Abstract. Habsyah TS, Basyuni M, Siregar LAM, Afandi D, Syahputra I. 2021. Distribution of polyprenol and dolichol in oil palm genotype (Elaeis guineensis) involving lipase activity. Biodiversitas 22: 830-837. The aim of this study was to determine the pattern of chemical compounds from the polyisoprenoid group in oil palm plants that were contributing to the lipase activity. The study was conducted in two locations. A sampling of leaves and fruit mesocarp from 90 progenies was conducted at PT. Socfin Indonesia Aek Loba Plantation, Asahan District, North Sumatra, Indonesia. Biochemical analysis of polyisoprenoid was carried out at the Forest Biotechnology Laboratory, Faculty of Forestry, University of Sumatera Utara, Medan. The results showed that the total lipids in leaves tissue ranged from 0.40 to 1.032 mg/g, with the lowest total lipid found in SL-4966 progeny and the highest total lipid found in SL-9737 progeny. Polyisoprenoid values ??ranged from 1.63 to 10.82 mg/g dry weight. The highest polyisoprenoid content was obtained in SL-10682 progeny, and the lowest was in SL-5045 progeny leaf tissue. The polyisoprenoid ??in fruit mesocarp tissue ranged from 1.79 to 4.14 mg/g. The lowest polyisoprenoid content (1.79 mg/g) of fruit mesocarp was found in SL-4233 progeny, while the highest (4.14 mg/g) was found in SL-5010 progeny.  Dolichol and polyprenol are equally dominant in palm leaves.  Carbon polyprenol chain lengths ranged from C45-C105, and dolichol ranged from C50-C110. The polyprenol carbon chain lengths in the mesocarp tissues were C65-C120 and dolichol was C65-C125. Polyisoprenoid analysis on oil palm leaf and fruit mesocarp tissue showed that lipid patterns of oil palm leaf and mesocarp were categorized as type II lipid patterns. It provides important information for selecting genotypes of low lipase activity of oil palm plants.

Abstract. Wibowo WA, Maryanto SD, Daryono BS. 2021. Phenotypic characters and identification CYPs (Cyclophilin) gene in Cucumis melo L. cv. Gama Melon Parfum. Biodiversitas 22: 3007-3014. Cucumis melo L. cv. Gama Melon Parfum is a new cultivar with a very strong fragrance as its main character. As a new cultivar with unique characters, it is necessary to characterize phenotype and molecular related to the fragrant aroma. The research aimed to study the phenotypic of the fruit of C. melo cv. Gama Melon Parfum (F3) and to identify the CYPs gene as one of the genes that act in encoding volatile compounds. Analysis of qualitative characters was based on International Plant Genetic Resources Institute (IPGRI) and Plant Variety Protection (PVP), phenotypic characters in melons observed by abiding the Rules for Registration of Varieties from the Indonesian Minister of Agriculture Decree No. 700/Kpts/OT.320/D/12/2011, while analysis of quantitative characters was using the ANOVA methods and software PKBT-STAT-2. The methods of molecular characterization included RNA isolation, cDNA synthesis used Reverse Transcriptase-PCR, amplification of DNA target used PCR, visualization of DNA target used electrophoresis, and DNA sequencing. Analysis of the in silico approach was carried out on the CuGenDB Melon database. Analysis of protein sequences and classification was obtained from InterPro. Phylogeny analysis using MEGA-X Software. The results were 18 qualitative characters and 11 quantitative characters were stable and uniform, whereas the molecular characterization of the genes was predicted Cyclophilin with peptidylprolyl isomerase (PPIase) activity and located in chromosome 1 (17059021-17058899).

Abstract. Pratomo B, Lisnawita, Nisa TC, Basyuni M. 2021. Short Communication: Digital identification approach to characterize Hevea brasiliensis leaves. Biodiversitas 22: 1006-1013. The purity of clones is a key factor to obtain uniform growth of Hevea brasiliensis Muell Arg in the field. Crop uniformity has an impact on production patterns that can be predicted in latex content. Basically, H. brasiliensis tree leaves show similarities to each other such as the color, texture, or shape of the leaves. Identification of clones that have been very common by the morphology of H. brasiliensis leaves. This study links the results of previous research on digital methods used. A total of 27 clones were sampled in this study which came from the garden collection of entres, which can be used as a source of elders for assisted pollination or source of planting materials for the needs of production-scale seeds in the field. Here, we used a database of 540 leaves from 27 clones and 10 quantitative characters to address these issues The analysis data used a randomized complete block design (RCBD) factorial. The result showed the identification method digitally on characterizing of leaves by using Software Adobe Photoshop, depicting that the characteristics contained in H. brasiliensis leaves are very proper to be used as the first character in identifying an H. brasiliensis clone. The present work provided a piece of important information on the characterization of H. brasiliensis leaves to enhance the selection of parent clones.

