Abstract

This thesis discusses the detection of koi harpes virus (KHV) in koi carp (Cyprinus carpio L) Application Method Using the Polymerase Chain Reaction (PCR). The purpose of this study is to determine the level of virus attacks causing harpesviridae group KHV Koi carp (Cyprinus carpio L.) using PCR applications in the Central Fish Karantian Hasanuddin Makassar. The research method used is descriptive with pictures PCR of Polaroid photographs. The results obtained with the results of attacks lightly infected 290 bp, the attack was infected with the results of 290 bp and 440 bp, with the result of severeinfection attack 290 bp 440 bp and 630 bp.Keywords: Koi Harpes Virus (KHV), Koi carp (Cyprinus carpio L) and PCR.

Abstract

Molecular Identification of bacterial L-asparaginase activity from Sargassum sp were isolated from Barrang Lompo Island South Sulawesi in 2013. Among them three bacterial strains exhibiting glutaminase free L-asparaginase activity were identified as Pseudomonas putida, Enterobacter asburine dan Marinobacter arcticus on the basis of 16S rRNA gene sequencing. The Highest Max Score 1358 and Query Cover 98 % were Pseudomonas putida furthermore identified as bacterial L-asparaginase activity from Sargassum sp. Pseudomonas putida has Identities 756/765 (99%) with 778 bp Query length, Query cover 98% and Gaps 5/765 (0%).Key words: L-Asparaginase, Sargassum sp..

Abstract

This research aims to determine the highest protein content and the optimum activity of the enzyme bromelain extracted from pineapple stem. This research was conducted at the Laboratory of Microbiology Makassar Alauddin State Islamic University in May 2013. Variable in this study was the isolation and measurement of bromelain enzyme activity as the independent variable while the crude extract of pineapple stem by variations in pH as the dependent variable. The type of this research was experiments with bradford method for determination of protein content enzyme bromelain, with ammonium sulfate concentration variation for precipitation was 10-60%, as well asthe determination of the enzyme bromelain activity at variation pH was 4.0; 5.0, 6.0; 7.0 and 8.0, at 650C temperature with incubation time was 10 minutes. Each stage has done three times in repetition and analyzed spectrometry. The results showed the highest levels of protein precipitation with ammonium sulfate at 60% was 37,785 mg/ml. While the optimum pH of the enzyme bromelain activity at pH 6.0 was 1.021 activity units /gram.Keywords: bromelain enzyme, Pineapple stem (Ananas comosus), precipitation with ammonium sulfate, protein contents, pH

Abstract

This study was conducted to isolate the bromelain enzyme from pineapple weevil (Ananas comosus) and measure the protein and enzyme activity of bromelain with gelatin substrate. Research stage involves determining the protein content of the bromelain enzyme in the treatment of ammonium sulfate precipitation by 10-60% concentration and determination of bromelain enzyme activity at various pH 4.0; 5.0; 6.0; 7.0 and 8.0, with the incubation time of 10 minutes at a temperature of 650 C. Each stage performed three repetitions and analyzed using spectrometry. The results obtained showed the highest protein content in precipitation with ammonium sulfate 60% in the amount of 37.214 mg / ml and the optimum pH of bromelain enzyme activity at pH 7.0 with the value of the activity of 1,081 units / gram.Keywords: ammonium sulfate precipitation, bromelain enzyme, pineapple weevil (Ananas comosus),pH

Abstract

Kunyit merupakan salah satu tanaman obat tradisional yang umum digunakan oleh masyarakat. Kunyit berfungsi menurunkan kadar kolesterol serta berperan sebagai zat antioksidan dan antitoksin sehingga diharapkan dapat mencegah kerusakan kandungan nutrisi pakan dari pertumbuhan dan aktivitas mikroba. Kunyit mengandung minyak atsiri, kurkuminoid, protein, fospor, kalsium, dan vitamin C. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian kunyit dalam pakan terhadap pertumbuhan Mencit putih (Mus musculus L.) hasil perkawinan Outbreading. Penelitian dilakukan di Laboratorium Genetika Fakultas Sains dan Teknologi, Universitas Islam Negeri Alauddin Makassar. Peubah yang diamati adalah litter size, Pertambahan bobot badan, Bobot pra sapih, bobot sapih dan konsumsi pakan. Penelitian bersifat eksperimental dengan empat taraf perlakuan yaitu perlakuan P1 (pakan 100% + 6 g kunyit), P2 (pakan 100% + 9 g kunyit), P3 (pakan 100% + 12 g kunyit) dan P0 atau kontrol (pakan 100% tanpa penggunaan kunyit). Data dianalisis dengan sidik ragam atau Analysis of Variance (ANOVA), jika perlakuan berpengaruh nyata terhadap peubah yang diamati maka dilanjutkan dengan uji Duncan. Hasil mencit menyatakan bahwa penggunaan pemberian kunyit pada pakan tidak berpengaruh nyata terhadap litter size lahir, pertambahan bobot badan, bobot pra sapih, bobot sapih dan konsumsi pakan.  Kata Kunci: kunyit (Curcuma domestica), mencit (Mus musculus L.), outbreading, pakan,pertambahan bobot badan

