Abstract

Research on substances as new antibiotics to inhibit or kill bacteria that are resistant to antibiotics needs to be done. One of the simplicia has an antibacterial effect is the leaves of green tea (Camellia sinensis). Antibacterial content the leaves of green tea (Camellia sinensis), namely alkaloids, saponins, tannins, and catechins (polyphenols). Tea one of the favorite products from Indonesia’ estate. The purpose of this study was to determine optimization of the inhibitory power the leaves of green tea’s (Camellia sinensis) extract against the growth of Escherichia coli and Staphylococcus aureus. This research is an experimental laboratory conducted at Bacteriology”s Laboratory, Department of Medical Laboratory Technology Surabaya in November 2020 - April 2021. This study uses the Kirby Bauer disk diffusion method, namely Mueller Hinton Agar (MHA) media which has been planted with Escherichia coli and Staphylococcus aureus with disks that have been prepared. soaked in the leaves of green tea’s (Camellia sinensis) extract at concentrations of 25%, 50%, 75%, and 100% and then incubated for 24 hours in an incubator. The results can be concluded the leaves of green tea’s (Camellia sinensis) extract can be used as an antibacterial. Optimal of results in this research were Escherichia coli at a concentration of 25% with an average diameter of 10,25 mm and Staphylococcus aureus at a concentration of 100% with an average of 22,5 mm. This is because content the leaves of green tea’s (Camellia sinensis) is catechins and alkaloids as strong antibacterials

Abstract

Soil worm infection transmitted Helminths (STH) is a group of parasitic nematodes that cause infection in the human gut which is transmitted through contaminated soil eggs or larvae. STH often infects people with poor environmental sanitation and lack of public awareness of the importance of maintaining personal and environmental hygiene. An increase in the value of eosinophils and a decrease in the value of blood hemoglobin are markers of infection by the STH parasite. The purpose of this study was to determine the relationship between the incidence of soil-transmitted helminths (STH) infection with the value of eosinophils and blood hemoglobin in the 1001 night village community, Surabaya. This type of research is an analytical observational study and the samples in this study were blood and feces taken from the people of 1001 Malam Surabaya village as many as 25 people with a random sampling technique using the native method for stool examination and using a hematology analyzer for blood examination. Based on the results of research conducted on the 1001 night village community, it was known that the prevalence of worm infection was Ascaris lumbricoides. The prevalence of STH infection with eosinophilia was 15 samples. A significant relationship after being tested statistically with the Spearman Rank Correlation test was found between STH infection and the eosinophil value of 0.034. Then on the relationship between hemoglobin values and STH infection, a significant value was obtained, namely 0.00. There is a significant relationship between STH infection with the value of eosinophils and blood hemoglobin.

Abstract

Acute Myocardial Infarction (AMI) is a heart disease condition due to heart muscle cells not getting enough blood flow and oxygen due to atherosclerosis of the coronary arteries. When the condition of atherosclerosis gets worse, it will cause an inflammatory reaction, causing an increase in neutrophils. Inflammation due to atherosclerosis will worsen blood flow to the heart, resulting in heart muscle necrosis which causes an increase in biochemical markers, namely cardiac troponin. The aim of this research is to determine the relationship between the number of neutrophil cells and troponin I levels in patients with Acute Myocardial Infarction.This type of research is a cross-sectional analytic observational with a retrospective approach. This research was conducted on patients with Acute Myocardial Infarction at the RSPAL dr. Ramelan Surabaya who performed a complete blood count and troponin I level with a period of January 1st 2020 – May 3rd 2021, taken by purposive sampling. The results showed that male respondents were 29 patients (63%) while female respondents were 17 patients (37%). Respondents aged 65 years amounted to 28 (61%) people, while respondents aged 65 years were 18 (39%) people. The results of the hypothesis test have a sig value of 0.000 < (0.05), so there is a relationship between the number of Neutrophil cells and Troponin I level in patients with Acute Myocardial Infarction. In hypothesis testing, the correlation coefficient value is 0.594, which means that there is a moderate relationship with a positive direction.

