Abstract

An experiment was conducted to identify the effectivity of antioxidant supplemented in ration for fat and fatty acid consentration and sensory quality of male local duck meat. Two hundred and eighty day old ducks (dod) reared for ten weeks were allotted to either one of 10 treatments in 4 replications with 7 ducks/replication for each treatment. The treatments were: (R0) basal-diet (RB) without antioxidant (control); (R1) RB+50 ppm santoquin (Sq)+100 IU vitamin E (VE); (R2) RB+50 ppm Sq+200 IU VE; (R3) RB+50 ppm Sq+300 IU VE; (R4) RB+100 ppm Sq+100 IU VE; (R5) RB+100 ppm Sq+200 IU VE; (R6) RB+100 ppm Sq+300 IU VE; (R7) RB+150 ppm Sq+100 IU VE; (R8) RB+150 ppm Sq+200 IU VE; (R9) RB+150 ppm Sq+300 IU VE. The experiment was conducted based on completely randomized design with one of the treatments as a control and the others were with antioxidants supplementations. The variables observed were: fat concentration, fatty acid composition and concentration, as well as sensory quality (off odor intensity and description values) in male duck meat. Result showed that Sq+VE supplementation could reduce fat content in duck meat. R1 and R4 were the best level to reduce fat contents in fresh meat, but in boiled meat R8 and R9 treatment was the best. Total composition and the ratio of unsaturated fatty acids were higher then those of saturated fatty acids. The fatty acid concentrations of linoleat (C18:2) and arakidonat (C20:4) increased parareled with  the antioxidant supplementations in fresh and boiled duck meat. Sq and VE supplementations significantly decreased (P<0,05) intensity off odor (fishy odor) in fresh or boiled duck meat, and 50 ppm Sq+100 IU VE or 100 ppm Sq+100 IU VE was the best treatment to reduce the intensity off odor (fishy odor)on fresh meat, while 150 ppm Sq+200 IU VE or 150 ppm Sq+300 IU VE was on boiled duck meat. It is concluded that lipid oxidation was effectively protected by Sq+VE supplementations resulted is reduced intensity off odor, while the sensory quality of local duck meat increased. Abstract

Kuantan cattle is one of local beef cattle breed of Riau Province which its origin was unknown. Kuantan cattle are commonly found in Indragiri Hulu and Kuantan Singingi Regency. Based on phenotype characterizations, kuantan cattles are similar with pesisir cattle (West Sumatera beef cattle). Historically, kuantan cattle were pesisir cattle brought by “minang” immigrants (Immigrant from West Sumatera) to this region. The purpose of this study was to analyze the origin of the kuantan cattle through genetic diversity analysis using DNA barcode. DNA barcode used was Cytochrome oxidase subunit I gene which was found in the mtDNA. DNA isolation was done on 25 kuantan’s blood samples and 18 pesisir blood samples. Amplification of COI gene segment used Polymerase Chain Reaction technique. The forward primer sequence used in this study was F’5 TTCTCAACCAACCATAAAGATATTGG-3’ and the reverse primer sequence used was reverse 5’-TAGACTTCGGGGTGTCCAAAGAATCA-3. It squeezed kuantan and pesisir sequence 5711 - 6420 base (GeneBank accession number NC_005971) with length by 710 bp. Analysis result of sequence using MEGA 5.2 Program showed that there were 6 polymorphic sites establishing 7 haplotypes on kuantan cattle and 9 polymorphic sites establishing 12 haplotypes on pesisir cattle. Based on genetic distance and phylogeney tree, kuantan and pesisir cattle were in same group with Bos indicus. Mutation in the COI gene segment in this study was too small and was not able to distinguish the difference of those breeds. The result of neighbor joining analyze indicated that kuantan cattle origin was from Bos indicus just like pesisir cattle.Key Words: COI Gene, Polymorphic, Kuantan Cattle, Genetic Distance, Phylogenetic Tree

Abstract

Oxidative stress (a condition where oxidant -free radical- activity dominates the antioxidant activity) in poultry may be caused by heat stress from high environmental temperature. In this research, antanan (Centella asiatica) and vitamin C was utilized as anti heat-stress agents for broilers. One hundred and twenty male broilers of 2 – 6 weeks of age were kept at 31.98 ± 1.940C poultry house temperature during the day and 27.36 ± 1.310C at night. The treatments consist of two kinds: two levels of vitamin C (0 and 500ppm) and three levels of antanan (0, 5 and 10% of diets). The data colected was analized for variance with a factorial in completely randomized design of 2 x 3 (2 levels of vitamin C, 3 levels of antanan) and continued with contrast-orthogonal test when applicable. The results indicated that the treatments of antanan 5% without vitamin C (A5C0), antanan 10% without vitamin C (A10C0), vitamin C 500ppm without antanan (A0C500), combination of A5 and C500 (A5C500), and combination of  A10 and C500 (A10C500) significantly (P<0.05) decreased the level of liver malonaldehida (MDA), carcass lipid and plasma cholesterol. Abstract

