Abstract

Bahan kontrol penting digunakan untuk mencapai mutu pemeriksaan laboratorium. Bahan kontrol dapat diperoleh dari serum komersial atau dapat dibuat sendiri berupa pooled sera. Bahan kontrol yang biasanya digunakan di laboratorium klinik adalah bahan kontrol komersial berupa liofilisat. Namun, bahan kontrol ini harganya cukup mahal, sehingga kurang efisien digunakan di laboratorium yang mempunyai rata-rata jumlah pemeriksaan sedikit. Bahan kontrol liofilisat yang dibuat sendiri dapat digunakan sebagai alternatif dalam mengganti kontrol pabrikan yang cukup mahal. Penelitian ini bertujuan untuk menganalisis stabilitas serum liofilisat buatan sendiri sebagai bahan kontrol terhadap parameter kolesterol dan trigliserida selama 8 minggu. Penelitian ini bersifat eksperimental menggunakan rancangan penelitian time series design yang dilakukan di Laboratorium Kimia Klinik Jurusan Teknologi Laboratorium Medis Poltekkes Kemenkes Surabaya pada bulan Oktober 2021−Mei 2022. Serum darah diproses dengan teknik freeze dryed untuk mendapatkan bentuk liofilisat. Serum bentuk liofilisat disimpan pada suhu 2-8°C dan dilarutkan menggunakan aquabides setiap minggunya untuk pemeriksaan kadar kolesterol dan trigliserida. Dalam grafik Levey-Jennings kadar kolesterol dan trigliserida tidak menyimpang dari 2SD selama masa penyimpanan. Uji regresi menunjukkan bahwa lama penyimpanan berpengaruh sebesar 0,4% terhadap kadar kolesterol dan berpengaruh 55% terhadap kadar trigliserida. Hasil dari penelitian dapat disimpulkan bahwa serum liofilisat buatan sendiri stabil selama 8 minggu penyimpanan.

Abstract

Ikan cakalang adalah salah satu jenis ikan pelagis dengan kandungan protein tinggi. Kandungan air yang cukup tinggi pada tubuh ikan menjadi media yang cocok untuk kehidupan bakteri dan mikroorganisme lain serta daging ikan memiliki sedikit tendon, sehingga proses pembusukan akibat aktivitas bakteri lebih cepat dan mutu ikan berkurang serta tidak layak konsumsi, dimana untuk mengetahui adanya cemaran mikroba pada pangan dengan pemeriksaan mikrobiologis yaitu menguji total bakteri pada ikan cakalang yang dihitung berdasarkan Angka Lempeng total (ALT). Penelitianinibertujuanmengetahuijumlahmikrobapadaikancakalang (Katsuwonus pelamis) yang dijual dipasar Gedangan Sidoarjo. Jenis penelitian ini adalah deskripsi dan penelitian dilakukan di Laboratorium Mikrobiologi Jurusan Analis Kesehatan Politekknik Kesehatan Surabaya. Sampel penelitian adalah ikan cakalang sebanyak 5 buah yang diambil secara total sampling. Metode pemeriksaan menggunakan hitung cawan dengan metode tuang. Hasil penelitian menunjukkan bahwa pengujian Angka Lempeng Total pada ikan cakalang (Katsuwonus pelamis) yang dijual dipasar Gedangan Sidoarjo setelah dilakukan uji secara mikrobiologi yaitu dari ke lima sampel yang dilakukan analisis terdapat 4 sampel yang termasukdalambatasnormalyangsesuaidenganStandarNasionalIndonesia(SNI)7388:2009 tentang batas maksimum mikroba dalam pangan yaitu 5x105koloni/g yang artinya ikan tersebut aman untuk dikonsumsi. Sedangkan 1 sampel termasuk dalam cemaran, karena tidak sesuai dengan Standart Nasional Indonesia (SNI) 7388:2009 tentang batas maksimum cemaran mikroba dalam pangan yaitu 5x105koloni/g yang artinya ikan tersebut tidak boleh untuk dikonsumsi berdasarkan Standart Nasional Indonesia (SNI) 7388:2009 tentang batas maksimum cemaran mikroba dalampangan.

