Analysis of the effect of ENSO and IOD on the productivity of yellowfin tuna (Thunnus albacares) in the South Indian Ocean, East Java, Indonesia. Biodiversitas 24: 2689-2700. Yellowfin tuna (Thunnus albacares) is one of the fish that migrates through the Indian Ocean and is primarily caught in the south Java waters which are directly adjacent to the Indian Ocean. Fish abundance and migration are influenced by oceanographic factors, including climatic factors which affect annual and interannual variations, such as the Indian Ocean Dipole (IOD) and the El Nino Southern Oscillation (ENSO). This study aimed to determine the effect of climate anomalies on the productivity of yellowfin tuna in the Indian Ocean south of East Java, Indonesia. Data collection was carried out at Coastal Fishing Port of Pondokdadap East Java, involving 58 respondents consisting of fishermen with yellowfin tuna catches. The boundaries of the research area are at coordinates 110.9°-114.5° East Longitude and 8°-11° South Latitude. The data used in the analysis consisted of Sea Surface Temperature (SST) data, chlorophyll-a data, Nino 3.4 data, Dipole Mode Index (DMI) data, and yellowfin tuna catch data for the year 2017-2021. The results showed that of the fifteen GAM models, the combination of variables that most affected fish productivity was the distribution of chlorophyll-a and the ENSO phenomenon with values ??of AIC (1503.33) and DE (64.30%). Pearson correlation analysis showed that the IOD phenomenon was influenced by SST and chlorophyll-a, while SST and chlorophyll-a did not significantly influence the ENSO phenomenon. These results indicated that the phenomenon of climate anomalies and the oceanographic conditions in the waters indirectly affect fish productivity through the food chain process.

Abstract. Aryani F, Kusuma IW, Meliana Y, Sari NM, Kuspradini H. 2023. Potential antibacterial and antioxidant activities of ten essential oils from East Kalimantan, Indonesia. Biodiversitas 24: 665-672. This study aimed to evaluate ten aromatic essential oils' antibacterial and antioxidant activity. Ten aromatic herbs were distilled using the water and steam distillation method. The essential oils obtained was tested for its physical properties, color, refractive index, and specific gravity. The refractive index was measured using a refractometer. On the other hand, specific gravity was measured using a pycnometer. Antioxidant activity testing using the free radical DPPH (2,2-diphenyl-1-picrylhydrazyl) and ascorbic acid was used as the control. Antibacterial activity testing using the agar diffusion method and pathogenic bacteria, namely Salmonella typhi, Escherichia coli, Propionibacterium acnes, and Staphylococcus aureus. Chloramphenicol was used as a positive control and acetone as a negative control. The results obtained essential oils with a slightly yellowish to brownish-yellow color. The highest refractive index and specific gravities were Syzygium aromaticum oil, and the smallest was Litsea elliptica and Eucalyptus pellita oils. The results of the antioxidant activity test carried out at a concentration of 500 µg/mL range from 4.82 % to 85.06 % inhibition and showed that the highest inhibition was Cymbopogon citratus and Syzygium aromaticum by 55.76 % and 85.06%, respectively. Antibacterial activity of ten essential oils showed a varied inhibition zone (0-60 mm) against S. typhi, E. coli, P. acnes, and S. aureus. The oil from Cymbopogon citratus and Citrus hystrix had the highest activity against S. typhi (60 and 21.67 mm, respectively), E. coli (60 and 20.33 mm, respectively), P. acnes (60 and 31.11 mm, respectively) and S. aureus (60 mm and 17.89 mm, respectively). The research results showed that the essential oils in this study had the potency for development as a natural antibacterial and antioxidants agents.

