Abstract

Mangosteen fruit with latin name Garcinia mangostana L. got the nickname Queen of tropical fruit (Tropical Fruits queen). Most of the mangosteen fruit is consumed in fresh condition. Mangosteen rind purple black. Mangosteen rind allegedly containing anthocyanin compounds. This study aimed to determine antioxidant activity of bark extracts of the mangosteen fruit and the total anthocyanin content in the rind of the mangosteen fruit. Object of research is that the mangosteen rind has been good to consume. Mangosteen rind mashed and then soaked with methanol containing 1% HCl. Antioxidant activity test mangosteen rind extract includes a qualitative test of antioxidant activity of extracts of TLC plates sprayed mangosteen rind 0.1 mM DPPH reagent produces patches of pale yellow with a purple background. Quantitative test of antioxidant activity based on the parameters EC50 using DPPH method with the comparison of vitamin C. Obtained EC50 value of 8.5539 tg/mL and EC50 values of vitamin C is 3.3676 tg/mL. Average total anthocyanin content in the skin of the mangosteen fruit is 59.3 mg in 100 grams of the mangosteen rind.

Abstract

Quorum sensing is a communication system among bacterial cells which correlates with biofilm formation. Biofilm can protect bacteria from environment including antibiotic of which can cause higher antibiotic concentration of 100 up to 1000 times. Inhibition of quorum sensing is expected to inhibit the biofilm formation. The cinnamon bark (Cinnamomum burmanii Ness. Ex Bl.) has been known to have antibacterial and antibiofilm activities. Leaves are available abundantly which urges a research to find out the activity as anti quorum sensing against Pseudomonas aeruginosa. The succesive maceration of dried pulverized leaves produced hexane, ethyl acetate, and methanol extracts.  Antibacterial activity was observed by microdillution method with MTT assay. Afterwards, the active extract was examined for anti quorum sensing activity by diffusion method in cetrimide Agar. Quorum sensing activity was shown by dark zone (opaque) growth around sample application, observed under UV light of 366 nm. TLC bioautography method was done to observe the active spots by using silica gel F254 as the stationary phase, chloroform-methanol (6:1 v/v) as the mobile phase, loading sample used was 1.25 mg and 30 min of plate contact duration. The ethyl acetate extract inhibited growth of  P. aeruginosa with shown by MIC at 8 µg/µL Quorum sensing as well as growth inhibition activities were observed at loading samples 12,5 and 25 mg/wells, while at 6,25 mg the extract only exhibited quorum sensing inhibition. Presences of substances having phenolic, flavonoid, alkaloid and aldehyde/keton as functional groups were detected by TLC method of the extract but no active spot identified on bioautography.

Abstract

Taman Nasional Gunung Merapi (TNGM) has various biodiversities which need to be explored. This study aims to analyze antioxidant activity four plants from Asteraceae, Rubiaceae and Polygalaceae which were collected from TNGM. Antioxidant activity is analyzed by DPPH radical capturing method using spectrophotometer. Sampel is extracted gradually by wasbenzen and methanol. DPPH radical capturing analysis shows that IC50 score of Cynara scolimus L. wasbenzen extract is 496,16g/mL; Artemisia china L. 407,86g/mL; Borreria repens DC. 984,04 g/mL and Polygala paniculata L. 633,34 g/mL. The IC50 score of methanol extract Cynara scolimus L. is 334,94g/mL; Artemisia china L. 199,99g/mL; Borreria repens DC. 402,28 g/mL and Polygala paniculata L. 77,00 g/mL . Thin layer chromatography analysis shows that methanol extract of Polygala paniculata L. contains flavonoid.

