Biodiversitas 22: 3049- 3056. Cassava leaves have various macro and micro-nutrients with varying levels, such as magnesium, iron, and zinc. The differences in genotypes of cassava may result in different mineral content in cassava leaves. This study aims to determine the estimator phenotypic markers and contrast genotype candidate selection related to target minerals. This study used a one-factor treatment composed of 12 cassava genotypes and arranged in a randomized block design (RBD) which each treatment was repeated 3 times. The observation was conducted on the leaves' qualitative and quantitative characters such as plant growth, leaf morphology, and leaf mineral content characters. The observation was performed at 3 and 6 months after planting. The results showed that the quantitative and qualitative characters that could be used as estimator for mineral content in cassava leaves are the Soil Plant Analysis Development (SPAD)- Chlorophyll value and leaf color. Genotypes G6-1-15-4-3, G6-2-15-1-1, G6-2-15-3-3 (lowest) and Malang (G4D0), G4D1-222, G3D2-413 (highest) could be categorized as contrast genotypes based on SPAD-value and mineral content. The strong positive correlation between Mg and Fe content with SPAD suggested that SPAD is a possible marker for high Mg and Fe content in leaves. Adversely, a weak relationship between SPAD and Zn content in leaves showed that SPAD is a weak marker for Zn content in leaves. Further research related to molecular markers development can be carried out based on these results.

Differential hierarchical metabolites expression of red/white Salacca sumatrana Arillus and its molecular docking studies. Biodiversitas 22: 1014-1024. Salak Sidempuan (Salacca sumatrana) is one of the medicinal plants originating from tropical countries. However, there is very little research on the study of finding bioactive compounds in S. sumatrana using the metabolomic approach. Here, we use two population types of S. sumatrana, e.g., fruit with red and white arillus. We extracted the compounds from the two tissues using the GC-MS technique and further analyzed their antiviral potential using an in-silico molecular docking technique. Fold change analysis showed that in white arillus there were 21 upregulated compounds and 25 downregulated compounds. Metabolites that have high expression in white arillus tissue are ranxinic acid, nicotinic alcohol, stearic acid, 2-furan carboxaldehyde, and others. Six accessions from S. sumatrana based on PCA analysis and separate dendrogram according to the type of arillus, i.e., red and white arillus. Two compounds that act as antivirals found in this study are stearic acid and palmitic acid based on preliminary molecular docking studies. These two compounds could be used for further studies as a preventive measure against SARS-Cov-2 in the future, however, it needs further comprehensive and clinical analysis the future. In conclusion, metabolites in white arillus were distinctly different from red arillus of Snake fruits. In addition, studies on differential metabolite expression on two types of arillus (red/white) can be used as markers in identifying arillus color early by plant breeders.

Molecular identification of Gracilaria species (Gracilariales, Rhodophyta) obtained from the South Coast of Java Island, Indonesia. Biodiversitas 22: 3046-3056. The study of seaweeds diversity and species identification is an important component of marine resource management. However, seaweed identification based on morphological characteristics has several limitations. Besides, DNA barcodes or the partial sequences of cytochrome c oxidase I (COX1) have been proved to identify seaweeds at the species level. To date, Gracilaria species molecular diversity has not been studied in the South Coast of Java Island. Hence, this study aimed to identify the Gracilaria spp. obtained from 6 different beaches along the South Coast of Java, based on the COX1 gene. COX1 gene utilization for identifying and observing the genetic diversity, both intraspecific (genetic variation within species) and interspecific (variations between species) of Gracilaria species in this study has produced good results. A total of 13 seaweed samples collected from beaches in this study were identified as Gracilaria salicornia, G. edulis, G. firma, and G. textorii. The results of genetic diversity analysis conducted using the COX1 gene showed the intraspecific diversity of G. edulis obtained from the beaches of Kondang Merak, Kukup, Nusakambangan, and Karapyak was included in the moderate diversity category. Also, the intraspecific diversity of G. salicornia obtained from the beaches of Kondang Merak, Kukup, and Nusakambangan was included in the moderate diversity category, while the intraspecific diversity of G. textorii from Menganti and Karapyak Beach had no diversity.

