Abstract

This study aimed to investigate the effectivity of goat milk fermentation as tyrosinase inhibitory with Lactobacillus rhamnosus TW 2. The examination of fresh milk contained the density, protein, and fat content. The culture of Lactobacillus rhamnosus TW 2 acid lactid bacteria starter was added with the concentration of 3, 4, and 5%, inoculated in pasteurized goat milk, then incubated at 37° C for 24 hours. Fermented milk were extracted by centrifugation, then supernatant was collected and was used for inhibition of tyrosinase enzymes activity on L-tyrosin and L-dopa substrate. The result showed that the density, fat content, and protein content of Etawah crossbred goat milk are 1.028, 3.73, and 5.45%, respectively. Re-identification of lactic acid bacteria showed similar morphology, physiology, and bio-chemistry with the isolated lactic acid bacteria. The growth curve of TW 2 was observed in 12 hours. The 5 % of Lactobacillus rhamnosus TW 2 was the best concentration to inhibit tyrosinase activity in L-Tyrosin substrate. In conclusion, fermentation of goat milk using Lactobacillus rhamnosus TW 2 at concentration of 5% as starter is effective to inhibit tyrosinase activity significantly.Key words: goat milk, fermentation, tyrosinase inhibitory

Abstract

The purpose of this study was to examine the molecular basis of Tursiops sp. originated from Java sea based on gene sequences ofCytochrome C Oxidase sub-unit II. Samples of blood were collected from five male bottlenose dolphins from PT. Wersut Seguni Indonesiacaptivity. Total deoxyribonucleic acid (DNA) was isolated from blood samples, then amplified by polymerase chain reaction (PCR), sequenced,and the data were analyzed using MEGA version 5.1 program. The amplification result of the PCR product obtained 824 base pairs (bp), andDNA sequencing obtained 684 nucleotides constituent of COX II genes that encode 228 amino acids making up the COX II. Nucleotide andamino acid sequence of COX II bottlenose dolphins originated from Java sea has a very high homology to Tursiops aduncus that are 99.13%and 100% respectively. Phylogram by neighbour joining method based on the nucleotide sequence of COX II gene indicating bottlenose dolphinsfrom Java sea belongs to group of Tursiops aduncus.Key words: bottlenose dolphin, COX II gene, nucleotide, Jawa Sea

Abstract

This study was aimed to find out the effect of ethanolic extract of ant plant (Myrmecodia sp.) to increase the activity of enzyme lipoprotein lipase (LPL) serum and to observe the histopathological changes of hypercholesterolemic rat liver. This study used 20 male rats grouped into 4 treatment groups, namely negative control group (K1), hypercholesterolemic group (K2), and hypercholesterolemic group that administered with ethanolic extract of ant plant 100 (K3) and 200 mg/kg bw (K4). The LPL enzyme activity were measured by the titration method and histopatological changes of liver were observed by calculated fatty degeneration and fatty infiltration. The data were analyzed using one way anova followed by Duncan test. The average of LPL enzyme activity on group K1, K2, K3, and K4 were 0.80±0.06, 0.45±0.10, 0.83±0.11, and 0.76±0.03 unit, respectively. The average number of fatty degeneration on hepatocyte and fatty infiltration were 1.80±0.83, 3.60±1.14, 23.00 ±1.22, and 40.20±1.30; and 9.20±0.84, 16.40±1.14, 2.60±0.54, and 4.80±0.83, respectively. The results showed that theraphy ethanolic extract of ant plant effects significantly (P0.01) on the increase of enzyme LPL and improve liver damage in hypercholesterolemic male rats. To conclude the administration of ethanolic extract of ant plant increases the LPL enzyme activity and improves liver damage on hyperch olesterolemic rats.Key words: Myrmecodia sp., LPL activity, histopathological liver, hypercholesterolemia

