Abstract

Kefir is a probiotic drink act as therapeutic agents in liver damage caused by exposure of formaldehyde. Therapeutic activities were estimate based on the content of kefir like antioxidants, lactic acid bacteria with yeasts, and essential amino acids. The purpose of this study was to prove the influence of kefir on the histological and levels of malondialdehyde (MDA) in liver of white mice (Mus musculus) male Balb C strain that exposure by formalin. The method used is experimental method, with three treatment groups, namely: (1) normal group, (2) the induction of formaline without kefir therapy group, (3) the induction of formaline with kefir therapy group. Histological of liver viewed by HE staining and MDA levels were measured by TBARS method. There are differences in the average number of significant cell damage induced by formalin group (p = 0030, p < 0.05). There are significant differences in MDA levels induced by formalin group (p = 0023, p < 0.05). These results are based on the ability of vitamin C, E and carotene (antioxidant) against Reactive Oxygen Species (ROS) due to exposure of formaldehyde, thus preventing the MDA levels increasing. Lactic acid bacteria and yeasts can preclude the possibility of  healthy cells to be attacked by ROS with increasing the body’s immunity. Essential amino acids can repairing damaged cells by stimulating cell regeneration. It can be concluded that kefir with a dose of 107 CFU / ml act as therapeutic agents in liver damage due to exposure to formaldehyde.

Abstract

The Dayak Benuaq tribe is one of the tribes in Indonesia who live in East Kalimantan. They have various knowledge related to raw materials and medical treatment traditions for generations that have never been documented, especially in Intu Lingau Village, Nyuatan District, West Kutai, East Kalimantan. This study aims to documentation of medicinal plants and herbs of the local communities. The finding shows that there are 104 plants used, which is divided into 34 families. The most common plant parts used in the treatment were leaves, the most common processing method was boiling, and the most utilized method was by drinking.

Abstract

Pengetahuan penggunaan tumbuhan sebagai obat tradisional hanyalah berdasarkan empiris. Perubahan pola pengobatan modern ke pengobatan tradisional, menyebabkan kebutuhan terhadap obat yang berasal dari bahan alam menjadi meningkat. Bahan alam yang digunakan sebagai obat sebaiknya berpegangan kepada pedoman bahwa bahan obat tersebut tidak menimbulkan keracunan baik akut maupun kronis dan terbukti bisa menyembuhkan penyakit atau berkhasiat sebagai obat serta dapat diperoleh secara kontinu. Secara empiris masyarakat kota Malang menggunakan umbi binahong (Anredera cordifolia (Ten) Steenis) untuk mengobati nyeri pada gigi yang disertai dengan pembengkakan yang keluar nanah, gastritis akut, nyeri kepala, panas dalam yang disertai sariawan, mengobati luka bekas operasi, mengurangi nyeri setelah operasi dan lain-lain. Ciri-ciri morfologi umbi binahong berbentuk silindris dengan tekstur permukaan yang tidak rata, panjangnya antara 4-7 cm dengan diameter 0,5-2 cm dan berdaging lunak. Secara anatomis tampak jaringan dasar (parenkim), berkas pengangkutan (xylem dan floem) dan benda ergastrik berupa amilum. Tumbuhan ini mudah tumbuh di dataran rendah maupun dataran tinggi. Kata kunci: obat tradisional, umbi, Anredera cordifolia (Tenn) Steenis,.