Abstract

Dalam rangka mendorong adopsi inovasi biosekuriti oleh peternak ayam ras petelur, maka diperlukan pemahaman tentang adopsi inovasi biosecurity ayam ras petelur dan faktor-faktor yang mempengaruhinya. Penelitian ini bertujuan untuk mengetahui tingkat adopsi inovasi biosecurity ayam ras petelur serta faktor-faktor yang mempengaruhinya. Metode penelitian ini adalah pembobotan biosecurity yang terdiri dari biosecurity sumber ayam, biosecurity hewan pengganggu, biosecurity tamu dan pekerja, biosecurity hewan sakit, biosecurity pakan, biosecurity limbah dan biosecurity rak telur, sedangkan untuk faktor-faktor yang mempengaruhi adopsi inovasi biosecurity menggunakan statistika infrensial dengan uji F dan uji t. Hasil penelitian diketahui tingkat adopsi inovasi biosecurity ayam ras petelur di kabupaten Sidrap rendah pada biosecurity terhadap ternak pengganggu sebanyak 59 orang atau 67,82% dan biosecurity tamu dan pekerja sebanyak 59 orang atau 67,82% sedangkan untuk faktor yang mempengaruhi tingkat adopsi inovasi adalah skala usaha dan kontrol perilaku. Kesimpulan dari hasil penelitian adalah ada dua faktor yang mempengaruhi tingkat adopsi inovasi ayam ras petelur di kabupaten Sidrap adalah untuk meningkatkan adopsi inovasi biosecurity dapat dimulai pada skala usaha yang lebih besar dan kemampuan peternak untuk mengkontrol perilakunya.Kata Kunci: biosecurity, faktor adopsi inovasi

Abstract

Penelitian ini dilakukan untuk mengukur pertumbuhan berat badan mencit (Mus musculus L.) dari perkawinan Inbreeding dengan pemberian pakan AD1 dan AD2. Jenis penelitian tersebut yaitu penelitian eksperimental yang menguji pertumbuhan Litter size, bobot prasapih, bobot sapih, pertambahan bobot badan, dan konsumsi pakan sebagai variabel terikat dan perkawinan inbreeding sebagai variabel bebas. Tahap penelitian yaitu penggunaan mencit (Mus musculus L.) 6 ekor jantan dan 6 ekor betina dewasa. Mencit betina berumur 56 hari dengan rataan bobot 24,00 g/ekor dan mencit jantan berumur 56 hari yang digunakan untuk mengawini betina dengan rataan bobot 26,00 g/ekor. Masing-masing 3 pasang untuk perlakuan AD1 dan 3 pasang untuk perlakuan AD2. Pakan yang digunakan pada penelitian ini yaitu pakan AD1 yang mengandung air 13,5%, protein kasar min 20,5%, lemak kasar min 7%, serat kasar max 5%, abu max 7%, calcium 0,9 dan 1,2%, phosphor 0,7dan 0,9%, mengandung antibiotika dan Coccidiostat. Sedangkan AD2 mengandung air 13,5%, protein kasar min 17%, lemak kasar min 7%, serat kasar max 6%, abu max 7%, kalsium 0,9 dan 1,2%, fosfor 0,7 0,9% dan mengandung antibiotika. Hasil yang diperoleh menunjukkan bahwa Liter size, bobot sapih, bobot badan dan konversi pakan tidak ada kecenderungan terhadap pertumbuhan bobot badan mencit (Mus musculus L.) dengan pemberian pakan AD1 dan AD2. Sedangkan bobot prasapih memberikan adanya kecenderungan terhadap pertumbuhan bobot badan mencit dengan pemberian pakan AD1 dan AD2.Kata Kunci: inbreeding, Liter size, mencit (Mus musculus L.), Pakan AD1 dan AD2.

