Abstract. Baimuratov, A, Bastaubayeva S, Arslan M, Yeraliyeva Z. 2021. Effect of fertilizer application on winter wheat productivity under precision agriculture in Kazakhstan. Biodiversitas 22: 1558-1563. The effect of mineral fertilization of light chestnut soils of Almalybak, Kazakhstan on the three winter wheat varieties grain yields and NDVI was carried out in 2017/18, 2018/2019 as well as 2019/2020 growing seasons using precision agriculture technologies. The experiments were set up in an incomplete factorial design with rates of NPK N90P60K45, N120P90K60, and N150P120K90. Agrochemical cartograms were ordered for the substance of effectively hydrolyzable nitrogen, extractable phosphorus, and exchangeable potassium. The total variation of NDVI was from 0.62 in the minimum (Bezostaya 100 variety, 2017/18) to 0.88 in the maximum (Aliya variety, 2019/20) in the flowering phase of wheat plants. The highest index of NDVI was observed in the N150P120K90 in the three winter wheat varieties for the three seasons. Studies directed in multivariate analyses built up the connection between fertilizer application, soil fruitfulness, and grain yield. As per the genuine heterogeneity of the fields, standards were determined and differential use of the fundamental phosphorus fertilizer was completed. The yield for the winter wheat varieties varied between 5.6-5.9 t/ha for Aliya variety, 6.5-7.0 t/ha for Grom, and 6.2- 6.7 t/ha for Bezostaya 100, which presents that P120 doses were more suitable in experimental plots for winter wheat varieties. The grain yield on mineral fertilization of NPK for the studied varieties varied from 4.2 to 7.6 t/ha for Aliya variety, from 4.0 to 6.8 t/ha for the Bezostaya 100, and from 4.5 to 7.9 t/ha for Grom. The highest grain yield was observed in the N150P120K90 for three winter wheat varieties in all three seasons.

Wood for use in building and industrial materials is generally supplied from plantations and natural forests. Timber demand has been increasing as the population grows, but the availability of timber from natural forests is declining. Therefore, to meet timber needs, wood from community forests is becoming a solution. Community forests are widely planted with fast-growing trees, such as Elaeocarpus angustifolius Blume. It offers the advantages of a short cutting cycle and large-diameter wood. This study aimed to determine the demarcation area between juvenile and mature wood on 6-year-old E. angustifolius based on anatomical, physical, and mechanical properties. The parameters used to determine the demarcation area included the following properties: anatomical (fiber length, cell wall thickness, and microfibril angle [MFA]), physical (moisture content and green density), and mechanical (modulus of elasticity [MOE], modulus of rupture [MOR], and hardness). Those parameters were measured from pith to bark. Data analyses were based on segmented linear regression models using PROC NLIN in SAS. Fiber length, cell wall thickness, green density, MOE, MOR, and hardness values increased from pith to bark. Results from the segmented linear regression models on anatomical properties allowed extrapolating that mature wood would begin at the 18 cm (based on fiber length), the 20 cm (based on cell wall thickness), and the 21 cm (based on MFA) distance from the pith. the proportion of juvenile wood in E. angustifolius was 100%. Fiber length, cell wall thickness, and MFA can be used to determine the transition point of juvenile wood and mature wood in E. angustifolius.

