Abstract

Background: Gracilaria Seaweed extracts are known to have an antibacterial content thatcan inhibit the growth of Porphyromonas gingivalis bacteria. P.Gingivalis are dominant inchronic periodontitis as a cause of periodontal tissue damage that can be inhibited by usingmaterials that contain antibacterial effect. Terms of materials to be applied in the oral cavitymust be biocompatible, through toxicity tests, one with bioviability test. In this studybioviability test performed on media mesenchymal stem cells. Purpose: To determinebioviability Gracilaria seaweed extract against mesenchymal stem cells as adjuvant therapyof periodontitis. Materials and Methods: study design was post test only control groupdesign. Mesenchymal stem cells in 96 wells were divided into a control group of cells (n=6),media controls (n=6), and treatment (n=6). The treatment group was given Gracilariaseaweed extract with a concentration of 2.5%, 5%, 10%, 20% and 40%. Stem cells wereincubated 24 hours before and after treatment. Once given MTT, optical density read with anELISA reader then calculated the percentage of living cells. Toxic material when living cells<50% and not toxic when living cells >50%. Living cell data were analyzed by statistical testKruskal Wallis and Man Whitney. Result: Viability cell at 2.5% concentration is 68.59%, 5%concentration = 29.67%, 10% concentration = 4.43%, 20% concentration = 2.5% and 40%concentration = 0.96 %. Conclusion: Extract of seaweed Gracilaria proven not toxic at 2.5%concentration and proved toxic at 5%, 10%, 20% and 40% concentration

Abstract

Background: Alveolar osteitis was one complication after tooth extraction. This conditionremains a common post operative problem that results in severe pain and inhibit sockethealing. Nannochloropsis oculata’s extract contains alkaloids, carotenoids, flavonoids,terpenoids and tocopherols that could accelerate the wound healing process. Purpose: Examine the effectivity of Nannochloropsis oculata extract to the increased density ofcollagen in alveolar osteitis healing process. Materials and Methods: This research designwas post-test only control group design, using 64 male Wistar rats then divided into twomajor groups. Group 1 (irrigation) and group 2 (dressing). In each group contains a negativecontrol group (irrigation PBS or dressing PBS), positive control group (irrigation Saline0,9% or dressing Alvogyl), and the treatment group (irrigation or dressing usedNannochloropsis oculata extract 2,5% and 5%). After treatment on the 3rd and 7th day, ratswere euthanazied than collagen density was measured at histopathology slide used Masson’sTrichom with magnification 100x under light microscope, than it what measured. Data wasanalyzed with Kruskal-Wallis and Mann-Whitney test. Result: There were significantdifferences of collagen density beetwen irrigation treatment groups (0,045), but there werenot significant differences beetwen dressing treatment groups (0,243). Dressing method wasmore effective to increase collagen density than irrigation groups (0,026). Conclusion:Dressing treatment used Nannochloropsis oculata extract concentration 5% was mosteffective to increased the collagen density in alveolar osteitis healing process on 7th days.

Abstract

Background: Bone healing on defect requiring the cells to form new bone. Fibroblast growth factor -2 belong to one of signaling control that play part in differentiation of fibroblast to osteoblast cell. Hydroxyapatite application on bone healing will aid bone regeneration as structure. Application of Sardinella longiceps oil that contain EPA and DHA as immunodulator could accelerate and optimize inflammatory phase in bone healing. Purpose: The aimed of this study to determine the effectivity of the combination of Sardinella longiceps oil and application of hydroxyapatite on the expression of fibroblast growth factor -2 (FGF-2) in bone healing process. Materials and Methods: The experimental units (male wistar rat, n=20) were devided into two groups, control (K) and treathment (P) groups. Treathment groups were given Sardinella longiceps oil per oral seven days before the surgery to make the defect, and continued for seven days later. Control groups were not given Sardinella longiceps at all. All of the wistar rat were have two defects in their right femur, which is use hydroxyapatite (K+ and P2) and no use hydroxyapatite (K- and P1). Defect was made as big as half diameter of round bur in femur. Rats were sacrificed seven days after surgery, and femur were sliced transversally and performed immunohistochemistry staining with Anti-bFGF Polyclonal antibody (Bioss®). Examination were done under light microscope. Data were analyzed using one-way Anova for FGF-2 expression. Result: Sardinella longiceps oil increased the mean of FGF-2 expression on P2 group : 15,29±5,251, compare to K- : 5,57±2,070 , K+ : 12,14±3,976 and P1 : 8,14±1,676 (P<0.05). Conclusion: The combination of Sardinella longiceps oil and hydroxyapatite application effective the expression of fibroblast growth factor-2 in the process of bone healing.

