Abstract

Tumbuhan Libo (Ficus variegate Blume) merupakan salah satu tumbuhan liar, dan bagian daunnya tidak pernah dimakan oleh ulat. Pada Tahap ini dilakukan optimalisasi teknik ekstraksi dan menguji metabolit sekunder daun libo. Optimalisasi dilakukan dengan mengubah berbagai variabel yaitu metode ekstraksi, variasi jumlah pelarut, dan lamanya waktu ekstraksi. Efektivitas ekstraksi dilihat dari %rendemen yang dihasilkan. Metode yang digunakan adalah maserasi dan soxhlet, sedangkan pelarut yang digunakan adalah metanol dengan variasi jumlah pelarut 1:10, 1: 20, 1:30 serta variasi waktu ekstraksi (1,2,3 4 dan 5 hari untuk maserasi dan 1,2,3,4 dan 5 siklus untuk metode soxhlet). Hasil yang didapat dari penelitian ini adalah metode terbaik untuk ekstraksi daun libo adalah metode soxhlet dengan % rendemen 34,00 dengan perbandingan pelarut 1:30 dengan lama ekstraksi sebanyak 5 siklus. Dari pengujian metabolit sekunder diketahui daun Libo ini mengandung fenol, flavonoid dan alkaloid.

Abstract

Has been done Isolation, Characterization and Secondary Metabolite Endophytic Fungal Isolate from Peronema canescens Jack Leave and Coptosapelta tomentosa Valeton K. Heyne Root. The aim of this research is to know the number of fungal isolates, chromatogram profile and secondary metabolite group of endophytic fungal isolates from P. canencens leaves and C. tomentosa root. Characterization of endophytic fungal isolates was done macroscopically and microscopically. Identification of secondary metabolites endophytic fungal isolates were performed by chemical reaction test and TLC (Thin Layer Chromatography) method with specific spray reagents. The data of this study were obtained based on the number of endophytic fungal that can be isolated, observing macroscopic and microscopic morphological profiles, chromatogram profile and secondary metabolites of each endophytic fungal isolated. The results showed that endophytic fungal that can be isolated from P. canencens leaves four isolates, and two isolates from C. tomentosa root. Morphological profile macroscopic endophytic fungal of the six isolates showed a greenish-colored colony, white gray, clear black. Microscopic profile of each fungal isolate having spores, sprangiosphora, sporangium, conidia, hyphae and stolon. The identified secondary metabolites are: alkaloids, terpenoids, and flavonoids, and polyphenols.

Abstract

The bioactivity of some species of rattan fruit has been previously studied and it was found that it has potential antioxidant agents. This study was conducted to determine the antioxidant content in three species of edible rattan fruit, namely Calamus manan Miq., Calamus ornatus Bl. and Calamus caesius Bl by using the DPPH method, whereby the test was carried out on the pericarp, flesh, and seeds of each fruit. The research findings revealed that phytochemical compounds, i.e. flavonoids, tannins, and triterpenoids are found almost in all parts of the fruit, while steroid is not found in any of the three species of the fruit. The antioxidant activity found in the fruit of C. caesius Bl is strong, meanwhile the antioxidant activity in the fruit of C. manan Miq only strong in its pericarp and seeds, but it is moderate in its flesh. The antioxidant activity found in the fruit of C. ornatus Bl. is strong in its pericarp and seeds, but it is moderate in its flesh. Therefore, these three species of rattan fruit are potential as antioxidant agents.
