Abstract

Sodium benzoate is a preservative that is permitted to be used in food and beverages. The maximum limit for using sodium benzoate in food is 600 mg/L of ingredients calculated as benzoic acid. In this study, High-Performance Liquid Chromatography (HPLC) method using a PDA detector was used for sodium benzoate levels analysis in fruit juice drink circulating in Malang. The mobile phase composition used in this study was methanol pro-HPLC:aqua pro injection (70:30) and added glacial acetic acid to reach pH 3. The analysis was conducted at a wavelength of 245 nm, and the flow rate was set at 1.00 mL/minute. This method was validated against the parameters of selectivity, linearity, accuracy, and precision, and the results obtained to meet the validation requirements. The application of the HPLC method after being validated in the determination of sodium benzoate levels in fruit juice drink circulating in Malang resulted in selectivity Rs ≥ 1.5, linearity obtained in the range 0.1-5 μg/mL with a correlation coefficient (r) = 0,9999, recovery at a concentration of 80%, 100%, and 120% was 105.31%, and precision with an RSD value of 1.40%. Based on the results, it can be concluded that the validation method of analysis shows that the method can be used to determine sodium benzoate level using HPLC with PDA detectors for routine use in fruit juice drink samples.

Abstract

Liver is an the important organ, it plays an essential role in maintaining the biological equilibrium of vertebrates. Reactive oxygen species (ROS) play an important role in pathological changes in the liver. The research has been carried out to know the potential use of buah merah (Pandanus conoideus) extract as hepatoprotrctive agent against CCl4 poisoning in mice. In this study, the hepatoprotective activity was determined by the reduction of SGPT (serum glutamic pyruvate transaminase) and SGOT (serum glutamic oxaloacetic transaminase) level. Application of buah merah extract significantly (p< 0,05) reduced SGPT and SGOT level. Microscopically, several changes were found, such as severe hydrofic degeneration and necrosis at liver cell that treated by CCl4. Base on histological examination of application of buah merah extract, could inhibit damage and reduced the degeneration and necrosis of liver cell. Keywords: buah merah, SGPT, SGOT, hepatoprotective.

Abstract

The aim of this research was to study the effect of a-pinene on number of cell that express iNOS (inducible Nitric Oxide Synthase) on lipopolysaccharide (LPS)-induced neuron-glial culture. Neuron-glial culture were divided into eight groups: (1) normal; (2), (3) and (4) incubated with a-pinene (0,2; 0,4 and 0,8 μg/mL respectively, for 24 hours); (5) incubated with LPS 1 μg/mL; (6), (7) and (8) incubated with a-pinene (0,2; 0,4 and 0,8 μg/mL respectively, for 24 hours) and then induced with LPS (1 μg/mL, on 1 last hour of treatment period). The expression of iNOS in neuron-glial culture was detected by immunocytochemistry using anti-iNOS rabbit polyclonal IgG and goat anti-rabbit IgG Biotin Conjugated. The result shown that iNOS were expressed on normal group. Lipopolysaccharide 1 μg/mL induction causing iNOS expression increased approximately three times higher than normal (from 9,7% became 28,7%). Inducible NOS expression on non LPS-induced neuron-glial culture did not affected by a-pinene. Whereas a-pinene 0,2 and 0,4 μg/mL could suppress iNOS expression on LPSinduced condition. a-pinene 0,2; 0,4 and 0,8 μg/mL could help neuronglial cells survived on LPS-induced condition, but normal neuron-glial culture’s density was decreased when incubated with a-pinene 0,8 μg/mL.  Keywords: alpha-pinene, cell culture, glial, iNOS, LPS, neuron

Abstract

Natural product that has developed as anticancer drugs are consists from secondary metabolites that has isolated from plant, ocean plant and microorganisms. To maintain its productivity, must be concerned to the cultivation of the medicinal plants. Medicinal plants need time to grow and producing secondary metabolites that would be isolated, so that the production of secondary metabolites compound from plant is very limited.             The plants that infected by endophytic fungi be able to grow faster than the normal plants. Endophytic fungi also have ability producing secondary metabolites compound that produced by the host plant. Taxol, podophyllotoxin and camptothecin are compounds that proved has anticancer activity. Endophytic fungi have also proved increasing the production of the compound.

Abstract

Celery (Apium graveolens L.) is commonly used to lower blood pressure, antirheumatic, relaxant, mild diuretic, antiseptic for the urinary tract, antioxidant, and anti-inflammation. According to previous studies, a number of the phytochemicals found in the plant show cytotoxicity toward some types of cancer cells. However, studies on the cytotoxic effects of celery herb ethanolic extract (CEE) on breast cancer cell (T47D), colon cancer cell (WiDr), and cervix cancer cell (HeLa), however, has not been done yet. Our research aims at doing so. Cytotoxicity test was conducted using MTT assay and its absorbance was read using ELISA reader at λ = 595 nm. Results of the assay show that CEE reduces cell viability at concentrations of 100-750 µg/ml on HeLa cells, while reduction of T47D and WiDr cell viability was not achieved until concentrations of 500-750 µg/ml. Based on these results, we conclude that CEE hold many potentials for further developments as preventive and therapeutive agent in cancer treatment.

