Abstract

Natural sources for hydrolytic enzymes produced by bacteria in demand in industry were increased. Hydrolytic bacteria can be found in agricultural and household waste, like black glutinous wash water. This study aims to identify and evaluate the presence and hydrolytic potential of bacteria in washing waste from fermented black glutinous rice. Fermented black glutinous rice washing water was used as the sample for bacterial isolation. The obtained isolates were screened for hydrolytic activity using modified Bushnell–Haas Agar supplemented with specific substrates to assess cellulolytic, amylolytic, and pectinolytic activities. Hydrolytic activity was expressed as a hydrolytic index to determine the most potent isolate, which was subsequently identified at the molecular level. A total of eight bacterial isolates were successfully obtained, among which one isolate, designated NKH4, showed the highest hydrolytic potential based on screening results. Molecular identification revealed that NKH4 is closely related to Bacillus tequilensis, with a similarity value of 97.96%. This species has previously been reported to produce various hydrolytic enzymes. The selected isolate NKH4 exhibited a qualitative hydrolytic index of 0.89, demonstrating its ability to hydrolyze starch, cellulose, and pectin. Therefore, NKH4, identified as Bacillus tequilensis, holds strong potential as a producer of hydrolytic enzymes and may serve as a promising candidate for further biotechnological applications.

Abstract

Tofu wastewater contains total nitrogen (N), phosphorus (P), and potassium (K) at levels of 43.37 mg/L, 114.36 mg/L, and 223 mg/L, respectively. In addition, lamtoro leaves have potential as an organic fertilizer; every 100 g of fresh lamtoro leaves yields approximately 85% dry matter, containing 20–25% protein, 20–30% free nitrogen, 5–10% fat, 3.89% energy, 1.5–2.5% tannins, 0.8–1.8% calcium, and 0.23–0.27% phosphorus. These two materials can be processed into liquid organic fertilizer (LOF) and applied to red spinach cultivated in polybags. This study aimed to analyze C-NPK content, determine the optimal concentration of LOF to enhance red spinach growth, and identify the best growth parameters influenced by LOF application derived from tofu wastewater and lamtoro leaves. The independent variable was LOF concentration with four replications and five treatments: K (0%), A (5%), B (10%), C (15%), and D (20%). The dependent variables included plant height, number of leaves, leaf width, root length, root fresh weight, root dry weight, stem fresh weight, and stem dry weight. The results showed that the LOF contained 9.64% organic C, 0.57% N, 0.042% P, and 0.358% K. The optimal concentration was treatment C (15%). The best growth parameters were observed in root length, root fresh weight, stem fresh weight, and stem dry weight.

Abstract

Papaya seed endophytic bacteria have been reported to exhibit antibacterial activity against pathogenic microorganisms, including Staphylococcus aureus and Escherichia coli. However, their antibacterial production remains suboptimal. This study aimed to determine the optimal conditions for antibacterial production and to identify the most potent bacterial isolate through 16S rRNA gene-based molecular analysis. Three endophytic bacterial isolates (CPA2, CPA8, and CPA9) were evaluated. Characterization was performed based on pH variations (5, 7, and 9) and growth media (Nutrient Broth (NB) and Tryptic Soy Broth (TSB)), followed by antibiotic sensitivity testing against ciprofloxacin, amoxicillin, gentamicin, and tetracycline. Antibacterial activity was assessed using the Kirby–Bauer method against S. aureus and E. coli. The results showed that all isolates grew optimally in TSB medium at neutral pH (7). Ciprofloxacin and tetracycline exhibited stronger inhibitory effects compared to gentamicin and amoxicillin. Among the isolates, CPA2 in TSB media pH 7 demonstrated the highest antibacterial activity, producing inhibition zones of 30.2 ± 1.77 mm against S. aureus (very strong) and 11.8 ± 0.02 mm against E. coli (strong). Molecular identification revealed that isolate CPA2 was closely related to Bacillus cereus strain JCM 2152 with 99.90% similarity. In conclusion, papaya seed endophytic bacteria, particularly isolate CPA2, show significant potential as a source of antibacterial compounds under optimized culture conditions. These results have the potential as an alternative source of antibacterial agents

Abstract

Riparian vegetation plays a crucial role in maintaining aquatic ecosystem stability by acting as a natural buffer against anthropogenic pollution and erosion. This study analyzed the composition, structure, and ecological functions of riparian vegetation in Lake Tondano, North Sulawesi, Indonesia, with emphasis on pollutant filtration and sediment stabilization. Field surveys were conducted from May to August 2025 at ten riparian stations surrounding Lake Tondano (4,638 ha). Vegetation sampling used a stratified line transect method with fifty plots of varying sizes according to vegetation strata. Quantitative analysis included relative abundance, frequency, dominance, Importance Value Index (IVI), and the Shannon–Wiener diversity index (H’). Ecological functions were assessed through correlations between vegetation structure and reported nutrient and sediment reduction efficiencies from tropical riparian studies. A total of fifteen dominant riparian species were identified, mainly from the families Poaceae and Cyperaceae. Cyperus papyrus (IVI = 21.81%) and Metroxylon sagu (IVI = 29.65%) functioned as key ecological species, while Ficus septica showed the highest IVI (90.83%), indicating strong structural dominance. The Shannon–Wiener diversity index (H’ = 2.65) reflects moderate to high diversity. Riparian vegetation was estimated to reduce nutrient loads by 40–60% and erosion by 30–50%, highlighting its importance for ecosystem-based lake management.
