Abstract

Generally, Bacteria in the intestine consist of beneficial bacteria and bacteria that have the potential to disrupt animal health such as pathogenic bacteria. The presence of pathogenic bacteria is one of the factors that cause gastrointestinal dysfunction, but can be suppressed by maintaining the presence of beneficial bacteria for the digestive tract, especially the type of Lactic Acid Bacteria (LAB) (Astuti, 2016). LAB has antagonistic activity because it is able to inhibit pathogenic bacteria so it can compete to maintain normal flora balance in the digestive tract.  The main inhibitory activity of LAB is caused by the accumulation of primary metabolites such as lactic acid, acetic acid, ethanol, and carbon dioxide. In addition, LAB is also capable of producing antibacterial compounds such as bacteriocins (Furtado et al., 2014). Naturally LAB has existed in the digestive tract of chickens, but it is easy to experience changes in the amount due to the influence of given the feed (Widodo et al., 2015). Feed and the environment can affect the composition of microbes in the digestive tract in chickens(Ghadban, 2002; Apajalahti et al., 2004). Provision of fermented feed can improve the composition of intestinal microflora and increase the number of LAB. Fermented feed is generally easily biodegradable and has a higher nutritional value than the original ingredient and it can reduce the pH of the digestive tract.AKBISprob is an alternative product to increase poultry production in the form of supplement made from a mixture of soybean wate, and palm kernel meal which is fermented with Aspergillus niger.  The fermentation process using Aspergillus niger can reduce the crude fiber contained in AKBIS, because the mold can produce amylase, pectinase, amyloglucosidase and cellulase enzymes which can degrade cellulose so that it is easier to digest. In addition, Aspergillus niger also produces metabolites in the form of citric acid which is a component that can reduce intestinal pH so that it is suitable for LAB growth. Based on research conducted by Nurliana et al. (2016; 2017), supplementation of 4% AKBISprob can maintain the chickens production and health as well increase the number of LAB and reduce the number of pathogenic bacteria such as Escherichia coli and Salmonella sp. in chickens intestine. Thus creating a balance of microflora in the digestive tract of laying hens. Based on the above reasons, it is necessary to conduct research on antibacterial detection of BAL in laying hens that have been given AKBISprob.

Abstract

Aceh cattle are one of the many cow’s species that are widely preserved and distributed in Aceh Province. This species is the result of a cross between Bos sundaicus and Bos indicus and has been designated as a native Indonesian class of cattle in 2011 by the Minister of Agriculture of Indonesia through the decision of the Minister of Agriculture number 2907 / Kpts / OT.140 / 6/2011 [1]. Aceh's cattle population in 2017 in total was 627,629 species [2]. Slaughtering the cows results in the obtaining of byproduct or waste such as ovaries. The ovarium of female cows is one of the complement results of animal slaughter house which is infrequently used. In fact, the ovaries comprise follicles which contain many active compounds inside the follicular fluids such as insulin-like growth factor I (IGF-I), follicle stimulating hormone (FSH), luteinizing hormone (LH), estrogen and progesterone [3]. Freeze drying/lyophilization is one of the preservative process which are generally used in food and non-food processing. This study was conducted to determine the effect of these methods on the concentration of estrogen in follicular fluid.

Abstract

Monitoring of physiological stress in wild and/or endangered animals living in their natural habitat can be generated via measuring faecal glucocorticoid metabolites (FGCMs) through enzyme immunoassay technique. However, a careful validation of each enzyme immunoassays (EIAs) and methodological issues such as samples preservation, and extraction procedures must be a significant concern and validated because  it may influence FGCMs levels. In this present study, the aim was to carry out a biological validation to find reliable EIAs that could be used for non-invasive measurement of glucocorticoid levels in crested macaques. Moreover, we compared three different techniques of preservation: lyophilisation, oven drying and field extraction and three different solvents for extraction: 80% methanol, 90% pure ethanol and 90% commercial alcohol (isopropyl alcohol) to find the simple and practical techniques for sample processing. For hormone analysis, we colIected 189 faecal samples from wild crested macaques living in the Tangkoko-Batuangus Nature Reserve, North Sulawesi, Indonesia. The results show that biological validation can be used to validate measurement of faecal glucocorticoid which 3α,11β-dihydroxy-etiocholanolone was the most reliable EIAsfor this species. Different preservation and solvent extraction were not significantly influenced levels of FGCMs (p0.05). In conclusion, Our study highlights the importance of biological validation of FGCMassays and presents practical techniques for the non-invasive monitoring of physiological stress in crested macaques.

