Biodiversitas 22: 4308-4319. The herpetofauna discovery in West Java is still low and has not yet described the herpetofauna community's species diversity entirely and thoroughly in West Java. It experiences high anthropogenic impacts both from infrastructure and tourism activities. If left further, herpetofauna species will become extinct locally because of anthropogenic factors. Therefore, it is essential to reveal the diversity and distribution of herpetofauna species. This study aimed to show diversity and distribution based on the elevational range in West Java. Visual Encounter Survey in combination with the Auditory Encounter Survey was applied in this study. We found 74 species where the Colubridae family were found most commonly in the study sites. Of 74 species, four species have high conservation status, including Rhacophorus reinwardtii, Ophiophagus hannah, Varanus salvator, and Malayopython reticulatus. Besides, ten species were found to be endemic to Java and Java-Bali. Based on elevation, herpetofauna is mainly located at elevations < 1000 m asl. The higher the height, the fewer the number of species found. In Amphibians, the highest number of species tend to be found at 400-1200 m asl and 1400-1700 m asl, while reptiles tend to be located at 400-900 m asl. Keywords: Auditory Encounter Survey, West Java

Short communication: Algal genotypes in White Syndrome infected coral Acropora muricata from the Karimunjawa Islands, Indonesia. Biodiversitas 23: 415-423. The physiological equilibrium of the coral holobiont may help the corals to survive under various environmental threats. The symbiont genotype is thought to play an important role in the host's susceptibility to thermal stress and coral disease incidence. We identified the Symbiodiniaceae diversity of coral Acropora muricata infected by White Syndrome (WS). Samples were collected from Cemara Kecil Island and Barakuda Beach, Kemujan Island, Karimunjawa Islands. We applied DNA barcoding of 28S nuclear ribosomal RNA gene of the healthy colonies, healthy tissue, and lesions of diseased colonies to perform their symbiont type identification. There were two different symbiont types observed from the phylogenetic analysis, which formed two clades. A clade consisting all Cemara Kecil Island samples while the other with all Barakuda Beach samples except for one. All samples harbored symbiont types from the genus Cladocopium. The members of the genus were reported to have various physiological properties which may support the host resilience when facing the disease. A robust study is needed to reveal the influence of symbiont genotypes on the occurrence of coral disease and the susceptibility of the coral host.

Biodiversitas 22: 5195-5202. Cultivation of Kappaphycus alvarezii seaweed has been carried out in Ekas Bay of Jerowaru Sub-district, East Lombok, West Nusa Tenggara, Indonesia but its production has decreased in the last three years due to its slow growth. In 2014, seaweed production was 1,882,875.50 tons and in 2017 it was 1,037,000 tons. Efforts are needed to increase growth by providing growth triggers from marine natural ingredients, namely Sargassum aquifolium. These brown algae can be found in the waters of Ekas Bay, the numbers are abundant and untapped. The purpose of this study was to analyze the growth of K. alvarezii given S. aquifolium extract. This research was conducted in the waters of Ekas Bay. The method used is experimental. The experimental design used was a completely randomized design with different treatments of K. alvarezii soaking time using S. aquifolium extract. The results showed that the growth of K. alvarezii given S. aquifolium extract increased and was significantly different between treatments. The highest absolute weight was 479 g in treatment A (soaking for 2 hours) and the lowest absolute weight was 181.25 g in treatment K (control). The highest growth rate occurred on the 18th day, namely 9.24% and the lowest was in the K treatment (control) which was 1.3%.

Biodiversitas 22: 5274-5279. Birds belong to the mangrove forest ecosystem, which is important biodiversity for life and must be maintained. This study aimed to examine the diversity of bird species in Air Telang Protected Forest (ATPF) which has experienced massive degradation. Birds' observation was carried out using the purposive sampling method by exploring predetermined areas with binoculars and cameras and direct observation. Birds were identified by analyzing their morphology, sound, and color. The research data obtained a total of 38 bird species that consist of 13 orders and 22 families. In addition, the results of this study obtained nine species of migratory birds and two species with near threatened (NT) status based on the IUCN Red List Category. This study is the first bird species inventory and identification research conducted in ATPF.

