Abstract

Research on substances as new antibiotics to inhibit or kill bacteria that are resistant to antibiotics needs to be done. One of the simplicia has an antibacterial effect is the leaves of green tea (Camellia sinensis). Antibacterial content the leaves of green tea (Camellia sinensis), namely alkaloids, saponins, tannins, and catechins (polyphenols). Tea one of the favorite products from Indonesia’ estate. The purpose of this study was to determine optimization of the inhibitory power the leaves of green tea’s (Camellia sinensis) extract against the growth of Escherichia coli and Staphylococcus aureus. This research is an experimental laboratory conducted at Bacteriology”s Laboratory, Department of Medical Laboratory Technology Surabaya in November 2020 - April 2021. This study uses the Kirby Bauer disk diffusion method, namely Mueller Hinton Agar (MHA) media which has been planted with Escherichia coli and Staphylococcus aureus with disks that have been prepared. soaked in the leaves of green tea’s (Camellia sinensis) extract at concentrations of 25%, 50%, 75%, and 100% and then incubated for 24 hours in an incubator. The results can be concluded the leaves of green tea’s (Camellia sinensis) extract can be used as an antibacterial. Optimal of results in this research were Escherichia coli at a concentration of 25% with an average diameter of 10,25 mm and Staphylococcus aureus at a concentration of 100% with an average of 22,5 mm. This is because content the leaves of green tea’s (Camellia sinensis) is catechins and alkaloids as strong antibacterials

Abstract

Soil worm infection transmitted Helminths (STH) is a group of parasitic nematodes that cause infection in the human gut which is transmitted through contaminated soil eggs or larvae. STH often infects people with poor environmental sanitation and lack of public awareness of the importance of maintaining personal and environmental hygiene. An increase in the value of eosinophils and a decrease in the value of blood hemoglobin are markers of infection by the STH parasite. The purpose of this study was to determine the relationship between the incidence of soil-transmitted helminths (STH) infection with the value of eosinophils and blood hemoglobin in the 1001 night village community, Surabaya. This type of research is an analytical observational study and the samples in this study were blood and feces taken from the people of 1001 Malam Surabaya village as many as 25 people with a random sampling technique using the native method for stool examination and using a hematology analyzer for blood examination. Based on the results of research conducted on the 1001 night village community, it was known that the prevalence of worm infection was Ascaris lumbricoides. The prevalence of STH infection with eosinophilia was 15 samples. A significant relationship after being tested statistically with the Spearman Rank Correlation test was found between STH infection and the eosinophil value of 0.034. Then on the relationship between hemoglobin values and STH infection, a significant value was obtained, namely 0.00. There is a significant relationship between STH infection with the value of eosinophils and blood hemoglobin.

Abstract

Acute Myocardial Infarction (AMI) is a heart disease condition due to heart muscle cells not getting enough blood flow and oxygen due to atherosclerosis of the coronary arteries. When the condition of atherosclerosis gets worse, it will cause an inflammatory reaction, causing an increase in neutrophils. Inflammation due to atherosclerosis will worsen blood flow to the heart, resulting in heart muscle necrosis which causes an increase in biochemical markers, namely cardiac troponin. The aim of this research is to determine the relationship between the number of neutrophil cells and troponin I levels in patients with Acute Myocardial Infarction.This type of research is a cross-sectional analytic observational with a retrospective approach. This research was conducted on patients with Acute Myocardial Infarction at the RSPAL dr. Ramelan Surabaya who performed a complete blood count and troponin I level with a period of January 1st 2020 – May 3rd 2021, taken by purposive sampling. The results showed that male respondents were 29 patients (63%) while female respondents were 17 patients (37%). Respondents aged 65 years amounted to 28 (61%) people, while respondents aged 65 years were 18 (39%) people. The results of the hypothesis test have a sig value of 0.000 < (0.05), so there is a relationship between the number of Neutrophil cells and Troponin I level in patients with Acute Myocardial Infarction. In hypothesis testing, the correlation coefficient value is 0.594, which means that there is a moderate relationship with a positive direction.