Abstract

This study was conducted to validate a commercial testosterone enzyme-linked immunosorbent assay (ELISA) kits (DRG EIA-1559) inanalytic and biological manner for measuring serum testosterone concentrations in kacang goats. This study used 18 healthy kacang goats, six bucks (2 years), six kids (6 months), and six does (2 years). Blood samples were collected from jugular vein and prepared as serum. Two validation tests were performed, an analytical validation comprises a parallelism, accuracy, precision and sensitivity and a biological validation by comparing testosterone concentration from bucks, kids, and does. Testosterone concentrations were measured using ELISA technique. Data of analytical validation were analyzed descriptively and test of equality of slope was performed to see the parallelism between samples and standard curves. Analysis of variance (ANOVA) was used for biological validation data. Results of parallelism showed that sample curve was parallel to the standard curve. Accuracy, precision (% CV of intra-and inter-assay) and sensitivity of the assay were 99.65±4.27%, 10%, 15% and 0.083 ng/ml, respectively. Results of biological validation showed that the assay used were accurately measured testosterone which testosterone concentrations in bucks were significantly higher compared to kids and does (P0.05). In conclusion, a commercial testosterone ELISA kits (DRG EIA-1559) is a reliable assay for measuring serum testosterone concentration in kacang goats. Abstract

 The research was aimed to evaluate feeding effect of powder and extract of dahlia tuber as a sources of inulin prebiotic on protein digestibility and productivity of crossbred local chicken. A total of 280 unsex local crossbred chicken with the age of 22 days and average weight of 180.46±1.21 g was assigned in a completely randomized design with 7 treatments and 4 replications (10 birds each). Dietary treatments consist of T0 (basal ration), T1 (T0 + 0.4% dahlia powder), T2 (T0 + 0.8% dahlia powder), T3 (T0 + 1.2% dahlia powder), T4 (T0 + 0.39% dahlia extract), T5 (T0 + 0.78% dahlia extract), T6 (T0 + 1.17% extract dahlia). Treatment was conducted for 8 consecutive weeks. Variables observed were protein digestibility, short chain fatty acid (SCFA), muscle protein mass, muscle calsium mass, and body weight. The data were analyzed using ANOVA and followed by Duncan test at the level of 5%. The results showed that feeding inulin in the form of powder or extract of dahlia tuber significantly (P0.05) increased protein digestibility, SCFA level, and body weight. Treatments T2 until T6 significantly (P0.05) increased muscle calsium mass but did not increase muscle protein mass. In conclusion, the used of 1.2% dahlia powder and 1.17% extract dahlia tuber as an inulin source increase protein digestibility and productivity of crossbred local chicken.Key words: inulin, dahlia tuber, crossbred local chicken

Abstract

The aims of the research were to evaluate resistance of Staphylococcus aureus against several antibiotics and to detect the gene encoding ofmethicillin resistant Staphylococcus aureus (MRSA) in broiler. Twenty three Staphylococcus aureus isolated from infected broiler in Yogyakartawere used. Genotypic identifications were done based on the occurrence of gene nuc and 23SrRNA. The resistance assay to several antibioticsrevealed that Staphylococcus aureus were resistant to penicillin 78% (18/23), doxycyclin 56% (15/23), gentamicin 26% (6/23), tetracyclin 22% (5/23), erythromycin 13% (3/23), and methicillin 9% (2/23). Detection of gene encoding MRSA on 23 isolats of S. aureus showed that 8 (34.8%) isolates contain gene mecA.Key words: Staphylococcus aureus, broiler, resistant, methicillin, mecA

Abstract

The purpose of this research was to identify avian influenza (AI) virus using serological and molecular methods on poultry which suspected as AI infected in Aceh province. This study used 37 samples of tracheal and cloacal swabs and organs from various species of poultry that were collected from several districts/cities in Aceh. Samples were collected and put into transport media and stored at 4° C before sending to the laboratory. Samples were inoculated in specific pathogen-free of embryonated chicken egg with the age of 9-11 days for further serological and molecular examination. From 37 samples which infected to embryonated chicken egg then followed by hemagglutinin agglutination test/hemagglutinin inhibition revealed that 7 samples were positively infected with AI virus. The amplification result of specific matrix gene primer was followed by electrophoresis on 2% agarose gel which were obtained in the form of a deoxyribonucleic acid (DNA) band at 276 bp for matrix gene and 1.725 bp for H5 gene for all isolates test. In conclusion, the virus which caused the death of various types of poultry in Aceh province is avian influenza A virus subtype H5.Key words: avian influenza virus, H5N1, serologic, matrix, heamaglutinin

