Abstract

Bloodstains is an important body fluids as evidence, because the blood has hemoglobin structure consisting of hemeand polypeptides. Perpetrators will attempt to clean the bloodstain that found at the crime scene by using cleaningagents and chemicals. Sodium hypochlorite is a chemical that found in cleaning fluids and bleach. This study aims toknow the effects of sodium hypochlorite to bloodstain and confirmatory test. Bloodstain on the objeck glasses andthe fabrics exposed by sodium hypochlorite concentration 5,25% and 0,15%, and examined with Teichmann’s testand Takayama’s test, and seen under microscope. In this study, show no crystal hemoglobin of the bloodstain on theobjeck glasses and fabrics exposed by sodium hypochlorite concentration 5.25% and concentration 0,15%. Thismight caused by sodium hypochlorite destructed structure of heme, so there were no reaction between heme, sodiumchloride and acetat glacial acids on Teichmann’s test and no reaction between heme and Takayama’s reagen onTakayama’s test. The conclusions of this study, crystal hemoglobin were not found on bloodstain exposed by sodiumhypochloryte using Teichmann’s test and Takayama’s test.

Abstract

Talaromycosis marneffei is a mycotic disease caused by Talaromyces marneffei which primarily infect immunocompromised patient. This disease is endemic in Southeast Asia, and Indonesia consider as endemic area due to a case of tourist who got the infection after visiting Indonesia. Diagnosis was made based on clinical suspicion with laboratory confirmation. Clinical manifestations are not specific, such as fever, anemia, weight loss, lymphadenopathy, hepatomegaly, splenomegaly, respiratory disorder, and cutaneous manifestation. Histopathology and culture are the most common examination performed for diagnosis. In histopathology examination, fungi appear as fission arthroconidia cells which shape round to oval, with cross wall formation inside or outside macrophage and histiocyte. Thermal dimorphism characteristic can be observed in fungal culture. Fungi grows as mold at 25oC-30oC and as yeast at 37oC. Definitive identification of T. marneffei was done by molecular examination using primers derived from ITS region as primary marker and beta tubulin region as secondary marker. Talaromyces marneffei is sapronosis, that transmitted via inhalation of conidia from environment.

Abstract

TNF is important in rheumatoid arthritis (RA) and spondyloarthritis (SpA) pathogenesis. Recently reports showedsTNF levels were significantly lower in SpA than RA synovial fluid (SF). Therefore, the differential regulation of TNFin both diseases were investigated focusing on the balance of sTNF-tmTNF and both TNF receptors expression. Synovialfluid from 22 RA and 25 SpA were analyzed by ELISA. sTNF was confirmed significantly higher in RA than SpA.Total TNF of synovial tissues assessed by qPCR showed similar levels, suggesting a higher tmTNF/sTNF ratio in SpA.The levels of sTNF-R1 and sTNF-R2 were significantly lower in SpA than RA SF. Interestingly, sTNF-R2/sTNF-R1increased in SpA. qPCR on synovial tissue showed similar mRNA levels for TNF-R2 but decrease of TNF-R1 in SpA.IL-6R levels did not showed differences and TIMP-3 tended to be higher in RA than in SpA SF. In conclusion, weobserved a shift from sTNF/TNF-R1 in RA to tmTNF/TNF-R2 in SpA.

Abstract

Accumulation of triglycerides associated with  insulin resistance in type 2 diabetes mellitus (T2DM). Alpha lipoic acid (ALA) has hypotriglyceridemic effects by lowering triglyceride levels in blood and liver. This study aims to analyze effect of ALA on triglyceride levels in  type 2 diabetic rats. Fifteen Wistar male rats (±8-12 weeks, 200-250 gram) divided into 3 groups (control, T2DM  and  T2DM–ALA given for 3 weeks). Diabetes was induced by giving streptozotocin (50 mg/kg.bw) and nicotinamide (110 mg/kg.bw) after 15 minutes intraperitoneally. Diagnosis of T2DM was determined if the rats have perifer blood glucose >250 mg/dL after 3 days. The results showed that triglyceride levels in  ALA group (p=0,003) lower than T2DM group, T2DM group (p=0,031)  had higher triglyceride level than control and no difference of triglyceride level between  T2DM-ALA given group with control (p=0,696). This study concluded that ALA can affect triglyceride levels in T2DM thus prevents occurrence of hypertriglyceridemia.

Abstract

The brain is the central organ of stress adaptation, and is also a target of stress. Chronic stress may result in abnormalchanges in brain plasticity; include dendritic retraction, neuronal toxicity, and suppression of neurogenesis andaxospinous synaptic plasticity. Repetitive stress exposure will gradually change the electrical characteristic, morphologyand proliferative capacity of neurons. Among brain region, the cerebellum is known to be severely affected by oxidativedamage associated with glucocorticoids level. It is believed due to the highest levels of glucocorticoid receptorslocalized in the external granular layer. BDNF, a member of neurotrophin family, is known to be a strong survivalpromoting factor, and plays a critical role in cell proliferation and differentiation, neuronal protection, and the regulationof synaptic function in the central nervous system. BDNF is highly expressed in the cerebellum, mainly in granulecells. Both acute and chronic stress change BDNF expression in the brain. Although the impact of stress on BDNFlevels showed the different results, BDNF is believed to protect neurons from injuries caused by stress.
