Abstract

Dengue keeps spreading through Aedes aegypti, and creeping resistance to synthetic larvicides such as temephos (Abate) has renewed interest in botanical alternatives. This study examined the larvicidal potential of ethanol extract from langsat leaves (Lansium domesticum var. pubescent), a plant already known to contain alkaloids, saponins, and terpenoids. Leaves were dried, macerated in ethanol, and concentrated by rotary evaporation before being diluted into four test concentrations. A completely randomized design was used with six groups — a negative control, a positive control (1% abate), and extract concentrations of 20%, 30%, 40%, and 50% — each with three replicates of 25 third-instar Aedes aegypti larvae, observed over 24 hours at six time points. Abate eliminated its entire larval population almost immediately, confirming assay validity. The extract itself produced a modest, dose-dependent effect: cumulative mortality rose from 38.67% at 20% and 30% concentrations to 43.99% at 40% and 53.77% at 50%. Two-way ANOVA followed by LSD testing showed that both concentration and observation time significantly affected mortality; the negative control differed significantly from the positive control and the 50% extract group, while the three lower concentrations did not differ from each other. Probit analysis placed LC50 at 45.24% (95% CI 42.67–48.44%) and LC90 at 67.57% (95% CI 60.25–84.83%), classifying the extract as moderately toxic on both measures. Langsat leaf extract thus shows measurable, if still limited, promise as a plant-based supplement to synthetic larvicides for dengue vector control.

Abstract

Background: Cigarette smoke has the potential to disrupt hormonal balance, affect spermatogenesis processes, damage spermatozoa viability, and create the presence of toxic substances in spermatozoa. On the other hand, avocados contain protein and various vitamins, such as A, B, B1, B2, E, and C, as well as other substances, such as beta-carotene, linoleic acid, lecithin, calcium, iron, pantothenic acid, potassium, phosphorus, niacin, and natural plant oils. Vitamins C and E in avocados are believed to increase the number of Leydig cells and spermatozoa. Objective: This study aims to evaluate the potential of avocado juice to improve reproductive health in mice exposed to kretek cigarette smoke. Methods: Male mice (Mus musculus) aged 2 months, weighing ±20g each, were divided into 5 randomly assigned groups: K- (control without treatment), K+ (positive control exposed to cigarette smoke), P1 (mice exposed to cigarette smoke and given avocado juice at a dose of 0.5 ml/20 g BW), P2 (mice exposed to cigarette smoke and given avocado juice at a dose of 0.7 ml/20 g BW), and P3 (mice exposed to cigarette smoke and given avocado juice at a dose of 0.9 ml/20 g BW). Cigarette smoke exposure was conducted for 14 days. Results: The results showed that regular avocado juice consumption could increase sperm production and reduce spermatozoa abnormalities in mice exposed to kretek cigarette smoke. Conclusion: The clinical implications of these findings may contribute to a better understanding of avocado juice's potential as a supplement to improve reproductive health, especially in the context of kretek cigarette smoke exposure.

Abstract

Background: People have long utilized various herbs for traditional medicine, including the suruhan or sirih cina plant (Peperomia pellucida). This plant is known by numerous local names such as rumput cacing, sirih bumi, pansit-pansitan, rumput gelas, and daing siheah cino. It is widely distributed in tropical and subtropical regions, including Indonesia, making the raw materials easily accessible for herbal medicine. Objective: This study aims to review the ethnobotany and the pharmacological potential of Peperomia pellucida as a natural ingredient for herbal medicine. Methods: This article is a narrative review that synthesizes various literatures regarding the traditional uses, phytochemical content, and biological activities of the suruhan plant. Results: The suruhan plant is used in traditional medicine through various preparation methods depending on the ailment, with the leaves being the most frequently utilized part. Phytochemical analysis reveals that it contains abundant bioactive compounds, including alkaloids, flavonoids, sterols, tannins, reducing sugars, amino acids, saponins, triterpenoids, carbohydrates, phenols, azulene, carotenoids, and quinones. These compounds contribute to its wide-spectrum antibacterial, antifungal, and anticancer activities. Conclusion: Peperomia pellucida holds significant potential as a herbal medicine ingredient due to its rich bioactive content and its long-standing history of traditional use. Accelerated industrial development and government support are needed to optimize its utilization in the healthcare sector.

