Abstract

The major problem faced by farmers in Central Java is the fluctuation of feed availability throughout the year, with feed surplus in the rainy season and less in the dry season. The objective of this study was to evaluate the support ability of agricultural by product as ruminant feed in Central Java. Survey method was used in this study, primary and secondary data were descriptively analyzed. The results showed that based on total population of ruminants and carrying capacity of the potential production of agricultural by product, there was an excess of support ability to 3,699,674 Animal Unit (AU) based on Dry Matter (DM); 1,419,142 AU based on Crude Protein (CP) and 1,825,567 AU based on Total Digestible Nutrients (TDN). However, based on carrying capacity of effective production of agricultural by product, there was an excess of support ability of feed only for 2,003,470 AU based on DM, 335,459 AU based on CP and 687,215 AU based on TDN. Abstract

The study was conducted to determine the critical component of a Total Quality Management (TQM) Program. The objectives of this research were: (1) identifying the problems of TQM implementation and, (2) analyzing of TQM performace in PT Madu Pramuka. Data were collected from PT Madu Pramuka through an expert survey instrument. Data analysis was done through Analytical Hierarchy Process (AHP). Results of this study indicated that the most important problem was the quality of raw material (honey) which were still under the standard. This is due to limited honey quality control tools. The TQM performance was not satisfying because it was only storage and packaging division used standard as the critical component of TQM. Abstract

Equine semen are far less tolerant in the freezing and thawing process than bull semen. The stallion spermatozoa are known susceptible to cold-shock relating with the content of their fatty acid on the plasma membrane. The extender is one of determining factors in the success of stallion semen cryopreservation, as an energy source and protector the cell from harmfull effect of cold shock. The common cryoprotective agent (CPA) for mammalian spermatozoa was glycerol, but for stallion semen cryopreservation, dimethyl formamide (DMF) was more suitable. This research was conducted to compare the success of the stallion semen cryopreservation in skim milk and dimitropoulos (DV) extender with DMF as cryoprotectant. Semen from three sexualy mature stallions was collected twice a week using an artificial vagina. Semen was evaluated macro- and microscopically and then divided into two tubes, diluted each of them with skim milk dan DV extender (1:1), centrifuged at 3 000 rpm for 15 minutes. The supernatant was removed and the pellet (spermatozoa) was re-diluted in skim DMF (SDMF) and DVDMF extender with the concentration of spermatozoa was 200x106 ml-1. The semen then packed in 0.3ml minitub straw equilibrated for two hours at 4-50C and frezee in liquid N2 vapor for 10 minutes. The assessment of sperm quality was conducted based on the percentage of sperm motility and viability. In this research, post-thawed semen in DVDMF showed the percentages of the motility (36.2%) and the viability (59.3%) higher (P < 0.05) than SDMF (28.5 and 48.0 %). In conclusion, the DVDMF extender provided better post-thawed semen quality than SDMF. Key words: cryopreservation, stallion semen, skim, dimitropoulos, dimethyl formamide

Abstract

An in vitro study was conducted to determine the effect of Acacia mangium Willd supplementation to Pennisetum purpureum on fermentation characteristics, protozoal numbers, nutrients degradability and in vitro methane production. Treatments consisted of four composite substrates with P. purpureum and A. mangium Willd ratios at 100:0 (0%), 85:15 (15%), 70:30 (30%), and 55:45 (45%). Crude saponin and total tannin contents of A. mangium were 1.67% and 4.51%, respectively. Methane and gas productions decreased linearly (P < 0.01) in response to acacia levels. Addition of A. mangium at 15%, 30% and 45% decreased CH4 production by 16.2%, 26.8% and 61.1%, respectively as compared to the control. There were linear decreases in total VFA and acetate concentrations (P < 0.01), and propionate production (P < 0.05) in response to increase in acacia addition. Total protozoal populations increased linearly (P < 0.05) with added acacia. In vitro dry matter and organic matter degradabilities of substrate decreased linearly (P < 0.01) with acacia addition. It is concluded that methane production is not essentially associated with protozoal population. A. mangium has a potential use for mitigation of enteric methane production. Abstract

Phytate is commonly high in cereals grain. This compound can be extracted with acid solution. The purpose of this study is to extract phytic acid from pollard using acetic acid at the concentration of 0.1, 0.5, 1, and 5%. Pollard was fractionated using vibrator ball mill to evaluate the distribution of phytic acid in the particle of pollard. One percent of acetic acid was used to extract phytic acid from pollard at ratio of 1:1; 1:1.5; 1:2; and 1:3. The fraction having particle size of 16, 30 and 50 mesh had similar phytic acid content. The results indicated that phytic acid content of these fractions was higher than that of 100 and 400 mesh fractions. Whole pollard had higher phytic acid content than its fractions. Phytic acid in pollard can be extracted by 1% acetic acid solution. Extraction of phytic acid from pollard using 1% acetic acid solution at ratio of 1:3 produced 23.83 g/kg. Abstract