Biodiversitas 22: 5106-5114. Peatland ecosystems provide a variety of environmental services and biodiversity with their ability to store and absorb carbon. The increase of land clearing followed by the construction of drainage or canal has resulted in the degradation of peatlands and is prone to fire incidences. Canal construction, whether we realize it or not, has a negative ecological impact on the in situ (local) environment, namely land degradation and subsidence, as well as a wider (global) impact in the form of loss of carbon stocks, increased GHG emissions that can trigger global warming. This study aims to identify the distribution of carbon in the area where drainage is made where the distribution of carbon stock in starting at a distance of 10 m, 50 m, 100 m, 250 m, and 350 m from the canal. The study was conducted on the community plantations including unburned areas (rubber plantations and secondary forests) and burned areas. The influence of land use and drainage development on carbon stock distribution is rarely known. The results showed a decrease in C-Stock with a decrease in drainage distance. The quantity of carbon stock in secondary forests ranged from 13.275 ton ha-1 - 24.839 ton ha-1, Burnt Area-1 ranged from 6.995 ton ha-1 - 11.59 ton ha-1, and Burnt Area 2 ranged from 4.677 ton ha-1 - 12.580 ton ha-1. In contrast, rubber plantations had higher C-stock in the initial observation distance, around 9.064 ton ha-1 - 11.805 ton ha-1, which may be due to intensive land use near the canal.

Abstract. Navia ZI, Suwardi AB, Baihaqi. 2021. Ethnobotanical study of medicinal plants used by local communities in Sekerak Subdistrict, Aceh Tamiang, Indonesia. Biodiversitas 22: 4273-4281. Local communities in Aceh Tamiang have gained a wealth of medicinal knowledge through practice and experience in their long-term battles with the disease. However, because of a lack of written records and rapid economic development, their traditional medicinal knowledge is under threat. This study investigated medicinal plants and related traditional knowledge of local communities in the Sekerak subdistrict, Aceh Taming, Indonesia. Field surveys, plant collections, and interviews with communities were used in this study. The Snowball Sampling technique was used to select 60 informants for the interviews. A total of 46 medicinal plant species belonging to 40 genera and 26 families were used for medicinal purposes by local communities to treat 28 different diseases. The itch, swelling, cough, and nosebleed were the most commonly treated. With a use-value index of 0.98, Tagetes erecta was the most commonly used medicinal plant by local communities. In addition, local communities identified the leaf (50%) as the most widely used plant part and oral administration (65%) as the most common method of administering traditional medicine. Elders have more knowledge of medicinal plants than younger generations, indicating that traditional knowledge is eroding across generations. However, initiatives to promote and conserve medicinal plants must be improved, particularly among the younger generation. This is required to ensure the availability of medicinal plants and the preservation of traditional knowledge in the future.

Genetic analysis for designing an ideotype of high-yielding sorghum based on existing lines performance. Biodiversitas 22: 5286-5292. Sorghum is a potential crop as an alternative and substitutes food source for rice or wheat. The ideal type of sorghum design for a high-yielding trait is needed to streamline the process of selecting the best genotype that can be developed into pure lines and released as varieties. The research purpose was to design the ideal type of sorghum based on the morphological, physiological, and agronomic characters that contribute to a high-yielding trait. The research was carried out at the Agriculture Research Center for Biotechnology and Genetic Resources, Bogor, West Java, Indonesia and experimental field of IPB University, Bogor, Indonesia. The genetic materials used in this study consisted of 56 F5 sorghum lines advanced from three segregating populations and 8 check varieties. The ideal type of sorghum to obtain a genotype with high grain weight per panicle was to have a phenotypic value of 150-235 cm for plant height (medium plant height). The physiological and agronomic characters obtained to design the ideal type of sorghum were stem diameter, leaf number, flag leaf area, leaf greenness, green leaf percentage, and panicle diameter with the minimum value for the traits are 17 mm, 10, 209 cm2, 44 CCI units, 48%, and 49 mm, respectively. This ideotype is going to be used as selection criteria for selection of high-yielding sorghum genotypes.