Abstract

Background: The prevalence of diabetes mellitus (DM) is quite high in the world and Indonesia. Diabetes mellitus with poor glycemic control periodontitis as a complication the largest in the oral cavity which causes damage to the alveolar bone. Osteoblasts synthesize osteoid collagen to form bone as raw material. Oxygenation high pressure is able to increase the amount of alveolar bone osteoblasts in patients with DM. Sea cucumber contains growth factors that play a role in the differentiation of osteoblasts to bone formation. Purpose: To determine the effectiveness of combination hyperbaric oxygen therapy 2.4 ATA and sea cucumber gel 3% in the increasing of osteoblasts in rats with DM induced P.gingivalis. Materials and Methods: The study design was factorial design group. Rats are inducted with STZ single dose 65 mg/kg BB to make DM and are inducted with P.gingivalis 1x109 sel/ml to make them suffer periodontitis. Twenty male Wistar rats were divided into five groups. K-(without treatment); K1 (Streptozotocin + P.gingivalis); K2(Streptozotocin + P.gingivalis + sea cucumber gel); K3 (Streptozotocin+ P.gingivalis + HBO); K4 (Streptozotocin +P.gingivalis +HBO + sea cucumber gel). Result: One-way ANOVA here were significant differences (p<0.05) in K2 (24.97 ± 4.99), K3 (22.80 ± 6.45), K4 (28.47 ± 3.20) that compared with K1 (9.22 ± 4.95). Conclusion: Each hyperbaric oxygen therapy doses of 2.4 ATA 100% O2 3x30 minutes with 5 minute intervals for 7 days and sea cucumber gel (Stichopus hermanii) concentration of 3% and a combination of both therapies showed an increase of osteoblasts significantly in rats with diabetes mellitus induced P.gingivalis.

Abstract

Background: Candida albicans acts as a trigger to denture stomatitis. Prevention of denture stomatitis can be done by cleaning dentures with mechanic and chemical methods. Bromelin enzymes from pineapple can be use as a chemical substance to purify dentures for its ability to prevent the growth of Candida albicans with its proteolytic activity. Objectives: to determine the influence of soap from pineapple peel extract as acrylic resin heat cured denture cleanser in Candida albicans impure. Materials and Methods: The experiment is post test only group design. The subject is divided into 6 groups. Control group (aquadest), Positive control group (soap with lime extract brand X), and four treatment groups (soap from pineapple peel extract with 0%, 3,5%, 7%, and 14% concentration). Each group is brushed with an electric tooth brush for 5’. Then each sample is inserted into Saboraud Dextrose Broth and 0,1 ml is taken and is vortexed for 30’ then is incubated in Saboraud Dextrose Agar for 24 hours. The colony is counted with the colony counter in CFU/ml. The data will be tested with One Way ANOVA and continued with LSD test. Result: The mean of Candida albicans colony in K-, P1, P2, P3, P4, K+ is 44.00; 32.25; 2.25; 6.00; 11.50; 19.00. There is a difference between the control groups and all the treatment groups (p<0,05). Conclusions: Soap from pineapple peel extract with the concentration 3,5% is the most effective soap as an acrylic resin heat cured denture cleanser to influence the growth of Candida albicans

Abstract

Background: One of the prevention for Early Childhood Caries (ECC) is maintain daily food through limiting cariogenik food’s consumption, controlling eating frequency, improving oral and dental health. Dental caries would be prevented by controlling eating frequency, improving oral and dental health, so pH plaque can also be controlled. Purpose: Analyze the relationship between the pH plaque to ECC occurrence in an endemic area of malnutrition status. Materials and Methods: Cross section study on 287 children aged 6-48 months in an endemic area of malnutrition in the district Neglasari, Tangerang city in 2013, interviewing the mother, and intraoral examination in children, multistage cluster random sampling method. Result: ECC highest proportion found in plaque pH from 6.0 to 6.4 as much as 79.2% with a value of p = 0.000 (p<0.05) and is one of the better predictors of the occurrence of ECC. Conclusions: There isa significant relationship between pH plaque to ECC occurrence, this variableis accordant with the prevention of ECC because itis included inthe model predictors of ECC.

Abstract

Background: Wound is destruction of part of the body's tissues caused by agent like germs, parasites, fungi, sharp force trauma, temperature changes, chemicals, electric shock. Inflammatory phase in the process of wound healing has very important role. In the chronic inflammation there are chronic inflammatory cell infiltration, namely macrophages, lymphocytes, plasma cells, eosinophils, mast cells, basophils accompanied by the proliferation of blood vessels and fibrosis. Ibuprofen is anti-inflammatory and analgesic drugs that usually given after tooth extraction. Purpose: This study was aimed to examine the effect of preoperative ibuprofen against the spread of chronic inflammatory cells in the process of wound healing after tooth extraction. Materials and Methods: The experimental unit used were 20 male Wistar rats divided into two groups, group1 (NaCMC 0.2%), group2 (Ibuprofen 7,2mg / 200grBB). NaCMC 0.2% for the group K and Ibuprofen for group X given 1 hour before incisor tooth extraction and then the bottom left tooth of mice were be lifted. The Socket were then sutured and left up to 7 days. Data were analyzed by Chi-Square test. Results: Chi-Square test showed significant difference between group K and X p = 0.003 (p <0.05). Conclusion: Preoperative administration of ibuprofen were lower the spread of chronic inflammatory cells in the process of wound healing after tooth extraction.