Abstract. Antonio MA, Buot Jr. IE. 2021. Characterization and delineation of two infraspecific taxa of Dioscorea esculenta (Lour.) Burkill: The leaf architecture approach. Biodiversitas 22: 1783-1789. The current taxonomic treatment for the wild and cultivated species of Dioscorea esculenta Lour. (Burkill) presented them as two different varieties. However, there are confusions among botanists and farmers. So, leaf architecture analysis was done to investigate whether leaf architecture data would support their current taxonomic treatment. The varieties showed variability in several characteristics indicating their distinct identity from each other. Based on general leaf characters, they differ in all the parameters observed. But on venation characters, evident variation was seen only on the number of basal veins and secondary vein angle of divergence. Nevertheless, the two varieties resemble each other in a number of characters, i.e. campylodromous primary veins, brochidodromous secondary veins, presence of intermarginal and intersecondary veins, admedially ramified tertiary veins, and well-developed areolation. Dendrogram generated from cluster analysis agrees with and supports above leaf architectural examination, suggesting the two varieties to be recognized as separate taxa. But the current taxonomic treatment naming them as var. spinosa and var. fasciculata seemed to have dragged the cultivated plant into the classification system generally used for wild plants. Thus, the proposed name for the cultivated plant is D. esculenta ‘fasciculata’ while that of the wild plant remains as D. esculenta var. spinosa.

The cytotoxic and apoptotic effects of wild and polyploidy genotype of Artemisia cina extracts on the WiDr colon and HTB-183 lung cancer cell lines. Biodiversitas 22: 2844-2852. Artemisia belongs to the Asteraceae family, usually used for traditional treatments of various diseases in China. Artemisia cina Berg ex Poljakov, which lots found in Indonesia, has not been studied much like a cancer drug. The research aims were (i) to compare the bioactive content of hexane and ethyl acetate extract of wild (TWN and KJT) and polyploidy type (J and M) of A. cina and its cytotoxicity on WiDr and HTB-183 cancer cells, (ii) to evaluate the cytotoxicity mechanism of the most substantial extract of it. All of the extracts were prepared by maceration methods using hexane and ethyl acetate separately. The research method used was quantitative with experimental design. The determination of quercetin, kaempferol, and artemisinin used HPLC. The cytotoxicity of the extract determined using the MTT method. The assay of the specific protein related to apoptosis using ICC assay. The content of three bioactive compounds on ethyl acetate extracts higher than hexane extract, in which the extract of J and KJT genotypes had higher levels than M and TWN genotypes. The IC50 value (µg/mL) of extracts against cancer cell lines tested was about 400-700 hexane extracts and 200-500 ethyl acetate extracts. The BCl-2 expressions of both cancer cell lines decreased by treating TWN-EA and M-EA, while the P53 expressions increased. The TWN-EA induced apoptosis of HTB-183 cell lines through caspase-8 and caspase-9 pathways, but M-EA induced caspase-9 pathway only. The TWN and M genotypes were potential to use as anticancer agents on colon and lung cancer.

Cryptobrachytone C is an active compound isolated from leaves of Cryptocarya pulchrinervia, an indigenous plant from Indonesia. Cryptobrachytone C is cytotoxic against P388 leukemia cancer cell lines. A profile of cryptobrachytone C accumulation in leaves based on physiological age is necessary to study cryptobrachytone C production in the plant since it has anticancer potential. In situ profiling of cryptobrachytone C accumulation in leaves was carried out by imaging techniques using matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI). The cryptobrachytone C ionization efficiency was carried out by derivatization using Girard-T reagent and coating with ?-cyano-4-hydroxycinnamic acid (?-CHCA) matrix. Scanned leaves show higher accumulation intensity in the second leaf. Quantitative accumulation profiling was conducted using GC-MS, where the highest amount of cryptobrachytone C was on the second leaf at 87.07 ± 47.21 mg. This showed that the most effective isolation of cryptobrachytone C would be obtained from young leaves of Cryptocarya pulchrinervia. This profile would play a role in cryptobrachytone C production in-situ or ex-situ using tissue culture.

Abstract. Arbiastutie Y, Diba F, Masriani. 2021. Short Communication: Ethnobotanical and ecological studies of medicinal plants in a mangrove forest in Mempawah District, West Kalimantan, Indonesia. Biodiversitas 22: 3164-3170. The mangrove forest Polaria Tanjung Pagar in Mendalok Village, Mempawah District, West Kalimantan, Indonesia, is a mangrove forest maintained by the local community. Currently, this mangrove forest has become one of the tourist attractions, not only visited by local and national, but also foreign tourists. This study aims to identify plants in Polaria Tanjung Pagar mangrove forest Kalimantan with the potential for medicinal uses. The research used a survey method in the mangrove forest by conducting an inventory of the medicinal plants found in the mangrove forest. Observations were made using a transect line with the total area understudy of 0.012 ha and sampling intensity of 0.4%. The investigation was based on cross-checking the traditional use of medicinal plants by the surrounding communities and the scientific literature. The results showed 13 types of medicinal plants (including tree and understorey plants) from 6 different families, i.e., Acrostichum aureum, Aegiceras corniculatum, Avicennia marina, Avicennia alba, Bruguiera parviflora, Bruguiera gymnorrhiza, Bruguiera cylindrica, Ceriops decandra, Rhizophora apiculata, Rhizophora stylosa, Rhizophora mucronata, Xylocarpus moluccensis, and Sonneratia caseolaris. The community structure of the mangrove forest had moderate diversity for tree species and low diversity for understorey plant species, with an H' valued at 2.39 and 0.25, respectively.