Zinc and Cu have important roles in growth and immune system, but their bioavailability are low due to the phytic acid content in poultry diet. This experiment was conducted in order to obtain the best combination of phytase, Zn, and Cu as growth stimulants for broiler fed diet containing phytic acid. Two hundred eighty eight Day Old Chicks unsexed were allocated into nine treatment diets with four replications (eight chicks in each replicate). The birds were raised up to 42 days old. Combination of the treatment diets were: R1 (positive control), R2 (negative control), R3 (R2 + 132.70 ppm ZnO), R4 (R2 + 286.16 ppm CuSO4), R5 (R2 + 132.70 ppm ZnO + 286.16 ppm CuSO4), R6 (R2 + phytase 1000 FTU/kg), R7 (R2 + phytase 1000 FTU/kg + 132.70 ppm ZnO), R8 (R2 + phytase 1000 FTU/kg + 286.16 ppm CuSO4 ), R9 (R2 + phytase 1000 FTU/kg + 132.70 ppm ZnO + 286.16 ppm CuSO4). The results of this research indicated that the phytase supplementation (1000 FTU/kg), ZnO (132.70 ppm), and CuSO4 (286.16 ppm) in poultry diet 1) improved the body weight and feed conversion, but did not significantly influence the feed consumption, 2) increased serum Zn, and 3) increased the alkalin and phosphatase activity. Abstract

The objectives of the present study were to estimate the genetic distance and to determine the discriminant variables among fighting and meat types sheep of Garut (Garut sheep). A total of 413 heads of Garut sheep from five sub-populations were used in this study. Data were collected from the Regional Animal Research Center (BPPTD) as follows: 102 heads of common type sheep from Margawati (M), 69 heads from meat type sheep from Wanaraja (P), 72 heads of meat type from Sukawening (D), 81 heads of fighting sheep from Wanaraja (T) and 89 heads of fighting sheep from Sukawening (A). Data obtained were analyzed by simple discriminant with SAS package program version 7.0 and MEGA2 program to get the construction of phenogram tree. The results showed that in general fighting sheep had body weights and body measurements significantly higher (P < 0.05) than meat type sheep. However they were almost the same as common type sheep Margawati. The length and width of ears, tail width and chest width were the most discriminant variables to determine the differences of fighting type and meat type sheep. The highest similarities between individuals inside the group were obtained from Margawati common type sheep (71.57%), followed by Wanaraja meat type sheep (69.57%), Sukawening meat type sheep (69.44%), Wanaraja fighting type sheep (69.14%) and Sukawening fighting type sheep (59.55%). The Wanaraja fighting type sheep (T) had the closest genetic distance (1.16) to the Sukawening fighting type sheep (A), while the longest genetic distance (6.17) was between the Wanaraja meat type sheep and the Margawati common type sheep. The phenogram tree showed that the Margawati common type sheep was outside Wanaraja and Sukawening fighting type and meat type sheep, but the genetic distances were closer for the Wanaraja and Sukawening fighting type sheep. Abstract

Mannan containing polysaccharides could be used as an alternative to replace antibiotics due to their capacity to block the colonization of pathogenic bacteria in the intestine of poultry. The aim of this study was to investigate mannan containing polysaccharides from palm kernel meal (PKM) and its inhibitory effect against Salmonella typhimurium. Hot water extractions were used to isolate mannan containing polysaccharides from cell wall of PKM. In vivo studies were conducted using broiler and layer chicks that were challenged orally with 104 cfu Salmonella typhimurium on third day. Split plot design was used as experimental design with strain as main plot and level of mannan polysaccharides as sub plot. The levels of mannan containing polysaccharides that were used consisted of 0 (R0); 1000 (R1); 2000 (R2); 3000 (R3); 4000 (R4) ppm, in term of total sugar. The results indicated that compared to the control group, feeding PKM containing mannan 4000 ppm decreased (P < 0.01) Salmonella typhimurium incidence. The addition of mannan did not affect feed consumption. On the contrary, the addition of 4000 ppm mannan gave significantly higher feed/weight gain ratio of the chicks (P < 0.05). The administration of feed supplemented with mannan from PKM did not influence weight gain of poultry. It is concluded that mannan from PKM can prevent the colonization of Salmonella typhimurium in poultry. Key words: palm kernel meal, mannan, Salmonella typhimurium, performance, poultry

Abstract

A total of 134 Pesisir cattle were genotyped for growth hormone (GH) gene by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The genotype and allele frequencies of the GH MspI and GH AluI Pesisir cattle were determined. The GH MspI gene frequencies for the C and T allele were 0.209 and 0.791 respectively, while GH AluI gene frequencies for the L and V allele were 0.992 and 0.008 respectively. The chi-square analysis indicated that this population is not in Hardy-Weinberg Equilibrium status. Expected heterozygosis value (He) for GH MspI and GH AluI were 0.3306±0.0266 and 0.0149±0.0073 respectively. The PCR-RFLP GH MspI marker has higher genetic variability compare to PCR-RFLP AluI marker. This finding showed that GH MspI T allele was favorable as a GH marker for Bos indicus breeds. Abstract

The experiment was conducted to investigate the adaptive grasses growing under shading albizia, rubber and oil palm canopy. Three species of tropical grasses (Brachiaria humidicola (Rend.) Sch., Digitaria decumbens Stent, dan Stenotaphrum secundatum (Walter) O.Kunt. were cultivated for at least four months. The parameters of research were productivity (including tillering rate) and nutrient quality of each grass. Data were analyzed using Split-plot design and for the significant differences were further tested by Least Significant Different (LSD). The result showed that B. humidicola production was better than other species in plantation and forest shaded. This species producting higher fresh yield and crude protein, but its tillering rate was poor. S. secundatum, although its production was not as great as B. humidicola, this grass had the greatest tillering rates compared to the others. It can be recommended that S. secundatum is persistent species, and has a good productivity for a long periode of time under shading.