Abstract

The role of Laboratory examination as a diagnostic of disease is very important, including in determining the amount of uric acid contained in the body. The POCT (Point Of Care Testing) method and the Enzymatic Colorimetric method using a Photometer are several ways that can be done to determine the amount of uric acid content. This study aims to determine the differences in the results of uric acid examination in the elderly using the POCT method with the Enzymatic Colorimetric method at the Bangunsari Health Center, Madiun Regency.This type of research is an analytic observational study with a cross sectional approach. The research population is the elderly who seek treatment at the Bangunsari Health Center, Madiun Regency. The study was carried out in January-May 2021. The research sample consisted of 30 people who were taken by purposive sampling. Research data in the form of primary data obtained directly from the results of laboratory examinations. The data analysis technique used the normality test with the Kolmogorov Smirnov Test and the Independent-Sample T Test.The results showed that the data on uric acid levels in the elderly at the Bangunsari Public Health Center, Dolopo District, Madiun Regency, which was measured using the POCT (Point Of Care Testing) method showed that the mean and standard deviation values ​​were 5,650±1,283 while those measured using the Enzymatic Colorimetric method were 6,340±1,332 indicating that there was an average difference The average results of measuring uric acid levels in the elderly are between those using the POCT (Point Of Care Testing) method and those using the Enzymatic Colorimetric method. Based on the results of the Independent Sample T-test, it is known that the value of Sig. (2-tailed) of 0.046 <0.05, it can be concluded that H0 is rejected and Hi is accepted. That is, there is a significant difference in uric acid levels using the POCT method with the Enzymatic Colorimetric method.

Abstract

The consumption rate of Monosodium Glutamate (MSG) in Indonesia has increased every year. Uncontrolled use of MSG in Indonesia for a long period of time can cause toxic effects on the body. The free glutamate content produced by MSG can affect the work of the immune system, especially in the innate immune system and cause oxidative stress. To determine the effect of exposure to Monosodium Glutamate (MSG) on the viability of monocyte cells. This study is a laboratory experimental in vitro with a post test only control group design. A total of 10cc of peripheral venous blood was isolated using the ficoll gradient centrifugation method. The results of monocyte cell isolates were exposed to Monosodium Glutamate (MSG) according to groups. Group I: negative control, group II: monocyte cells + MSG 3%, group III: monocyte cells + MSG 6%, group IV: monocyte cells + MSG 9%. Subsequently incubated for 24 hours at 37 °C in 5% CO2. Then the viability test was carried out using trypan blue staining. Monocyte cell viability calculations were carried out under an inverted microscope with a magnification of 400x per 100 cells. The data obtained were analyzed statistically using the one-way Anova test followed by the LSD test. The average viability in each group was obtained as follows, monocyte cell viability in the control group was 63%, group II was 47%, group III was 45% and group IV was 35%. There is an effect of exposure to Monosodium Glutamate (MSG) on the viability of monocyte cells with the most significant effect being the 9% MSG concentration with an average viability of 35%.

Abstract

Cadmium is one of the chemicals contained in cigarettes, when consumed by humans in the long term and with sufficient intensity often can damage human organs such as the liver. This study aims to determine the exposure of cadmium to the levels of SGOT and SGPT in active smokers and passive smokers in a coffee shop in East Surabaya. This study used a correlational type of research, with a cross-sectional research design. There were 30 respondents who were divided into 2 groups, namely 15 active smokers and 15 passive smokers in a coffee shop in East Surabaya. The examination of cadmium used an Atomic Absorption Spectrophotomter (AAS), while the examination of SGOT and SGPT used the IFCC method (kinetic) with a photometer. The data were analyzed by Kolmogorov-Sminorv test and r-Pearson test. The results showed that there was no effect of high cadmium exposure in active smokers and passive smokers on SGOT and SGPT levels. The average cadmium in active smokers and passive smokers were 2.93 g/L and 1.30 g/L. while the levels of SGOT and SGPT in active smokers were 27.26 U/L and 36.86 U/L. The average levels of SGOT and SGPT in passive smokers were 19.73 U/L and 17.86, respectively. In these results it can be seen that the increase in cadmium levels does not affect liver function cells.

Abstract

Lead (Pb) is a heavy metal that can cause poisoning and accumulate in the human body. Determination of lead levels by using AAS the condition is that the sample must be a solution, then it is necessary to destruct first. There are two types of destruction that can be done, namely wet destruction using acid reagents to decompose samples and dry destruction by heating or destruction using very high temperatures. HNO3 oxidizing agent and HNO3 mixture with HClO4 are the main oxidizing agents that can provide the best destructive results compared to other oxidizing agents. This study aims to determine the best oxidizing and destructive substances present in the hair using Atomic Absorption Spectrophotometry (AAS). The research was conducted at surabaya regional health laboratory in April - May 2021. This research is comparative research. The sample used is the hair of gelam gas station operator officer, Candi. There are two ways of preparing samples that can be done in pb analysis on hair, namely proses wet and dry destruction. The result of destruction is determined by the level of lead using Atomic Absorption Spectrophotometry (AAS). The results of the analysis pno dry destructionHNO3 as a whole has rentang lead levels 0.1281-0.1958 mg / L. Dry destruction HNO3 : HClO4 secara overall has rentang lead levels 0.1457-0.2159 mg /L. HNO3 closed wet construction has lead content range 0.1431-0.2034 mg/L and wet destructive closed HNO3 : HClO4 has lead content range 0.1080-0.5371 mg/L. Based on these results, it can be known that all hair samples tested in this study contain lead levels that are still within normal limits or within the tolerance limit of < 12 μg/g.

