Abstract

Empirically, fermented milk product has been proven to improve and cure for several certain diseases. Bioactive peptides were produced through a fermentation process in goat milk promotes many benefits on body health. The present study revealed that goat milk was fermented using starter commercial 3% to produced yoghurt increased protein content significantly. By using SDS- PAGE, showed that the decomposition of protein fraction was better than the fresh goat milk. Results then were analyzed by LC-MS/MS and found out that there were three kinds of bioactive each peptide consisted of 16 amino acids and were protected from the action of the protease enzyme. Goat's milk yoghurt was treated per oral in mice after hypercholesterolemic diet for 14 days with dose range of 300 mg/kg; 600 mg/kg; and 900 mg/kg for 4 weeks (28 days). The results demonstrated three type of bioactive peptides performed activity as anti-hypercholesterolemia on mice models which showed highly significant (P <0.01) on the production of malondialdehyde (MDA) compared to control. Furthermore, goat milk yogurt also reduced MDA level and decreased fats accumulation in mice. Goat's milk yoghurt at dose of 600 mg/kg was able to provide the best therapeutic effect in lowering MDA level and with dose of 900 mg/kg also gave the best therapeutic effect in reducing the fat accumulation on liver. Based on histopathology observation, it was revealed that fermented goat milk reduced cells damage in liver. In summarize, these findings suggest that fermented goat milk promotes another activities as anti-hypercholesterolemia based on in vivo study.Keywords: Fremented goat milk, hypercholeterolmia, malonyldialdehyde, liver hispathology

Abstract

Cancer is the leading cause of death in Indonesia. Generally, cancer therapy use chemotherapy, radiation, surgery or combination of those. Various side effects caused by cancer therapy make the use of additional drugs. This leads to an increase in total cost incurred to cancer patients. The purpose of this study was to describe the treatment of cancer therapy’s side effects as well as the average cost of side effects treatment, and its percentage in the total cost of cancer patients in Rumah Sakit Margono Soekarjo. This research was a descriptive study with retrospective data by total sampling method obtained from patients’ medical and receipts records of five most common cancers (breast, nasopharynx, Non Hodgkins Lhympoma (NHL), ovarian, and cervix) during January-December 2010. The average cost of patient’s side effects treatment was calculated by the average cost of side effects compared to the average total direct costs incurred to cancer patients. The samples was obtained from the population of 2261 patients who met the inclusion criteria, that are 73 patients consist of 62 female patients and 11 male patients. The most common side effects of cancer therapy were nausea and vomiting (59.80%), nausea-vomiting, anemia, and pain (16.44%), and nausea-vomiting and anemia (15.07%). The most used therapy was vitamin B complex followed by histamine-2 inhibitor. The average cost of side effects for breast cancer was Rp 1,715,373 (8.73%), nasopharynx was Rp 1,149,049 (6.98 %); NHL was Rp 360,778 (1.97 %); ovary was Rp 911,118 (3.61 %) and cervical was Rp 1,416,027 (8.87 %). The results indicated that the hospital needs to pay more attention on nausea-vomiting effect of chemotherapy, and also cervical cancer side effects therapy as the most costly.Keywords: Cost Analysis, Cancer, Side Effects, Prof. Dr. Margono Soekarjo Hospital

Abstract

Curcuma (Curcuma xanthorriza Roxb.) and Awar-awar ((Ficus septica Burm.f.) are well known to have anticancer potential especially for breast cancer with low toxicity. This study aims to examine the potential effect of the combination of Curcuma ethanolic extracts (CEE) and awar-awar leaves ethanolic extract (AEE) to increase the efficacy of cisplatin toward T47D breast cancer cells. The combination activity was done using 3 series of concentration, 1/3; 1/6 and 1/12 of IC50, to determine the combination index (CI) of cisplatin, CEE and AEE under MTT assay. The result showed that the combination of 2.5 µM, 5 µg/ml, 1 µg/ml concentrations of  cisplatin, CEE and AEE respectively result in synergistic effect with CI values less than 1. The treatment exhibited the cell accumulation in S phase (27.7%) against T47D breast cancer cells confirmed through cell cycle examination by flow cytometry. These results provided the evidence that the CEE and the AEE can be developed as co-chemotherapeutic agents combined with cisplatin to improve the effectiveness of breast cancer treatment.Keywords : Curcuma xanthorriza Roxb., Ficus septica Burm.f., cisplatin, cell cycle

Abstract

Propolis contains arginine (amino acids), ferulic acid (a derivative compound cinnamic which promotes the formation of collagen), flavonoids (phenolic compounds), and other compounds that can prevent the growth of bacteria, virus, and fungi. The purpose of this study was to measure the most concentration of propolis extracts that effective to heal burns of New Zealand rabbit's.  This was an experimental study with the post-test only control group design. As many as 15 male white rabbits were divided into 5 groups which treated by  propolis extract  with concentration of 50%, 75%, 100%, positive control by bioplacenton and negative control with propylene glycol. The macroscopic observation was done with detached necrosis tissue and the growth of collagen tissue as the parameter. Data were analyzed by using the one-way ANOVA and continued by the SNK test. The results of this study indicated that the propolis extracted have potency on healing burn wound with the significance of 0.00 (p≤0,05). In the concentration of 100%, extract of propolis has  potency on healing burn wound that proportionates with bioplacenton with the significance value of 1.000 (p≥0,05). It is concluded that by increasing dose, the healing time of burn wound will be shortened.Keywords : Propolis, healing burns, rabbit