Ethnoecological study on the utilization of plants in Ciletuh-Palabuhanratu Geopark, Sukabumi, West Java, Indonesia. Biodiversitas 22: 661-674.  Plants can provide benefits for community as food, medicine, fuel, building materials, furniture, and others. Plant utilization should be conducted sustainably in a region with high importance of geological, biological, and cultural diversity, such as in Ciletuh-Palabuhanratu Geopark, Sukabumi, West Java, Indonesia. This study aimed to reveal: (i) plant utilization by local community in Ciletuh-Palabuhanratu Geopark based on land cover; and (ii) community dependence on the plants being utilized. Method used in this study was mixed of qualitative and quantitative methods using direct observation, semi-structured and structured interviews data collection techniques. The result of study showed that the community in Ciletuh-Palabuhanratu Geopark still uses plants for food (80 species), medicinal plants (73 species), livestock fodder (9 species), firewood (27 species), building materials, furniture (38 species), and others (23 species). The highest plant utilization came from gardens (131 species), home gardens (99 species), natural forests (77 species), dry-paddy fields, and wet-paddy fields (7 species). Generally, various agroecosystems and also forest in Ciletuh-Palabuhanratu Geopark have an important role in providing plant products to fulfill the daily needs of rural people.

Spenochlea zeylanica and Ludwigia octovalvis are dominant in Trimurjo paddy fields in Central Lampung Regency, Lampung, Indonesia and difficult to control with 2,4-Dichlorophenoxyacetic acid (2,4-D) and metsulfuron-methyl herbicides that had been used for more than 12 years. This study aims to determine the resistance status of S. zeylanica and L. octovalvis and physiological activity in resistance and susceptible of both weeds species exposed to  2,4-D and metsulfuron-methyl herbicides. The study was conducted from August to December 2019 in the Greenhouse Botanical Garden of Sumatra Institute of Technology. The study used a split-plot design, the origin of weed species as the main plot and herbicide dose as the subplot. S. zeylanica exposed has low-level resistance to 2,4-D and metsulfuron-methyl herbicides with resistance Index (RI) of 2.19 and 2.87, respectively. L. octovalvis exposed was still classified as susceptible to 2,4-D and metsulfuron-methyl herbicides with RI of 1.00 and 1.59, respectively. The physiological activities (carbon assimilation, stomatal conductance, and transpiration rate) of S. zeylanica resistant to 2,4-D and metsulfuron-methyl herbicides is higher than S. zeylanica susceptible 2,4-D and metsulfuron-methyl herbicides

Abstract. Badiaraja PH, Zubaidah S, Kuswantoro H. 2021. Maternal effect of agronomic and morphological characters on cluster structure of F3 soybean lines. Biodiversitas 22: 969-982. The study was to determine the maternal effect on the agronomic and morphological characters and their impact on F3 soybean lines cluster structure. The results showed the maternal effect was found in agronomic and morphological characters. In agronomic characters, the maternal effect was shown by plant height, number of productive nodes, number of filled pods, seed weight per plant, and 100-seed weight. In morphological characters, pod width, pod thickness, seed width, and seed thickness were shown by pod width. Based on cluster analysis results, at Grobogan/UM 3-2 cross, the best individuals for the number of branches, the number of productive nodes, the number of filled pods, the number of unfilled pods, the number of seeds per plant, and the seed yield per plant were obtained in Cluster 2. For the 100-seed weight, pod length, pod width, pod thickness, seed width, and seed thickness, the best individuals were in Cluster 1. Meanwhile, at UM 3-2/Grobogan cross, the best individuals for the plant height were in Cluster 2, while seed length was in Cluster 3. For the number of seeds per pod, the best individuals can be obtained from Cluster 6. The maternal-effect led to the cluster structure differed between cross and its reciprocal.