Abstract

ABSTRACTCigarettes contain various harmful substances that can endanger health. One of them is nicotine which causeslipid metabolism disorders, including a decrease in HDL cholesterol levels and an increase in total cholesterollevels. Smoking habits in people with comorbidities such as diabetes mellitus can exacerbate existingdyslipidemia conditions. If not managed properly, it can cause complications that lead to increased mortality inpeople with diabetes mellitus. This study aims to determine the differences in HDL cholesterol and totalcholesterol levels in active smokers and passive smokers patients with diabetes mellitus at PuskesmasTanjunganom, Nganjuk Regency. This research uses observational study with a cross sectional approach whichwas conducted in January-May 2021 on 30 patients with diabetes mellitus at Puskesmas Tanjunganom, NganjukRegency, consisting of 15 active smokers and 15 passive smokers. Checking total cholesterol levels was carriedout at Puskesmas Tanjunganom, Nganjuk Regency. While the examination of HDL cholesterol levels wascarried out at UPT Labkesda, Nganjuk Regency. Based on the independent sample t-test statistical test, it wasfound that the p-value of difference HDL cholesterol levels in active smokers and passive smokers with diabetesmellitus was 0,000 and the difference total cholesterol levels in active smokers and passive smokers withdiabetes mellitus was 0.012. So it can be concluded that statistically there are differences in HDL cholesteroland total cholesterol levels in active smokers and passive smokers who suffer from diabetes mellitus atPuskesmas Tanjunganom, Nganjuk Regency.Keywords: Diabetes Mellitus; Cholesterol Levels; Active Smoker; Passive SmokerABSTRAK Rokok mengandung berbagai zat berbahaya yang dapat menggangu kesehatan. Salah satunya nikotin yangmengakibatkan gangguan metabolisme lipid antara lain penurunan kadar kolesterol HDL dan peningkatan kadarkolesterol total. Kebiasaan merokok pada orang dengan penyakit penyerta seperti diabetes melitus dapatmemperburuk kondisi dislipidemia yang sudah ada. Apabila tidak dikelola dengan baik dapat mengakibatkankomplikasi yang menyebabkan peningkatan mortalitas penderita diabetes melitus. Penelitian ini bertujuan untukmengetahui perbedaan kadar kolesterol HDL dan kolesterol total pada perokok aktif dengan perokok pasif yangmenderita diabetes melitus di Puskesmas Tanjunganom Kabupaten Nganjuk. Penelitian ini merupakan penelitanobservasional dengan pendekatan cross sectional yang dilakukan pada bulan Januari-Mei 2021 terhadap 30pasien penderita diabetes melitus di Puskesmas Tanjunganom Kabupaten Nganjuk yang terdiri atas 15 orangperokok aktif dan 15 orang perokok pasif. Pemeriksaan kadar kolesterol total dilakukan di LaboratoriumPuskesmas Tanjunganom Kabupaten Nganjuk. Sedangkan pemeriksaan kadar kolesterol HDL dilakukan di UPTLabkesda Kabupaten Nganjuk. Berdasarkan uji statistik independent sampel t-test diperoleh p-value perbedaankadar kolesterol HDL pada perokok aktif dengan perokok pasif penderita diabetes melitus sebesar 0,000 sertaperbedaan kadar kolesterol total pada perokok aktif dengan perokok pasif penderita diabetes melitus sebesar0,012. Sehingga dapat disimpulkan bahwa secara statistik terdapat perbedaan kadar kolesterol HDL dankolesterol total pada perokok aktif dengan perokok pasif yang menderita diabetes melitus di PuskesmasTanjunganom Kabupaten Nganjuk.Kata Kunci : diabetes melitus; kadar kolesterol; perokok aktif; perokok pasif

Abstract

BSTRACTThe researcher saw that the level of cleanliness and hygiene of the sellers at traditional markets in Surabaya was still low, and the researchers also observed that the need for vegetable food was quite high. Coliformbacteria contamination in drinking water can cause various diseases such as diarrhea, fever, dysentery, andothers. The parameter is the water used for processing soy milk, namely PDAM water and refill water. Thisstudy aims to determine the presence or absence of coliform bacteria contamination in soy milk sold in thePucang market in Surabaya, as well as whether or not the soy milk is consumed. This study aims to determinethe presence or absence of coliform bacteria contamination in soy milk sold in the Pucang market in Surabaya,and whether or not the soy milk is consumed. This research used 10 samples of soy milk obtained from 10different sellers. This analysis used the MPN method with a combination of 5-1-1 tubes, and each was incubatedon Lactose Broth (LB) Single Strength, Lactose Broth Double Strength, Brilliant Green Lactose Broth (BGLB)and Eosin Methylene Blue Agar (EMBA) media. The data were presented descriptively and compared with SNI7388:2009. The results showed that 7 of 10 samples were contaminated with faecal coliform bacteria (57.14%).It can be concluded that there is coliform bacteria contamination in the sample, but the MPN value is still belowthe SNI limit.

