Plants diversity and socioecological functions of homegarden in Sundanese rural area: a case in Sumedang district, West Java, Indonesia. Biodiversitas 24: 156-175. Homegarden is one of the traditional agroforestry systems in Indonesia, which is traditionally planted with various annual and perennial plants. It has provided social, economic, cultural, and ecological functions. The objective of this study is to elucidate the vegetation structure and function of the homegarden system of two groups of rural communities who are affected by the development project of Jatigede Dam, Sumedang, West Java. The first community group of residents who lost their village, their homegarden, and another agricultural land due to the Jatigede dam and settled in new resettlement provided by the local government in Cihegar Mekar Hamlet. The second community group, residents living in the neighboring Kampung Cihegar Mekar, whose settlements and homegarden were not inundated by the Jatigede Dam, but the Jatigede Dam inundated their agricultural lands. Qualitative and quantitative methods were used with an ethnobotanical approach. The results showed that the diversity of species of the homegarden of Cihegar Mekar and Ciboboko Hamlets affected by the Jatigede dam project was high. In Cihegar Hamlet, the number of species, genera, and family in 2018 and 2022 was recorded at 63 and 155, 58 and 130, and 39 and 59, respectively. While in Ciboboko, Hamlet in 2018 and 2022 recorded the number of species, genera, and family at 81 and 144, 76 and 114, and 44 and 55, respectively. These variables have significantly increased in the past 4 years (2018-2022). The residents are intensifying the cultivation of their homegarden because other agricultural land types, such as wet-rice fields, have been dramatically reduced or even lost due to inundation by the Jatigede dam. The 224 plant species found in the homegarden consist of 127 (56.70%), 30 (13.39%), 27 (12.05%), and 13 (5.80%) ornamental, fruit, vegetable, and spice species. At least 20 species and varieties (landraces) of the homegarden have been considered to fulfill the daily household need, and some surpluses are sold to obtain household income. So, the high diversity of plant species in the village homegarden has essential functions to support food security and the population well-being of those affected people of the development project of Jatigede Dam. Homegardens appear to be an important strategy for attaining the rural development program and can be accomplished with proper planning.

Biodiversitas 24: 399-414. Weeds are considered undesirable plants that grow where they are not wanted, as they often outcompete crops and other beneficial species. This study aimed to compare species composition and diversity of weedy plant communities in fields planted with herbicide tolerant maize and associated field margins in the Eastern Cape Province, South Africa. A total of 88 weed species, representing 31 families were recorded from 244 plots for maize fields and field margins combined. The maize fields and margins across the four sites shared 53% of species. Non-metric Multidimensional Scaling grouped maize fields and field margins separately based on weed species composition. Moreover, maize fields and margins differed significantly in terms of Simpson (p=0.044) and Shannon indices (p=0.003), with diversity being higher in the fields. This study confirms that maize fields have a dissimilar composition of weedy species and dissimilar diversity, which differs from those of field margins. This result enables farmers to know whether current herbicide use is sufficient for targeted weed control. The findings and knowledge will benefit maize farmers and future researchers working on weeds in Sub-Saharan Africa when it comes to the development, implementation, and prioritization of various herbicide-based weed management protocols to ensure maximum crop productivity.

Sidikalang M3 generation through induction of gamma-ray irradiation. Biodiversitas 24: 200-207. Upland rice cultivar Sidikalang is one of the local upland red rice cultivars of North Sumatra, which has the potential to be developed. However, this rice has limited planting area due to factors such as its higher plant height and long duration compared to other rice cultivars. Plant characteristics can be improved through induced mutation using gamma-ray irradiation. This study aimed to determine gamma rays' effect on changes in morphological, biochemical, and molecular characters and to obtain a superior Sidikalang cultivar mutant plant M3 generation. Observations of morphological changes were carried out quantitatively and qualitatively, biochemical analysis (amino acid content) was done using a High Performant Liquid Chromatography analyzer, and molecular analysis was performed using Random Amplified Polymorphism DNA markers. The results showed that 600 Gy gamma-ray irradiation improved quantitative morphological characters (shorter plant height, higher number of tillers and panicles, faster panicle release age) and qualitative morphological characters (flag leaf angle in dwarf mutants became upright) compared to no irradiation. 400 Gy gamma-ray irradiation also increased almost all amino acid content compared to no irradiation. The presence of polymorphic bands in the M3 generation Sidikalang cultivar mutant plant evidenced changes in morphological and biochemical characters.

