Several studies have been carried out on crustacean larvae diversity, but they focused on the eastern and central regions of the Segara Anakan lagoon/estuary, Cilacap District, Central Java Province, Indonesia. Furthermore, one study has explored crustaceans in the western region but focused on adult individuals and used morphological characteristics. Currently, there is no internet data on the crustacean larvae diversity from the estuary's western region, and there is a high possibility that some species are unidentified. This study aims to describe crustacean larvae diversity in the western region of the Segara Anakan lagoon in Indonesia by using DNA barcoding. Sequence divergence of 3% to databases in GenBank and BOLDsystems was used as a genetic threshold for species border. A total of 14 morphotypes were barcoded, but only nine were successful, of which three were identified as bacteria and six as crustaceans. Based on the predetermined genetic threshold, all crustacean morphotypes can be identified at the species level, and six species were obtained. Meanwhile, a previous study on the adult individual only identified specimens at the genus level, and only one genus was similar to these obtained results. A total of 5 out of 6 species were new records to the western region Cilacap, while 2, namely Caridina gracilipes and Ptychognathus altimanus, are newly recorded in the Segara Anakan lagoon. These results contribute significantly to the information on crustacea larvae diversity, which is essential for sustainable management.

Human-wildlife conflict has emerged as one of the major threats to the global population of large carnivores. This condition applies to the endemic and Critically Endangered Sumatran tiger (Panthera tigris sumatrae). Although efforts to resolve conflict have been made from time to time, a sustainable solution is still very much needed, particularly given the unique characteristics of each case and area of conflict. A community-based approach to human-tiger conflict management can become a local-based solution towards a more sustainable management. This study aims to identify needs, gaps, and next steps related to collective action and sustainable community-based human-tiger conflict management in Seluma, Bengkulu, using a qualitative approach with literature review followed by semi-structure interview method. This study’s findings show that in the context of community-based human-tiger conflict management, the needs and gaps are still identified, particularly in: training and capacity building, adaptive management and applied research, communication and information exchange, policy frameworks and legal instruments, as well as funding. To make community-based human-tiger conflict management sustainable, the strength and sustainability of: coherent legal instruments; commitment, involvement, active role, communication, and support from all stakeholders, and; comprehensive environmental, social and economic data-based planning; need to be ensured.

Biodiversitas 23: 6049-6055. Marine organisms have become essential candidates for discovering novel compounds that can be medically and commercially important. They are believed to yield novel compounds with unique chemical structures and significant biological activities since they managed to survive a milieu of pathogenic microorganisms and essential predators. The quest against fungal infections has been challenging due to acquired resistance to commercially available drugs hence, novel sources to treat infectious diseases caused by fungi have been investigated. Sea cucumbers have been recognized in Asian countries as a rich food source. Apart from that, they have also been studied for their medicinal properties. This study aimed to evaluate the antifungal activity of the selected sea cucumber species found in Brgy. Sugod, Tukuran, Zamboanga del Sur, Mindanao, Philippines using the modified SPOTi assay. Methanol: ethyl acetate crude extracts of Holothuria scabra, Stichopus sp., and Holothuria atra were tested for their antifungal activity against Aspergillus niger and Candida tropicalis. A 96-well plate assay was used, and the growth of fungi was assessed visually for up to 72hrs. The concentration resulting in ‘no fungal growth’ was observed and taken as the Minimum Inhibitory Concentration (MIC) value. The average MIC was calculated. Results showed that sea cucumber crude extracts have MIC values of 0.0333-0.1 mg/mL. Tukey’s posthoc test confirmed that the antifungal activity of the sea cucumber extracts was comparable to the positive control Fluconazole (p>0.05). The present study revealed a potentially effective natural antifungal agent from sea cucumbers.

Antimicrobial resistance characteristics of multidrug resistance and extended spectrum beta-lactamase producing Escherichia coli from several dairy farms in Probolinggo, Indonesia. Biodiversitas 23: 215-221. Escherichia coli bacteria initially reside in the digestive tract of humans and animals but are able to adapt to new environments that are different from their initial habitat. The pathogenicity of E. coli can occur when these bacteria grow more than normal limits, produce toxins, and are resistant to certain types of antibiotics. The purpose of this study was to investigate the antimicrobial resistance characteristics of MDR and ESBL-producing E. coli from several dairy farms in the Probolinggo district of East Java province, Indonesia. A total of 150 samples consisting of 109 milk and 41 environmental samples from 41 dairy farms were used for isolation. TSIA and IMViC biochemical tests were used to identify E. coli bacteria. Escherichia coli resistance profile was obtained through disc diffusion test on several antibiotics, namely tetracycline, streptomycin, trimethoprim, chloramphenicol and aztreonam. Escherichia coli that was resistant to 3 or more antibiotics was defined as MDR. The results of isolation and identification obtained 124 (82.6%) isolates characterizing E. coli bacteria. The antimicrobial susceptibility test of E. coli showed 9 (7.26%) MDR isolates and 2 (22.22%) ESBL isolates by double-disc synergy test (DDST). MDR E. coli was dominated by the pattern of antimicrobial drug resistance TE-S-W (tetracycline, streptomycin, trimethoprim) with a total of 8 (38.10%) isolates, followed by antimicrobial drug resistance pattern TE-S-W-ATM (tetracycline, streptomycin, trimethoprim, aztreonam) with one (4.76%) E. coli isolates. The pattern of antimicrobial drugs of ESBL E. coli showed in one (11.11%) sample of ESBL E. coli from a milk sample with the pattern of TE-S-W-ATM (tetracycline, streptomycin, trimethoprim, aztreonam) and one (11.11%) sample ESBL E. coli (AL 30) from the environmental sample with a pattern of TE-S-W pattern (tetracycline, streptomycin, trimethoprim). The discovery of MDR E. coli isolates and ESBL E. coli from milk and environmental samples at several dairy farms in Probolinggo district, East Java, Indonesia is a matter of concern and requires real action to reduce antibiotic resistance.