The objective of this research is to evaluate an application of an inactive Newcastle disease (ND) vaccine derived from velogenic strain of local Newcastle disease virus (NDV). In this research . the Ira strain of velogenic ND virus was grown in specific pathogen free (SPF) eggs and then was inactivated by formalin at a final concentration of 1 :1,000 at 4°C. The inactive antigen was then emulsified with an oil adjuvant or aluminium hydroxide gel before being administered for vaccination in layers and compared to a commercial inactive ND vaccine . Results indicated that application of these inactivated ND vaccines for booster vaccination following vaccination with an active lentogenic ND virus in pullets nearly producing eggs, resulted in high antibody titre which persisted for considerable long period of time and capable of protecting layers from sick of ND and from reducing egg production . Hence, it could be concluded that the inactivated vaccine emulsified in either oil-adjuvant (lanolin-paraffin) or aluminium hydroxide gel were considered to be highly immunogenic and capable of protecting layers from sick of  ND and from reducing egg production   Keywords : Newcastle disease, inactive vaccine, oil adjuvant . aluminium hydroxide, immunogenicity, protection

Abstract

chemical composition (nitrogen/N; neutral detergent tibre/NDF; acid detergent fibre/ADF; hemicellulose/HC; lignin and silica) and IVOMD (in vitro organic matter digestibility). Straws from eight varieties obtained from Yanco Agricultural Institute, Leeton, N.S.W. was used. Straws were cut into upper and lower part in equal length, then chopped. Chopped straw from each varieties and each part was treated with urea at 4% DM. The experiment used an 8x2x2 factorial design. Results showed that the chemical composition and IVOMD varied between varieties. Before treatment with urea, in all varieties the N content was higher in the upper (8.1-11.1 g/kg) than the lower part (5.8-8.3 g/kg). The IVOMD of the lower part was higher than that of the upper part except for lIb, and Yrl varieties. The IVOMD of the lower part untreated straw ranged from 325 - 498 g/kg whereas in the upper part it ranged from 325-439 g/kg. Urea treatment consistently increased the N content and IVOMD of both parts in all varieties. After urea treatment there was no significant difference in IVOMD between upper and lower part in any variety. The increase of IVOMD in response to urea treatment was higher when the original quality of straw is low where the increase IVOMD is 53% (from 325 to 499 g/kg). There was no consistent effect of urea treatment on the other chemical components. This study concluded that the urea treatment would be more beneficial for use with low quality rice straw.   Key words: Rice straw, variety, urea treatment, chemical composition, in vitro organic matter digestibility

Abstract

To investigate the effect of intravenous glucose infusion on nitrogen (N) retention, two experiments were conducted in 2 phases . The first phase (Exp 1) was to establish the time required for a stable glucose entry and the second phase (Exp 2) was designed to study the effect of different levels of glucose infusion on N retention . In Exp l, four wethers were used, each prepared with catheters in the left and right jugular veins, continuously fed luceme chaff at calculated maintenance energy level . The animals were infused continuously with saline for 2 days and thereafter with glucose solution at the rate of 21 .8 mmole/h for 6 days and again with saline on the seventh day. Glucose entry rate (GER) were measured using constant infusion of D-[6-'H] glucose, on the second day of saline infusion (Day 0) and at the same period each day for the next seven days. Infusion of glucose reduced endogenous glucose entry by 30% on Day 1 and 2, by 13% on Day 3, but by Day 4 onwards endogenous glucose entry had returned to normal levels. Plasma insulin, reached its peak value on the first day of glucose infusion and subsequently declined . Plasma urea concentration declined with ensuing days of glucose infusion . It was concluded that for sheep fed maintenance energy level infused with glucose, it takes approximately 4 days for glucose metabolism to reach equilibrium. In Exp 2, four wethers were used, each prepared with catheters and fed luceme pellets at calculated maintenance energy level . The sheep were infused continuously for five days with glucose at a rate of either 0 (saline), 10, 20 and 30 mmole/h in a Latin Square design (4 infusion rate x 4 periods) . A large increase in endogenous glucose entry was found with the highest level of infusion . Glucose entry rates were 28 .8, 48.3, 54 .7 and 86.1 mmole/h for glucose infusion of 0, 10, 20 and 30 mmole/h respectively . No significant changes in both plasma glucose and urea concentration with infusion rate of glucose. N retention increased with increasing level of glucose infused (P<0.05) and was mainly due to the reduction of urinary N. Abstract

In ovo immunisation of chickens with flagellin and whole cell protein antigens of Campylobacter jejuni was examined to determine Campylobacter infection. Four groups of embryonated chicken eggs (10 eggs per group) were immunised in ovo at day 17 of incubation and booster was given at 7 days post-hatch. Group I was immunised in ovo and oral booster with whole cell protein of C. jejuni, group II was immunised in ovo and oral booster with C. jejuni flagellin protein, group III was immunised in ovo and intraperitoneal booster with whole cell, and group IV was treated as control. The humoral immune responses were determined by enzyme-linked immunosorbent assay (ELISA) and the mucosal immune responses were examined by a direct fluorescent histology antibody technique. Immunised chickens of Group I, II, and III shown to have higher antibody titers than  those of control chickens (group IV). The titres of anti-campylobacter antibodies of all isotypes in serum, bile, and intestinal scrapping after challenge were not significantly different in all groups. In addition, when immunised chickens were orally challenged with a homologous strain of viable C. jejuni organism, the chickens remained infected throughout the experiment based on cloacal swabs and caecal contents. These findings indicated that although in ovo immunisation resulted in increasing of the mucosal and humoral immune responses in chickens, it is not strong enough to protect the Campylobacter colonisation in the intestinal tract.   Key words : In ovo immunisation, Campylobacter jejuni, whole cell, flagellar, chickens

