Abstract

Chloramphenicol (CAP) is a broad spectrum antibiotic that has been banned in many countries due to its serius side effect to human. Detection of CAP residue in food has been determined to a minimum required performance limit (MRPL) of 0.3 ng/g. The purpose of this research was to conduct the analysis of CAP residue in bovine meat by using LCMS and to study the presence of CAP residue in marketed bovine meat samples. LC separation was done on a Shimpack column C18 with ammonium acetate 10 mM/water as mobile phase, and ESI-MS analysis in negative ion mode. The coefficient of determination, R2 = 0.9981 at concentration of 0.125, 0.25, 0.63, 1,00 and 2.00 ng/g. Recovery at three fortification levels (0.25, 0.50 and 1.00 ng/g) was in the range 77.5, 97.3 and 83.4%. The decision limit and the detection capability were 0.15 ng/g and 0.17 ng/g respectively. Analysis results of 52 marketed samples showed that CAP residue were detected in 9 samples in the concentration range of 0.14 to 2.70 ng/g and 6 among those positive samples were above the MRPL value. Therefore, it is important to increase the awareness and also to monitor regularly CAP residues in food originated from animal to provide safe food for the consumers. Abstract

A preliminary study related on vaccine safety and vaccination effectivity for controlling avian influenza (AI) subtype H5N1 was carried out at Virology Laboratorium, Indonesian Veteriner Institute, Bogor. A Prototype of inactivated vaccine was made using AI H5N1 local isolate (A/Chicken/West Java/67-2/2003). The vaccine was then tested for safety and protection in DOC of layers. Antibody response, protection and shedding virus challenge were observed in the experiment. Result showed that the vaccine was saved and protected against virulent viral challenge. Efective vaccination was achieved at 3 weeks chicken old started with low level of antibody. Antibody titre increased gradually and reached the top at 8 weeks post vaccination. Challenge test using AI virulent at the age of 4 and 8 weeks post vaccination showed that the vaccine gave high protection (90%). Viral shedding was not longer expressed than 7 days after challenge. It is concluded that this prototype is a satisfied AI vaccine in laboratory level.     Key Words: Vaccine, Avian Influenza, H5N1, HPAI

Abstract

In processing of buck semen, seminal plasma is a problem because it contains a phospholipase A enzime produced by the Cowper gland. If this enzime interacts with egg yolk, it causes semen coagulation, and consequently death of spermatozoa. The purpose of this research was to examine the effect of Priangan ram seminal plasma on viability of Peranakan Etawah (PE) buck spermatozoa preserved at 3–5oC. Semen was collected using artificial vagina once a week. Fresh semen was divided into three tubes then centrifuged at 3,000 RPM for 30 min. Supernatant of the first tube was mixed again with Pasteur pipette (treatment A or control). Supernatant of the second tube was removed (treatment B or without seminal plasma). Supernatant of the third tube was removed and changed with Priangan ram seminal plasma in the same volume (treatment C). Semen was diluted with Tris extender containing 20% egg yolk and stored in refrigerator at 3–5oC. Quality of diluted-semen including percentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) was evaluated every day during storage at 3–5oC for three days. Results of this study showed that mean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS, spermatozoa abnormal, and IPM of PE buck fresh semen, respectively was 0.68 ml, cream, thick, 7, ++/+++, 4,148.57 million cell/ml, 70%, 83.89%, 7.12% and 84%. At day-4 of storage, percentages of MS, LS, and IPM for treatment C (40, 52.2 and 51.6%) was significantly (P<0.05) higher than that of: treatment B (31, 44.8 and 45.2%) and treatment A (11, 15.6 and 14.8%). In conclusion, seminal plasma of Priangan ram could maintain the quality of PE buck semen preserved at 3–5oC for three days, and it prevent semen from coagulation. Abstract

An experiment was conducted to evaluate the effect of the inclusion of different levels of shrimp waste meal fermented with Aspergillus niger (LUF) in diets on growth performance and digestive organ weight of broilers. A total of 75 d-old chicks were randomly allocated to 5 (five) treatments in 3 replication pens of 5 birds each. Treatments consisted of LUF inclusion of 0 (control), and 5, 7.5, 10 and 12,5% (P0, P1, P2, P3 and P4) in the diets. Birds were raised under standard condition and provided with feed and water ad-libitum. Feed and birds were weighed weekly up to 35 days to determine body weight, feed intake and feed conversion. At the end of experimental period, the birds were slaughatered and dressed up to determine carcass percentage and digestive organ weight. There was a significant negative linear response in body weight, feed consumption and feed conversion with increase of LUF more than 7.5% in the diets until 35 days of age, but % carcass was almost the same for all treatment. There was no significant response in digestive organ weight with increasing levels of LUF. The present result indicated that LUF could be considered as a potential feed ingredient as protein source of broiler but its inclusion should be limited until 7.5% of the diet to maintain growth performance and digestive organ weight. Abstract