Detection of multidrug-resistant  Staphylococcus aureus and coagulase-negative staphylococci in cow milk and hands of farmers in East Java, Indonesia. Biodiversitas 24: 658-664. Cow milk is a nutrient-dense food containing various nutrients, i.e., carbohydrates, fat, and protein. However, it can potentially spread several pathogenic bacteria, such as staphylococci. Staphylococci cause public health problems. Staphylococcal strains were divided into two groups based on coagulase characteristics, namely coagulase-positive and coagulase-negative staphylococci (CoNS). Coagulase-positive was represented by Staphylococcus aureus. S. aureus and CoNS can become resistant to several antibiotics, known as multidrug resistance (MDR) bacteria. S. aureus and CoNS bacteria are known as causal agents in human infections. This study aims to identify the presence of multidrug resistance S. aureus and CoNS bacteria in several dairy farms in East Java, Indonesia. We collected 332 milk and 125 swab samples from the hand of farmers from several dairy farms in Probolinggo, Tulungagung, and Blitar of East Java, Indonesia. The collected S. aureus and CoNS were tested for antibiotic susceptibility using the disc diffusion method. The antibiotic used were cefoxitin, erythromycin, gentamicin, tetracycline, and oxacillin. The results of isolation and identification based on morphological and biochemical characteristics showed that out of 457 samples, 291 samples (63.68%) were positive for S. aureus, and 166 samples (36.32%) were positive for CoNs. There were 15 S. aureus isolates (5.15%) and 7 CoNS isolates (4.22%) confirmed as MDR; therefore, it is confirmed that S. aureus and CoNS resistance to various drugs was very high in the province of East Java. Intensive livestock systems, high livestock densities, and excessive use of antibiotics in modern dairy systems may predispose the developing growth of MDR S. aureus and CoNS infections. In conclusion, the results of this study indicate the presence of very high multidrug resistant S. aureus and CoNS in the East Java provinces. MDR bacteria can be prevented from spreading by improving biosecurity, hygiene in the milking process, and regular health care for animals and dairy workers.

Stakeholder and social networks analysis of conservation partnership in Bantimurung Bulusaraung National Park, South Sulawesi, Indonesia. Biodiversitas 24: 1017-1024. Social forestry or community-based forest management is increasingly promoted to increase the level of welfare of communities living around the forest, especially in many tropical countries, including Indonesia. One form of social forestry in Indonesia is Conservation Partnership (Kemitraan Konservasi) which is implemented in conservation areas and nature reserves. Since this scheme is relatively new, not extensive knowledge has been accumulated. This study aimed to identify the stakeholders involved in Conservation Partnerships in Bantimurung Bulusaraung National Park, focusing on the communities in Rompegading Village, Cenrana Sub-district, Maros District, South Sulawesi, Indonesia, and to analyze the pattern of social networks of each stakeholder in the management of the national park. Data were collected using observation, interviews, and literature research. We identify eight stakeholders involved in the Conservation Partnerships, namely Bantimurung Bulusaraung National Park, Sonrea Farmer Group, Rompegading Village Government, an NGO named Community Forest Service Team (TLKM), Center for Social Forestry and Environmental Partnerships (BPSKL Sulawesi), Bulusaraung Forest Management Unit (FMU), Faculty of Forestry Hasanuddin University, and a private company named PT. Adimitra. Each stakeholder involved in the Conservation Partnerships has different interests, goals, power, and legitimacy. The three stakeholders with the most significant interest, power, and legitimacy are Bantimurung Bulusaraung National Park, BPSKL Sulawesi, and TLKM. The results of the social network analysis indicate that the interaction among stakeholders is incomplete since not all stakeholders interact with each other. Thus, this study recommends the stakeholders to facilitate training on forest management and institutional strengthening, and also conduct stakeholder meetings to comprehend institutional roles in conservation partnerships as well as a medium for equal information exchange.

Abstract. Al-Miyah SAF. 2023. Estimation the levels of two genes expression and their effects on tetracycline resistance of Acinetobacter baumannii isolated from different sources. Biodiversitas 24: 176-181. Acinetobacter baumannii can cause severe infections of the blood, brain, and lungs. It may also results in infections of the urinary tract and wounds. It can be found on the skin, in food, drink, or soil, and can be transferred by direct contact. The pathogen Acinetobacter baumannii spreads quickly. This study aimed to find a relationship between the infection with Acinetobacter baumannii and the corresponding IL-17 level in patients' sera. To determine the effect of tetracycline exposure on the gene expression levels of the TetA and AdeB genes. This study included 150 samples from different patients suffering from burns, wounds, blood, and sputum. Samples have been tested by Vitek for the presence of Acinetobacter baumannii. Each isolate was then cultured on two separate Mueller-Hinton Agar (MHA) dishes, one with and one without tetracycline. RNA was extracted from all the cultured dishes and converted to cDNA. Quantitative real-time polymerase chain reaction (qRT-PCR) has then been done to evaluate the gene expression level of the TetA and AdeB genes and normalized to the results of the 16sRNA gene. The results of gene expression showed that isolates grown with tetracycline in the medium had significantly higher gene expression than isolates grown without the antibiotic for both the TetA (2.49 vs. 1.023, respectively) and AdeB (5.051 vs. 1.023, respectively) genes. The results of IL-17 showed a higher level in infected patients (21.37 pg/mL) compared to non-infected subjects (14.76 pg/mL).

