Abstract

Heterotrophic bacteria in marine ecosystems have important as decomposer of organic material into the elements essential. This group of bacteria can be found in a variety of habitats including marine sediment. This research aims to isolate the heterotrophic bacteria and the activity against pathogenic (Vibrio alginolyticus, Aeromonas hydrophila and Pseudomonas sp). The research was conducted from November until December 2016. Samples were obtained from the waters of Tanjung Medang, District of North Rupat, Riau Province. Isolation and antagonistic test were conducted at the Laboratory of Marine Microbiology and Biotechnology Department of Marine Sciences Faculty of Fisheries and Marine. In this research was a survey method where heterotrophic bacteria from the sediment were isolated on marine agar medium by pour plate and streak quadran techniques. Activity of the heterotrophic isolate against bacterial pathogens was conducted using diffusion agar on Mueller Hinton Agar (MHA). The number of heterotrophic bacteria ranged from 4,4 × 106 up to 13,5 × 106 CFU/g.Twelve isolates of the sediment bacteria were selected for the antagonism test. However, only nine isolates (Isolate A, C, E, F, O, S, V, W, Z) were able to inhibit the growth of pathogenic bacteria. One isolate (E) performed the highest ability to inhibit the growth of patogens (V. alginolyticus, A . hydrophila and Pseudomonas sp). Keywords: Isolation, Heterotrophic, Sediment, Antagonism, Pathogenic Bacteria.

Abstract

Parathelphusa pardus is a new freshwater crab species found in the swamp area in the Sawah Village. A study purpose to understand the type of food consumed by the crabs has been conducted in October-November 2016. Crab stomach was removed and its content was analyzed by using a microscope. Type of food present was identified and noted and the Index of Preponderance was calculated. The crabs were captured using a traditional fishing line and traps. Results shown that the stomach of P. pardus contains detritus (64,87%), phytoplankton (28,60%), crustacean remains (5,88%), and algae (0,49%). In each molting status, as well as in male and female crab, the food composition of P. pardus is not different. These crabs may accelerates the degradation of litter particles in the swamp, provide feces as natural food for other small organisms and improve the soil oxygenation through their burrows. Keywords : Parathelphusa pardus, stomach content analyis, Index of Preponderance

Abstract

Abstract             There are several human activitiessuch as agriculture and palm plantation are conducted around the Lubuk Siam Lake and thus the remains of fertilizer may enter to the water and affects trophic state in general. To understand the trophic state of the Lubuk Siam Lake’s watersbased on Vollenweider’s Trophic Index (TRIX), a research was conducted in February - March 2017.There were 3 sampling stations namely Station 1 (inlet area), Station 2 (the middle of the lake), and Station 3 (outlet area). In each station there were 3 sampling points, in the surface (50 cm), 2 Secchi depth (126 cm) and 3 Secchi depth (189cm). Sampling were conducted 3 times, once/week. Water quality parameters measured were DIN (Dissolved Inorganic Nitrogen), total phosphorus, dissolved oxygen, chlorophyll-a, transparency, temperature and pH.Results shown that the transparency was 63 – 89 cm, temperature 28.5 – 30.530C, pH 6.83 – 7.27, total phosphorus 0.02 – 0.08 mg/L, DIN  0.45 – 0.73 mg/L, chlorophyll-a12.76 – 44.86 mg/m3, oxygen saturation 52.78%– 103.29%. The TRIX value indicates that the Lubuk Siam Lake can be categorized as mesotrophic to hypereutrophic. Keywords : Water quality, Trophic state, Lubuk Siam Lake, TRIX