Abstract

Meanings of animal origin are often contaminate by microorganism, one of it is chicken eggs. Free range chicken is often consumed raw for medicinal purposes without knowing any contamination in it. Free range chicken are often associated with the genesis of foodborne diseases that can cause ailments in the form of diarrheal and gastroenteristic diseases. According to data provided by the 2018 ministry of health, there are approximately 7 million cases of diarrheal disease every year. The bacteria responsible for diarrhea is Salmonella sp, Escherichia coli and Staphylococcus aureus. This type of research is laboratory experimental research through culture. The population of this study was eggs obtained from the village of Tanggul Kulon RT 01/RW 07 Jember. The sample of this research is free range chicken taken by purposive sampling based on criteria. The place of this research was conducted at the Bacteriology Laboratory of Poltekkes Surabaya from January to April 2021. The result showed 4 out of 12 positive samples identified as Escherichia coli and 8 out of 12 positive samples identified as Salmonella parathypi A bacteria

Abstract

The Erythrocyte sedimentation rate is a routine blood test performed to monitor the course of the disease where high values ​​are likely to be associated with the presence of inflammation. ICSH (International Council for Standardization in Hematology) recommends the Westergren method as a method for examining LED with several developments in terms of the anticoagulants and diluents used. Anticoagulants that can be used are Sodium Citrate 3,8% anticoagulant and EDTA (Ethylene Diamine Tetra-Acetid-Acid) anticoagulant. This study aims to determine the differences in the erythrocyte sedimentation rate of the westergren method using EDTA anticoagulants and sodium citrate 3.8%.This study uses primary data collection techniques with comparative research type. The research was conducted at the Health Analyst Laboratory of Poltekkes Kemenkes Surabaya, in January-April 2021. The sample of the study was 50 menstruating women who had met several criteria, who would be examined for LED values ​​using the Westergren method using EDTA anticoagulants in purple vacuum tubes and sodium citrate 3,8 % on a black vacuum tube.The results showed that the mean of sedimentation rate with EDTA anticoagulant was 27.66 mm/hour and 22.04 mm/hour for sodium citrate 3,8% anticoagulant. The statistical test was carried out by using the paired sample t-test which showed the sig (p) value = 0.000 0.05, so it could be concluded that there was a difference between the EDTA anticoagulant LED and the sodium citrate anticoagulant LED. Thus the EDTA anticoagulant can still be used as an anticoagulant in examining the erythrocyte sedimentation rate

Abstract

Actinomycetes are gram-positive bacteria that have aerobic properties. Actinomycetes have the ability to produce antibiotic compounds. Actinomycetes are the second largest organisms that live in soil with a population of 1-10 million per gram of soil. Actinomycetes also make up 10-50% of the microbes in the soil. The purpose of this study was to determine the antifungal activity of actinomycetes isolates isolated from the soil of the Mangrove Wonorejo forest in Surabaya against the fungus Trichophyton rubrum. This type of research is an experimental laboratory with quantitative analysis. Actinomycetes isolates were taken from 3 different soil locations, pretreatment by heat treatment at 90°C for 15 minutes. Actinomycetes were isolated on casein starch agar medium which had been added with 0.002% Nystatin. Selection of antimicrobial compounds was based on the disc diffusion method which was characterized by the formation of a zone of inhibition around the disc. The results showed that from 3 locations of soil sampling, 8 actinomycetes isolates were obtained, but only 6 isolates were able to inhibit the fungus Trichophyton rubrum. showed that isolates C3-2, B3-1, C4-1, were categorized as having very strong inhibitory power. While isolates A5-1, A3-2 were categorized as having inhibitory power and isolates A4-1 and A4-2 were categorized as having moderate inhibitory power against the fungus Trichophyton rubrum.