Abstract

The experiment was conducted in Cilebut Experiment Station, Bogor, with the aim to improve the productivity of Kacang goats. Twenty male goats, 9-10 months old, with an average initial liveweight of 16.27 kg were used. The goats were randomly divided into four treatments with five animals each, and were kept by grouped pen system. The dietary treatments were: (R0) = Elephant grass (EG) + concentrate (C0), (R1) = EG + (95% C0 + 5% molasses), (R2) = EG + (90% C0 + 10% molasses), (R3) = EG + (85% C0 + 15% molasses), respectively. The Elephant grass was offered ad libitum while the concentrate was offered at 3% of total body weight per treatment group. The experiment was carried out for 14 weeks. Feed intake per treatment group, individual weight gain, and feed conversion were analyzed. Average daily gain was analyzed in a completely randomized design. The results indicated that R0 (0% molasses), R1 (5% molasses), R2 (10% molasses) and R3 (15% molasses), did not significantly affect the average daily weight gain (P>0.05) with an overall mean of ADG to be 55.6 g/head/day. Feed intake and feed conversion were not significantly different among dietary treatments with an average dry matter intake of 2,621 g/5 heads/day or 524 g/head/day; protein intake was 370 g/5 heads/day or 74.0 g/head/day with energy intake of 10.929 Mcal/5 heads/day or 2.186 Mcal/head/day, and feed conversion of 9.43. In conclusion, the fact that molasses addition to rations did not affect feed intake, daily weight gain and feed conversion indicating that the molasses addition did not improve the Kacang goats fattening productivity.   Key words : Productivity, goats, molasses

Abstract

Fermentation processes increase the protein of palm oil sludge and produce some useful enzymes. However, drying process, especially with heating often affects the nutritive values of feed ingredients and the enzymes activity. Therefore, this experiment was designed to study the responses of native chickens when fed ration containing fresh or dried fermented palm oil sludge (FPOS). Experimental diets with different levels of fresh or dried FPOS (5, 10 and 15% equal to dried FPOS) were formulated with similar nutrient contents. A control diet with no FPOS was also included. Each treatment was replicated 5 times for 12 weeks. Performances (body weight, feed consumption, feed conversion and mortality) of chickens were recorded. Carcass yield, abdominal fat content, weight of liver and gizzard were measured at the end of feeding trial. Data were subjected to analysis of variance and different between means were tested by orthogonal procedures. Results during the starting period showed that, chickens fed FPOS diet gain more weight significantly than the control birds (298.1 vs. 264.7). The dry matter intake of the dry FPOS diet was significantly better than the fresh FPOS diet (2.88 vs. 3.32). The FPOS dietary levels did not affect body weight gain and dry matter intake of native chickens, significantly. However, increasing the FPOS levels worsen the dry matter conversion (DCR) significantly. Data during 12 weeks trial showed mat the body weight gain was not significantly affected by treatments. The dry matter intake of the FPOS diets were significantly higher than the control diet (3469 vs. 3065 g/bird), hence the DCR of the control diet was significantly better than the FPOS diets (3.28 vs. 3.62). Feeding dry FPOS resulted in a better DCR as compared to fresh FPOS (3.48 vs. 3.76), but not affected the dry matter intake. The FPOS dietary levels did not affect body weight gain and dry matter intake of native chickens, significantly. However, inclusion of 5% FPOS in me diet gave better DCR significantly, as compared with 15% FPOS (3.51 vs. 3.83). Feeding dry or fresh FPOS (5 - 15%) did not significantly affect the mortality, carcass yield, abdominal fat levels, weight of liver and weight of gizzard of native chickens. It is concluded that dry FPOS was better than the fresh FPOS and could be included in me native chickens diet up to 10%.   Key words: Palm oil sludge, fermentation, native chickens

Abstract

One hundred and seventy six adult female cattle ticks, Boophilus microplus, were collected to observed the influence of their body development on their eggs fertility under laboratory conditions (22°-32°C and 84-92% relative humidity) . The regression analysis result shown that as the body developed, eggs production and eggs fertility increased, but there was no correlation between ticks weight and the incubation periode . The relation between ticks weight and eggs production was shown as Y = -309 .433 + 13 .868X and r = 98 .5 % whereas the influence of ticks body development on eggs fertility was shown as Y = 8 .224 + 0 .338X, r = 92 .0% and the incubation periode was about 21-26 days . The fertile eggs begin to be produced by ticks with body weight of about 35-49 mg and fertility 28.39%.
