Abstract

Diabetes mellitus (DM) is a chronic condition which caused by insulin secretion disorder, insulin resistance or both. High number of diabetics per year has led many researchers to conduct research to find the most safe and effective medicine for DM treatment. Flavonoid are known to act as inhibitors of α-amylase as a parameter of pancreatic damage and help lower blood glucose levels. One source of flavonoids is purslane. The aim of this study was to examine the effect of purslane ethanol extract on glucose and α-amylase levels in rat. Twenty rats Rattus novergicus L. strain Wistar are divided into 5 groups; control, positive control, and three test groups, each was given purslane ethanol extract of 100 mg/kgBW, 200 mg/kgBW, and 300 mg/kgBW. Blood glucose and body weight were measured before alloxan induction, after induction, as well as day- 7, 14, 21, and 28; while feed and drink consumption were measured everyday starting with the administration of purslane ethanol extract. The α-amylase test was carried out on the 28th day using enzymatic UV determination of degradation product of maltose and glucose method. The analysis result showed that rats blood glucose and α-amylase decreased in each group with purslane extract. The antioxidant content of flavonoids in purslane ethanol extract has been shown to have anti-diabetic effects by repairing pancreatic function.

Abstract

The Mixed Shih-Tzu dog, 3 years old and 4.3 kg body weight, had clinical signs of unable to stand, both rear limbs suffer from diplegic lameness so that they are only able to rely on both forelimbs. The owner said that her dog is traumatized by a four-wheeled vehicle. The results of blood profile before surgery showed physiological, only an increase in leukocyte cells 18.83 x103/µL. The radiographic examination with a ventral-dorsal position shows oblique fractures in the pelvic dexter and dislocation of the pelvic sacroiliac sinister bone. The method of handling used was open reduction internal fixation (ORIF) using a 2.0 veterinary cuttable plate (VCP) bone plate with a 2.0 mm cortical self-tapping screw for pelvic fractures and 2.7 mm cortical non-self-tapping for sacroiliac dislocations. The dog walked and showed a good progress in one week after ORIF, based on clinical and post-operative radiographic examinations, sacroiliac fracture-dislocation fixation using VCP with 2.0 mm self-tapping screw cortical for pelvic fracture and 2.7 mm non self-tapping for sacroiliac dislocation provides excellent stabilization and no visible complications.

Abstract

Growth Promoter Antibiotics are used to prevent disease and promote growth and production in poultry. Repeated admnistration of feed can have a microorganic resistance effect, accumulation of antibiotic residues in animal and environmental products and imbalance of normal microflora in the intestine. The antibacterial and carbohydrate content of some natural ingredients can be potential as a replacement candidate for AGP. This study aims to determine the role of a combination of wild ginger, honey, and probiotics (Bacillus subtilis and Lactobacillus acidophilus) as AGP candiate in Vitro. The antibacterial activity of the combination of wild ginger and honey against pathogens (E. coli) and their use against probiotics was tested by disk diffusion method, while the calculation of optical density values to determine the minimum inhibitory concentration and minimum bactericidal concentration was carried out on E. coli. The ability of inhibition of probiotics against pathogens is also done by the disk diffusion method. The disk diffusion test results showed the best combination was extract of 25% wild ginger aquades + 100% Lombok honey with inhibition zone diameter (8,53 ±, 03). Optical density values indicate this combination is able to inhibit and kill E. coli (DO 0,00 ±, 002) and support B. subtilis (DO 0,18 ±, 002) and L. acidophilus (DO 0.25 ±, 005) significantly better than positive control. MIC value of wild ginger aquades extract and honey combination against E. coli is wild ginger aquades extract 3.13% + Lombok honey 25%, while MBC value is wild ginger aquades extract 6,25% + Lombok honey 25%. The combination of B. subtilis and L. acidophilus showed the largest inhibitory zone diameter against E. coli (7,30 ±, 02 mm) compared to individual colonies. The combination of wild ginger and honey, in addition to inhibiting also able to kill pathogens and support the growth of probiotics, so this formula can be used as one of the replacement candidates for AGP.