Abstract

The role of Laboratory examination as a diagnostic of disease is very important, including in determining the amount of uric acid contained in the body. The POCT (Point Of Care Testing) method and the Enzymatic Colorimetric method using a Photometer are several ways that can be done to determine the amount of uric acid content. This study aims to determine the differences in the results of uric acid examination in the elderly using the POCT method with the Enzymatic Colorimetric method at the Bangunsari Health Center, Madiun Regency.This type of research is an analytic observational study with a cross sectional approach. The research population is the elderly who seek treatment at the Bangunsari Health Center, Madiun Regency. The study was carried out in January-May 2021. The research sample consisted of 30 people who were taken by purposive sampling. Research data in the form of primary data obtained directly from the results of laboratory examinations. The data analysis technique used the normality test with the Kolmogorov Smirnov Test and the Independent-Sample T Test.The results showed that the data on uric acid levels in the elderly at the Bangunsari Public Health Center, Dolopo District, Madiun Regency, which was measured using the POCT (Point Of Care Testing) method showed that the mean and standard deviation values ​​were 5,650±1,283 while those measured using the Enzymatic Colorimetric method were 6,340±1,332 indicating that there was an average difference The average results of measuring uric acid levels in the elderly are between those using the POCT (Point Of Care Testing) method and those using the Enzymatic Colorimetric method. Based on the results of the Independent Sample T-test, it is known that the value of Sig. (2-tailed) of 0.046 <0.05, it can be concluded that H0 is rejected and Hi is accepted. That is, there is a significant difference in uric acid levels using the POCT method with the Enzymatic Colorimetric method.

Abstract

Cadmium is one of the chemicals contained in cigarettes, when consumed by humans in the long term and with sufficient intensity often can damage human organs such as the liver. This study aims to determine the exposure of cadmium to the levels of SGOT and SGPT in active smokers and passive smokers in a coffee shop in East Surabaya. This study used a correlational type of research, with a cross-sectional research design. There were 30 respondents who were divided into 2 groups, namely 15 active smokers and 15 passive smokers in a coffee shop in East Surabaya. The examination of cadmium used an Atomic Absorption Spectrophotomter (AAS), while the examination of SGOT and SGPT used the IFCC method (kinetic) with a photometer. The data were analyzed by Kolmogorov-Sminorv test and r-Pearson test. The results showed that there was no effect of high cadmium exposure in active smokers and passive smokers on SGOT and SGPT levels. The average cadmium in active smokers and passive smokers were 2.93 g/L and 1.30 g/L. while the levels of SGOT and SGPT in active smokers were 27.26 U/L and 36.86 U/L. The average levels of SGOT and SGPT in passive smokers were 19.73 U/L and 17.86, respectively. In these results it can be seen that the increase in cadmium levels does not affect liver function cells.

Abstract

Lead (Pb) is a heavy metal that can cause poisoning and accumulate in the human body. Determination of lead levels by using AAS the condition is that the sample must be a solution, then it is necessary to destruct first. There are two types of destruction that can be done, namely wet destruction using acid reagents to decompose samples and dry destruction by heating or destruction using very high temperatures. HNO3 oxidizing agent and HNO3 mixture with HClO4 are the main oxidizing agents that can provide the best destructive results compared to other oxidizing agents. This study aims to determine the best oxidizing and destructive substances present in the hair using Atomic Absorption Spectrophotometry (AAS). The research was conducted at surabaya regional health laboratory in April - May 2021. This research is comparative research. The sample used is the hair of gelam gas station operator officer, Candi. There are two ways of preparing samples that can be done in pb analysis on hair, namely proses wet and dry destruction. The result of destruction is determined by the level of lead using Atomic Absorption Spectrophotometry (AAS). The results of the analysis pno dry destructionHNO3 as a whole has rentang lead levels 0.1281-0.1958 mg / L. Dry destruction HNO3 : HClO4 secara overall has rentang lead levels 0.1457-0.2159 mg /L. HNO3 closed wet construction has lead content range 0.1431-0.2034 mg/L and wet destructive closed HNO3 : HClO4 has lead content range 0.1080-0.5371 mg/L. Based on these results, it can be known that all hair samples tested in this study contain lead levels that are still within normal limits or within the tolerance limit of < 12 μg/g.

