Abstract

Pyrazolines have been reported to possess  various  biological activities such as antimicrobial,  antidepressant,  antidiabetic,  anti-inflammatory,  antipyretic,  anticancer, and  anticonvulsant  activity. In this research, a halogenated pirazoline 5-(4-bromophenyl)-3-(naphthalene-1-yl)-4,5-dihydro-1H-pyrazole was synthesized  by cyclization  reaction between chalchone with hydrazine  hydrate  using acetate acid as catalyst under microwave irradiation. The compound showed a good yield, and then was characterized by  UV,  IR, 1H-NMR, and 13C-NMR spectroscopy. The antibacterial activity of pyrazolines was determined using the agar diffusion method  against  Gram-negative bacteria:  Escherichia  coli  and  Salmonella  typhi  and  Gram-positive bacteria: Bacillus  subtilis  and  Staphylococcus  aureus. This compound  showed  inhibitory  activity against those four bacteria. Test results of antibacterial activity indicated that pyrazoline analogues were potential as antibiotics.

Abstract

In this research, chalcones  of  acetylpyridine  derivatives  were synthesized  using stirrer method  with  a  base catalyst  (NaOH)  at room temperature. The compound  produced from the  synthesis  was  (E)-3-(1H-indole-3-yl)-1-(pyridine-3-yl)prop-2-en-1-one  (1).  The synthesized compound was  characterized using UV,  IR, NMR and Mass  Spectroscopy.  Its antioxidant activity was tested using DPPH method which  compound  (1)  showed mild antioxidant activity proved by IC 50 > 250 µg/mL.

Abstract

Isolation of cellulolytic bacteria has been investigated because this bacteria producescellulase enzyme which has important role in various industry applications. In this research, isolation of cellulolytic bacteria was originated from sediment samples that was taken from 4 sampling points in estuary watershed of Siak River Bengkalis area and Dumai waters, which were 925 point (Siak River), 929 point (Pelabuhan Roro Pakning), 930 point (Bengkalis Strait), and 931 point (Dumai). Isolates of cellulolytic bacteria were identified with Gram staining method, the enzyme activities were measured with Nelson-Somogyi method. The results of identification showed 1 isolate was Gram positive and 15 isolates were Gram negative. The highest cellulase activity was obtained from 931-3S isolate (0.735±0.03) x 10 -3 U/mL, and it was not significant different with 929-1S, 929-2S, 929-3S, 930-3S isolates.

Abstract

Ferns weed is a plant that is widely available in the oil palm plantation that grows as a parasite on the trunks of palm oil and will result in decreased quality of fresh fruit bunches, These ferns can be used as compost material because it contains enough nutrients.The composting process is done by adding chicken manure and effective microorganism (EM-4) as well as liquid waste palm oil mill (LCPMKS) as an activator.Analysis of compost quality is determined by the parameters water content, pH and C/N ratio for 30 days and analyzed every 5 days composting time.The results showed that the addition of EM-4 activator on kompostidak mixture gives a real difference to the control sample on the value of C / N ratio with a P value> 0.05The compost produced memliliki pH value, water content and C/N ratio, respectively for 8, 40-52% dan17-19 after 30 days of composting time.

Abstract

Cellulolytic bacteria are bacteria that have ability to hydrolize cellulose into smaller oligosaccharides and into glucose units. Glucose is used as a source of carbon and energy for bacterial growth. The aims of this study wasto determine the ability of three bacillus sp. bacteria from rhizosphere of mustard plants (Bacillus sp. B1), rhizosphere of rice plants (Bacillus sp. B2), and peat soil of Giam Siak Kecil (Bacillus sp. B3) to produce cellulase enzymes based on clear zone which is seen surrounding the colonies. Cellulolitic activity of the bacteria was determined by their ability bacteria to degrade carboxymethyl cellulose (CMC) substrate. The results showed that all isolates tested were able to produce cellulase. Bacillus sp. B1 isolate had the highest cellulolitic activity [(6.502 ± 0.462) x 10 -3 U/mL] than 2 isolates based on 24 hourproduction time and120 rpmspeed of agitation.

Abstract

Beef is potential as a medium for microbial growth such as coliform bacteria since it has a high nutrients content. Coliform bacteria in a given amount could be used as a hygienic indicator and a sign for the presence of pathogenic bacteria. One species of coliform bacteria that often contaminate meat is Escherichia coli. Escherichia coli is a normal flora in gastrointestinal tract, but some strains of them are pathogens which cause acute diarrhea. The aim of this study was to calculate the number of coliformbacteria and to detect the presence of Escherichia coli in beef sold in traditional and modern markets in Pekanbaru using the Most Probable Number method. The results showed that all samples were found to be contaminated by Escherichia coli andcoliform bacteria. The number of coliform bacteria in all samples exceeded the maximum threshold of bacterial contamination according to Indonesian National Standard No. 7388:2009, i.e 1x102 MPN/g. The number of coliform bacteria in samplessold at traditional and modern markets were in the range of 0.11 x10 7 MPN/g to >24x10 7 MPN/g and 0.35 x10 8 MPN/g to >24x10 8 MPN/g, respectively.