The solid state fermentation technique on palm kernel meal by using Aspergillus niger wild type and NRRL 337 was studied. The fermentation was carried out at 30oC for 3 days continued with enzymatic process at room temperature and 40oC for 2 days. The result showed that at the third days of fermentation spores started to grow on the surface. The 3rd days fermentation can improve protein content and in vitro digestibility (IVDMD) with minimal loss of dry matter. The dry matter contents at 3 days fermentation were 48.88 and 48.83% for product using Aspergillus niger wild type and NRRL 337. Combination 3 days fermentation and 2 days enzymatic process at room temperature by using Aspergillus niger NRRL 337 type gave the best product with optimal IVDMD value and protein digestibility of 51.47 and 71.33%.   Key words : Palm kernel meal, fermentation, Aspergillus niger

Abstract

Observation on the antibody fluctuations of infected cattle with metacercariae of Fasciola gigantica and the effect of triclabendazole treatment were made by means of ELISA technique. Seven cattle were infected with 700 metacercariae and one cattle remained uninfected, as negative control animal. Treatment with triclabendazole was given to 6 cattle, when the mean antibody levels of infected cattle reached the peak, and the other one remained untreated, as positive control animal. One week after treatment the mean antibody levels started to drop and then decreased gradually. After eight weeks of treatment, the mean antibody levels of the treated cattle reached the lowest level or the same value as before infection, thereafter, 6 cattle were reinfected with different dosages of metacercariae of F. gigantica. Two cattle were infected with 400 metacercariae, the other two were infected with 600 metacercariae and the rest of them were infected with 800 metacercariae. The results showed that the immunological responses of re-infected cattle are quicker (5 weeks after infection) and the peak of the antibody levels are higher (ELISA OD = 1.7) than after the first infection (11 weeks after infection and ELISA OD = 1.2). However, after re-infection, there were no significant different on the antibody fluctuations and antibody levels among the infected group, although those cattle received different dosages.   Key words: Antibody, cattle, Fasciola gigantica, ELI

Abstract

Productive and reproductive performances of 85 heads of Ettawah-cross (Peranakan Ettawah or PE) does at first breeding were studied at the Research Institute for Animal Production, Bogor . They were fed freshly-chopped King grass (Pennisetum put7xtreophoides) and corn-husk (2 :1) ad libitum and concentrate feed was given at a rate of 300 - 400 g/head/day . During the last month of the pregnancy and lactation period the amount of concentrate feed was increased to 500 - 700 g/head/day . Sixty-five does were in pre-pubertal stage and 86 .2% reached puberty at liveweights between 16 - 21 .8 kg (mean 18 .8 t 0 .4 kg) which is about 60% of mature liveweight . Oestrus without ovulation was found in 6% of the animals which has contributed on the relatively low conception rate (64 .7%) . A high pre-weaning mortality of kids born (37 .5 %) caused production inefficiency that suggests the challenge to improve management practices . Milk yield of PE does varied widely (0 .3 - 0 .8 kg/day), hence, there is a chance for improvement towards increasing milk production in Indonesia through well planned selection program.