Abstract

Pasak bumi (Eurycoma longifolia) has the potential to be developed as antihypertensive, antipyretic, aphrodisiacs and health supplements. The use of E. longifolia as a traditional medicine needs to be pursued in the form of more effective and appropriate formulation. The capsule preparations are easy to make and cancover the bitter taste of E. longifolia. Clinical trials in this study use design pre-post treatment in healthy humans. Subjects used were male - healthy men and healthy women who met inclusion criteria and were subjected with formulated capsule for 14 days. The study resulted capsule formula comprising of the ethanolic extract of E. longifolia 300 mg, vivapur 101 300 mg, 58 mg maydis starch, aerosil 3%, talc 2%, and Mg stearate 1%. The results showed that the capsule of E. longifolia did not affect the value of heart rate, respiration rate, body temperature and weight (P > 0.05), based on paired t-test, but they causes a decrease in blood pressure of healthy human. The ethanol extract of E. longifolia caused vasodilation of blood vessels that can be used in antihypertensive therapy.

Abstract

Kecapi (Sandoricum koetjape (Burm.f.) Merr) was one of the traditional medicine from  family Meliaceae and had compounds such as flavonoids, saponins and polyphenols that can be used as antifungal. The objective of this research to know compounds contained from S. koetjape bark that has potential as an antifungal toward C. albicans.Research carried out two stages using  extraction by maceration with methanol and fractionation by column chromatography method used the solvent n-hexane, chloroform, ethyl acetate and methanol. The next step was examined its activity toward C. albicans and identification of fraction active compounds with the Gas Chromatograpy-Mass Spectrometry (GC-MS). The results of this research showed that methanol extract from S. koetjape bark had antifungal activity againstC. albicans (39,654%). Next, the methanol extract was fractionated and fractionation results were obtained six fractions with the greatest antifungal activity of fraction III, followed by fraction V (39.66% b/v), fraction IV (39.02% b/v), fraction II (37.91% b/v), fraction VI (35.48 % b/v) and fraction I (33.72% b/v). Identification of compounds with GC-MS in fraction III and compounds contained have potential as an antifungal are α-gurjunene, trans-caryophyllene, aromadendrene, α-humulene, β-caryophyllene, δ-Cadinene, alloaromadendrene, octadecanoic acid (as. stearat), hexadecanoic acid metil ester (metil palmitate), hexadecanoic acid (as. palmitate), 9-octadecenoic acid metil ester (metil elaidate), 9-octadecenoic acid (as. oleat).

Abstract

Sumbawa horse milk is one of pure milk of wild horses farmed by organic farmers in the island of Sumbawa, West Nusa Tenggara, Indonesia. The communities claim that milk has many benefits. In this study Sumbawa horse milk was effective as an antibacterial against Staphylococcus epidermidis (one of the bacteria caused acne). Antibacterial activity test performed by spread plate method and quantified by standard plate count. Percentage of the number of colonies after contact with  Sumbawa horse milk for 5 minutes (used acid control). The result of 25% is 99.96%, 12.5% is 99.99%, 6.25% is 99, 67% and 3.125% is 99.73%. This suggests that  Sumbawa horse milk has good antibacterial activity until 3.125%.

Abstract

Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower (Hibiscus rosa-sinensis L.) with a concentration of 0.6% w/v has mucolityc activity which is equivalent to 0.1% acetylcysteine based on the capasity to decrease viscosity of the mucus. The use of hibiscus as a new cough medicine done traditionally. Syrup of ethylacetate insoluble fraction containing alkaloids was the appropiate liquid dosage form that has some advantages for examples easy to use and fast absorption of the active substance. The study aims to determine the optimum formulation syrup ethanolic fractions containing alkaloids from hibiscus flowers using the method of Simplex Lattice Design. Ethylacetate insoluble fraction containing alkaloids from the hibiscus flower is obtained by using the Vacuum Liquid Chromatography (VLC). Simplex Lattice Design Methods used to optimize the formulation syrup ethylacetate insoluble fraction containing alkaloids from hibiscus flower with seven formulas with variations in the amount of glycerin, sorbitol solution 70%, and mucilago 0.5% CMC-Na. Physical stability was analized to find higher total response value (R total) as parameter of the optimum formula by Simplex Lattice Design Methods with Design Expert ® software version 8.0.2. Optimum formula syrup ethylacetate insoluble fraction containing alkaloids from the hibiscus flower obtained with the proportion of 25.376% glycerine, 51.985% of 70% sorbitol solution, and 22.639% of 0.5% CMC Na mucilago. Data obtained physical characteristics test compared with prediction value using Design Expert ® software version 8.0.2. Physical characteristics of optimum syrup ad the zero week compared with physical characteristics on the forth week using paired t-test. The result showed that physical properties of syrup formula optimum didn’t different significantly for easy for pouring and tasted response except for viscosity and acidity. Ethylacetate fraction syrup containing alkaloids from hibiscus flower optimization results are less stable during the four weeks of storage.