Abstract

This study was purposed to inquire the activities of benzopyran (chromones) and benzofuranones (coumaranones) derivatives against Steinernema feltiae (S. feltiae). The toxicity assay against S. feltiae showed that benzopyran derivatives 2, 3, 4, 6, and 9 have the highest activity on S. feltiae with viabilities percentage of 50%. The compound 9 demostrated the highest activity with LD 50 and LD values, 7.2 and 52.2 μM, respectively. The activities of compound 7 and 10 showed the lowest toxicity. Interestingly, the activity of benzofuranone derivatives showed significant activities against S. feltiae. Compare to benzopyran derivatives, the benzofuranone derivatives has the highest toxicity, in particular compound 13 with LD 5.45 μM. The nematicidal assay showed that benzofuranones (coumaranones) derivatives revealed higher activities than benzopyran (chromones) derivatives.Key words: chromones, benzopyran, coumaranones, benzofuranone, and Steinernema feltiae

Abstract

This research aimed to determine the optimum time for early pregnancy diagnosis in dwarf goat by trancutaneous ultrasonography and to determine ultrasonography imaging of dwarf goat pregnancy since insemination until embryo formation. Animal used in this research were 3 productive female goats with normal estrus cycle and 1male goat. The female goats were kept in separation pen from the male goat. The grasses were fed three times a day and additional feed were fed in the morning and afternoon with the water was given ad libitum. The goats were synchronized with intramuscular double injection of 1 ml estron®/each female goat with 11 days interval. The observation of estrus was examined using male goat following the second injection of estron®. The sign of estrus in female goats were characterized with standing immobile and allow the male goat to mount them. If the signs had been evident, the female goats were naturally mated. Pregnancy was determined by the presence of the embryonic vesicle by isoechogenic until hyperechogenic visualization surrounded by hypoechogenic of the embryonic fluid. The result of the transcutaneous ultrasonography in dwarf goat pregnancy showed the presence of embryonic vesicle on day 14 and embryo with isoechogenic visualization was detected on day 24 of pregnancy.Key words: dwarf goat, early pregnancy, transcutaneous ultrasonography

Abstract

This study was conducted to assess the effectiveness of a diet contained fresh anchovies and calcitriol supplementation for 6 weeks to prevent osteoporosis in ovariectomized rats. Fifteen Wistar rats aged 8 weeks were divided randomly into 3 groups (normal/K, ovariectomized/Ov, and ovariectomized + calcitriol/OVD), 5 mice each. Group K and Ov rats were fed with fresh anchovies, while the OVD group was fed with fresh anchovies + calcitriol. At the age of 15 weeks, all mice were done for euthanasia, then left femur was collected for immunohistochemistry examination of tartrate resistant acid phosphatase5b (TRAP5b). The detection of (TRAP5b) was conducted using a monoclonal ant ibody anti TRAP5b, and detected using a streptavidin-biotin. The results showed that the metaphysis part of distal femur bone of mice group K, Ov, and OVD were positive TRAP5b stained with brown color on trabecular bone in bone marrow cavity and trabecular spiculum surface, but rats in group K and Ov had extensive bone marrow cavity and normal trabecular spiculum, whereas OVD group showed bone marrow cavity dilation, accumulation of adipocytes in the bone marrow cavity, and shorter the spiculum of trabeculae. It can be concluded that fresh anchovies diet and calcitriol supplementation for 6 weeks are not effective in preventing osteoporosis in ovariectomized rats.Key words: calcitriol, femur, ovariohisterectomy