Abstract

Diabetes mellitus remains an expanding metabolic burden, and Indonesia ranks fifth globally with an estimated 10.7 million cases in 2019. Chronic hyperglycaemia damages the kidneys and pancreas through oxidative stress pathways, which keeps plant-derived antioxidants under active investigation as adjunct therapy. Red betel leaf (Piper crocatum) contains flavonoids and alkaloids that inhibit α-glucosidase in vitro, yet histopathological evidence on target organs remains limited and inconsistently reported. This experimental study evaluated the effect of boiled red betel leaf water at concentrations of 25%, 50%, 75%, and 100% on renal histopathology (tubular cell size and glomerular diameter) and pancreatic histopathology (nuclear and cytoplasmic clarity of the islets of Langerhans) in alloxan-induced diabetic mice (Mus musculus), given a single dose of 100 mg/kgBW. A Completely Randomized Design was used with two control groups (K+ and K−) and four treatment groups (P1–P4), totalling 18 mice with three visual-field replicates per group. The extract was administered by gavage at 0.13 mL per day for seven days. Multivariate Analysis of Variance (MANOVA) on renal parameters showed a significant concentration effect on tubular size and glomerular diameter (Wilks' Lambda = 0.011; F = 19.205; p = 0.000), with tubular size and glomerular diameter increasing proportionally with concentration, reaching 58.2% and 64.2% at P4 relative to K+. The Kruskal-Wallis test on pancreatic data was also significant (p = 0.015), and Mann-Whitney post-hoc analysis indicated that groups P3 and P4 differed significantly from K+ but not from K−. Concentrations of 75% and 100% produced histological features closest to the healthy control. Post-treatment blood glucose values for groups P1–P4 were incompletely recorded in this study, so the magnitude of glycaemic reduction cannot yet be confirmed; this gap should be closed in follow-up work before an effective concentration is recommended for the next stage of preclinical testing.

Abstract

Background: State policy on population growth carries broad implications for health development, with the Family Planning (KB) program as one of its principal instruments. Long-Acting Reversible Contraception (LARC) uptake, IUD especially, still lags behind short-acting hormonal methods in Indonesia, partly because couples of reproductive age (PUS) receive incomplete information when deciding (Gayatri, 2020; Mahendra et al., 2019). Objective: To examine whether adding leaflet media to routine Information-Education-Communication (KIE) counseling strengthens PUS decision-making certainty about IUD contraception, compared with KIE from health workers alone. Methods: A quasi-experimental, posttest-only, non-equivalent control group design was conducted in the working area of Puskesmas Gunungpati, Semarang. Fifty respondents (25 receiving KIE plus leaflet, 25 receiving KIE alone) were drawn from the reproductive-age population (15–40 years) through proportional cluster random sampling. Because the decision-certainty score is ordinal, group differences were tested with the Mann-Whitney U test. Results: The KIE-plus-leaflet group scored higher (median 5.00; mean 4.16) than the KIE-only group (median 4.00; mean 3.28), U=158.500, Z=-3.134, p=.002. The proportion who ultimately chose IUD did not differ significantly between groups (23/25 versus 22/25; Fisher's exact test, p=1.000). Conclusion: Leaflet media, added to standard KIE counseling, strengthens decision-making certainty around IUD contraception among PUS, even without a shift in how many ultimately accept the method. Health workers are encouraged to keep leaflets in routine family-planning counseling, given the gain shows up in decision quality rather than acceptance rates.

Abstract

Background: S. aureus and S. pyogenes are Gram-positive bacteria that are the main causes of impetigo, a skin infection marked by a red rash. Impetigo is commonly treated with antibiotics, but long-term use risks side effects, so a complementary alternative such as green tea leaf extract (Camellia sinensis) is needed. Aim: To determine the effect of liquid soap containing 7%, 10%, and 13% green tea extract on the inhibition zone of S. aureus and S. pyogenes. Methods: A true experimental study with a post-test-only with control group design used five treatment groups. Inhibition zone diameters were measured with a caliper and analysed using one-way ANOVA with Games-Howell post-hoc comparisons and Mann-Whitney to compare the inhibition zones between the two bacterial species. Results: The 10% concentration produced the highest mean inhibition zone, 5.53 mm against S. aureus and 6.33 mm against S. pyogenes (moderate category), significantly larger than the negative control (p<0.05) but still below the tetracycline positive control. No significant difference was found between the two species (p=0.381). Conclusion: Green tea extract liquid soap shows moderate antibacterial activity against S. aureus and S. pyogenes, with comparable potential against both pathogens as a topical complementary therapy for impetigo.