Abstract

Peningkatan pelayanan laboratorium kesehatan, sangat berhubungan dengan pemantapan mutu laboratorium kesehatan, serum kontrol merupakan bagian dari pemantapan mutu laboratorium kesehatan. Pooled sera merupakan suatu jenis serum kontrol buatan sendiri. Penelitian ini bertujuan untuk mengetahui adanya pengaruh suhu dan waktu penyimpanan pooled sera terhadap uji stabiltas kadar ureum dan kreatinin abnormal. Penelitian ini menggunakan metode eksperimental dengan bahan penelitian pooled sera parameter ureum dengan menggunakan metode barthelot dan kretinin metode jafe tanpa deproteinasi yang disimpan selama 6 minggu pada suhu refrigerator dan suhu. Pooled sera didapatkan dari RSU. Haji Surabaya dan pelaksanaan penelitian dilakukan di kampus Analis Politeknik Kesehatan Surabaya. Hasil penelitian Rata-rata hasil pemeriksaan kadar ureum pada pooled sera pada rerfirgator adalah 57,60 mg/dl SD adalah 8,381 mg/dl, dan CV 14%. Rentang hasil mean ± 2SD adalah 38,94 mg/dl-72,76 mg/dl. Rata-rata pooled sera kadar ureum pada suhu freezer adalah 79,77 mg/dl, SD adalah 19,936 mg/dl, dan CV 24%. Rentang hasil mean ± 2SD adalah 41,05 mg/dl-118,09 mg/dl. Rata-rata kadar kreatinin pada pooled sera pada rerfirgator adalah 4,00 mg/dl. SD adalah 2,717 mg/dl, dan CV 67%. Rentang hasil mean ± 2SD adalah -1,58 mg/dl-8, 44 mg/dl. Rata-rata kadar kreatinin pooled sera pada suhu freezer adalah 5,69 mg/dl. SD adalah 3,947, dan CV 69%. Rentang hasil mean ± 2SD adalah -2,58 mg/dl – 13,72 mg/dl. Kesimpulan penelitian ini adalah pooled sera ureum dan kreatinin abnormal yang disimpan selama 6 minggu yaitu mengalami peningkatan dan hasil penelitian menunjukkan mean dan standart deviasi diluar batas.

Abstract

Ascariasis adalah salah satu infeksi parasit yang banyak dijumpai di Indonesia, di sebabkan oleh cacing gelang Ascaris lumbricoides, Linn. Bawang putih tunggal (Allium sativum L.) merupakan salah satu tanaman alternatif yang dapat digunakan masyarakat untuk pengobatan. Kandungan bawang putih tunggal terdiri dari senyawa flavonoid, saponin, dan tannin. Tujuan penelitian ini untuk mengetahui pengaruh pemberian perasan bawang putih tunggal terhadap waktu kematian cacing Ascaridia galli. Metode dalam penelitian ini adalah eksperimental dengan rancangan post-test only grou design. Subjek dari penelitian ini adalah Ascaridia galli, penelitian ini dilakukan di Laboratorium Parasitologi Jurusan Teknologi Laboratorium Medis Poltekkes Kemeskes Surabaya pada bulan Januari – Mei 2021. Penelitian ini menggunakan 7 kelompok perlakuan yaitu NaCl 0,9% sebagai control negatif, pirantel pamoat 0,25% sebagai control positif dan perasan bawang putih tunggal dengan konsentrasi 10%, 30%, 50%, 70% dan 100%. Data yang dianalisis menggunakan uji Kolmogrov-smirnov, uji Kruskal Wallis, lalu dilanjutkan dengan uji Post Hoc untuk mengetahui perbedaan waktu kematian cacing setiap konsentrasi dan kontrol. Rata-rata waktu kematian Ascaridia galli pada konsentrasi 10% selama menit, konsentrasi 30% selama menit, konsentrasi 50% selama menit, konsentrasi 70% selama menit dan konsentrasi 100% selama menit. Konsentrasi yang paling efektif untuk membunuh cacing Ascaridia galli pada penelitian ini adalah 100%. Sehingga dapat disimpulkan bahwa perasan bawang putih tunggal memiliki efek anthelmintik terhadap cacing Ascaridia galli