Abstract

Most of the components of poultry feed (80%) of grains and meal that contains phytic acid which has anti-nutritional factor because it can bind minerals and reduce its availability. Phytic acid can be hydrolyzed by the enzyme phytase. Phytase enzyme naturally found in sugar cane juice, but its use as poultry feed supplements have not been done. The study was conducted using sugar cane juice PS 851 from Jatiroto PTPN XI, Lumajang, East Java in order to get the information potential of sugar cane juice as a liquid supplement and phytase enzyme carrier for poultry viewed from the aspect of nutrient content of sugarcane juice and phytase activity in the release rate of phosphorus. Research conducted at the Faculty of Animal IPB for 10 months. The rate of hydrolysis of phytase on P was tested using rice bran as a substrate. Sugar cane juice is added to the 2.5% level, using 4-level incubation (1, 2, 3 and 4 hours), each level consisting of 37°C and 42°C; pH 2; pH 4.5 and pH 5 with three replications. Study using a Two Factors Experiments in Completely Randomized Design and it was continued by DMRT test. P release rate was measured by spectrophotometry. The results showed that the sugar cane juice has a phytase activity of 0.0766 U / ml, brix level of 22.15%, containing water 73.03%, protein 0.47%, crude fiber 6.43%, minerals Ca 0.03%, P 0,02%, Co 0.14 mg / l, Fe 1.8 mg/l, Mn 1.55 mg/l, Zn 1.37 mg/ l, Cu 0.19 mg/ l, Se 12.63 mcg/100 g, vitamins B3 5.26 mg/100 g, C 0.72 mg/100 g, E 0.08 mg/100 g, sucrose 32.42%, fructose 2.41%, galactose 2% and glucose 1.58%. Supplementation of 2.5% sugar cane juice can increase the P release rate of 112-235% at optimum conditions of pH 5, at 37°C with a long incubation period of 1-4 hours. Abstract

Basic information on reproductive potency of Garut rams is necessary in order to identify the capacity of rams in producing chilled or frozen semen. Eight Garut rams (three to five years old) were used in this study. The male sexual behaviors were observed and semen was collected once a week using artificial vagina. Semen quality was evaluated and its potency to produce frozen semen was calculated. Results of this study indicated that first, second, and third ejaculations were at the 29, 87 and 176th seconds, respectively. Fresh semen volume, sperm concentration, motility, intact acrosomal cap, and intact plasma membrane were 0.99 ml, 3224 million/ml; 76.67; 86.13 and 87.73%, respectively. Protein value, fructose, vitamin C, vitamin E, sodiu, potassium, calcium, magnesium, phosphor, chloride, and mangan in seminal plasma of fresh semen were 4140, 180, 3.2, 24, 180, 117, 9, 6.12, 60, 104, and 5 mg/ml, respectively. Measurement of head length, width, and length of sperm tail were 6.59, 3.99, and 42.65 μm, respectively. Length and width measurement of right and left testes, and scrotal circumference were 12.71, 6.5, and 32.36 cm, respectively. Capacity of each Garut rams to produce frozen semen from three consecutive ejaculations are 35.88 mini straw with the cencentration of 200 million motile sperm per 0.25 ml.   Key words: Reproductive characteristics, Garut rams

Abstract

The effect of fermentation process and duration of storage of fermented coconut meal with Aspergillus niger on its peroxide number has been studied. First stage of experiment was arranged in a completely randomized design with four treatments (raw coconut meal, 0 and 4 day aerobic fermentation incubation and after 2 day anaerobic enzymatic incubation). The peroxide number was determined as potentiality for rancidity. The three incubation processes decreased the peroxide number significantly. The highest reduction was obtained at 4 day aerobic fermentation (49.7%). The decrease of peroxide number of fermented coconut meal was correlated with the decrease of lipid content. The regression coefficient is highly significant (r2 = 0.76, P<0.01). In the second experiment, the effect of storage on the peroxide number of fermented product was conducted by factorial design 2x3x4 i.e., two type of substrates (fermented and non fermented coconut meal), 3 storage temperatures (-13, 4, and 29oC) and 4 storage times (1, 2, 3, and 4 months). Statistical analysis for lipid contents showed that there was significant interaction (P<0.05) between kind of substrates and storage time. The lipid content of fermented product was decreasing from the first up to the third month of storage, then it was increasing at the fourth month, but the lipid content of non fermented coconut meal did not significantly changed (P>0.05). Statistical analysis for the peroxide number showed that there was highly significant interaction between the type of substrates and storage temperature. The peroxide number on the fermented products did not significantly increase, where as the non fermented products show significantly increased during storage course. The significant interaction between storage duration and temperature observed. At low temperatures (-13 and 4°C) storage the peroxide number increased 44%, while at high temperature (29°C) up to 95%. The highest peroxide number on the fermented product (43.5 ppm) was obtained at the temperature storage of 29°C for 4 month period. This value is still under rancidity limit (80 ppm). It could be concluded that fermentation process reduced the rancidity of coconut meal.   Key words : Coconut meal, fermentation, peroxide number, Aspergillus niger