Abstract

Diabetes mellitus type 2 is a metabolic disease that occurs due to abnormalities in insulin secretion, insulin action or both. One of the most common disorders in the body system is the red blood cell index consisting of MCV, MCH and MCHC. Spirulina platensis is a blue-green microalgae that contains phycocyanin so that it can increase hemoglobin levels, erythrocytes and blood profile in the bone marrow. The purpose of this study was to determine the difference before and after giving Spirulina platensis supplementation for 10 days on the erythrocyte index in patients with type 2 diabetes mellitus. This type of research was one group pretest-posttest. This research was conducted at the Waru Community Health Center, Sidoarjo Regency from December 2020 to May 2021. The results showed that the mean value of MCV decreased before: 80.19 fl and after: 80.1 fl. Meanwhile, the average value of MCH and MCHC increased, namely MCH before: 27.57 pg and after: 27.68 pg, then MCHC before: 34.33 g / dL and after: 34.51 g / dL. In the third statistical test, the significance value (p-value), namely the MCV (0.863), MCH (0.773) and MCHC (0.627) shows that (p> 0.05), it can be concluded that there is no significant difference before and after supplementation. Spirulina platensis on the erythrocyte index (MCV, MCH and MCHC) in people with type 2 diabetes mellitus.

Abstract

Lead is one of the heavy metals that can cause health problems and is widely found in the environment. Lead level examination can use specimens of hair, urine, and blood. Hair can be used as an indicator of examination for lead poisoning that is internal and external. While urine is a screening test on lead poisoning. And the blood specimens is an overview of the level of lead absorbed by the body. The purpose of this research was to look at the content of lead levels in specimens of hair, urine, and blood of TPS 3R Sutorejo garbage officers. This research is a descriptive research with cross sectional design conducted at Balai Besar Laboratoriun Kesehatan Surabaya in October 2020 – June 2021. The samples used were 10 garbage officers who were taken using purposive sampling techniques. The results showed that lead levels in the hair were between 0.108 μg/g – 0.239 μg/g, in urine 0.000095 mg/L – 0.00121 mg/L, and in blood 1,665 μg/dL – 4,785 μg/dL. All respondents (100%) have lead levels in specimens of hair, urine, and blood in the normal category. Examination using blood and urine specimens can be used to determine the level of lead that is collected in a short period of time. While in the long term can use hair specimens. Timbal merupakan salah satu logam berat yang dapat menyebabkan permasalahan kesehatan dan banyakditemukan di lingkungan. Pemeriksaan kadar timbal dapat menggunakan spesimen rambut, urin, dan darah.Rambut dapat dijadikan indikator pemeriksaan untuk keracunan timbal yang bersifat internal dan eksternal.Sedangkan urin merupakan screening test pada keracunan timbal. Dan darah menunjukkan gambaran kadartimbal yang terserap oleh tubuh. Tujuan dari penelitian ini adalah untuk melihat kandungan kadar timbal dalamspesimen rambut, urin, dan darah petugas sampah TPS 3R Sutorejo menggunakan Spektrofotometri SerapanAtom. Penelitian ini merupakan penelitian deskriptif dengan rancangan cross sectional yang dilaksanakan diBalai Besar Laboratorium Kesehatan Surabaya pada bulan Oktober 2020 – Juni 2021. Sampel yang digunakanadalah 10 orang petugas sampah yang diambil menggunakan teknik purposive sampling. Hasil penelitianmenunjukkan bahwa nilai kadar timbal dalam rambut antara 0,108 µg/g – 0,239 µg/g, dalam urin 0,000095mg/L – 0,00121 mg/L, dan dalam darah 1,665 µg/dL – 4,785 µg/dL. Semua responden (100%) memiliki kadartimbal dalam spesimen rambut, urin, dan darah dengan kategori normal. Pemeriksaan menggunakan spesimendarah dan urin dapat digunakan untuk mengetahui kadar timbal yang terabsorbsi dalam jangka waktu pendek.Sedangkan pada jangka waktu lama dapat menggunakan spesimen rambut.Kata kunci : Timbal; Petugas sampah; Rambut; Urin; Darah; Spektrofotometri Serapan Atom

Abstract

Gas stations are a source of lead exposure that comes from vehicle gas emissions and petrol fuel vapors. Gas station officers, both operators and nonoperators, have a risk level of lead exposure that can enter the body and then accumulate and excreted through urine by 75-80%. Lead exposure can endanger the health of gas station officers. The purpose of this study was to analyze differences in lead levels in the urine of gas station operators and nonoperators in Kademangan District, Probolinggo City. This research is an observational study with a cross sectional method which was conducted at the Surabaya Health Laboratory Center in October 2020 - June 2021. The sample used was 15 operators and 15 nonoperators who were taken by purposive sampling. The results showed the average value of lead levels in the urine for operators 0.0007213 ppm, indoor nonoperators 0.0006386 ppm and outdoor nonoperators 0.0008263 ppm. The conclusion of this study was that there was no significant difference in lead levels in the urine of gas station operators and nonoperators.