Biodiversitas 23: 6428-6436. Red snappers are an economically valuable fishery resource. Most of these snapper species are inhabitants of coral reef ecosystems. A variety of red snapper species can be found in the northern waters of Papua, particularly in the Yapen Regency. However, information regarding the biological and ecological aspects of these fish is virtually unavailable. On the other hand, the utilization intensity of red snapper as a source of community income continues to increase. By using DNA barcode sequences, this study attempted to examine species diversity and relationships among Lutjanidae family species. The study was carried out from June to August 2022 in the waters of Yapen Regency by taking fish samples from the fish caught by local hand-lining fishers. Red snapper samples were identified morphologically and molecularly. According to molecular analyses, it was identified nine red snapper species from the genera of Lutjanus, Aphareus and Pristipomoides. The morphological characteristics of the species from the genus of Aphareus are similar to those from the genus of Pristipomoides yet different from those of the species from the genus of Lutjanus. The phylogenetic tree consisted of four clades with significant bootstrap values ranging from 98 to 99%. Clades 1, 2 and 3 comprise the species from the genus of Lutjanus, while clade 4 contains species from the genera of Aphareus and Pristipomoides. The greatest genetic distance was found between Lutjanus fulvus and Pristipomoides multidens, while the smallest genetic distance was found between Lutjanus vitta and Lutjanus ehrenbergii. Based on the study results, some management implications are discussed. For example, information on species biodiversity maps is needed to determine the target stock of a managed species so that management objectives are more focused on the species level and not on the genus or family level.

Kanka P, Sukmasuang R, Duengkae P, Siripattaranugul K. 2023. Abundance and physical factors affecting the appearance of selected terrestrial birds in Khao Yai National Park using camera trapping. Biodiversitas 24: 222 232. This study used results from camera traps set for Dhole (Cuon alpinus) and their prey during 2017-2020 in Khao Yai National Park to assess the species richness of ground-dwelling birds and identify abundance and also temporal appearance of the species in the area. We selected only the Pheasant species that typically forage on the ground to investigate the physical factors affecting the appearance in Khao Yai National Park. Based on camera trapping data with a total of 4139 trap days, 115 camera locations showed 36 species of birds belonging to 21 families, and 13 orders of ground-dwelling bird species were recorded. Siamese Fireback (Lophura diardi), Red Junglefowl (Gallus gallus), and Silver Pheasant (Lophura nycthemera) were detected with an abundance index of 10.65%, 7.27%, and 1.84%, respectively. The ground-dwelling bird abundance index from the camera traps accounted for 26.85% of the total fauna. Although the Phasianidae were found to be diurnal species, this study also shows a statistically significant difference in the average time between Red Junglefowl and Siamese Fireback. Meanwhile, there was no statistically significant difference between Red Junglefowl and Silver Pheasant during their active time. The water sources showed a significant effect on the presence of the Siamese Fireback in data analysis; however, no physical environmental factors significantly affected the distribution of Red Junglefowl and Silver Pheasant. This study found that surrounding creeks and streams significantly impacted the appearance of the Siamese Fireback. As a result, area management needs to prevent and reduce disturbances along a creek in order to effectively manage the habitat of the Siamese Fireback and other wildlife.

