Abstract

Potato (Solanum tuberosum L.) is one of the five basic sources of carbohydrates. It consumed by many people world spreads. One of the constraints in potato production is the presence of fungal diseases. Fungi that cause diseases in potato crops include Phytophthora infestans which causes late blight, Fusarium oxysporum which cause fusarium wilt, Alternaria solani Sor. which cause brown spot disease, and Aspergillus niger which infect bulbs and produce aflatoxin. Identification of potatoes-contaminating fungi can lead the farmers to get a better potatoes production. This research goal is to identify potatoes-contaminating fungi in traditional market of Condong Catur, District of Sleman, Yogyakarta. This research was using descriptive method with laboratory examination. We took 30 defected potatoes suspected of being infected by fungi. Samples were taken from eachpotato aseptically and then cultured in Saboraud’s Dextrose Agar (SDA) media. Observation was held for the fungi growth after 24 hours incubation. From the laboratory examination, there were fungal infections in all of potatoes and identified as Phytopthora infestans (26.67%), Fusarium oxysporum (86.67%), Alternaria solani Sor. (6.67%), and Aspergillus niger (13.33%). Phytophthora infestans, Fusarium oxysporum, Alternaria solani Sor., and Aspergillus niger identified as potatoes-contaminating fungi in Traditional Market of Condong Catur, District of Sleman, Yogyakarta.Keywords: contaminant, fungi, potatoes

Abstract

Banana peels have not been utilized optimally, just disposed of as organic waste or used as animal feed. Ripe banana peel skin is rich of chemical compounds which are antioxidants, both flavonoid compounds and other phenolic compounds. Banana peel contains high antioxidant activity compared to meat, especially “Raja Bulu” banana skin  which has antioxidant equal to 97,85%. Natural source of antioxidants in banana peel skin is very beneficial for health. The purpose of this study was to determine the effect of various drying temperature on antioxidant activity and the optimum drying temperature to antioxidant activity. This study also aims to determine the level of panelist's preference for herbal drinks of “Raja Bulu” banana peel with various drying temperatures. The type of research used is True Experimental Research. The study design was using RAL with 4 treatment P0 (control), P1 (drying temperature 75°C), P2 (drying temperature 80°C), P3 (drying temperature 85°C). Data is included the amount of antioxidant activity content and the average of organoleptic properties of banana peel herbal drink. Data analysis techniques used are One Way Anova and qualitative descriptive. The results showed 1) Various drying temperature effect on the antioxidant activity of banana peel herbal drink. 2) The best treatment is P3 (drying temperature 85°C) with antioxidant activity of 84.87%. 3) The result of the panelist's favorite test for the best color, flavor and taste on P3 treatment with 85 ° C drying temperature.

Abstract

The twin pregnancy is a kind of pregnancy with two or more fetuses. Basically, the twins can be grouped of two, they are fraternal twins (dizygotic) and identical twins (monozygotic). Fraternal twin is a kind of twins that comes from two ova, which each of them will be fertilized by the sperm. While, identical twin comes from an ovum, which fertilized by a sperm. This research is conducted in order to observe the inheritance pattern from the pedigree chart, and to observe similarity and dissimilarity phenotype between twin couple on families observed. Methods used for this research are data collection to construct pedigree, phenotype analysis to compare the similarity and dissimilarity phenotype between the twins that are observed, paraffin method to observe the form of hair from cross section and agglutination reaction to observe the blood group from the twins. Based on pedigree chart of twins in Laweyan, Surakarta, it seems that the identical twins is passedfrom generation to generation, and on the family observed the inheritance pattern of identical twins birth process is come from the women of identical twins. The lobe of ear from each twins there is a twin couple showing the different lobe. There is any similarity and dissimilarity relate to form of hair cross section from each couple. The blood group of each couple are showing the same result.Keywords: identical twins, inheritance pattern, pedigree chart

