Abstract

Crossbreeding program was an option in order to improve Indonesian local goat productivity. In general, crossbreeding is aimed to utilize the benefit ofheterosis (hybrid vigor: HV); where better offspring performance compared to its parents phenotype is expected. Currently, crossbreeding between Boer bucks and  Jawarandu does is widely practised; resulting in a crossbred called Boerja. However, information regarding the HV effect were less available. Thus, this study aims to calculate HV in Boer × Jawarandu in relative to the mid-parental average, and relative to the dam phenotype. Mature body weight (BW) of 504 head of goat derived from Boer buck (n=6), Jawarandu doe (n=15), Boerja F1 (n=298), Boerja F2 (n=167), and Boerja F3 (n=18) were analyzed. Backcross system was applied to produce Boerja F2 (Boer ♂ × Boerja F1 ♀)  and Boerja F3 (Boer ♂ × Boerja F2 ♀). Result shows that BW in Boer, Jawarandu, Boerja F1, Boerja F2, and Boerja F3 were 79,00 ± 16,21; 29,07 ± 3,76; 41,84 ± 4,12; 41,69 ± 3,68 and 39,96 ± 4,97 kg respectively. The heterosis value of Boerja F1, Boerja F2, and Boerja F3 in relative to the parents average were -23, -31, and -34%, while in relative to the dam were 44, 0, and -4% respectively. According to HV value, it is concluded that the most optimum result of Boer × Jawarandu crossbreeding was found in Boerja F1. Higher Boer breed proportion in F2 and F3 were found to have lower BW.

Abstract

This research was conducted to assess the level of substitution of napier grass with brown seaweed (Sargassum polycystum) in diet of the goat on rumen metabolism product and nutrient digestibility in vitro. The experimental design used was completely randomized design with 4 treatments and 4 replications. Feed treatments consisted of T1 (control) = 50% napier grass + 50% concentrate; T2 = napier grass 37,5% + S. polycystum 12,5% + 50% concentrate; T3 = napier grass 25% + S. polycystum 25% + 50% concentrate; T4 = napier grass 12,5% + S. polycystum 37,5% + 50% concentrate. The result, substitution of napier grass with brown seaweed (S. polycystum) did not significant (P>0.05) effect on pH, N-NH3, VFA total, acetate, propionate, butyrate, adenosine triphosphate (ATP), dry matter digestibility, organic matter digestibility, and neutral detergent fiber digestibility. It is suggested that brown seaweed (S. polycystum) could replace napier grass on goat feed at up to 37,5%.

Abstract

The purpose of this research to determine the pH value and the antimicrobial activity of bilimbi extract (LBW) as a substitute for antibiotics. The method was field experiment using Completely Randomize Design with 4 treatments and 6 replications. The treatments were consist of P0(+) = antibiotic, P1= LBW 10%, P2= LBW 20% and P3= LBW 30%. Bilimbi extract used from bilimbi powder which is then dissolved. The measured variables were pH value and inhibition zone diameter of Escherichia coli, Salmonella sp. and Lactic Acid Bacteria. The data were analyzed by ANOVA and continued by Duncan’s Multiple Range Test (DMRT). The result of this research showed that LBW with higher concentration produces low pH value and the treatments between variation of concentration gave significantly different effect (P<0.01) on diameter of inhibition zone for Escherichia coli, Salmonella sp. and Lactic Acid Bacteria. The conclusion of this research was 30% concentration of LBW produces the lowest pH value, so that it can be inhibit the bacterial growth and has the potential as a substitute for antibiotics based on the result of antimicrobial activity that produces the best activity.

Abstract

Mayonnaise is popular sauce. It’s kind of semi solid oil in water (o/w) emulsion which containing oil, acidifier and pasteurized egg yolk as an emulsifier. The consumers have demanded that for natural, healthy, and low calorie food especially for development and manufacture of low fat mayonnaise. The common emulsifiers are egg yolk, casein, and whey. Kefir can replace egg yolk to develop low fat mayonnaise. The use of egg yolk be reduced and kefir was used to develop a low fat mayonnaise as emulsifier replacer to egg yolk. The purpose of this research attempts to observe the emulsion stability, antioxidant, and oxidation stability of low fat mayonnaise prepared using rice bran oil and kefir during storage. The research methods using experimental design. The result shows the addition of kefir can create low fat mayonnaise by increasing emulsion stability.

Abstract

Whey is liquid cheese waste produced from the cheese making process after being separated from the curd. Cheese production waste contains a carbon source, one of which is lactose as an energy source in the development of fermented biotechnology, especially the manufacture of probiotics for livestock. Lactose (4-7%) and protein (0.6-1%) in whey cheese can be used as a medium for the growth of lactic acid bacteria and yeast in producing secondary metabolites which could be applied as nutraceutical feed additives that were useful for medical or health, including prevention and treatment of diseases and improve livestock performance.The research purpose was to determine the effect of adding kefir grain 1-5% into whey cheese based on chemical quality and microbiological. Whey Cheese is mixed with 10% molasses and then heated at 80oC for 10 minutes later the temperature is lowered between 35-40 oC and added starter according to treatment. The next step is incubating anaerobic kefir fermentation for 24 hours with temperatures between 35-40oC. The experimental method was designed with a completely randomized design with 6 starter level treatments and 3 replications, namely 0%, 1%, 2%, 3%, 4%, 5%. The results showed that the addition of 1-5% kefir grain was very significant (P<0.01) increasing protein, lactic acid bacteria, yeast and reducing water, fat, Total Plate Count (TPC). It can be concluded study concludes that the addition of 5% kefir grain into whey cheese added with 10% molasses affects the increase in lactic and yeast acid bacteria, protein and decreases fat, water, Total Plate Count  (TPC).