The diversity of utilizations, tapping flow discharge, and conservation of sugar palm (Arenga longipes Mogea) cultivated in Langkat, North Sumatra, Indonesia. Biodiversitas 23: 122-132. Sugar palm (Arenga longipes Mogea) is one of the Non-Timber Forest Products (NTFPs) cultivated by the forest community in the village of Sei Limbat, Selesai Sub-district, Langkat District, North Sumatra. It is a plant with several benefits for economic development and a source of livelihood. Therefore, this research aims to determine the morphology of sugar palms through the identification of the species developed by the community, potential use, economic value, and tapping productivity. The morphological identification was used to determine the species of sugar palm developed by the community. The multiple utilization and economic values were identified and analyzed using the result of in-depth interviews through questionnaires. In this study, it was found that the diversity of uses of the palm plant included the utilization of palm sugar (consisting of large palm sugar, small palm sugar, liquid sugar and powdered sugar), utilization of black palm fiber, and conservation purposes. Based on the morphological identification, the species of sugar palm cultivated by the forest community was Arenga longipes Mogea. Furthermore, the tapping productivity was influenced by the panicle circumference, tree height, and flower height from the ground. The tapping productivity was calculated using daily production, which was correlated with sugar palm tree morphology. Based on the result, it can be concluded that the species identified is A. longipes which has a variety of uses and the tapping productivity was influenced by plant morphology.

Efforts to meet rice needs in Indonesia can be made through an extensification program. New paddy fields generally have a high FeSO4 content, so rice varieties resistant to these conditions are needed. This study aims to evaluate the growth of lowland rice varieties under FeSO4 stress and to analyze the expression of a gene that plays a role in iron storage, namely the OsFER1. Six lowland rice varieties, namely Ciherang, Inpari 42, Inpari 30, Sunggal, Logawa, and Cibogo were grown on media with 0 and 400 ppm FeSO4 applications. The plant height, leaf bronzing score, and the number of filled grains produced were observed to determine the effect of FeSO4 administration on the agronomic performance of rice. The OsFER1 gene expression ratio was analyzed using real-time PCR. The best plant height at 400 ppm FeSO4 was found in the Inpari 30 variety (40.3 ± 0.49 cm), with a leaf bronzing score was 1. The Inpari 30 also produced grain at 400 ppm FeSO4. Molecular analysis showed that the highest OsFER1 expression was found in the Inpari 30 grown at 400 ppm FeSO4. Based on morphological and molecular observations, Inpari 30 is tolerant to FeSO4 stress.

Abstract. Natsir M, Ulya Z, Fitriani R. 2022. Mangrove forest utilization policies reconceptualized with a view to improving the regional economy in Aceh Tamiang District, Indonesia. Biodiversitas 23: 6570-6578. One of the Aceh's (Indonesia) districts with mangrove forests is Aceh Tamiang District. Manyak Payeud, Bendahara, Seureuwey, and Banda Mulia are the four sub-district in the Aceh Tamiang region that have the most potential tourist sites. However, due to the widespread illegal logging of mangroves carried out by the community to suit their daily requirements, the preservation of mangrove forests in the four sub-districts above is seriously damaged. As a result of illegal logging, mangroves are becoming less common in Aceh Tamiang District. Therefore, a unique policy, which can incorporate both community needs and economic development, is needed to manage the mangroves of Aceh Taminga District. The expectation is that mangrove management will enable Aceh Tamiang District to develop tourist attractions in this study. The normative juridical research methodology with SWOT analysis was applied in this study to understand the potential of mangrove management in improving the regional Economy in Aceh Tamiang District. The findings of this study are based on a SWOT analysis of four subdistricts in Aceh Tamiang District. The result suggests that the notion of forestry policy can support both local community interests and regional economic development. Additionally, it can be carried out through the Village Qanun to raise community awareness of the need to maintain and manage mangroves to enable the implementation of regional plans. To harmonize the understanding of the significance of mangroves as one of the regional tourist destinations, institutional coordination between the Aceh Tamiang district government and the provincial and central governments can also be used to realize the development of Aceh Tamiang tourism policies.