Ethyl Methane Sulfonate (EMS) mutant based on morphological, physiological and molecular characters. Biodiversitas 24: 531-538. The initial generation of Ethyl Methane Sulfonate (EMS) random mutations usually still shows high variation due to allele segregation. This research aimed to confirm the genetic differences between the M4 generation of chili pepper (Capsicum frutescens L.) mutant resulted from EMS mutations (G7/01) and the initial line (G7), and the stability of the mutant based on morphological, physiological, and molecular characters. The morphological characters and the capsaicinoid content of the mutants G7/01 with the initial line (G7) were compared. The capsaicinoid content and fruit spiciness were measured by spectrophotometer ? 280 nm. Molecular characterization was conducted using Random Amplified Polymorphic DNA (RAPD) genetic markers. Based on the morphological characters, the G7/01 mutants have some superior properties compared to the initial line G7. All the G7/01 mutant plants contain higher capsaicinoid compounds than the initial line plants. The dendrogram developed based on RAPD profile showed that all the mutant plants were positioned apart from the initial line plant, suggesting that there are some genomic changes in the mutants compared to the initial line plants. All the mutant plants, except T1, showed insignificant variation in morphological characteristic, capsaicin content, and RAPD profile, indicating genetic stability.

Characteristic and activity of cellulolytic bacteria isolated from mangrove soil in Northern Coast of Aceh Province, Indonesia. Biodiversitas 23: 6587-6599. The cellulolytic bacteria as decomposing cellulose bacteria is related to the soil fertility and productivity of the mangrove ecosystem, and the presence and activity of enzymes produced can evaluate them. The study aims to analyze the cellulolytic bacteria characterization, cellulase enzyme activity, and production and to evaluate the molecular biology of cellulase bacteria isolated from the mangrove soil on the northern coast of Aceh Province, Indonesia. This study was conducted at six locations, and soil samples were collected randomly with three replications. The results showed that the purified bacteria produced 39 isolates that grew in CMC selective media. The common bacteria discovered have an irregular colony shape, undulate colony edge, raised colony elevation, and cream pigmentation. The bacilli cell is the most common shape, while 22 bacteria out of the 39 isolates showed cellulase activity. The cellulolytic index (CI) ranged from 0.31 to 4.82, and the highest CI was BTM533 (4.82), followed by BTM622 (2.09). The quantitative analysis showed that the highest specific cellulase activity of BTM533 and BTM622 were at 36 hours and 42 hours with the value of 0.042 U mg-1 and 0.129 U mg-1. Among the bacteria isolates, two cellulolytic bacteria with high cellulase activity were identified as Bacillus safensis and B. altitudinis using the 16S rRNA gene molecularly.

Abstract

The root-knot nematodes Meloidogyne incognita poses a serious threat to horticultural food and plantation crops. One of the effective and efficient measures to overcome this nematode is using bacteria as a biological control agent. This study aimed to test the effectiveness of Bacillus subtilis bacteria grown on media with additional micronutrients as a biological control agent for M. incognita. The test was carried out by growing bacteria in 100 mL of nutrient broth (NB) added with FeCl2, MnCl2, and CuCl2. Each type of micronutrient was administered at concentrations of 35 ppm, 40 ppm, 45 ppm, 50 ppm, and 55 ppm. As a control, NB media was used without the addition of micronutrients. The test results showed that the best performances on bacterial growth were 55 ppm MnCl2, 45 ppm MnCl2, 40 ppm MnCl2, 40 ppm CuCl2, 50 ppm MnCl2, and 35 ppm MnCl2. These six treatments were then used in an antagonism test. A total of 150 J2 M. incognita in 4 mL of suspension was put into a petri dish with a diameter of 5 cm. After that, 1 mL of each bacterial suspension was put into a petri dish following the treatment. The mortality rate of M. incognita in each treatment was 77.5% in 55 ppm MnCl2, 74.5% in 45 ppm MnCl2, 77.25% in 40 ppm MnCl2, 73.25% in 40 ppm CuCl2, 78.75% in 50 ppm MnCl2, 79% in 35 ppm MnCl2, and 53.25% in control. The same six treatments were also used to measure the chemotaxis index of M. incognita on tomato roots. Tomato seeds of the Tantyna variety were soaked for 60 minutes in a bacterial suspension grown on micronutrient-enriched media. After seven days, the tomato seed roots were removed and placed in 5 mL 20% Pluronic F-127 Gel which had been infested with 100 J2 M. incognita. After 6 hours, the number of nematodes approaching the roots was observed. The chemotaxis index in the treatments ranged from 16% to 24%, while that in the control was 33%. This study corroborates that the addition of micronutrients MnCl2 and CuCl2 to the growth medium of B. subtilis can increase antagonistic activity against M. incognita and suppress its chemotaxis response in tomato roots soaked in the bacterial suspension.