Diversity of Bacillus spp. from soybean phyllosphere as potential antagonist agents for Xanthomonas axonopodis pv. glycines causal of pustule disease. Biodiversitas 22: 5003-5011. Pustule disease caused by Xanthomonas axonopodis pv. glycines (Xag) is an important disease in soybean. Bacteria from soybean phyllosphere is one of the potential biological agents against this disease. This is because the microorganisms have similarity. This study aimed to determine the diversity and species of bacteria from the soybean phyllosphere that have potential as biological agents. The research was carried out by morphological observation, physiological - biochemical testing,  and molecularly with BOX AIR primer. Molecular identification was carried out by amplifying the 16S-rRNA gene with 27F and 1492R primers. The results showed that 11 isolates could inhibit Xag in vitro which showed morphological, biochemical, and molecular diversity. These bacteria were identified as Bacillus spp. which consisted of 4 groups, namely Bacillus siamensis, B. subtilis. B. amyloliquifaction and B. velezensis. The diversity of phyllosphere bacteria allows them to be used as biological agents because they do not inhibit each other and have diverse living abilities in various environmental conditions.

Morphometric variation of the horseshoe crab Tachypleus gigas (Xiphosura: Limulidae) from the Banyuasin estuarine of South Sumatra, Indonesia. Biodiversitas 22: 5061-5070. Morphological studies are essential for fish resource management, ecology, conservation, and stock assessment. This research was conducted to characterize the morphological variation of the coastal horseshoe crab (Tachypleus gigas) from the Banyuasin estuarine, South Sumatra, Indonesia. The body weight (BW), prosoma width (PW), carapace length (CL), telson length (TEL), and total length (TL) were measured for describing the morphometric variability of T. gigas. A total of 70 T. gigas (32 females and 38 males) were found using trammel net fishing and bottom gillnet during the survey. Multivariate and allometric methods analyzed their morphometric characters. Based on Kruskal- Wallis tests, the result showed a significant difference in somebody measurements (TL and CL) between males and females. The principal component analysis (PCA) results showed that all morphometric characters had a strong correlation in both sexes, so the first principal component (PC1) values were 92% and 72%. In comparison, the second principal component (PC2) values were 5% and 20% in females and males, respectively. The results of discriminant function analysis (DFA) suggested that only one (BW) of the five morphometric characters was significant in separating both sexes. Both sexes were also revealed variations in growth patterns based on the allometric analysis results. These results were expected to be used as a basis for managing the horseshoe crab population-based conservation.

Lesser-known ethnic leafy vegetables Talinum paniculatum grown at tropical ecosystem: Morphological traits and non-destructive estimation of total leaf area per branch. Biodiversitas 22: 4487-4495. Talinum paniculatum known as Java ginseng is an ethnic vegetable in Indonesia that has also been utilized as a medical plant. Young leaves are the primary economic part of T. paniculatum, which can be eaten fresh or cooked. This study was focused on characterizing morphological traits of T. panicultaum and developing a non-destructive yet accurate and reliable model for predicting total area per leaf cluster on each elongated branch per flush growth cycle. The non-destructive approach allows frequent and timely measurements. In addition, the developed model can be used as guidance for deciding the time to harvest for optimum yield. Results indicated that T. paniculatum flourished rapidly under wet tropical conditions, especially if they were propagated using stem cuttings. The plants produced more than 50 branches and more than 800 leaves, or on average produced more than 15 leaves per branch at the age of nine weeks after planting (WAP). The zero-intercept linear model using a combination of two traits of length x width (LW) as a predictor was accurate and reliable for predicting a single leaf area (R2 = 0.997). Meanwhile, the estimation of total area per leaf cluster was more accurate if three traits, i.e., number of leaves, the longest leaf, and the widest leaf in each cluster were used as predictors with the zero-intercept linear regression model (R2 = 0.984). However, the use of a single trait of length (L) and width (W) of the largest leaf within each cluster as a predictor in the power regression model exhibited moderately accurate prediction at the R2 = 0.883 and 0.724, respectively.