Abstract

Kontaminasi timbal akan menimbulkan dampak negatif jika masuk kedalam tubuh dalam jumlah yang berlebihan yang dapat merugikan tubuh. Salah satu orang yang berisiko terkena kontaminasi timbal yaitu pengepul sampah, pada TPS banyak sampah B2 rumah tangga khususnya yang mengandung timbal (Pb) sehingga terkena kontaminasi langsung sampah B2 Penelitian ini bertujuan untuk mengetahui perlakuan yang paling efektif antara destruksi basah secara terbuka atau tertutup dan mengetahui larutan pendestruksi terbaik dalam pemeriksaaan kadar timbal pada urin petugas sampah. Penelitian ini merupakan penelitian eksperimental dengan rancangan posttest only design yang dilakukan di Laboratorium Kesehatan Daerah Jl. Gayungsari Barat No. 124 Surabaya. Bahan uji dalam penelitian ini adalah urin petugas sampah. Sampel penelitian bersifat purposive sampling. Penetuan kadar timbal dilakukan dengan alat AAS (Atomic Absorption Spectrophotometry). Sampel harus berupa larutan yang jernih sehingga dilakukan preparasi. Terdapat dua cara preparasi sampel yang dapat dilakukan dalam analisis Pb pada urin yaitu proses destruksi basah secara terbuka dan tertutup dengan menggunakan zat pengoksidasi Asam Nitrat (HNO3), Asam Sulfat (H2SO4) dan Hidrogen Peroksida (H2O2). Hasil pengukuran rata – rata kadar timbal pada destruksi basah secara terbuka dengan zat pengoksidasi HNO3 dan H2SO4 (3:1) yaitu 0,4348 mg/L dan zat pengoksidasi HNO3, H2SO4 dan H2O2 (6:2:1) yaitu 0,4138 mg/L, rata – rata kadar Pb dalam destruksi basah secara tertutup dengan zat pengoksidasi HNO3 dan H2SO4 (3:1) yaitu 0,4018 mg/L dan destruksi basah secara tertutup dengan zat pengoksidasi HNO3, H2SO4 dan H2O2 (6:2:1) yaitu 0,3897 mg/L.

Abstract

Pemerintah memiliki fasilitas pelayanan kesehatan berupa Pusat Kesehatan Masyarakat (Puskesmas) yang memberikan pelayanan kesehatan tingkat pertama kepada perorangan dan masyarakat, salah satunya yaitu laboratorium puskesmas yang dimana dalam pelaksanaannya melakukan pengukuran, penetapan, dan pengujian terhadap bahan uji dari manusia untuk penentuan jenis dan penyebaran suatu penyakit, kondisi kesehatan, atau faktor yang dapat berpengaruh pada kesehatan perorangan dan masyarakat. Untuk menjamin tingkat akurasi dan presisi hasil pemeriksaan maka dilakukan Pemantapan Mutu Eksternal dengan tujuan sebagai bahan evaluasi dalam meningkatkan kinerja laboratorium. Dalam pelaksanaan kualitas pemantapan mutu seringkali terjadi perbedaan hasil serta dalam pelayanannya masih terbatas, karena kurangnya partisipasi laboratorium pada kegiatan Pemantapan Mutu Eksternal. Jenis penelitian deskriptif dengan populasi 39 Laboratorium Puskesmas Wilayah Kabupaten Malang dengan jumlah sampel 15 Puskesmas. Sampel yang digunakan dalam penelitian ini adalah serum kontrol normal untuk menilai pemeriksaan kolesterol dan trigliserida. Hasil analisis data menunjukkan persentase pemeriksaan kolesterol berdasarkan nilai VIS kriteria baik sebannyak 60 %, kriteria cukup sebanyak 33,33 %, kriteria kurang sebanyak 6,66 %, kriteria buruk sebanyak 0 %. Sedangkan pemeriksaan trigliserida berdasarkan nilai VIS kriteria baik sebannyak 73,33 %, kriteria cukup sebanyak 6,66 %, kriteria kurang sebanyak 6,66 %, kriteria buruk sebanyak 13,33 %.