Nutrients content of four tropical seaweed species from Kelapa Beach, Tuban, Indonesia and their potential as ruminant feed. Biodiversitas 23: 6191-6197. Indonesia is one of the tropical countries with the largest diversity of seaweed, but studies on identifying the nutrients content of numerous seaweed species from tropical oceans and their potential as ruminant feed have not been widely reported. This study was designed to evaluate the nutrient, macro and micromineral content of brown (Laminaria sp. and Padina australis) and red seaweed (Eucheuma cottonii and Gracilaria sp.) from Kelapa Beach, Tuban, East Java. All tests were carried out in duplicate from individual seaweed and data were analyzed descriptively by calculating the average of data obtained. The result showed that all of the seaweed species in this experiment had a high content of water (69.41-86.33%) and ash/mineral (23.42-65.63%). The brown seaweed Padina australis had the highest dry matter (30.59%) and crude protein (12.57%). The red seaweed of Gracilaria sp. had the highest ash (65.63%) and the lowest organic matter (34.37%), ether extract (0.21%), crude fiber (2.49%), nitrogen-free extract (19.95%), neutral detergent fiber (21.74%), acid detergent fiber (5.46%), and gross energy (1,083 Cal/g); however, in contrast, Eucheuma cottonii had the highest organic matter (76.58%), ether extract (2.85%), crude fiber (8.80%), nitrogen-free extract (56.38%), neutral detergent fiber (56.93%), acid detergent fiber (11.44%), and gross energy (2,911 Cal/g) but the lowest dry matter (13.67%) and crude protein (8.55%). The brown seaweed Padina australis had the highest Na (0.89%), Mg (4.90%), and Cd (3.73 ppm) contents. The red seaweed of Gracilaria sp. had the highest K (2.18%), Ca (5.80%), Fe (7,596 ppm), Mn (818 ppm), Cu (5 ppm), Zn (52 ppm), and Pb (60.38%); however, in contrast, Eucheuma cottonii had the lowest P (0.01%), K (2.02%), Ca (0.64%), Mg (1.09%), Fe (400 ppm), Mn (38 ppm), Pb (17.31 ppm), and Cd (1.53 ppm) but the highest S (0.46%). Based on this study, brown seaweed (Laminaria sp and Padina australis) and red seaweed (Eucheuma cottonii and Gracilaria sp.) from Tuban Regency, East Java, Indonesia have the potential as ruminant feed, especially as soluble carbohydrates and an organic mineral source that requires attention for heavy metals to prevent toxicity to ruminants.

Abstract. Setyati WA, Pringgenies D, Soenardjo N, Pramesti R. 2023. Enzyme-producing symbiotic bacteria in gastropods and bivalves molluscs: candidates for bioindustry materials. Biodiversitas 24: 20-25. Biotechnology plays a vital role in modern civilization because almost all aspects of human life have benefited from the development of biotechnological applications, especially in the bioindustry. Therefore, the present study aims to obtain bacterial isolates from molluscs and identify those with the potential to produce protease, amylase, and cellulase enzymes using bimolecular methods and bacterial biochemical analysis. The samples were collected in April 2022 from the coastal waters of Krakal, Special Region of Yogyakarta, Indonesia. The symbiotic bacteria were isolated, and screened for different enzymatic activity. The molecular method (16S rRNA gene sequence) was used to identify acterial isolates with the highest potential for enzymatic activity. Biochemical analysis of isolates that are potential candidates for industrial materials was carried out using the thin-Layer Chromatography (TLC) method to detect the compounds produced by bacteria. The results showed that the bacterial isolates GS 1-4, GS 2-1, and GS 2-12 had similarity with Alcaligenes faecalis (99.83%), Alcaligenes faecalis (99.74%) and Alcaligenes aquatilis (98.51%) respectively. The three isolates showed the potent enzymatic activity and contained alcohol, amines, aldehyde, ketone and conjugated aromatics, as well as amino acids for potential candidates of bioindustry materials.