Abstract

An experiment to induction shoots and rooting of the shoot of yellow bamboo Bambusa vulgaris have been carried out. Single nodal segments with axillary buds were the starting material. After sterilization, nodal segments were directly inoculated on modified Murashige & Skoog (MS) medium, supplemented with cytokinin Benzylaminopurine (BAP) with various concentration (0,0 ppm, 0,5 ppm, 1,0 ppm and 2,0 ppm) and auxin Indole butyric acid (IBA) with various concentration (0,0 ppm, 2,5 ppm, and 5,0 ppm). The results of the experiment showed the combination BAP 1,0 ppm and IBA 2,5 ppm has effectively induced shoot multiplication. Meanwhile, the used BAP 2,0 ppm and IBA 2,5 ppm resulted in the best shoot elongation. On the other hand, rooting of the shoots in vitro have not been successful yet.Keywords: BAP, IBA, induction shoots and rooting, in vitro, yellow bamboo

Abstract

Phalaenopsis amabilis (L.) Blume or Moth Orchid is Indonesia’s native orchid. In its cultivation, the crown rot which is caused by F. oxysporum is one of diseases that usually attack P. amabilis. The aim of this research is to assess the potency of endophytic fungi Sistotrema sp., Ceratorhiza sp. and Moniliopsis sp., that were isolated from healthy P. amabilis roots, in inhibiting F. oxysporum growth in vitro. The experiment of antagonist test was arranged in complete randomized design by three replications. The in vitro antagonist test was done on PDA and the parameter was the percentage inhibitation of F. oxysporum growth by endophytic fungi. The collected data were analyzed by one way anova and DMRT. The result showed that the three endophytic fungi isolated from P. amabilis roots had the potency in inhibiting F. oxysporum. In vitro antagonism test result showed that Moniliopsis sp. had the highest inhibition percentage (71,80%) among Ceratorhiza sp. (67,70%) and Sistotrema sp. (54,87%).

Abstract

Dental caries or commonly known as “Tooth Decay” is one of the most common infection on human. Dental caries begins by the formation of dental plaque and it is a multifactorial disorder. One of the efforts to prevent the formation of dental caries is by utilizing plants which contain antibacterial substance. Pineapple contains bromelain, saponin, flavonoid, and tannin enzymes which functions as antibacteria. This research aimed to discover the antibacterial effectiveness of pineapple juice toward the isolate growth of dental plaque bacteria by using three different pineapple juice concentrations (25%, 50%, and 75%). The method was experimental by using dental bacteria isolate from a patient of Dental and Oral Clinic in Puskesmas Kecamatan Tanah Abang. The result shows that the higher concentration shows the bigger inhibition zone diameter formed around the plate’s paper. Consequently in concentrations of 25%, 50%, and 75%, each has its own inhibition zone rate as much as 2,90 mm (weak); 3,89 mm (weak); and 5,89 mm (moderate).

Abstract

Melon (Cucumis melo L.) is one of the fruits commodity priorities and require more attention. Plant breeding efforts led to improved varieties like TACAPA .TACAPA is a new melon cultivar derived from crosses between ♀ PI and ♂ Action with cross test PI. Since 2012, superior melon reassembled from crosses between ♀ TACAPA and ♂ PI become TANIA and ♀ TACAPA and ♂ Action become TALITA which projection can growth in critical lands karst. The objective of this research was to know ability to grow and morphological characteristics TALITA and TANIA melons were tested to determine nutrient levels of CaCO3. This research started from a field survey conducted in Hills of Mountain Sewu, South Mountain hills Sentolo Kulon Progo, and Dlingo Bantul, Yogyakarta.Melons grown on soil with fertilizer mixed media which further watering treated CaCO3 solution with a concentration of 0.5 % , 1%, 3%, 4%, and 5%. Observations were made until the time of harvest (± 10 weeks). TANIAand TALITA melon could grow on critical lands karst compared with parental. TANIA melonfruits have a weight, round, D - horizontal, D - vertical lower than two parental. TALITA melon fruits have a weight, D-horizontal higher than two parental, and D-vertical is lower than two parental.Keywords: critical land karst, Cucumis melo L., TALITA, TANIA

