Abstract. Zulfahmi, Parjanto, Purwanto E, Yunus A. 2021. Genetic diversity and population structure of Eurycoma apiculata in Eastern Sumatra, Indonesia. Biodiversitas 22: 4431-4439. Information on genetic variation within and among populations of Eurycoma apiculata plants is important to develop strategies for their conservation, sustainable use, and genetic improvement. To date, no information on genetic variation within and among populations of the E. apiculata has been reported. This study aims to assess genetic diversity within and among populations of E. apiculata based on RAPD markers, and to determine populations to collect E. apiculata genetic material for conservation and breeding programs. Young leaves of E. apiculata were collected from six natural populations. Fifteen RAPD primers were used to assess the genetic diversity of each population. The data obtained were analyzed with POPGEN and Arlequin software. The amplification results of 15 selected primers produced 3-16 loci with all primers 100% polymorphic. At the species level, the mean allele per locus (Na), number of effective alleles (Ne), percentage of polymorphic loci (PPL), Nei’s gene diversity index (He) and Shannon information index (I) were 2.000, 1.244, 100%, 0.167, and 0.286, respectively. At the population level, the mean values for Na, Ne, PPL, He and I were 1.393, 1.312, 39.27%, 0.119, and 0.186, respectively. The highest value of gene diversity within population (He) was found in the Lingga-1 population and the lowest value was found in the Rumbio population. The value of genetic differentiation among populations (GST) of E. apiculata is 0.284, consistent with the results of the AMOVA analysis which found that genetic variation among populations was 23.14%, indicates that the genetic variation of E. apiculata was more stored within populations than among populations. The gene flow (Nm) value of E. apiculata was 1.259 migrants per generation among populations. The Nm value of this species was high category, and could inhibit genetic differentiation among populations. The clustering of E. apiculata population based on the UPGMA dendrogram and PCA was inconsistent with its geographic distribution, reflecting the possibility that genes migration occurred between islands in the past. The main finding of this study was the genetic variation of the E. apiculata mostly stored within the population. Therefore, the population with the highest genetic diversity is a priority for in-situ conservation, and collection of E. apiculata genetic material for ex-situ conservation and breeding programs should be carried out minimum from Lingga-1 and Pokomo populations.

Abstract. Pratiwi RA, Nurlaeni Y. 2021. The potency of Myrtaceae Family from Cibodas Botanic Gardens (Cianjur, Indonesia) as botanical pesticide. Biodiversitas 22: 4648-4664. Cibodas Botanic Gardens (CBG) is a biodiversity reservoir that can be explored for the discovery of new candidates for botanical pesticides. Myrtaceae has been reported to provide biological activity against pests or pathogens due to their essential oil contents. This research was conducted to inventory and categorize Myrtaceae collection in CBG that had the potency to be botanical pesticide. The list of Myrtaceae collection of CBG obtained from Unit Registration and Collection CBG per 2021. Database fulfilling regarding the bactericide/fungicide/herbicide/insecticide potency of the Myrtaceae of CBG’s collection was carried out through digital references search. Our result showed that there were 73 species of Myrtaceae (from 18 genera) that are potential to be botanical pesticide sources. There were 17 species that are considerably had a high potency. Most of them belong to the Eucalyptus and Melaleuca, followed by Backhousia, Leptospermum, Psidium, and Syzygium. The data resulted from this study is expected to serve as baseline information for further research about the formulation, efficacy, and conservation management of botanical pesticides from Myrtaceae for sustainable use. Furthermore, the development of biological pesticides is a step to improve the quality of Indonesian export products so as to increase national competitiveness in the globalization era nowadays.

Abstract. Truong DD. 2021. Estimating residents' willingness to pay for wetland conservation using contingent valuation: the case of Van Long Ramsar Protected Area, Vietnam. Biodiversitas 22: 4784-4793. Willingness to pay (WTP) for wetland conservation is an important basis for designing market-based wetland protection strategies and sustainable wetland management. The main objective of this study is to estimate villagers' WTP for wetland conservation in Van Long Wetland Protected Area, Vietnam, and analyze factors influencing WTP. A questionnaire survey based on the dichotomous contingent valuation method (CVM) was conducted at seven communes surrounding Van Long Ramsar Protected Area (VLPA). The results showed that the local people in the studied area appreciate the roles of the wetland in preserving landscape values, supporting livelihoods, and transmitting values ??to future generations. The value of biodiversity conservation and the value of water filtration and regulation are perceived to a lesser extent in terms of the importance of conservation. Of the 384 respondents, 96% are WTP for wetland conservation at different levels. With the parametric model, the average value of WTP ranges from 300,000 to VND 328,000 VND /family/year. In the non-parametric model, the average of WTP is from 338,000 to 359,000 VND/family/year. The probability of environmental response' that willing to pay for conservation is closely related to their awareness, family income and payment amount. The results also showed optimistic points that the local people are willing to contribute to improving wetland quality. In a general sense, the results of this study make good contributions to the literature related to WTP for wetland conservation in developing countries.