Abstract

The aim of this study was to determine increasing of chitinase crude enzyme activity produced by Trichoderma viride at different gamma irradiation doses and to know the degradation on the eggshell of Haemonchus contortus. Eggs of Haemonchus contortus were isolated from sheep feces using buoyant centrifugation method. The qualitative test of chitinase was conducted by inoculating Trichoderma viride irridiated by  variation gamma doses control, 250 Gray, 500 Gray, 750 Gray and 1000 Gray on the media of colloidal chitin agar. The quantitative test was conducted by measuring chitinase activity using colorimetric method based on specific enzyme activity and determination of dissolved protein levels. Shell changes were observed microscopically while N-acetyl glucosamine levels and dissolved protein levels were measured using spectrophotometer. Data were analyzed using One Way Analysis of Variance (ANOVA) with significance of the 95% and continued with Duncan’s Multiple Range Test  (DMRT test). The results showed that gamma irradiation doses treatment increase of chitinase crude enzyme activity produced by Trichoderma viride. At a dose of 500 Gray, Trichoderma viride had the optimum chitinase specific activity i.e. 0.30 U/mg. Furthermore, chitinase crude enzyme effective to degrade all layers of the eggshell of Haemonchus contortusand the higher the concentration of crude enzyme given, the greater the level of egg damaged.

Abstract

Subclinical mastitis in Ettawa crosbreed goat (PE) is economic disease which lost milk yield that bacteria caused. The aim of these research was to isolatian and identification bacteria that caused subclinical mastitis in PE goat from Kemiri Kebo, Turi, Sleman, Yogyakarta. A total of 24 goat milk PE subclinical mastitis samples, were used in these research. Determination of subclinical mastitis in PE goat used CMT test which (++) and (+++) results. Isolation and identification conventional bacteria based on biochemical. Subclinical mastitis PE goat in Kemiri Kebo, Turi, Sleman, Yogyakarta was caused Staphylococcus intermedius (38%), Staphylococcus aureus (32%), Staphylococcus epidermidis (25%), Staphylococcus hycus (12%), Streptococcus sp (22%), Corynebacterium sp (12%), Pseudomonas aeruginosa (18%), and E. coli (2%). The research showed that Gram positive bacteria is dominant that caused subclinical mastitis in PE goat compared than Gram negative bacteria.

Abstract

The objective of this study was to determine the relation between morphometric of different parity and litter size in Ettawa Grade goat. The material of this study was 47 heads of Ettawa Grade goat. Purposive sampling was applied to determine sample based on population and breeding policy. The Ettawa Grade used were does from1-4 parities and having litter size records from first kidding. Morphometric characteristics measured were chest depth, chest width, hip width, hip height, heart girth, body length, and withers height. The data obtained were analyzed using General Linear Model (GLM) and Principal Component Analysis (PCA) of Statistycal Analysis System (SAS) Ver 6.12. The results showed that morphometric of Ettawa Grade goat was vary and different in various parity. The PC1 of body length and heart girth were 0.685 and 0.530 respectively, indicating the most discriminant variabel to determine the differences among parity. Body index of Ettawa Grade goat does did not affect the litter size. In conclusion, body length and heart girth can be used as the differentiation parameter among parity. Litter size will increase with increasing parity.

Abstract

Ongole Crossbred cattle or more popular known as Peranakan Ongole (PO) cattle is one of local breeds cattle in Indonesia. For improving the population and genetic quality, superior bull with high capacity to produce semen is seriously required. This study aimed to find out the estimation value of semen production of PO bull based on body weight and scrotal circumference. A total sampling of 9 bull candidates of PO cattle available at National Artificial Center (NAIC) Singosari, Malang, Indonesia were used in this study. The method used was case study. Scrotal circumference was directly measured as a primary data. While body weight, semen volume, semen concentration and sperm motility were collected from BBIB Singosari from secondary data. Data was analyzed using simple linear regression. The results showed that with the limited number of samples, there is no significant correlation (P>0.05) between body weight and scrotal circumference on semen volume, semen concentration, and sperm motility.  It was concluded that with limited number of samples involved in this study, there was no significantly on semen production based on body weight and scrotal circumference on PO bull. The suggestion was to improve the correlation value and significant result. The number of samples should be increased.

Abstract

Coconut water extender is the solution to the difficulty and high cots of procurement of diluents. The objective of this study was to examine the effect of various formulations of coconut water extender on quality of PO bull sperm during storage in 4-5 oC. This study was carried out at Animal Reproduction Laboratory of Faculty of Animal Husbandry Brawijaya University and Laboratory of Beef Cattle Research Center Grati Pasuruan on March to April, 2018. The study method used laboratory experimental. The experimental design was Factorial Randomized Completely Block Design and the data were analyzed by Analyze of Variance. There are four treatments in this study (P0 = CEP-3 + 20 % of yolk, P1 = Coconut Water + 20% of yolk, P2 = Coconut Water + 20% of yolk + 0,4% white egg + fructose 1 mg/ml, dan P3 = Coconut Water + 20% of yolk + 0,4% white egg + fructose 2 mg/ml) and ten replications of each treatment. There were significant differences in sperm motility between CEP-3 extender and coconut water-based with various formula extenders during chilled storage at 2-5 oC. But the treatment between coconut water-based formulas did not show a significant difference. Coconut water extenders are able to maintain sperm motility up to day 5 with values above 57%.