Abstract

Kanker payudara menduduki peringkat kedua setelah kanker leher rahim yang menyerang wanita di seluruh dunia. Eksplorasi terhadap tumbuhan yang mudah ditemukan, seperti herba Hedyotis corymbosa, sebagai alternatif pengobatan kanker payudara perlu dilakukan. Dilakukan uji in vivo terhadap lima kelompok tikus Galur Sprague Dawley terdiri atas kelompok perlakuan DMBA (20 mg/kgBB), perlakuan ekstrak etanolik herba H. corymbosa (1500 mg/kgBB), perlakuan DMBA + dosis I (750 mg/kgBB), perlakuan DMBA + dosis II (1500 mg/kgBB) dan perlakuan pelarut CMC - Na 0,5%. Kelompok perlakuan DMBA, DMBA + dosis I, dan DMBA + dosis II diinduksi DMBA 10 kali selama lima minggu. Kemudian setelah diistirahatkan selama empat minggu, dilakukan pemberian ekstrak selama 10 hari. Pada akhir percobaan dilakukan pengorbanan kemudian diambil jaringan payudara untuk diamati aktivitas proliferasinya. Dilakukan pengamatan imunohistokimia terhadap protein c-Myc pada jaringan payudara. Pengamatan pada metode imunohistokimia terhadap protein c-Myc menunjukkan adanya perubahan yang positif pada kelenjar payudara kelompok perlakuan DMBA + dosis I. Hasil yang diperoleh menunjukkan bahwa ekstrak etanolik herba H. corymbosa dosis 750 mg/kgBB sudah mampu menghambat proliferasi sel kanker payudara dan tidak bersifat dose dependent. Kata kunci: kanker payudara, Hedyotis corymbosa, proliferasi sel, c-Myc

Abstract

Chronic wounds are still problem is one of the most frequent among the community. Pressure ulcers (decubitus) are examples of types of chronic wounds in which the maintenance costs of the injury fairly expensive. The use of plants is an appropriate alternative for the treatment of decubitus chronic wounds. One among the many plants that are known efficacious drugs for the health of Aloe vera. With the various components contained therein, aloe vera is known to affect the decubitus chronic wound healing. This study is purely experimental posttest-only design approach. The sample uses a white rats strain wistar female, weight 150-250 gr and 2,5 to 3 months of age. The variable is the concentration of the extract of Aloe vera and scores healing pressure ulcers (decubitus) degree II in white rats. The data obtained using observation, then analyzed with the assumption test (Lilliefors test and the Barlett test), one way Anova test and different test real distance of Duncan with significansy values of 0,05. The results indicate that there is the influence of various concentrations of leaf extract of Aloe vera to diameter decubitus chronic wound healing in rats and the concentration of leaf extract of Aloe vera 100% to give a good effect on the diameter of decubitus chronic wound healing on white rats (á 0.05).

Abstract

Cervical cancer is one of leading cause of cancer death in women in the developing countries. One of the strategy to prevent cervical cancer based on cytotoxic agents are now being developed. Ciplukan (Physalis angulata L.) is one of potential plant as chemopreventive agent due to Physalin and Withangulatin constituent in this plant. This study was aimed to know cytotoxic effect  of ciplukan ethanolic extract (CEE) in human cervical carcinoma Hela cell line. Evaluation of cell viability value was determined using MTT assay. The expression of p53 as cell proliferation regulator was observed with immunocytochemistry assay. Ethanolic extract of ciplukan showed cytotoxic effect in HeLa cell line with IC50 of 158 µg/ml. Further observation of cell proliferation regulator showed that CEE induces expression of p53 that inhibit cell proliferation. The result showed that CEE has potential activity to be developed as anticancer agent in human cervical cancer.

Abstract

Most of probiotic product as functional food were developed from dairy product. However, milk protein content cause allergy and make it limited to non vegetarians only. To solve these problems, some efforts were tried to produce probiotic drink from fruit juice, such as pineaple, tomato, papaya and aple. The aims of this research were to know the quality of probiotic fruit juice organoleptically and the viability of probiotic in it. Lactobacillus plantarum were used as probiotic and the fermentation process were last for 5 days at 30°C. The results showed that the best quality performed by fermented aple juice, with sour taste, pale brown color, aple fermented aroma and good homogenity. The probiotic viability also showed highest number, 1,7x108 cfu/ml. This research can be developed to produce fermented fruit juice with better quality.