Abstract

The adsorption ability of activated sulfuric acid Cengar clays (LC0.2, LC0.4 and LC0.6) has been studied using cation of Pb (II) in aqueous solution. The adsorption experiments were observed at each contact time i.e 5, 15, 30, 60, 70, 80, 90 and 120 minutes. All of the experiments were done at a temperature of 30°C and initial concentration of 20 ppm. The amount of Pb (II) adsorbed by the adsorbents was measured by Atomic Absorption Spectrophotometer (AAS). All data was then applied into kinetic models of the pseudofirst order, pseudo-second order and Elovich to determine the adsorption behavior. We found that 0.759, 0.799 and 0.827 mg g-1 Pb were adsorbed by LC0.2, LC0.4 and LC0.6 at 90 minutes contact time, respectively. All of the activated sulfuric acid Cengar clays which adsorbed Pb (II) cations according to pseudo second-order kinetics with  k values (g mg -1 min -1 ) of each adsorbent were 0.156, 0.228 and 0.311, respectively. Fulfillment of the pseudo second-order models has indicated that the process occured was chemicaladsorption.

Abstract

Coliform bacteria are often referred as indicator organisms to indicate the potential presence of disease-causing bacteria. Escherichia coli is a type of Coliform bacteria. Most Escherichia coli are harmless and commonly found in the intestines of humans and animals. However, some strains can cause illness. Coliform bacteria and Escherichia coli contamination in chicken could be detected using Most Probable Number (MPN) multi-step assay which consists of presumptive, confirmed, and completed test. The aim of this study was to determine the number of Coliform bacteria contamination and to detect Escherichia coli in 15 chicken meat samples collected from 3 traditional markets and 3 modern markets in Pekanbaru. Estimated numbers ofColiform bacteria in samples were varied from 2.1x10 4 MPN/g to >11x10 MPN/g. We found that chicken samples were contaminated by Escherichia coli, which showed colour changes and bubles production in EC broth and the appearence of dark centered colony with metallic shine on EMB Agar.

Abstract

Microwave assisted hydrodistillation (MAHD) is the improvement of hydrodistillation (HD) technics which uses microwave energy as source in its refining process. The purpose of this research was to determine the best method of two extraction methods forpatchouli leaves (Pogostemon cablin Benth.). MAHD and HD methods have been compared and evaluated for its effectiveness to isolate the essential oil of fresh patchouli leaves. The results showed MAHD method offered a very important advantage exceeded HD within energy storage and isolation term. MAHD method took time for 1.5 hours whereas HD for 6 hours. The essential oil composition of extraction that determined by GC-MS. The main compounds of essential oil of  Patchouli leaves were patchouli alcohol with levels 53.00% HD and 53.10% MAHD. The density was 0.959 g/mL of HD and 0.963 g/mL of MAHD. Antimicrobial test showed te HD essential oil effectiveness was better than those of MAHD and te  effectiveness on E.coli bacteria. However, the MAHD method was to be considered enviromental friendly.

Abstract

Cananga odorata (Lam.) Hook.f & Thoms plants were one of essential oil sources wich has an antibacterial and antioxidant activity. In the research, isolation of essential oil from Cananga odorata (Lam.) Hook.f & Thoms was carried out with traditional hydrodistillation method and microwave method. The optimalcondition in the traditional method was 4 hours, while the optimal condition in the microwave method were 100o C, 40 minutes and 600 W. Isolation of the essential oil from Cananga odorata (Lam.) Hook.f & Thoms using microwave method produced higher yield (1,08%) compared to the traditional method (0,62%). The essential oil from Cananga odorata (Lam.) Hook.f & Thoms was indentified by GC-MS and resulted 35 compounds for traditional method and 53 compounds formicrowave method. Antibacterial activity assay showed equal result for both methods, that the Gram-negative bacteria (Salmonella enteritidis, Staphylococcus aureus) were more active in inhibiting the growth of bacteria compared to Grampositivebacteria (Escherichia coli, Bacillus subtilis). The antioxidant activity assay of the essential oil from Cananga odorata in the microwave method (884,31 µg/mL) was better than that of the traditional method (883,78 µg/mL).