Abstract

Abstract.  Cereals and legumes used as animal feed containing phytic acid which cannot be absorbed by the digestive tract of monogastric animals because phytic acid (C6H18O24P6) is an antinutrient that binds approximately 80% P in feed, also binds to proteins, vitamins and minerals (Mg++, Fe++, Zn++, Mn++, Ca++). Then, one option to overcome this problem is the application of phytase enzymes from various sources, including those produced by bacteria. Phytase enzymes can hydrolyse phytic acid in the feed. This study was aimed to determine the growth phase of Burkholderia sp. HF.7 and optimisation of production of phytase activity from variations of Phytate Production Media (PPM) media. This research used as a descriptive approach. The study design used a completely randomised design (CRD) with a factorial pattern consisting of 2 factors, each variation in phytate sources: calcium (Ca) phytate, rice bran, corn bran, and soybeans. Nitrogen source: (NH4) 2SO4, yeast extract, and peptone. Burkholderia sp. HF.7 Growth phase as a standard for phytase production is the 62-hour stationary phase with an OD value of 2.060 log/cell. The optimum phytase production in the variation of PPM media is soybean-peptone with a protein content value of 46.5 mg/mL and an activity value of 8.20 U/mL under conditions of pH 7 with incubation of 37oC for 62 hours. So, phytase activity produced by PPM of cereal crops has a higher current value compared to PPM Ca-phytate media with low activity value.Keywords: Burkholderia, Phytate, Phytase, enzymes.

Abstract

Indonesia has a very high biodiversity, which has later become one of the natural products of interest to the international community, including fishery products. One of the high-demand Indonesian fishery products is whiteleg shrimp Litopeaneus vannamei. However, safety food Exported whiteleg shrimp products must meet the criteria, including free from bacterial contamination such as Escherichia coli, Salmonella, Vibrio cholera. This study attemptted to analyze E. coli, Salmonella, V. cholerae contamination in 3 ponds in Bojo, Cilellang, and Palanro Village in District Malusetasi, Barru Regency, South Sulawesi. Two samplings for each pond were conducted in the morning were pond water and  fresh whiteleg shrimp. SNI 2728-2018 specifies the quality and safety requirements for fresh shrimp. This standard applies to whole or headless fresh shrimp handled from fresh shrimp and does not apply to fresh shrimp that has undergone further processing. Based on SNI 2728-2018, the E. coli test showed positive in Cilellang Village (sample A) with 11 MPN/g, negative in Palanro Village (sample B) and in Bojo village (sample C) with the value of <2 MPN/g. Escherichia coli test showed positive in sample D (Vannamei shrimp in Cilellang Village) and sample E (Vannamei shrimp in Palanro Village) with 2.0 MPN/g, 17 MPN/g, respectively. Only sample F (Vannamei shrimp in Bojo village ) showed a negative result. As for the Salmonella test, positive results showed in sample A, while sample B and sample C showed negative results. The Vibrio cholerae test showed negative at all samples. . This study concludes that Whiteleg shrimp from ponds in Mallusetasi District is classified as safe for consumption.

Abstract

This study aims to analyze the physical and chemical properties of soil from paddy fields, garden lands and city area also determine the soil inhibitory test of dog saliva bacteria. The results showed that these three soil types have different physical chemical properties and have different inhibitory zones against dog saliva bacteria. The physical chemistry of soil from paddy fields, garden lands and city area (0 cm) are as follows: for color; dark chocolate; grayish brown; and blackish ash, for texture: clay; clay; and sandy clay, for moisture content: 7.14; 6.53; 2.13. for pH close to neutral around 6.52-7.03; Total N is low (0.08-0.23); organic C compounds are classified as moderate; nutrient content of Ca/Mg is classified as moderate (9.55 /1.70 - 14.89/2.22); cation exchange capacity is classified as high (25.32-29.03) in paddy fields and gardens and compounds. The inhibitory zone of the highest dog saliva bacteria is in garden soil (3.7 cm), then paddy soil (2.86 cm) and land in the city area (1.6 cm).