Abstract. Patana P, Alikodra HS, Mawengkang H, Harahap RH. 2023. State of human tiger conflict around Gunung Leuser National Park in Langkat Landscape, North Sumatra, Indonesia. Biodiversitas 24: 837-846. The Sumatran tiger (Panthera tigris sumatrae) is one of the key species which is considered an endangered species by the IUCN (2008). There has also been a recent threat to their population due to conflict with humans. Information about landscape characteristics and livelihood related to human-tiger conflict (HTC) is needed for a proper mitigation strategy based on local parameters. Therefore, this study was carried out around Gunung Leuser National Park (GLNP) using quantitative and qualitative methods. Principal component analysis (PCA) was used to analyze and map the risk of HTC using landscape characteristics. The physical factors, including land cover, slope, elevation, distance from rivers, and settlement, were analyzed. The livelihood aspect related to land use change was described descriptively and became a major analysis. The study area is located in the buffer zone of GLNP in Langkat District. The results showed that the Eigen weight value for the slope, elevation, and land cover factor was greater than the distance to the river and settlement. The spatial analysis also revealed that Langkat Landscape was in the medium risk of HTC by covering 60% area, while 40% was in the interval of high to very high risk. Furthermore, the high dependence on land for agriculture and the activity of livestock has become a trigger for more massive HTC in the last three years. It is a challenge in human-tiger conflict mitigation to involve the resolution of people’s livelihoods surrounding the national park. The livelihood approach could be a future solution that bridges human and tiger aspects, in addition to habitat management and protecting conservation areas.

Ethnobotanical identification of mango (Mangifera indica L.) and other fruit trees mentioned in Old Javanese Ramayana (10th century Java, Indonesia). Biodiversitas 24: 609-616. Among the 2778 stanzas in Old Javanese Ramayana (OJR), only 46 (1.6%) mentioned fruit trees. There are 63 Old Javanese phytonyms for fruit or fruit tree and only 12 (19%) of them are Sanskrit in origin. Fifty-six species from 44 genera and 26 families are identified, higher than the number of fruit trees carved in reliefs of Borobudur Temple. Of the 56 species, 13 are exotic, specifically from the Indian subcontinent. One of the exotic trees from India that has been naturalized in Java for centuries is Mangifera indica, which is the most often mentioned in the text. Meanwhile, 39 of them are expressly described as fruits that can be consumed raw as food/dessert. It was also found that there are four landscapes, namely hermitage, countryside, forest, and royal garden, which depict the presence of fruit trees. The forest is the landscape with the most mentioned fruit trees in its description. Furthermore, the fruit tree present in the depiction of all landscapes is Mangifera indica. The prevalence of mango trees in these landscapes is probably related to the mango fruit’s edibility as well as its symbolic and religious significance. Other fruit trees, such as Aegle marmelos, Garcinia xanthochymus, Phyllanthus emblica, and Tamarindus indica also have religious significance in Hinduism.

Predicting the current potential geographical distribution of Baccaurea (B. lanceolata and B. motleyana) in South Kalimantan, Indonesia. Biodiversitas 24: 930-939. Baccaurea lanceolata and B. motleyana are underutilized species of Kalimantan fruit tree but are a potential source of food and medicine. However, little is known about the occurrences and potential geographical distribution of B. lanceolata and B. motleyana. This study aimed to predict the potential geographical distribution of B. lanceolata and B. motleyana using MaxEnt, and understand the key factors which influenced their distribution. In addition to 19 bioclimatic factors, occurrence data for 57 B. lanceolata and 87 B. motleyana were gathered from field surveys. Solar radiation, altitude, and slope were then utilized to estimate the distribution of these species. Jackknife test was used to evaluate the importance of the environmental variables for predictive modeling. The Area Under Curve (AUC) values of B. lanceolata and B. motleyana were 0.927 and 0.851, indicating that the model is a good and informative model for species distribution of these species. The models for B. lanceolata suggest that the distribution is mainly influenced by altitude, temperature seasonality, precipitation of the wettest month, and precipitation of the driest quarter. Temperature, annual range, precipitation of the wettest month, precipitation of the coldest quarter, and solar radiation in July were the key environmental factors influencing the distribution of B. motleyana. The potential geographic distribution of B. lanceolata and B. motleyana can be useful information to help the researcher in restoration and conservation planning.