Abstract

One of the native catfish to Indonesia is the baung fish (Hemibagrus). Baung fish are found in rivers in Sumatra, Kalimantan and Java. The catfish population is declining, thus conservation is needed to prevent extinction. To conduct an effective conservation efforts, molecular studies are needed to confirm the fish species from the three islands. Mitochondrial DNA is one of the markers that is often used to see the lineage and kinship of animals for conservation purposes. The purpose of this study is to determine the genetic diversity of the NADH Dehydrogenase Subunit I (ND1) gene catfish from each of these locations, can be used as a genetic marker. Samples were obtained from their natural habitat, namely Magelang (5), Palembang (3), Riau (2), Samarinda (2), 2 from Sintang, and 3 from Banjarmasin. The DNA of the fish sample was then isolated and then used as a template for amplification of DNA fragments using PCR techniques. Amplikon (PCR product) was then purified by gel extraction and then sequenced to determine the DNA sequence. The potential of DNA sequences as catfish genetic markers was proven by analyzing genetic diversity between species using the MEGA version 7.0 program (Kumar et al., 2016). The sequencing results 972 nucleotides composing the ND gene, and found differences in 268 nucleotide sites and 47 amino acid sites. Based on the nucleotide sequence of the ND1 gene, catfish from the Progo river (Magelang, Central Java), the Musi river (Palembang, Sumatra), the Kampar river (Riau), the Kapuas river (Sintang, Kalimantan), the Martapura river (Banjarmasin), the Mahakam river (Kalimantan) is included in the Hemibagrus sp.; catfish from the Elo river (Magelang, Central Java) belongs to the genus Mystus sp.; catfish from the Bengawan Solo river (Bojonegoro, East Java) belongs to the Pangasius sp.

Abstract

Abstract              The crop is the part of the chicken's digestive tract that is more likely to break during processing. This research aimed to isolate Salmonella sp. from crop in local chickens and broiler from the Peunayong Chicken Market in Banda Aceh. Samples were swab contents of crop from 15 crops of local chickens and 15 crops of broiler. This research is a Cross sectional study with Carter method, starts with a swab of crop contents and then is inserted into the Selenite Cystine Broth (SCB) media, if the color of the media becomes orange, then followed by streaking bacteria on Salmonella Shigella Agar (SSA) media. Then observed the morphology that grew on SSA media including size, shape, surface, elevation, edges, aspects and colony color, followed by Gram stain to observe bacterial morphology microscopically. The research data were analyzed descriptively, displayed in figures and tables. The results of the isolation showed that 7 from 15 samples (46%) of local chicken cropping and 15 from 15 samples (100%) of broiler were positive for Salmonella sp. Therefore, it can be concluded that Salmonella sp. can be isolated from local chicken and broiler, Salmonella sp. prevalence is higher in broiler than in local chicken.Keywords : Eviserasi; Food borne disease; Gram negatif; Zoonotic disease Abstrak              Tembolok merupakan bagian saluran pencernaan ayam yang lebih mungkin pecah selama pemrosesan. Penelitian ini bertujuan untuk mengisolasi Salmonella sp. pada tembolok ayam buras dan ayam ras yang berasal dari Pasar Ayam Peunayong Kota Banda Aceh. Sampel berupa swab isi tembolok dari 15 tembolok ayam buras dan 15 tembolok ayam ras. Penelitian ini merupakan Cross sectional dengan metode Carter dimulai dari swab isi tembolok kemudian dimasukkan kedalam media Selenite Cystine Broth (SCB), apabila warna media menjadi orange dilanjutkan dengan penanaman bakteri pada media Salmonella Shigella Agar (SSA). Kemudian diamati morfologi yang tumbuh pada media SSA meliputi ukuran, bentuk, permukaan, elevasi, tepi, aspek dan warna koloni, dilanjutkan dengan pewarnaan Gram untuk mengamati morfologi bakteri secara mikroskopis. Data hasil penelitian dianalisis secara deskriptif, ditampilkan dalam bentuk gambar dan tabel. Hasil isolasi didapatkan bahwa pada 7 dari 15 (46%) sampel tembolok ayam buras dan 15 dari 15 sampel (100%) sampel tembolok ayam ras positif terdapat Salmonella sp. Oleh sebab itu, dapat disimpulkan bahwa Salmonella sp. dapat disolasi dari tembolok ayam buras dan ayam ras, prevalensi Salmonella sp. lebih tinggi pada ayam ras dibandingkan ayam buras.Kata kunci : Eviserasi; Food borne Disease; Gram negative; Zoonotic disease