Breed and age effects on concentration of adiponectin and reproductive performance in Anglo Nubian, Etawah grade and its crossbred bucks. Biodiversitas 22: 1112-1119. The purpose of this study was to evaluate the effect of differences in breed and age on reproductive performance and adiponectin and testosterone production in Anglo Nubian, Etawah grade, and crossbred (Anpera) bucks. A total of 12 bucks with four individuals of each breed were used. This study collected five data points from each buck regarding adiponectin, testosterone, and reproductive performance (libido and semen characteristics). Data were analyzed with factorial analysis of variance and Duncan's test. The result shows that adiponectin concentration between breeds was significantly different (P <0.01). There were also significant differences (P<0.05) in adiponectin concentrations based on buck age. There were also significant interactions with adiponectin concentrations (P <0.01). Furthermore, testosterone concentrations showed significant differences based on breed (P <0.05) and age (P <0.05). There were also significant age-breed interactions affecting testosterone concentrations (P <0.01). Libido and semen characteristics had no significant differences based on breed and age group, and no significant age-breed interactions (P> 0.05). The heterosis effect on adiponectin concentration (48.05%), testosterone concentration (27.68%), libido (-0.61%), semen volume (-1.93%), sperm motility (0.49%), sperm normal morphology (0.18%), and sperm concentration (0.00%) was measured. In conclusion, there is a significant effect of breed, age, and age-breed interactions on the concentration of adiponectin and testosterone in bucks, and both of these variables have a high heterosis effect on crossbred bucks.

Biodiversitas 22: 1750-1756. Lowland forest in Java Island Indonesia is dominated by teak forest. However, for some decades, the forest has degraded and decreased teak wood production. Rehabilitation of the degraded forest is needed to accelerate the forest land cover. Vegetative propagation of selected clones is one of the techniques used in the asexual reproduction of plants to maintain superior genetics from ancestors to their offspring. In addition, adding organic fertilizer can improve the chemical and physical properties of degraded sites. In this study, vegetative propagation techniques of selected teak clones and addition of organic fertilizer were used to rehabilitate the degraded forest. The objectives of the research were to determine the best propagation technique and appropriate dosage of organic fertilizer toward the early growth of clonal teak plantations. The results showed that the height and diameter parameters did not significantly differ for vegetative propagation and interaction with organic fertilizer dosage (P > 0.05). However, the dosage of organic fertilizer significantly increased the DBH of the teak (P < 0.05), although the height parameter did not significantly differ among dosages of organic fertilizer (P > 0.05). The best dosage of organic fertilizer was 5 kg/individual, under which the average growth in height and diameter of teak at 14 months was 3.79 and 5.60 cm, respectively. This suggests that vegetative propagation of selected teak clones and addition of 5 kg/individual organic fertilizer could be implemented to support the successful rehabilitation of degraded forests.

Abstract. Pin TG, Supriatna J, Takarina ND, Tambunan RP. 2021. Mangrove diversity and suitability assessments for ecotourism in Cimalaya Wetan Coast, Karawang District, Indonesia. Biodiversitas 22: 803-810. Mangrove forest has been a popular ecotourism destination recently. One of the methods to assess the mangrove potential for ecotourism is by using Tourism Suitability Index (TSI). This study was aimed to assess the ecotourism potentials of mangrove forest sizing of 56.43 hectares in Cilamaya Wetan Sub-district, Karawang District, West Java Province, Indonesia using the TSI method. To assess the mangrove variables, 10 sampling stations have been selected across the coast. In each station, 10 sample plots sizing of 10 m x 10 m have been established. The TSI was measured based on mangrove variables including species diversity, density, and thickness. We recorded six mangrove plant species belonging to three plant families. Avicennia alba, Bruguiera gymnorrhiza and Rhizophora stylosa, were common species found along the coast. Shannon-Wiener index (H’) showed that the west and east parts of Cimalaya Wetan had high mangrove diversity compared to the central part. The TSI analysis showed that the most suitable areas of mangrove forests for ecotourism were 32.18 hectares (57.11% of the total area), mainly located in the east part of the coast. Moreover, the moderate and least suitable areas were 19.12 hectares (33.94%) and 5.03 hectares (8.93%), respectively, and located in the central part of the coast.