Abstract

Catechins , isolate from green tea  GMB4 clones could be developed as a potential preventive agent for atherosclerosis. This activity is thought to be based on the structure epimerisation, hydroxy phenolic  and galloil group of these isolates. The purpose of this study is to prove the influence catechins ,isolate from green tea, on the inhibition of the decrease eNOS expression in male wistar rats induced by  high  fat diet. The method used in this study consisted of ELISA for measurement of eNOS expression. The results showed that  catechins inhibits the decreasing expression of eNOS at a dose of 6 mg / day and 24 mg / day was significantly but not dose 3mg / day. This suggests a dose 6mg / day and 24 mg / day is able to repair the damage signaling insulin on PI3K pathway caused by insulin resistance. These results are based on the ability of catechins as antioxidants. Catechins with hydroxy phenolic group  is a potential electron donor to bind  free radicals. Polyphenol catechins may also modulate  signaling cellular processes such as NFkâ, activator protein 1 DNA binding, glutathione biosynthesis, PI3K / Akt, MAPK pathway and the Nrf2 transcription factor that regulates expression of several antioxidant genes. It can be concluded catechins  inhibits  decreasing expression of eNOS  at a dose of 6 mg / day and 24 mg / day and can be developed as an agent and vasoprotective ateroprotective.

Abstract

Cyclooxygenase is an enzyme that plays a role in the formation of prostaglandins, which can cause inflammation and pain when overexpressed. This study aims to determine the interaction between COX-2 macromolecules (receptors) with their ligands, namely indomethacin and diclofenac in silico using the Molecular Docking method. The COX-2 receptor was downloaded in the form of a 3D structure from the RCSB GDP with code 5F19. Diclofenac Ligand and Indomethacin were downloaded in the form of a 3D structure from the RCSB GDP with 4ZBQ code and 4IK7 code. The results showed that the interaction between COX-2 and indomethacin was the interaction of hydrogen, which linked indomethacin with amino acid Leu531 and steric interactions between indomethacin and amino acids Trp387, Tyr385, Tyr355, Leu352, and Val523. The interaction of [COX-2 – Indomethacin] produces a value of ∆G of -103.136 kcal/mol, and the value of RMSD is 1.244 Å. Whereas, the interaction that occurs in COX-2 with diclofenac is the steric interaction that happens between diclofenac with amino acids Leu390, Trp387, Gln203, His388, His207, and Thr206. The interaction parameter between [COX-2-Diclofenac] obtained ∆G value of -7.843 kcal/mol and an RMSD value of 2.07851 Å.

Abstract

Penelitian bertujuan mengetahui pengaruh sari seduh teh hitam terhadap pengekspresian PPAR ã sel adiposa jaringan lemak visera Rattus norvegicus strain Wistar. Duabelas ekor tikus (Rattus norvegicus strain Wistar) jantan umur 6-8 minggu, berat 200 gram, diberi 4 macam perlakuan, yaitu A (diet tinggi lemak + SSTH 0 g/hari), B (diet tinggi lemak + SSTH 0,015 g/hari), C (diet tinggi lemak + SSTH 0,030 g/hari) dan D (diet tinggi lemak + SSTH 0,045 g/hari) selama 90 hari. Setelah masa perlakuan, tikus dibedah dan diambil lemak viseranya. Jaringan tersebut kemudian dibuat preparat dengan metode parafin. Jumlah sel yang mengekspresikan PPAR ã dianalisis dengan menggunakan pewarnaan immunohistokimia dan untuk mengkonfirmasi bentuk sel adiposa digunakan pewarnaan Hematoxylene&Eosin. Antibodi primer yang digunakan adalah anti PPAR gamma poliklonal antibody rabbit IgG dan antibodi sekunder biotin-goat-anti rabbit IgG. Hasil penelitian menunjukkan jumlah sel adiposa yang mengekspresikan PPAR ã mengalami penurunan seiring dengan penambahan dosis SSTH. Hal tersebut mengindikasikan SSTH dapat menurunkan pengekspresian PPAR ã pada sel adiposa jaringan lemak visera. Kata kunci: jaringan lemak visera, PPAR ã, diet tinggi lemak, sari seduh teh hitam

Abstract

Communications is process transfer of information signing the forwarding of message of someone to others, either through indirect and also direct that is media.Television as a mass media technically capable of informing public in numerous amount in the same time. One of television program offering various product to fulfill requirement and satisfaction of physical is commercial. Such commercial is influenza drug commercial, because met many people. This matter of tendency make people to do self-medication as solving of from problem of health which they recognize by self.             Data collecting uses purposive sampling method in 102 respondents . Instrument test is done by validity test and reliability testing . Data analysis tool used is correlation analysis,t-test and regression analysis. Based on the result of the research, correlation coefficient is 0.645 so r table is 0,195, means there is a relation between influenza drug commercial and self-medication drug of choice. Based on the test t, t calculation is 2,376 so t table is 1,980, means there is influenza drug commercial in television has significant effect to self-medication drug of choice with equation regression Y = 4.803 + 0.522 x, means if influenza drug commercial in television indicator increasing 1 so self-medication drug of choice increasing 0.522. Value Sig. Of 0.000 <0.05 then the correlation coefficient deduced influenza drug commercial to self medication ‘s drug of choice in people in Malang is significant.

Utilizing of antibiotics to cure operation infection actually not differ from gift antibiotics in general. The difference was antibiotic treatment be addition therapy after surgery. The aim of antibiotics treatment was prevent or cure infection by reducing or eliminating bacteria until human body can handle this pathogenic organism.