Involvement of purple acid phosphatase gene into nitrogen uptake of oil palm (Elaeis guineensis). Biodiversitas 22: 1385-1390. Nitrogen is the most important nutrient element in terms of plant growth. Plant purple acid phosphatases (PAPs) are known to participate in the phosphate (Pi) acquisition and utilization. Moreover, PAP gene plays an important role in nitrogen fixation. A single nucleotide polymorphism (SNP) was previously detected in the exon 7 of EgPAP3, based on SNP mining analysis of oil palm (Elaeis guineensis Jacq.). genome database. This study was aimed to obtain a Cleaved Amplified Polymorphic Sequences (CAPS) marker based on SNP within EgPAP associated with efficient nitrogen uptake oil palm progenies. Primer pairs were designed and used for PCR amplification of 3 oil palm progenies that showed low N-content, 3 progenies with moderate N-content, and 3 progenies with high N-content. The amplicon was purified prior to single-pass DNA sequencing analysis. Based on Pearson’s chi-square and odds ratio statistical analysis, the SNP has strong positive correlation with the phenotype. The SNP is located at chromosome 13 with a distance of 17.7771 cM from start codon. The sequencing analysis revealed that three progenies with high N-content samples had GG allele motif, while moderate N-content progenies had GA allele and low N-content progenies had AA allele motifs respectively. In addition, a restriction site of NIaIV was found to be adjacent to the SNP location, thus the PCR products of all samples were digested with NIaIV restriction enzyme. NIaIV was able to distinguish between high, medium and low efficient DNA samples. Whole high N-content progenies with GG allele motifs were undigested indicating a single band size of 670 bp identical to the untreated PCR product (control). Moderate N-content progenies produced a 670 bp, 550 bp, and 120 bp bands because of digested by NIaIV. Low N-content progenies also resulted in double bands of 550 bp and 120 bp due to digested by NIaIV. Furthermore, NIaIV restriction enzyme was applied to digest other 54 oil palms DNA samples with unknown genotypes. Whole GG samples were consistently shown to have single band, GA and AA samples were also consistent in producing two bands with different lengths. Based on this result, CAPS marker based on SNP in EgPAP3 was successfully developed to screen between high and low efficient N-uptake of oil palm progenies.

Abstract. Hartanti AT, Susanti FN, Prasasty VD, Radiastuti N. 2021. Culturable endophytic fungal diversity in cassava tubers of Indonesia. Biodiversitas 22: 1250-1260. Fungal endophytes are found in plant tissue and do not harm the host plant. Cassava (Manihot esculenta Crantz) is the third staple food besides rice and corn. This study aimed to identify the diversity of endophytic fungi in the cassava from several regions in Indonesia, including Bogor, Lembang, Makasar, Serang, Singaraja (Buleleng), Subang, Wonogiri, and Wonosobo. The endophytic fungi were isolated and were molecularly characterized using the internal transcribed spacer (ITS) rDNA sequence (ITS4 and ITS5). Phylogenetic analysis was performed by using the MEGA 7 with a bootstrap value of 1000 repetitions. All 14 isolates of endophytic fungi were isolated from different regions. Of these six isolates belonged to genus Fusarium (F. solani FS5, F. lichenicola FS2, F. lichenicola FS4, F oxysporum FS1, F. falciforme FS3, F. falciforme FS6), and four to the genus Aspergillus (Aspergillus sp. FS7, Aspergillus sp. FS8, Aspergillus sp. FS9 and A. fumigatus FS10). In addition, Nectria pseudotrichia, Lasiodiplodia sp., Penicillium citrinum, and Schizophyllum commune were also isolated from cassava tubers. In conclusion, it suggested that diverse endophytic fungi colonize tubers of cassava growing in Indonesia.