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian gonadotropin releasing hormone (GnRH) terhadap peningkatan kualitas spermatozoa sapi bali. Penelitian ini menggunakan dua ekor sapi bali berumur 2 dan 4 tahun dengan rancangan pola bujursangkar latin 2 perlakuan dan 2 periode perlakuan. Perlakuan yang dilakukan adalah P1 (NaCl fisiologis) dan P2 (GnRH 100 µg) dengan interval periode waktu perlakuan adalah satu minggu. Sampel semen dikoleksi menggunakan vagina buatan tiga hari setelah perlakuan. Data hasil pemeriksaan kualitas spermatozoa sapi bali dianalisis dengan uji Wilcoxon Signed Ranks Test. Berdasarkan uji statistik menunjukkan bahwa pemberian GnRH 100 µg tidak berpengaruh nyata (P>0,05) terhadap pemeriksaan warna, pH, konsistensi, volume, konsentrasi dan motilitas spermatozoa sapi bali. Hasil pemeriksaan menunjukkan bahwa warna semen pada kedua perlakuan adalah krem dan putih susu. Derajat keasaman (pH); konsistensi; volume semen (ml); konsentrasi (10⁶/ml); dan motilitas spermatozoa (%) sapi  bali pada P1 vs P2 adalah 7,0±0,0 vs 7,0±0,0; 2,5±0,71 vs 3,0±0,0; 4,5±2,8 vs 4,8±2,8; 925±106,07 vs 1210±155,56; dan 69,0±1,41 vs 71,5±0,71 (P>0,05). Disimpulkan bahwa pemberian GnRH 100 µg tidak memengaruhi kualitas spermatozoa sapi bali.

Abstract

One of the effort to provide superior rams is to pay attention to the provision of feed that has good substances and is able to improve the quality of sperm. Based on studies, snail flour has a fairly high protein content and snails can be an alternative source of protein in additional feed. This study aims to improve the quality of sheep sperm using an additional feed of snail flour (Achatina fulica) which can increase the use value and economic value. Eight sheep used in this research were divided into one group as the control and two groups as the treatment. The control group was given standard sheep forage, while the treatment groups were given standard sheep forage added with flour snail of 5% and 10%. Sperm collection using castration method. Parameters observed were motility, viability, and concentration of sperms. The results of the study showed that the addition of snail flour as additional feed for sheep affected sperm quality with a significant increase in concentration parameters, while motility and viability did not show a significant increase

Abstract

Ket-A-Xyl® (Ket-A-Xyl® 20 ml, AgroVet, Peru) merupakan sediaan anastetika jadi yang telah banyak dipasarkan di Indonesia untuk induksi anastesi pada anjing. Studi ini menguji penggunaan obat Ket-A-Xyl® untuk kastrasi pada kucing. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh pemberian Ket-A-Xyl® terhadap parameter fisiologis kucing jantan domestik. Sebanyak 53 ekor kucing jantan domestik dengan berat badan berkisar 1,5 – 5,68 kg dipuasakan selama 8 jam kemudian ditimbang untuk menentukan dosis anestesi. Kucing diinjeksikan sediaan Ket-A-Xyl® secara intramuskular, kemudian diamati dan dicatat perubahan fisiologisnya. Demikian juga untuk onset dan durasi obat dicatat saat kucing memasuki stadium anestesi I hingga tahap recovery. Analisis data parameter fisiologis dilakukan menggunakan aplikasi SPSS dengan uji paired sample t-test. Kelompok yang dibandingkan dalam uji paired sample t-test tersebut adalah kelompok data fisiologis kucing pada tahap stadium III dengan tahap recovery. Hasil analisis data menunjukkan bahwa sediaan Ket-A-Xyl® menimbulkan efek yang signifikan terhadap frekuensi napas (p<0,05), tetapi tidak pada suhu dan frekuensi pulsus. Rata-rata onset dan durasi obat Ket-A-Xyl® menunjukkan waktu yang lebih singkat dibanding studi sebelumnya yang menggunakan obat atropin-ketamin-xylazin. Sediaan anastesi Ket-A-Xyl®  berpotensi  sebagai  obat anestetika yang baik  pada  kucing  domestik  karena  lebih  efisien,  onset dan durasi singkat, serta tidak menimbulkan respon fisiologis yang fatal selama penggunaannya.