Abstract

Improving health laboratory services is very much related to strengthening the quality of health laboratories. The control serum is part of the quality assurance of the health laboratory. Lyophilized serum is a type of homemade control serum. This research aims to determine the homemade lyophilized serum used as a control material for the parameters of Blood Urea Nitrogen (BUN) and creatinine which were analyzed for 2 months at a temperature of 2 - 8°C. This type of researvh is a quasi-experimental conducted at the Laboratory of Clinical Chemistry, Department of TLM Poltekkes, Ministry of Health, Surabaya and the pramita laboratory. Samples were obtained from the serum of students mahoring in TLM Poltekkes Kemenkes Surabay that has been qualified and were collected in one container for lyophilization. The average results of the BUN (Blood Urea Nitrogen) examination from week 1 to week 8 were 7.15 mg/dL, 7.15 mg/dL, 6.82 mg/dL, 7.12 mg/dL. dL, 7.12 mg/dL, 7.33 mg/dL, 7.15 mg/dL, 7.33 mg/dL, and 7.33 mg/dL. The average results of the creatinine examination from week 1 to week 8 were 0.69 mg/dL, 0.69 mg/dL, 0.70 mg/dL, 0.67 mg/dL, 0.72 mg/dL, 0.72 mg/dL, 0.68 mg/dL, 0.70 mg/dL. From the results of the examination, it can be concluded that the levels of BUN (Blood Urea Nitrogen) and creatinine in homemade serum lyophilized until the 8th week are still stable. Storage time for 8 weeks has no effect on BUN (Blood Urea Nitrogen) and creatinine levels in homemade lyophilized serum. Abstract

Pengguna smartphone setiap tahun semakin meningkat dengan berbagai manfaat dan kemudahan bagi penggunanya, khususnya bagi mahasiswa. Suhu hangat pada smartphone merupakan tempat yang cocok untuk bakteri dapat berkembang biak sehingga dapat menyebarkan penyakit infeksi dikarenakan seringnya kontak dengan tangan ditambah dengan orang yang memiliki higenitas pribadi yang buruk. Staphylococcus aureus adalah bakteri flora normal pada kulit, dapat bersifat sebagai pathogen oportunistik. Tujuan penelitian ini adalah untuk mengisolasi dan mengidentifikasi bakteri Staphylococcus aureus dari usap layar smartphone dan menganalisa kebersihan tangan. Jenis penelitian penelitian ini adalah deskriptif menggunakan teknik pengumpulan data primer dengan memperoleh data secara langsung dari 25 sampel smartphone. Populasi dalam penelitian ini adalah mahasiswa D4 Teknologi Laboratorium Medis poltekkes Kemenkees Surabaya. Spesimen penelitian ini adalah smartphone. Penelitian ini dilakukan di Laboratorium Bakteriologi Poltekkes Kemenkes Surabaya dan dilakukan pada bulan Oktober 2021 – Juni 2022. Dari 25 sampel smartphone (100%) diperoleh 11 sampel (44%) positif dan 14 sampel (56%) negatif tidak teridentifikasi bakteri Staphylococcus aureus. Diharapkan mahasiswa dan pembaca agar lebih meningkatkan personal hygiene dengan mencuci tangan setelah menggunakan smartphone.

Abstract

Demam tifoid disebabkan oleh bakteri Salmonella typhi yang menyerang saluran pencernaan. Berdasarkan data angket, sebagian besar kebiasaan buruk yang dilakukan oleh mahasiswa adalah tidak menjaga pola makan dan makan sembarangan yang belum tentu higienis yang dapat menyebabkan infeksi demam tifoid. Kadar leukosit pada penderita demam tifoid umumnya dalam kondisi normal dan di bawah batas normal. Akibat penyumbatan pembentukan darah di sumsum tulang oleh Salmonella typhi yang menandakan jumlah leukosit di bawah batas normal.