Biodiversitas 22: 5425- 5431. Research on the development of black rice has not received much attention. The anther culture technique can shorten the plant breeding process and produce doubled haploid (DH) lines. The target of this study was to evaluate DH black rice lines with early maturity, good agronomic performance, and high yields. Twenty-seven doubled haploid lines and three check varieties (Aek Sibundong, Inpari 24, and Jeliteng) were evaluated at two locations (Kediri, East Java, and Bogor, West Java) from January to July 2020. A randomized complete block design was applied to this study with three replicates. The combined analysis results showed that the genotype effect (G) was highly significant (p < 0.01) for all characters. In addition, all characters had a high broad-sense heritability. Characters that were positively correlated with yield based on genotypic and phenotypic correlation were the number of productive tillers, the number of vegetative tillers, panicle length, the number of filled grains, and the number of total grains. The 1000-grains weight and panicle length had a positive and the largest direct effect on yield. Principal component analysis showed that the number of productive tillers and the 1000-grains weight were related to yield. The selection index model that can be formulated is as follows I = (3*0.375*Yield) + (0.723* 1000-grains weight) + (0.280* Number of productive tillers). Based on the selection index, 14 genotypes were selected.

Ghimire S, Koju NP. 2021. Short Communication: Fish diversity and its relationship with environmental variables in Kamala River, Nepal. Biodiversitas 22: 4865-4871. Kamala River originates from the lower part of the Mahabharat range and flows through inner to outer Terai, providing a broad range of ecosystem services; provisioning, regulating, supporting, and cultural. However, the information regarding the environmental factors and species diversity in the river is not available enough. Hence the study aimed to explore baseline information on the diversity and abundance of fish and their relation to environmental variables. The fish diversity was assessed in five sections of Kamala River and correlated with different environmental variables. The study was carried out during September- October 2019, marking the post-monsoon season of Nepal. Fish samples were collected using cast nets, and physiochemical parameters were analysed onsite and in the laboratory. Altogether 19 freshwater fish species belonging to 5 orders, 8 families, and 15 genera were recorded. Cyprinids were the most dominant in the river, while Channids, Mastacembelids, Botiids, Sisorids, Gobiids were represented less. The Redundancy Analysis (RDA) ordination method revealed that species variation was correlated with temperature, Electrical Conductivity (EC), Total Dissolved Solids (TDS), and nitrate. Nonetheless, different river sections were disturbed due to mining, deforestation, and construction activities, which could pose a real threat to fish diversity and population, and other aquatic organisms.

Sukmarani D, Proklamasiningsih E, Susanto AH, Ardli ER, SudianaE, Yani E. 2021. Superoxide dismutase (SOD) activity of Ceriops zippelianaBlume in Segara Anakan Cilacap (Indonesia) under heavy metal accumulation. Biodiversitas 22:5627-5635. Ceriops zippeliana Blumeis a true mangrove species that occupy habitats exposed to environmental stress, such as salinity and heavy metal contamination, which is the case in Segara Anakan Cilacap, Central Java. Both salinity and heavy metal stress can lead to the production of reactive oxygen species (ROS), which can be detrimental to plants if they are present in excess. Plants have defense mechanisms to prevent excessive ROS, one of which is by the use of superoxide dismutase (SOD) enzyme. This study aims to explain the accumulation of heavy metals (Pb, Cd, Cu, and Zn) in C. zippeliana as well as the translocation of the metals. In addition, the correlation between heavy metal concentrations and SOD activity in C. zippeliana is reported. Plant samples were randomly collected from Segara Anakan, Cilacap. The analysis was conducted using the bio-concentration factor (BCF), translocation factor (TF), and linear regression in the SPSS program. It was found that Pb, Cu, and Zn levels in Segara Anakan Cilacap sediments were still within acceptable limits, while Cd level was moderately polluting. Metal accumulation was higher in the branches of C. zippeliana compared to those in roots and leaves. The SOD activity of C. zippeliana in Segara Anakan is seemingly not related to metal contents in plant parts.

Short Communication: PCR detection of Avibacterium paragallinarum from layers with infectious coryza symptoms in poultry farms of Sleman District, Indonesia. Biodiversitas 22: 4890-4894. Infectious coryza (IC), caused by Avibacterium paragallinarum, is a contagious and infectious respiratory tract disease that affects the commercial poultry industry. Molecular techniques, such as species-specific PCR, HPG-2 PCR are mostly used for the detection of  A. paragallinarum. The current research was carried out to isolate A. paragallinarum from the layers of infectious coryza signs in Sleman District, special region of Yogyakarta, followed by PCR confirmation of the identified bacteria. Nine field isolates were observed and determined based on their colony and cell morphology. All isolates were characterized biochemically and confirmed with species-specific HPG-2 PCR for A. paragallinarum. Out of 9 isolates, 6 (66.7%) isolates were biochemically identified as A. paragallinarum and confirmed by HPG-2 PCR.