DNA barcode of Enggano hill myna, Gracula religiosa enganensis (Aves: Sturnidae) based on mitochondrial DNA cytochrome oxidase subunit I. Biodiversitas 22: 1635-1643. The sharp decline of the Enggano hill myna population due to illegal trading and habitat degradation needs to be our concern to prevent this bird from extinction. Taxonomically, Enggano hill myna is referred to as a sub-species, but this has not been confirmed by genetic data. We have sequenced seventeen Enggano hill myna mitochondrial DNA COI genes to describe their genetic identity (barcode), genetic distances, and phylogeny. DNA genome from seventeen blood samples was isolated with DNeasy® Blood and Tissue Kit Qiagen, while PCR amplification was performed using a pair primers, namely COIGRF (5'-TTCTGATTCTTTGGCCATCC-3') and COIGRR (5'-GTTGGAAGGCTTTGCGTTTA-3'). We used Clustal W alignment in MEGA 10.2.2 software to search single nucleotide polymorphisms. Genetic distance was analyzed by using the Kimura 2-parameter, and the phylogenetic tree was reconstructed with Neighbor-Joining models. We found 98.60% conservative sites, 0.69% parsimony sites, and 0.83% singleton sites from the 716 bp sequence. The highest nucleotide composition was cytosine (32.20%), and the lowest was guanine (16.80%), followed by 49% GC content. Seven SNP sites were found in 716 bp COI gene sequences of seventeen individuals. The genetic distance between Enggano hill myna individuals was ranged from 0.0-0.8%, and all Enggano hill myna individuals separated from Chinese and Indian populations in the phylogenetic tree with a genetic distance of 0.9% and 1.1%. Our data suggest that the Enggano hill myna population is still classified as a sub-species. The COI gene sequences that we found can be used to quickly identify this species and are also important to prevent illegal trading in Indonesia.

Abstract. Hartati S, Samanhudi, Manurung IR, Cahyono O. 2021. Morphological characteristics of Phaius spp. orchids from Indonesia. Biodiversitas 22: 1991-1995. Orchid is one of the ornamental plants with great economic value in various communities. Characterization and grouping of several orchid species can be used in breeding efforts to obtain hybrid with the desired traits. This study is aimed to characterize the morphological appearance of Phaius orchids and to cluster the species based on their similarity in morphological characteristics. Five species of Phaius (P. tankervilleae, P. indigoferus, P. montanus, P. amboinensis, and P. callosus) of the collection of the Bogor Botanical Garden were observed based on 30 morphological characters including the characters of stems, pseudobulbs, leaves and flowers. The data obtained were converted into binary data and computed using the Numerical Taxonomy and Multivariate Analysis System (NTSYS) Spc 2.02 program to produce dendrogram. The results show that based on the morphological characterization of the five species of Phaius spp., they have similarities from 63% to 80%. Among the five Phaius species, P. amboinensis, P. tankervilleae, and P. montanus had 80% similarity and can be used as parents for crossing.

Effect of single and mixed inoculation of arbuscular mycorrhizal fungi and phosphorus fertilizer application on corn growth in calcareous soil. Biodiversitas 22: 1920-1926. Low availability of soil phosphorus (P) in calcareous soil in West Timor has become a constraint in improving plant productivity. A field experiment was carried out in the calcareous soil to investigate the effect of single and mixed indigenous arbuscular mycorrhizal fungi (AMF) inoculation and P fertilizer application on growth and yield of corn in Kupang District, East Nusa Tenggara Timur Province, Indonesia. The experiment was laid out in a split-plot design consisting of indigenous AMF inoculation as the main plots and inorganic P fertilizer rates as the sub-plot treatments. The main plot treatments were: without AMF inoculation (M0), inoculated with single AMF inoculum (M1) and inoculated with mixed AMF inocula (M2). Sub-plots consisted of four P rates namely 25% (P1), 50% (P2), 75% (P3) and 100% (P4) of the recommended inorganic superphosphate dose of 200 kg SP36 ha-1. The results showed that there were significant interactions between AMF inoculation and P fertilizer dose on AMF spore density, AMF colonization and soil available P. Results indicated a strong correlation between AMF colonization and soil available P. Inoculation of both M1 and M2 significantly increased soil available P, plant growth and yield compared to the uninoculated plant. Spore production in the two AM responded differently to P fertilizer. Low doses of inorganic P fertilizer favored spore production in M1, whereas P fertilizer sporulation in M2 was adversely affected. The percentage of AMF root colonization of M1 and M2 decreased with an increase in P fertilizer applied. Further studies need to be conducted to evaluate the effectiveness of indigenous AMF in improving plant yield at low level of inorganic fertilizer application to achieve more sustainable agricultural practice in the region.