Abstract

Background: Orthodontic tooth movement depends on bone remodeling involving osteoclasts and osteoblasts. RANKL has an important role in osteoclastogenesis and increased during orthodontic tooth movement. Caffeine contained in coffee, tea, and carbonated drinks can increase osteoclastogenesis. Purpose: To analyze the effect of caffeine on the expression of RANKL and the number of osteoclasts in orthodontic tooth movement. Materials and Methods: The experimental laboratories research used 16 rats were divided into 2 groups: the control group (K): rats given orthodontic mechanical force (OMF) and the treatment group (P): rats given OMF and caffeine at 1,37mg / 100 g twice a day. OMF was conducted by ligature wires were applied on permanent maxillary right first molar and both permanent maxillary incisivus .Subsequently, the permanent maxillary right first molar moved to mesial with Ni-Ti orthodontic closed coil spring. Observations were made by histological examination to calculate the number of osteoclasts and immunohistochemical examination to determine the expression of RANKL. Result: the administration of caffeine increased the number of osteoclast in both compression and tension areas (p<0.05). The number of osteoclast in compression area larger than those which in tension area (p<0.05), The administration of caffeine improved the expresion of RANKL in both compression and tension areas (p<0.05). The expression of RANKL in compression area larger than those which in tension area (p<0.05), Conclusion: The administration of caffeine increased the expression of RANKL and osteoclast numbers on orthodontic tooth movement, therefore it may be an alternative to accelerate orthodontic treatment.

Abstract

Background: Dental and oral diseases which are often found in children is dental caries. Streptococcus mutans is the main cause of caries. Caries can be prevented by using a topical application of fluoride. The Anchovy (Stolephorus insularis) contains protein, vitamins (A, B1, C), and minerals (Fe, Ca, K, F). Calcium fluoride (CaF2) within the anchovy can inhibit the occurrence of dental caries. Purpose: The aim of this study was to determine the antimicrobial ability of anchovy extract (Stolephorus insularis) to Streptococcus mutans. Materials and Methods: This study was a laboratory experimental research with post test only control group design. Diffusion method were applied with 2 controls: negative control used DMSO 1%, positive control used NaF solution, and 3 concentrations of anchovy extract (Stolephorus insularis) 3%, 6%, and 12%, each group were composed of 6 samples. Antimicrobial was assessed by measuring the diameter of the clear zone around the discs contained the anchovy extract (Stolephorus insularis). Data were analyzed by Kruskal-Wallis test followed by Mann-Whitney test. Result: The results from this study showed clear zone around the discs of the anchovy extract (Stolephorus insularis). The more concentration of the extract showed the more antimirobial zone diameter. The average zone of antimicrobial at the concentration of 3% were 7,11 mm, 6% 9,5 mm, 12% 10,78 mm, for the negative control DMSO 1% 6 mm and the positive control NaF solution 8,16 mm. The largest diameter of the clear zone was at concentration of 12% (P < 0,05). Conclusion: The anchovy extract (Stolephorus insularis) had an antimicrobial effect to the growth of Streptococcus mutans.

Abstract

Background: Periodontitis have a bidirectional relationship with diabetes mellitus, both can increase oxidative stress that trigger an increase of pro-inflammatory cytokines, such as TNF-α. Sea cucumbers have anti-inflammatory component that act to inhibit the release of inflammatory mediators. Hyperbaric oxygen therapy reduce oxidative stress. Purpose: The aim of this study is to investigate the effect of the sea cucumber powder and hyperbaric oxygen therapy on the expression of TNF-α in rats with diabetes mellitus induced by P.gingivalis bacteria. Materials and Methods: The study was an experimental laboratories research with factorial design. Twenty wistar rats were divided into 5 groups. K0 negative control, K1 positive control. K1-K4 groups were induced with streptozotocin for diabetes condition and P.gingivalis bacteria for periodontitis condition. K2 group was treated with seacucumber 3% for 7 days, K3 with OHB 2.4 ATA 3 x 30’ interval 5’ for 7 days, and K4 group was treated with combination of sea cucumber and OHB. At the 51 st day all rats were sacrified, then the expression of TNF-α on periodontal macrophages were examined by immunohistochemistry stain. Data were analized by Kruskal Wallis followed by Mannwhitney. Result: Expression of TNF-α were increased in K1 (11.50±1.291) compare K0 (p<0.05). Sea cucumber treatment, OHB, and combination treatment decreased expression of TNF-α significantly in amount of 2.50±0.577 (K2), 8.25±2.217 (K3), 3.00±0.816 (K4). Conclusion: Sea cucumber powder and OHB therapy affected the expression of TNF-α in rats with diabetes mellitus induced by P. gingivalis bacteria.