Aeromonas spp. is known to be pathogenic to freshwater fish, including catfish. Antibiotics are often used to overcome bacterial attacks. However, the indiscriminate use of antibiotics, especially at incorrect doses and frequencies, could result in the emergence of resistant bacteria. The presence of resistance genes is also suspected to trigger the resistance potential of Aeromonas sp. against antibiotics. The objective of this study is to characterize the resistance genes of Aeromonas spp. isolated from diseased walking catfish (Clarias sp.). Ten Aeromonas spp. were isolated from infected walking catfish cultivated in Java. The resistance genes assessed included tetA, strA-strB, and qnrA. The antibiotics tested consisted of ampicillin (AMP), erythromycin (ERY), oxytetracycline (OXT), kanamycin (KAN), enrofloxacin (ENRO), and chloramphenicol (CHL). Results showed that some Aeromonas spp. had at least one out of the three genes assessed (i.e., tetA and strA-strB) but none had the qnrA gene. Aeromonas spp. were sensitive to KAN, ENRO, and CHL but resistant to AMP, ERY, and OXT. Thus, KAN, ENRO, and CHL are effective antibiotics for treating fish infected with Aeromonas spp.

Biodiversitas 22: 5169-5180. Cellulolytic actinobacteria are cellulase-producing bacteria capable of degrading cellulose. This study aimed to isolate, characterize, evaluate the cellulolytic ability, and to determine physiological characterization of soil cellulolytic actinobacteria isolated from the Ujung Pancu area, Aceh Besar. Isolation of actinobacteria from soil samples was performed using serial dilution method on Yeast Malt Agar (YMA) medium. Morphological characterization was carried out by growing isolates on YMA, Oatmeal Agar (OA), and Yeast Starch Agar (YSA) media. Cellulolytic ability was determined by calculating the cellulolytic index (IS) on 1% carboxymethyl cellulose (CMC) medium after adding 0.1% congo red solution. Physiological characterization of cellulolytic actinobacteria tested in this study was salinity, pH, and carbon source in liquid Yeast Malt (liquid YM), and the growth was measured at a wavelength of 581nm. The results showed that a total of nine isolates of actinobacteria were isolated, which belonged to the genus Streptomyces. Cellulolytic test results showed that eight isolates had the ability to degrade cellulose. Isolates AUP-04, AUP-03, and AUP-01 had the highest cellulolytic index value. Physiological characterization results revealed that three isolates had different tolerances for salinity levels, pH, and types of carbon sources. AUP-03 isolate grew well at 10% salinity with an OD value of 0.88, isolate AUP-01 grew at 5% salinity with an OD value of 0.49, whereas isolate AUP-04 grew well on media that did not contain salinity. All three isolates grew well at pH 6 with OD values of 0.93, 1.12, and 1.27. AUP-03 and AUP-01 isolates grew well on media containing dextrose as carbon source with OD values of 0.154 and 0.17, respectively, while isolate AUP-04 grew well on glucose-containing media with an OD value of 0.22.

Abstract. Sari CP, Santosa SHMB, Marsono D. 2021. A portrait of the ecological clusters in the urban fringe area of Surakarta, Indonesia. Biodiversitas 23: 670-679. The population growth in Surakarta has led to agglomeration. To fulfill human needs, Surakarta has seen some major land-use changes across decades. The availability of natural resources and ecosystem services is very important for the sustainability of life. Biodiversity index-based area management is one of the best ways. Cluster methods will facilitate the planning, implementation and subsequent management of an area. This study divides the urban fringe of Surakarta into 48 ecological units. The biotic analysis of biodiversity index calculations is used to assess all ecological units. The value of the biodiversity indices is the basis for forming clusters. From the statistical analysis, 5 clusters were formed based on the level of similarity. The majority of the clusters were located in Karanganyar District. Cluster 3 was the cluster with the most members and the most value of the index of biodiversity. Cluster 2 had only 2 clusters. The range of biodiversity index in each cluster was not much different. The richness of the species in each cluster was in the medium-high range (tree level 0.945-4.500, saplings 0.908-4.889 and seedlings 0.638-4.440). The diversity of the species was at a moderate level with the tree level diversity index value of 2.044-2.290, saplings 1.610-2.294, seedlings 1.557-2.291 and was at a moderate level with the tree level index evenness value of 0.735-1.421, saplings 0.641-1.412, seedlings 0.730-1.645. In each cluster, the condition of the biodiversity was quite good and able to support the function and services of the ecosystem of urban areas and urban fringes. There was no dominance of plants, and the types were still diverse for the ecosystem stability. The development of Surakarta was faster in the south and the north. Biodiversity in eastern Surakarta was found to be more effective in supporting the ecosystem services of Surakarta City.