Determinants of symptom variation of Pepper yellow leaf curl Indonesia virus in bell pepper and its spread by Bemisia tabaci. Biodiversitas 24: 869-877. Pepper yellow leaf curl Indonesia virus (PYLCIV) is an important virus that infects chili pepper in Indonesia. PYLCIV has been reported in several regions in Indonesia on several host plants, such as chili pepper, tomato, sweet potato, and ornamental plants. PYLCIV infection has never been reported in bell pepper plants, in 2021 we found yellow mosaic symptoms on bell pepper leaves so this research aimed to conduct molecular characterization on the virus causing yellow leaf curl disease in bell pepper and its relationship to distribution and symptoms variations in the field. The research method includes symptom observation, disease incidence, the population of Bemisia tabaci in the field, and molecular detection via PCR on PYLCIV obtained from bell pepper leaf and on B. tabaci imago. PYLCIV infection in a bell pepper is differentiated into five types of symptoms: mottle, leaf curl and malformation, yellowing, green mosaic, and yellow mosaic. The symptoms type often found in bell pepper plants are yellow mosaic and leaf curl and malformation, with the incidence rates being 19.5% and 21.5%, respectively. The high whitefly population in both symptoms type caused the high incidence rate. Moreover, via PCR, whiteflies on bell pepper with those five symptom types were positive for Begomovirus (B. tabaci viruliferous). Bell pepper positive for PYLCIV appeared to have long fruit ripening periods with small sizes, elongated shaped, and abnormal. While healthy bell pepper showed rapid ripening and large and plentiful fruits. The five symptom types of PYLCIV from bell pepper showed the highest nucleotide homology with PYLCIV isolates from Selulung Village, Kintamani Sub-district, Bangli District, Bali Province, Indonesia, obtained from chili pepper with a homology of 99.7-99.8%. The phylogenetic tree showed that the five symptom types PYLCIV in bell pepper formed a group with PYLCIV isolate from Selulung obtained from chili pepper. This research is the first report of PYLCIV infection on bell pepper in Indonesia.

Morphometric characterization and zoometric indices of female Bali cattle reared in Lombok Tengah District, West Nusa Tenggara, Indonesia. Biodiversitas 24: 966-974. Livestock body conformation can be described by using their morphometric characteristics and zoometric indices. This study aims to provide information about female Bali cattle's (Bos javanicus) morphometric characterization and zoometric indices using principal component analysis (PCA). The study was conducted by using 149 heads of the female Bali cattle reared by smallholder farmers in Lombok Tengah District, West Nusa Tenggara Province, Indonesia. Morphometric characteristics in this study consists of body length (BL), withers height (WH), chest girth (CG), chest width (CW), chest depth (CD), rump height (RH), rump width (RW), head length (HW), and head width (HW). Therefore, the zoometric indices in this study consist of body index (BI), body ratio (BR), cephalic index (CeI), compact index (CoI), conformation index (CnI), depth index (DI), index of compression (IC), height index (HI), height slope (HS), length index (LI), over increase index (OII), thoracic development (TD), transverse pelvic (TP), and weight (W). Data were analyzed by descriptive analysis and factor analysis (principal component analysis/PCA) using SPSS 25.0 program. The average body measurements for BL, WH, CG, CW, CD, RH, RW, HL, and HW were 108.40, 108.35, 148.54, 34.54, 57.50, 106.28, 32.03, 34.66, and 19.10 cm, respectively. The highest correlation coefficient (r) in morphometric characteristics was shown between RH and WH (0.744) and in zoometric indices between BR and HS (0.997). The PCA for morphometric characteristics revealed two factors of PC1 (BL, CG, CW, CD, RW, and HL) and PC2 (WH, RH, and HW), which explain about 67.50% of the total variation. The PCA for zoometric indices revealed four factors of PC1 (CoI, CnI, DI, LI, TD, TP, W), PC2 (BR, HS, and OII), PC3 (BI, IC), and PC4 (CeI), which explain about 90.64% of the total variation. It was concluded that six morphometric characteristics (BL, CG, CW, CD, RW, and HL) were important to describe the body conformation of female Bali cattle and could be used as selection criteria.