Short Communication: Characteristics of the endangered Javan banteng (Bos javanicus) spermatozoa. Biodiversitas 24: 759-766. Morphology and morphometry of spermatozoa become essential fertility parameters in domestic and wild animals. Such morphological abnormalities can arise from genetic or environmental origins and cause fertility deprivation. Semen collection using transrectal massage has been used in several domestic and wild animals and is known to be simpler, safer, and less painful than artificial vaginas and electro-ejaculation. This study aimed to evaluate morphological and morphometrical features of the banteng spermatozoa head. The objective of this study was also, to evaluate the use of transrectal massage as a semen collection protocol. This study was conducted in Taman Safari Indonesia, Cisarua, Bogor, West Java, involving one banteng (Bos javanicus) bull. Semen samples were collected weekly, smeared, and stained with a modified Williams' staining method. This study found ten types of head morphological abnormality, comprising several major defects, with several types known to be the genetic origin. There were also nine types of spermatozoa tail abnormalities and teratoid form abnormalities. The total normal spermatozoa proportion in this study was 54.30±1.74%. Morphometric dimensions of length, width, and area head of banteng spermatozoa were 9.88±0.01 µm, 4.91 µm, and 38.12±0.05 µm2, respectively, with a tail length of 60.48±0.03 ?m. Although our result was limited by the number of samples available for the study, this first report on the banteng bull's sperm morphology and morphometry still need to be considered. The possibility of transrectal massage use for banteng semen collection protocol demonstrated in this study may be helpful to support the conservation approach in terms of breeding program strategy.

Abstract. Tambaru E, Ura’ R, Tuwo M. 2023. Diversity of herbal medicine in Mamasa District, West Sulawesi, Indonesia. Biodiversitas 24: 2013-2022. This study aimed to identify the species of traditional medicinal plants, the parts processed as medicine and the processing methods of the plants used in traditional medicine. This research was conducted in Mellangkenapadang Village and Paladan Village, Sesenapadang District, Mamasa Regency. This research method was descriptive exploratory. Qualitative data collection was carried out using the cruising method. Sampling of medicinal plants in the site was carried out using the Participatory Rural Appraisal method, an assessment process involving active roles of the community. The results showed that the number of medicinal plants growing in Mellangkenapadang Village and Paladan Village, Sesenapadang District, Mamasa Regency were 49 species belonging to, 30 families, the most widely used being Asteraceae, and 22 orders, the most widely used being Solanales. The number of medicinal plant species used in Mellangkenapadang Village was 46 species. The most frequently used plant was guava (93.33%) and the lowest pumpkin, pineapple, spoon leaf, and airplant (3.33%). Meanwhile, in Paladan Village, 43 species were used; the most frequently used was painted nettle (96.67%) and the least were airplant and galangal (3.33%). The most widely used plant part as traditional medicine was leaf (59.18%) and the least used were stems (2.04%) and seeds (2.04%). The processing of medicinal plants was mostly boiling (55.10%) and the least burning (2.04%). Medicinal plant knowledge needs to be preserved for the younger generation so that it can be used sustainably in the future. This can be done by conserving biodiversity and preserving the plant's genetic material for pharmaceutical development.

Diversity of arbuscular mycorrhizal fungi in asphalt post-mining land in Buton Island, Indonesia. Biodiversitas 23: 6327-6334. Studies on the presence and diversity of Arbuscular Mycorrhizal Fungi (AMF) on overburden (OB) and asphalt post-mining areas in Buton Island are still limited. The purpose of this study was to examine the presence and diversity of AMF on OB and asphalt post-mining stockpiles in Buton Island, Southeast Sulawesi Province. The sampling of soil and roots of successional plants was carried out at eight post-mining sites at the IUP Kabungka (Pasarwajo) (6 sites) PT. WIKA Aspal and IUP Lawele (2 sites), PT. WIKA Bitumen. The AMF spores were isolated using the wet filter pour method, followed by the identification of AMF which was done by observing the morphology of AMF spores. The results showed that all successional plants were in symbiosis with AMF, which was indicated by the existence of AMF structure in plant roots with the percentage of AMF colonization ranging from 5 - 95%. The number of spores per 50 g ranged from 1 to 9 spores. A total of 9 AMF species were found to belong to five genera, i.e., Claroideoglomus, Haloatospora, Glomus, Gigaspora, and Cetraspora from three families. Claroideoglomus claroideum was found to be the most dominant species isolated with the high occurrence and the highest Importance and Shannon-Wiener Diversity Indices followed by Haloatospora cf. pansihalos, Glomus sp. 5, and Glomus sp. 4. This study also showed that the AMF diversity index differed among locations. The presence of AMF can accelerate the emergence and development of adaptive plants on post-asphalt mining land in Buton Island.