Abstract

The purpose of this research was to study the effect of green tea polyphenol to TNF-á production on human trophoblast cell culture exposed by 33 mM glucose. Trophoblast culture isolated from human fetal placental tissue by sectio caessaria. Monolayer trophoblast cells that had been incubated for 3 days at 5% CO2; 37°C were divided into 2 groups: (1) normal glucose (5 mM) and (2) glucose 33 mM exposure, both divided into 2 sub groups: (a) without green tea polyphenol treatment, and (b) green tea polyphenol treatment 0,1; 0,2; and 0;4 mg/ml. Cells incubated for 3 days at 5% CO2; 37°C then analyzed cytotrophoblast cells characteristic. TNF-á level was measured by ELISA and analyzed with oneway ANOVA. Immunocytochemistry showed the number of cells that expressed TNF-á then analyzed descriptively. The results of this study showed that TNF-á level at 0,1; 0,2; and 0,4 mg/ml polyphenol were 2804,333 ñg/mL; 2513,222 ñg/ml; and 2739,889 ñg/ml respectively compared with 2739,889 ñg/mL normal glucose without polyphenol, and 2721,000 ñg/mL; 2612,111 ñg/mL; and 2566,555 ñg/mL compared with 2621,000 ñg/mL glucose 33 mM exposure without polyphenol. Number of cells that expressed TNF-á with 0,1; 0,2; and 0,4 mg/ml polyphenol treatment were 0%; 2%; and 5,5% compared with 0% normal glucose without polyphenol, and 82%; 0%; and 0% compared with 3% glucose 33 mM exposure without polyphenol. Green tea polyphenol exposure for 3 days at 0,1; 0,2; and 0,4 mg/ml didn’t significantly affect the decreasing of TNF-á production. Keywords: GDM, green tea, polyphenol, TNF-á, trophoblast

Abstract

Breast cancer represent one of the most second malignancy of the woman after cervix cancer. In Indonesia estimated ±100 new case per 100.000 population every year, meaning more or less 200.000 new case every year to all cancer type. While for the breast cancer ± 23.140 new case every year. If breast cancer found in early stage, it has high life expectancy, ranging from 85-95%. But actually 70-90% patient came to hospital after their disease were hard and advanced stage. Cancer medication at advanced stage was very difficult and its result very dissatisfactory. The aim of research is to know the epidemiology, clinical symptom, diagnosis, therapy, prognosis. Descriptive study with retrospective design. Total sampling was all outpatient in oncology deparment of Hasan Sadikin Hospital of Bandung with advanced stadium of breast cancer from January 2003 up to December 2008. 164 eligible respondens who had age below 35 yo 28 (17.1%) and above 35 yo 136 (82.9%). Average of menarche 11.77 yo. Location of right breast cancer 78 (47.6%), left 82 (50%), and both 4 (2.4%). A lot of stadium was T4bNoMx that is 18 (11%). Chest x-ray, laboratory, and USG were the most diagnostic procedure often used that is 19 (11.6%). The most common therapy was BE+VC+RM that is 17 (10.4%). The prognosis was healing 20 (12.2%),  not/not yet healed 30 (18.3%), not control 110 (67.1%), and who control in the other place 4 (2.4%). Conclusion was mostly diagnosis of breast cancer according to anatomy pathology, chest x-ray, laboratory, and ultrasonografi. A lot of breast cancer location was left side. Excision biopsy, and followed by radical mastectomy were the most therapy procedure. Therapy evaluation to assess the level of healing was difficult because most patient did not conduct the control after therapy.

Abstract

Curcumin, the major yellow pigment of Curcuma sp. has several interesting biological activities, such as anticarcinogenecity, antiinflamatory, and antioxidant. Curcumin naturally occurs as major compound in Curcuminoid substance beside its derivatives, i.e. Bisdemethoxycurcumin (BDC) and Demethoxycurcumin (DC). Quantitative analysis of Curcumin is difficult to carry out unless prior to completely resolved Curcumin from BDC and DC. A good resolution, reproducible, and accurate method for determination Curcumin in pharmaceutical product (capsule formed) by HPLC has been carry out. Using C18 colurnn and mixture of Acetonitrile: Acetic Acid glacial 1% (45:55 v/v) as eluent, BDC, DC, and Curcumin were retained with retention time of  10.79, 12.29, and 13.91 minutes, respectively. Curcumin was simultaneously separated from BDC or DC. The resolution factor (Rs) between Curcumin and DC was 1.73. Good linear relationship (r = 0.9997, Vxo = 0.99%) between peak area and concentrations was found at range of the Curcumin concentration used, i.e. 23.56-62.83 µg/mL. Curcumin recoveries were (93.60-118.17)% with the coefficient of variation (CV) 6.99%. Curcumin content in each capsule sample was found to be (8.89±0.06) mg. It showed that samples contained higher amount of Curcumin than that claimed on its label, 5 mg.
