 days. The results showed that the best media for acclimatization of Pencil orchid was fern root fertilized every four days, the best media for growing Pencil orchid was fern root and Wood Shavings, and the best fertilizing frequency to promote Pencil orchid growth were every 3 days or every 4 days.Keywords: Pencil orchid, acclimatization, planting media, fertilization

Abstract

Compost has been commonly used as the primary organic matter in organic plant production, especially in vegetable production.  Different sources of compost materials give different medium structures and chemical characteristics. The objectives of this research were: 1) to evaluate the response of growth and yield of chili pepper to a rate of compost and source of animal waste which were used as compost; 2) to determine the optimum rate of fertilizer for the highest growth and yield of chili pepper. The research was carried out from November 2015 until March 2016. Compost material treatments consisted of cow dung, empty palm oil bunches, and mixed of vegetable residue and weeds. The compost rate treatments consisted of 0 ton ha-1, 10 ton ha-1, 20 ton ha-1, and 30 ton ha-1. No interaction between the two treatments was found to influence plant growth and yield variables.   The type of compost materials affected plant height of 5, 6, and7 weeks after transplanting.  Cow dung as a source of compost showed the best effect in increasing chili plant height.  Plant height, the number of plant dichotomous, and plant canopy responded linearly  to the increase of the compost rate. The usage of compost at 20 ton ha-1 and 30 ha-1 resulted in higher fruit length, the number of fruit, and fruit weight per plant.   Keywords: chili pepper, compost, organic, growth, yield

Abstract

Crocidolomia binotalis Zell is an important pest in Brassicaceae vegetable crops. Vegetable insecticide is one potential alternative to controlling the pest. This study aims to determine the application time of noni fruit extract and to determine the effective concentration of noni fruit extract as a vegetable insecticide in controlling C. binotalis larva on cabbage plants in the field. this study used a Completely Randomized Design (RAL) consisting of two treatment factors and four replications. The first factor is the concentration of extract (5 ppm, 10 ppm, 15 ppm and 20 ppm) and the second factor is the time of application of vegetable pesticide before the pest is invested (W1) and after the pest is invested (W2). So there are 32 units of experiments and 4 units of experiments added as control. The results showed, noni fruit extract had no significant effect on mortality of C. binotalis larvae. The highest mortality rate is at 100% K2 (10ppm) concentration at the time of application before the pest is invested (W1). The interaction had significant effect on the wet weight of the plant canopy but had no significant effect on the mortality of the pest, the intensity of the attack, the percentage of pupa formed, and the present percentage of imago.