Abstract

Background: Green betel (Piper betle) and red betel (Piper crocatum) leaves are widely reported to contain flavonoids, tannins, alkaloids, and saponins, compounds empirically linked to free-radical scavenging activity, yet a direct, quantitative comparison of their antioxidant strength across two polar solvents remains scarce. Objective: To compare the phytochemical content and antioxidant activity of green and red betel leaf extracts using 70% ethanol and 70% methanol via the DPPH method. Method: This laboratory-experimental study was conducted at the Biomedical Science Laboratory, Universitas Karya Husada Semarang, from 22–28 February 2024 (Ethical Clearance No. 003/KEP/UNKAHA/SLE/2024). Antioxidant activity was measured across a 5–9 ppm concentration range, modeled with the linear regression curve y=ax+b, and compared across the four extract-solvent combinations using the Kruskal-Wallis test. Result: Phytochemical screening revealed a more complete compound profile in 70% ethanol than in 70% methanol for both leaf types. All four combinations exhibited DPPH inhibition above 70% even at the lowest tested concentration (5 ppm; range 72.10–97.46%), placing the true IC50 below 5 μg/mL for every combination — well under the 50 μg/mL “very strong” threshold. The Kruskal-Wallis test detected no significant difference in inhibition percentage among the four combinations (H=3.85, df=3, p=0.279). Conclusion: Both green and red betel extracts, in either solvent, qualify as very strong antioxidants with no statistically meaningful difference in potency, positioning either as an equally promising candidate for further natural antioxidant development.

Abstract

Background: Pneumonia caused by Klebsiella pneumoniae remains a considerable global health burden, and antibiotic misuse has driven the bacterium toward growing antimicrobial resistance, leaving plant-derived compounds such as red ginger (Zingiber officinale var. rubrum) worth investigating as alternative antimicrobial agents. Objective: This study aimed to determine the effectiveness of red ginger extract in inhibiting the growth of K. pneumoniae, combining in vitro testing with in silico target-gene prediction. Method: Red ginger rhizomes were extracted, tested for antimicrobial activity against K. pneumoniae at four concentrations, and the dominant bioactive compounds were subjected to in silico target-gene analysis. Result: The extract inhibited K. pneumoniae growth in a dose-dependent manner, with inhibition zones ranging from 6.7 mm at 25% concentration to 8 mm at 100%. The bioactive compounds were predicted to target multiple K. pneumoniae proteins across several cellular locations, most notably the acrR gene, a transcriptional repressor of the multidrug efflux pump operon acrAB. Conclusion: Red ginger extract inhibits K. pneumoniae growth and appears to act through the acrR-acrAB efflux pump pathway, a mechanism central to the bacterium's antibiotic resistance, indicating value as a candidate antimicrobial adjunct that warrants further optimization.

Abstract

Background: Pregnancy is a natural process that brings physical and psychological discomfort to pregnant women. One of the most common discomforts in the third trimester is lower back pain, which occurs due to increased production of the hormone relaxin, causing the pelvic bone joints to stretch and facilitate childbirth. Petrissage massage combined with lavender aromatherapy was administered to third-trimester pregnant women as a therapy to reduce this pain. Objective: This study aimed to determine the effect of a combination of petrissage massage and lavender aromatherapy on lower back pain in third-trimester pregnant women. Method: This quantitative study used a pre-experimental one-group pretest-posttest design without a control group, involving 15 samples of third-trimester pregnant women. The independent variable was the administration of petrissage massage and lavender aromatherapy, while the dependent variable was back pain. Pain was measured before and after the intervention using the Numeric Rating Scale (NRS). Result: The Wilcoxon test yielded a p value of 0.000, indicating that the combination of petrissage massage and lavender aromatherapy reduced lower back pain in third-trimester pregnant women. Conclusion: There is a significant effect of combined petrissage massage and lavender aromatherapy on lower back pain in third-trimester pregnant women; pregnant women can apply this combination as a nonpharmacological approach to reduce lower back pain.

Abstract

Background: At Muhammadiyah Rodliyah Achid Moga Hospital, approximately 110 postpartum mothers give birth each month; however, only 60% breastfeed their infants, while 40% do not. To date, hospital interventions have been limited to education on breastfeeding techniques and breast care, with no prior education provided regarding oxytocin massage. Objective: This study aimed to determine the difference in breastfeeding behavior among postpartum mothers before and after receiving education on oxytocin massage at Muhammadiyah Rodliyah Achid Moga Hospital. Methods: This quantitative study employed a pre-experimental design with a one-group pretest-posttest approach involving 86 respondents—determined using Slovin's formula from a population of 110 postpartum mothers—selected via accidental sampling. Data were analyzed using the Kolmogorov-Smirnov normality test and the Wilcoxon test. Results: The normality test indicated that the data were not normally distributed (p < 0.05). Following the education, there was an increase in both the mean and minimum scores for breastfeeding behavior. The Wilcoxon test yielded a p-value of 0.001 (p < 0.05), indicating a significant difference in the breastfeeding behavior of postpartum mothers before and after the oxytocin massage education. Conclusion: Oxytocin massage education contributes to improving breastfeeding behavior among postpartum mothers at Muhammadiyah Rodliyah Achid Moga Hospital; however, the pre-experimental design (lacking a control group) means these findings require further confirmation through studies that include a comparison group.