Abstract. Karyawati AS, Puspitaningrum ESV. 2021. Correlation and path analysis for agronomic traits contributing to yield in 30 genotypes of soybean. Biodiversitas 22: 1146-1151. To obtain more productive soybean genotypes, it is essential to know the contribution of each agronomic trait to the productivity of soybeans. The purpose of this study was to analyze the correlation and path analysis in thirty genotypes of soybeans (Glycine max (L.) Merrill) for eleven traits. The study was conducted in March-June 2019, in an experimental area located at Agro Techno Park, which belongs to Universitas Brawijaya, Malang, East Java Province, Indonesia. The experimental design was a randomized complete block design with three replications. Analysis of correlation showed that the number of branches, number of filled pods, number of seeds per plant, weight of 100 seeds, and weight of seeds per plant had a positive correlation value to the yield. Path analysis showed that the number of seeds per plant had a high positive, direct effect on the yield. The plant height, number of branches, fresh weight and dry weight had an indirect effect on the yield. Based on the result, the number of seeds per plant can be selected as one of the selection criteria in a soybean breeding program to obtain a high-yield soybean variety.

Abstract. Kurnia I, Arief H, Mardiastuti A, Hermawan R. 2021. The potential of bird diversity in the urban landscape for birdwatching in Java, Indonesia. Biodiversitas 22: 1701-1711. Birdwatching is a recreational activity of observing birds in the wild with the naked eye¸ using tools such as telescopes and binoculars, or listening to bird sounds. The observation locations can be either natural landscape or urban landscapes with bird diversity. However, the dominance of built spaces and man-made vegetation differed from natural landscapes. This difference will affect the composition of birds found in the urban landscape. This paper aimed to analyze bird diversity in urban landscapes and their potential for birdwatching. The research was conducted from February to April 2020 in five cities in Java (Bogor, Sukabumi, Bandung, Yogyakarta, Surabaya). Bird data were taken using the IPA method and were carried out in 85 green open spaces. Total bird species found were 75 species, with the largest number found in Bogor (66 species), followed by Surabaya (36 species), Bandung (28 species), Sukabumi (26 species), and Yogyakarta (19 species). The locations with the highest species richness in each city are Bogor Botanical Gardens (53 species), Cikundul Agrotourism Area Sukabumi (18 species), Bandung Zoological Garden (21 species), Gembira Loka Zoo (14 species), and Pakal City Forest Surabaya (20 species). Commonly seen bird species are dominated by generalist species typical of urban landscapes (e.g., Collocalia linchi, Passer montanus, and Pycnonotus aurigaster). Bird species richness has a positive correlation with the local area and habitat diversity. A total of 74 bird species are resident species that can be found throughout the year as birdwatching objects. Only one species are migrant species namely Merops philippinus.

Orchid (Orchidaceae) is one of the largest flowering plant families comprising more than 25.000 species. Knowledge of wild orchid diversity and their habitat preferences is useful for its conservation efforts. Research of wild orchid diversity in Indonesia has been done before, however, there is still few information about phenetic analysis and habitat preferences of wild orchids in Indonesia particularly in Gunung Gajah, Purworejo, Indonesia. Therefore, our research aimed to study species diversity and phenetic relationship of wild orchids and their habitat preferences in Gunung Gajah, Purworejo. Exploration was done using purposive sampling method. Phenetic analysis was carried out using MVSP 3.1, implemented by clustering method to understand phenetic relationships and ordination by PCA method to reveal morpho-anatomical characters which determine the clustering pattern. Thirteen orchid species of subfamily Vanilloideae, Orchidoideae, and Epidendroideae were found. Two clusters were formed on dendrogram and each of them was separated into two sub-clusters. PCA analysis based on stem, leaf, and flower characters showed different clusters compared to dendrogram. Habitat preferences of wild orchids in Gunung Gajah are alkaline soil, low soil moisture, air temperature 24-29°C, and low to high light intensity. Epiphytic orchids preferences on their host tree are dominated by median and upper part of main stem.

Abstract. Awang-Jamil Z, Aminuddin MF, Zaidi BQ, Basri AM, Ahmad N, Taha H. 2021. Phytochemicals and antimicrobial analysis of selected medicinal plants from Brunei Darussalam. Biodiversitas 22: 601-606. Medicinal plants have been widely used but their pharmacological properties are yet to be fully explored. This study was aimed to determine the phytochemicals and their antimicrobial activity of four selected medicinal plants: Melastoma malabathricum, Merremia borneensis, Pandanus amaryllifolius, and Senna alata from Brunei Darussalam. Phytochemical constituents were evaluated by using gas chromatography-mass spectrometry (GC-MS) and antimicrobial screening was carried out using agar well diffusion method. A number of phytochemicals were identified in the plant extracts, of which the following compounds were present in higher amounts (>10 %): coumaran, hentriacontane, hexatriacontane, 3-hydroxy benzyl alcohol, ?-linolenic acid, palmitic acid, catechol, squalene, and ?-tocopherol-?-D-mannoside. A concentration-dependent antimicrobial activity was observed in some extracts against Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Saccharomyces cerevisiae. Results of this study showed medicinal potential of the four selected plants.