Abstract

The need for medical albumin is currently at 91%, which is relatively expensive price of albumin causes many people to switch to a cheap source of albumin of snakehead fish. This led to the exploitation of snakehead fish on a large scale so that the fish populations in natural will experience deterioration. Until now, the snakehead fish has not been cultivated by farmers because the fish is not a commodity featured and high production costs of the feed sector. Meanwhile, the potential for additional material feed nutrients are available quite easily and cheaply from the waste tofu. Utilization of bagasse as a mixture of snakehead fish feed can be an alternative to reduce the cost of the feed without reducing the quality of fish in terms of content snakehead fish albumin. In addition, tofu is used as dietary supplement of snakehead fish is expected to reduce the rate of environmental pollution caused by waste agricultural products. The method was used in this research is experimental. Snakehead fish is reared for 17 days with one control and four treatments of feed that is, commercial feed with a mixture of 20%, 40%, 50% and 70% tofu waste. After 17 days of snakehead fish from each treatment were analyzed its albumin levels. The data analysis using regression and correlation to determine the relationship between the treatments of feed against the observed variables. Results of regression and correlation analysis shows that administration of tofu waste in different concentrations has no effect on the feed exhibited significantly to weight and body length of the fish, the oxygen content, pH, water temperature, weight of the test sample and albumin levels in fish flesh. But the determinant of constant value shows that there is one variable that have relevance to the treatment. Based on Pearson correlation test, it is known that high levels of tofu waste given to the feed associated with increased levels of albumin catfish at each treatment. The highest levels of snakehead fish albumin is 4,835 g/100 g of fish meat on the treatment of 70% tofu waste out on feed, while the lowest levels of albumin is contained in the 2286 control treatment that is 0% tofu waste to the snakehead feed.

Abstract

Pepper (Piper nigrum L.) is a commodity of high economic value, but the production is still low. The main reason is the lack of availability for good pepper seedling. Pepper has been commercially propagated by cuttings. To optimize pepper cutting growth could be done through the application of plant growth regulator (PGR). PGR can also be obtained from plant extracts. Some parts of the plant can be used as an exogenous growth regulator hence it contains many hormones that plants need. This experiment was conducted at Teaching and Research Farm, Universitas Jambi to select the best plant extract as an exogenous growth regulator that could induce root and growth of pepper cutting. Five plant extracts, animal urine, and synthetic growth regulator were used as treatment namely: Rootone-F as control, coconut water (Cocos nucifera L.), cow urine, extract of mung bean sprouts (Vigna radiata) + moringa leaf extract (Moringa sp.), young corn (Zea mays) seed extract + moringa leaf extract, young corn seed extract + moringa leaf extract, bamboo shoot extract + banana hump (Musa paradisiaca) extract. The experiment was set in the form of randomized complete design with three replications. Result revealed that synthetic plant growth regulators and plant extracts as exogenous growth regulator have a significant effect on the growth of pepper cuttings. Coconut water gave the best of pepper seedling growth based on the percentage of live seedling, percentage of cutting sprouts, shoot length, root length and shoot dry weight.

Abstract

This research aims to determine the highest protein content and the optimum activity of the enzyme bromelain extracted from pineapple stem. This research was conducted at the Laboratory of Microbiology Makassar Alauddin State Islamic University in May 2013. Variable in this study was the isolation and measurement of bromelain enzyme activity as the independent variable while the crude extract of pineapple stem by variations in pH as the dependent variable. The type of this research was experiments with bradford method for determination of protein content enzyme bromelain, with ammonium sulfate concentration variation for precipitation was 10-60%, as well asthe determination of the enzyme bromelain activity at variation pH was 4.0; 5.0, 6.0; 7.0 and 8.0, at 650C temperature with incubation time was 10 minutes. Each stage has done three times in repetition and analyzed spectrometry. The results showed the highest levels of protein precipitation with ammonium sulfate at 60% was 37,785 mg/ml.