Abstract

Cervical cancer is a type of cancer possessing the 3rd highest incidence among women worldwide. Today’s therapies against cancer are still ineffective, thus people are seeking for alternative method. Erythrina fusca Lour. has been used traditionally as anticancer. This experiment is conducted to study the ability of chloroform extract of E. fusca Lour. leaves and its isolates on HeLa cervical cancer cells. The cytotoxicity assay and doubling time assay were done using direct counting method, while the DNA staining assay was done using acrydine orange. The chloroform extract and isolate # 30 showed cytotoxic activity with IC50 of 16 and 5 µg/ml, respectively, while isolate # 2 and isolate # 8 didn’t. The effluent # 30 could not affect HeLa cells growth at the given concentration, but it might promote DNA fragmentation indicating apoptosis induction. Abstract

In order to screen natural antioxidant, the research about antioxidant of some Euphobiaceae herbs, have been conducted. The air-dried herbs  of Euphorbia heterophylla L, Phyllanthus acidus (L.) Skeels, and Phyllanthus buxifolius Muell Arg were extracted with metanol. The obtained extract was concentrated and then suspended to produce n-hexane, ethyl acetat and aqueous fractions. Free radical scavenger activity against DPPH (1,1-diphenyl-2-pycrylhydrazyl) measured by spectrophotometric method  and the IC50 value was determined. The compounds of  active fraction had been identified by TLC method. All of the herbs showed activity as DPPH scavenger. Among these herbs, Euphorbia heterophylla L. and Phyllanthus buxifolius Muell, Arg. exhibited a strong free radical scavenging of ethyl acetat fraction with IC50 value 5,88 µg/ml and 4,64 µg/ml. The result of TLC by mobile phase n-buthanol-acetic acid-water (4:1:5) and acetic acid 15% showed flavonoid compound.Keywords: Euphorbiaceae herbs, antioxidant, DPPH, flavonoid

Abstract

Human Epidermal Growth Factor Receptor 2 (HER2) plays critical role in the initiation of breast cancer cell proliferation. Inhibition of HER2 overexpression is a promising strategy in the treatment of breast cancer. One of the potential natural substances that possess cytotoxic in cancer cells is Secang (Caesalpinia sappan L.). Ethanolic extract of secang heartwood containing brazilein and brazilin shows cytotoxic effect on various cancer cells. Thus this research aims to determine the proliferative inhibition activity of secang heartwood ethanolic extract (SEE) through HER2 expression on MCF-7/HER2 positive. Cytotoxic activity of SEE on MCF-7/HER2 positive cells was observed by MTT assay, meanwhile the inhibition activity of  HER2 on MCF-7/HER2 cells was determined by immunofluoresence assay. Furthermore, to confirm the possible mechanism of brazilein and brazilin for its cytotoxic activity was studied through molecular docking using PLANTS software. Based on this study, SEE exhibited cytotoxic effect on MCF-7/HER2 positive cells with IC50 value of 25 µg/ml, and EES also inhibited HER2 expression, indicated that the antiproliferative effect of SEE was possible through the inhibition of HER2. Docking score of brazilein, brazilin, and native ligand on HER2 were -77; -73; -120 respectively, showed the affinity of brazilein in HER2 was lower than affinity of native ligand, but they had specific amino acid to interaction. Abstract

Heat Shock Protein 27 (Hsp27) is overexpressed in cervical cancer as a response to stress conditions. Hsp27 overexpression increase invasion, migration, and adhesion pathways of cancer cells. The Yogurt supernatant contains Short-Chain Fatty Acids (SCFA) include acetate, lactate, and butyrate which have anticancer activity. This study aimed to investigate supernatant of LBA-ST (Lactobacillusbulgaricus-acidophilus, Streptococcusthermophillus) Yogurt can decrease the expression of Hsp27 in HeLa culture cells. The mechanism on how supernatant yogurt inhibit invasion, migration, and adhesion was studied by immunocytochemistry. The data was then collected and analyzed using One-Way ANOVA. From this study, it can be concluded that the expression of proteins that play a role in invasion, adhesion, and migration of the Hsp27 was proven to be decreased (p< 0.05 ± 0.005).Keywords: HeLa cells, yogurt supernatant, Lactobacillus bulgaricus-acidophilus, Streptococcus thermophillus, Hsp27

Abstract

The Antiproliferative effects of methanol extract from Bruguiera gymnorhiza (B. gymnorhiza) barks were tested in vitro against three human cell lines: Hela, Raji and Myeloma cells. The extract was found to have antiproliferative effects against Hela, Raji and Myeloma cells with an IC50 value of 133, 504 and 384 µg/mL, respectively. The antiproliferative test was then performed on Hela, Raji, and Myeloma cells. Cytotoxicity assay of the extract was then determined using MTT method. There were some indications of apoptosis, such DNA fragmentation, as assessed by acrydine orange- ethidium bromide staining. These results indicate that extract from B. gymnorhiza barks can induce apoptosis in human cell lines.Keywords: Antiproliferative, Apoptosis, B. gymnorhiza

Abstract

Guttiferron F and essential oil were isolated from extract n-hexane of Garcinia cf bancana Miq. Antioxidant activity was done by DPPH method to  n-hexane, methanol, ethyl acetate, n-butanol extracts and Guttiferron F, showed activity with IC50  24.5, 22.4, 29.2, 37.6 and 25.8 µg/mL respectively.