Abstract. Serosero RH, Sulistiono, Butet NA, Riani E. 2021. Stock status of coconut crab (Birgus latro) in Daeo, Morotai Island District, North Maluku, Indonesia. Biodiversitas 22: 626-634. The populations of coconut crabs (Birgus latro) in nature continue to decline due to their simple capture for human consumption. This study aimed to determine the status of coconut crab stock in Daeo, Morotai Island Regency, North Maluku, Indonesia including estimations of growth parameters and population size. This research was conducted from October 2016 to September 2017. Growth parameters were estimated based on 12 monthly length-frequency samples. Data analysis was carried out using the ELEFAN I method on the FiSAT II program for estimating growth parameters, and the Schnabel method for estimating population size. The results showed that the level of exploitation of male and female coconut crabs in Daeo was high (E > 0.5). The asymptotic length L? for males was 56.96 mm thoracic length (TL) and that of females 52.50 mm TL. The growth coefficient K for males was 0.14 and that for females 0.16. Length to is -1.026 (male) and -0.913 (female). The results generated using the Schnabel method suggested that the population size of the coconut crab in Daeo was 7,466 to 73,948 individuals considering a confidence interval of 95%. The recapture of tagged coconut crabs was merely 0. 82%.

Hasani Q, Pratiwi NTM, Wardiatno Y, Effendi H, Martin AN, Efendi E, Pirdaus P. 2020. Phytoremediation of Iron (Fe) in Ex-sand Mining Waters by Water Hyacinth (Eichhornia crassipes (Mart.) Solms). The high concentration of iron (Fe) has allegedly caused the water quality of puddles at sand mining area. The poor water quality, in which high contentrations of iron will to a reduction in aquaculture production. This study verified the ability of water hyacinths (Eichhornia crassipes (Mart.) Solms.)) as a phytoremediation agent for iron (Fe) concentration in water at the sand mining area, in East Lampung Regency, Indonesia. The study was carried out with 3 treatments and 3 replication each, including the treatment of (i) 25% water surface coverage, (ii) 50% water surface coverage, and (iii) 75% water surface coverage of water hyacinth. Measurement of Fe concentrations, bioconcentration factor (BCF) and translocation factor (TF) once a week, until Fe concentrations in water are adequate for aquaculture. This was achieved for 21 days. The results showed that the highest percentage reduction of Fe (97.49%) was observed at water hyacinth cover area 50%. The highest value of BCF was 2385.51, while the highest TF was 1.14 in stems and 1.02 in leaves. The results of this study open up opportunities for use water in ex-sand mining areas for aquaculture by the community and its management by the Government of East Lampung Regency, Lampung, Indonesia.

Biodiversitas 22: 696-706.  Botanic gardens contain plant collections for the purpose primarily for ex-situ conservation, yet the public rarely appreciates these collections. As such, assessing the conservation value of botanic gardens collections is very important as an effort to increase public awareness of the importance of botanic gardens and their plant collections. This research aimed to assess the conservation value of plant collection in Bogor Botanic Gardens (BBG), Indonesia using quantitative and qualitative methods. We focused our assessment on medicinal plant collections with the habitus of tree. Conservation value of a plant collection was approached by preservation value and utilization value both quantitatively using monetary units and qualitatively using non-monetary index. Based on these two approaches, quantitatively most (91%) of the medicinal plant collections in BBG assessed in this study have a value of more than one million rupiahs, even 21% have a value of more than 100 million rupiahs, implying the high monetary value of medicinal plant collection in Bogor Botanic Gardens. Meanwhile, qualitatively, the conservation value score of a species will be strongly influenced by the existence of protection laws issued by the government and the cultural level of plants' use by the community. Our framework of assessing the conservation value of plant collections, especially those with medicinal purposes, can be replicated in other botanic gardens.