Abstract

This study was aimed to identify the presence of E. coli O157:H7 and analyze the association between factors and frequency of E. coli in chicken meat sold at Kaget Market, Pagi Market, and Pembangunan Market in Pangkalpinang City.  A total of 30 chicken meat samples were collected proportionally using simple random sampling.  This study was used a cross sectional method.  The examination of E. coli was carried out based on SNI 2897:2008 concerning The Testing Method of Microbial Contamination in Meat, Eggs, and Milk and Their Processed Products.  The E. coli O157 was confirmed with O157 latex agglutination test and H7 antiserum test.  The data were analyzed using a descriptive test and risk factors were analyzed using chi-square and odds ratio.  The results showed that E. coli O157:H7 was found in 2 of 3 samples (66,7%) isolated from Kaget Market.  The significant risk factors (P<0,005) for the presence of E. coli were traders with age of more 45 years, kiosk distance of less than 5 meters from the highway, and washing hands using unclean water.  The presence of E. coli O157:H7 will be a threat for consumer health.

Abstract

Penyakit Parkinson merupakan penyakit neurodegeneratif dengan gangguan motorik seperti bradikinesia, rigiditas, dan tremor yang dapat mempengaruhi keseimbangan tubuh.  Hal ini terjadi karena penurunan kadar dopamin yang dapat dipicu oleh adanya stres oksidatif. Kunyit (Curcuma longa) mengandung kurkumin yang mempunyai aktivitas antioksidatif dan neuroprotektif yang potensial sehingga dapat mencegah gangguan neurodegeneratif karena stres oksidatif. Tujuan penelitian ini adalah untuk mengetahui aktivitas ekstrak rimpang kunyit pada mencit model Parkinson yang diinduksi haloperidol. Ekstraksi rimpang kunyit dilakukan secara maserasi dengan pelarut etanol 96%. Sebanyak 60 ekor mencit dibagi menjadi 6 kelompok, masing-masing mendapat perlakukan sebagai berikut : Kelompok normal diberi CMC-Na per oral (p.o) dan NaCl fisiologis intraperitoneal (i.p), kelompok haloperidol diberi CMC -Na (p.o) dan larutan haloperidol i.p (1 mg/kg BB), kelompok levodopa diberi levodopa (p.o) dan larutan haloperidol i.p, Kelompok ekstrak 100 diberi ekstrak kunyit dosis 100 mg/kg BB dan larutan haloperidol i.p , Kelompok ekstrak 200 diberi ekstrak kunyit dosis 200 mg/kg BB dan larutan haloperidol i.p, Kelompok ekstrak 400 diberi ekstrak kunyit dosis 400 mg/kg BB dan larutan haloperidol i.p. Pemberian CMC-Na, levodopa dan ekstrak kunyit diberikan selama 21 hari. Lima belas menit setelah pemberian perlakuan hari terakhir, semua hewan uji kecuali kelompok 1 diinjeksi haloperidol dosis 1 mg/kg BB secara i.p. Kemudian dilakukan uji batang, uji roda berputar, uji refleks geotaksis, uji refleks menghindari jurang dan uji kemampuan penciuman pada menit ke 5, 60, 120 dan 180. Data yang diperoleh dianalisis menggunakan uji anova satu jalur dilanjutkan dengan uji post hoc LSD. Hasil analisis data menunjukkan bahwa pemberian haloperidol dapat meningkatkan waktu katalepsi, meningkatkan jumlah jatuh dan mempercepat waktu jatuh pertama kali, menurunkan refleks geotaksis, meningkatkan waktu refleks menghindari jurang serta menurunkan kemampuan indera penciuman. Sedangkan pemberian ekstrak kunyit dosis 200 dan 400 mg/kgBB dapat menurunkan waktu katalepsi, menurunkan jumlah jatuh dan memperlama waktu jatuh pertama kali, menurunkan refleks geotaksis, menurunkan lama waktu pada uji refleks menghindari jurang serta meningkatkan kemampuan indera penciuman jika dibandingkan dengan Haloperidol (P<0.05). Pemberian ekstrak 200 dan 400 mg/kg tidak berbeda bermakna (P>0,05) dengan pemberian levodopa. Berdasarkan hasil dapat disimpulkan bahwa ekstrak rimpang kunyit dosis 200 dan 400 mg/kgBB memiliki aktivitas untuk mencegah terjadinya Parkinson pada mencit yang diberi haloperidol.