Abstract

Penelitian ini bertujuan untuk mengukur konsentrasi hormon IGF-1 pada cairan folikel ovarium sapi aceh. Cairan folikel diaspirasi dari 30 pasang ovarium yang dikoleksi dari 30 ekor sapi aceh di Rumah Potong Hewan (RPH) Banda Aceh. Cairan ovarium dikoleksi berdasarkan ukuran folikel dan diidentifikasi ada tidaknya korpus luteum (CL). Ukuran folikel dikategorikan menjadi tiga (3) kategori yaitu folikel kecil (Ø1˗˗5 mm), folikel sedang (Ø5˗˗8,5 mm), dan folikel besar (Ø≥8,5 mm).  Pengukuran konsentrasi hormon IGF-1 dilakukan dengan metode Enzyme Linked Immunosorbent Assay (ELISA). Data dianalisis menggunakan uji Kruskal-Wallis. Hasil penelitian menunjukkan bahwa sebanyak 12 ovarium (40%) memiliki CL dan 18 ovarium (60%) tidak memiliki CL. Konsentrasi hormon IGF-1 pada ovarium yang memiliki CL dengan kategori folikel kecil dan sedang masing-masing 17,39±7,24 ng/ml dan 0,1 ng/ml. Konsentrasi hormon IGF-1 pada ovarium tanpa CL dengan kategori folikel kecil, sedang, dan besar masing-masing 8,81±2,73 ng/ml, 2,09±0,27 ng/ml, 1,38±0,93 ng/ml. Rataan IGF-1 pada folikel kecil terlihat lebih tinggi baik pada ovarium yang memiliki CL maupun pada ovarium yang tidak memiliki CL, namun secara statistik tidak berbeda (p0,05). Dapat disimpulkan bahwa konsentrasi hormon IGF-1 pada semua kategori folikel tidak berbeda baik pada ovarium yang memiliki CL maupun ovarium yang tidak memiliki CL. (This study was conducted to measure concentration of IGF- hormone in ovary follicular fluid of aceh cow. Follicular fluid was aspired from 30 ovarian pairs collected from 30 aceh cows in Banda Aceh animal slaughterhouse. Ovarian fluids are collected based on the follicle size and identified by the presence of the corpus luteum (CL). The size of the follicle are categorized in three categories: small (Ø1˗˗5 mm), medium (Ø5˗˗8.5 mm), and large (Ø≥8.5 mm) follicles. Measurement of IGF-1 concentration using an Enzyme Linked Immunosorbent Assay (ELISA) method. Data were analyzed using a Kruskal-Wallis test. The results showed that 12 ovaries (40%) have CL and 18 ovaries (60%) have not CL. The concentrations of IGF-1 in the ovaries that have CL in small and medium follicles  were 17.39±7.24 ng/ml and 0.1ng/ml, respectively. The concentrations of IGF-1 in the ovaries without CL in small, medium, and large follicles were 8.81±2.73 ng/ml, 2.09±0.27 ng/ml, 1.38±0.93 ng/ml, respectively. The mean of IGF-1 concentration in small follicles was higher in both ovaries with CL or without CL, but it was not significantly different  (p0.05). It can be concluded that the concentration of IGF-1 in all categories of follicles did not differ either in ovaries with  CL and without CL.)

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengetahui persentase penyimpangan  bobot badan sapi aceh jantan yang diukur menggunakan rumus Lambourne dibandingkan dengan bobot aktual. Sampel yang digunakan yaitu 50 ekor sapi aceh jantan berumur 2-2,5 tahun di BPTU-HPT Indrapuri. Parameter yang diamati yaitu Panjang Badan (PB), Lingkar Dada (LD), dan Bobot Badan (BB). Pengukuran PB dilakukan dengan cara mengukur jarak antara ujung samping tulang bahu (tuberculum humeralis lateralis) sampai dengan ujung tulang duduk (tuberculum ischiadium) menggunakan tongkat ukur. Pengukuran LD dilakukan dengan melingkari rongga dada di belakang sendi tulang bahu (os scapula) menggunakan pita ukur. Pengukuran BB dilakukan dengan dua cara yaitu menggunakan timbangan ternak digital (BB) dan menggunakan rumus Lambourne. Besar persentase penyimpangan BB sapi aceh jantan menggunakan rumus Lambourne terhadap bobot badan aktual adalah 3,41% dan didapatkan persamaan regresi Y = -350,30 + 2,27x1 + 2,06x2.ABSTRACTThe aim of this study was to investigate the percentage of deviation of body weight estimation using a Lambourne formula had compared with real body weight in aceh bull. Samples of this study were 50 aceh bulls 2-2.5 years old in BPTU-HPT Indrapuri. Observed parameters were body length, chest circumference, and body weight. Body length was measured from the side of the shoulder bone (tuberculum humeralis lateralis) until sitting bone (tuberculum ischiadium) using measuring stick. Chest circumference was measured from the chest cavity behind the shoulder bone joint (os scapula) using a measurement tape. Measurement of body weight was carried out in two ways, first it was known from digital cattle scales (real body weight) and second it was calculated using a Lambourne formula. The percentage of deviation of body weight estimation using Lambourne formula had compared with aceh bull’s real body weight was 3.41% and the regression formula was Y = -350.30 + 2.27x1 + 2.06x2.

Abstract

Penelitian ini bertujuan untuk mengkaji perilaku harian burung rhea (Rhea americana) di penangkaran Taman Safari Gurun Putih Lestari Jantho, Aceh Besar. Pengamatan perilaku harian dilakukan terhadap 12 individu burung rhea (6 jantan dewasa dan 6 betina dewasa) menggunakan metode scan animal sampling dengan interval pengamatan setiap 10 menit. Data perilaku harian dikelompokkan berdasarkan enam rentang waktu pengamatan, kemudian dianalisis menggunakan Friedman test dan dilanjutkan dengan Mann-Whitney U test untuk mengetahui perilaku harian signifikan per jenis kelamin. Hasil analisis menunjukkan bahwa persentase penggunaan waktu (time budget) melakukan aktivitas harian burung rhea berdasarkan waktu pengamatan menunjukkan perbedaan yang signifikan (p <0,05) pada perilaku minum dan istirahat, sedangkan perilaku lainnya tidak berbeda nyata (p >0,05). Berdasarkan jenis kelamin, perilaku makan dan minum betina secara signifikan (p <0,05) lebih tinggi dibandingkan jantan, sedangkan perilaku termoregulasi, agresi, dan courtship jantan secara signifikan (p <0,05) lebih tinggi dibandingkan betina. Kesimpulan,penelitian ini menunjukkan terdapat perbedaan variasi perilaku harian burung rhea pada penangkaran berdasarkan waktu pengamatan (perilaku makan dan istirahat), serta perbedaan variasi perilaku harian berdasarkan jenis kelamin (makan, minum, termoregulasi, agresi, dan courtship). Dalam penelitian ini juga teramati perilaku memakan feses (coprophagy) yang belum pernah dilaporkan sebelumnya untuk jenis ini.

Abstract

Interferon-tau (IFN-τ) is a pregnancy signal produced by embryonic trophoblast cells at the time of implantation in a mammal's endometrial wall, which is useful for conveying a message that the mammal can accept the presence of the foreign object (embryo). This study aims to determine the accuracy of early pregnancy diagnosis in Aceh cows by measuring the concentration of IFN-τ. This study involved eight cows aged 3-5 years, weighing 150-250 kg, clinically healthy, and having normal reproduction (have had calves and at least two regular cycles). All cows were estrous-synchronized twice using PGF2ɑ at a dose of 5 ml with 11 days intervals before artificial insemination (AI). Serum collection was performed on days 14, 15, 16, 17, 18 after AI. Based on ultrasound examination, out of eight cows, four (50%) cows were diagnosed as pregnant, and four (50%) cows were diagnosed as not pregnant. Mean (± SD) IFN-τ of pregnant vs. non-pregnant cows on day 14, 15, 16, 17, and 18 were 14.96±8.65 pg/ml vs. 6.14±5.54 pg/mL; 16.74±5.28 pg/mL vs. 4.44±3.51 pg/mL; 14.33±5.9 pg/mL vs. 5.78±5.20 pg/mL; 13.87±5.42 pg/mL vs. 4.38±3.76 pg/mL; and 13.93±6, 16 pg/mL vs. 3.24±2.63 pg/mL, respectively. The lowest IFN-τ concentration in cows that were successfully pregnant was 7.88±1.84 pg/mL. The best timing of IFN-τ for pregnancy diagnosis was on day 15 after AI, with an accuracy of 87.5%, specificity of 100%, and sensitivity of 80%.

Abstract

This study aimed to compare the progesterone concentrations in Waringin sheep estrus cycle which induced by PGF2α or the combination of PGF2α and GnRH. This study used six Waringin sheep those were divided into twice groups, K1 and K2. K1(n=3) were injected by 7.5 mg PGF2α intramuscularly for 10 days, while K2 (n = 3) were injected by PGF2α and GnRH. On the first day K2 were injected by 7.5 mg PGF2α, then followed by injection 50 µg of GnRH on day 8th and re-injected by 7.5 mg PGF2α on day 15th. On day 18th, sample was re-injected using 50 µg GnRH. Waringin sheep those showed estrus symptoms were detected visually and with signs showed by sheep stud. Blood samples were taken on the 7th, 14th, and 21st day after the peak heat. The measurement of progesterone concentration was conducted by enzyme linked immunosorbent assay (ELISA). The results showed those the concentrations of progesterone on day 7th, 14th, and 21st on K1 vs K2 were 1.324±1.079, 7.607±8.922, and 5.220±1.653  vs 4.705±3.369, 4.184±5.512, and 1.797±0.898 ng/mL (P>0,05), respectively. In conclusion, the concentration of progesterone of Waringin sheep after inducing with PGF2α or combination PGF2α and GnRH at different cycle periode did not show differences. Keywords: Estrus Cycle, GnRH, PGF2α, Progesterone, Waringin Sheep