Abstract

The role of TGF-β1 is known as the main immunosuppresor associated with tumor, but on the other opinion, it is associated with proliferation and tumor invasion. The increase and decrease of the secretion of TGF-β is to regulate the proliferation, differentiation, and death of various cell types. Now we all know the extract of Jatropha curcas L seed serves as antitumor. Allegedly, it can regulate the expression of TGF-β1 in control of cell number. The purpose of this study is to determine the effects of Jatropha seeds to the regulation of gene expression of TGF-β1 as a tumor marker. The method is performed by giving a dose groups the extract of jatropha seed (0, 5, 25, 50, 250 mg/BB) in mice. Then measurement of mRNA expression (RT-PCR), the protein of TGF-β1 levels (ELISA), and qualitative observations of liver histology were done. The expression of TGF-β1 mRNA is significantly 4.39 to 7.34 times higher than (ANOVA, p 0.05) than the control. Histological observation of liver showed the extract of jatropha seed induces damage nucleus of hepatocytes cell and sinusoidal. The effects extract of jatropha seed increased the level of TGF-β1 mRNA but not followed by increasing protein of TGF-β1 levels, and it was stimulated necrosis and apoptosis of hepatocytes cell.

Abstract

The purpose of this study is to determine the effect of sugar level variations (sucrose and glucose) to the physical properties and flavor, as well as to determine the level of consumer preferences on hard candy lozenges of Legundi leaf extract. Hard candy lozenges are produced using the fusion method to create three formulas contain combination of sucrose and glucose, which is FI (70%: 30%), FII (50%: 50%), and FIII (30%: 70%). Hard candy lozenges were tested the physical properties include: uniformity of weight, hardness, moisture content test, a soluble and TLC-densitometry and responsive taste test using a questionnaire with scoring assessment by respondents. The test results were analyzed using SPSS 11.0 for windows with a significance level (α) = 0.05. The results showed that the variation of sucrose and glucose levels affect the physical properties of hard candy lozenges of Legundi leaf extract that increases the hardness level, and increase the time to dissolve (p <0.05) but did not affect the water content of the tablet (p> 0.05). The more sucrose is used it increase the hardness of the tablets and dissolve into the longer time. Based on the perceptive sense test results subjectively of hard candy lozenges of Legundi leaf extract, Formula III is the most favored by respondents with sucrose and glucose ratio of 30%: 70%.

Abstract

The increasing therapy problem including multidrug-resistant tuberculosis (TB) has made it important to discover a new anti-TB drug candidate. The aim of this study was to acknowledge the activity of ethyl acetate extracts of kenikir (Cosmos caudatus H.B.K) and sendok (Plantago major L.) leaves against Mycobacterium tuberculosis (M. tuberculosis) H37Rv. This research used Middlebrook (MB) 7H9 media and observed the growth of M. tuberculosis using Lowenstein Jensen (LJ) media. The concentration of extracts were 0.25 mg/ml, 0.50 mg/ml, and 1.00 mg/ml. The result of this study showed that ethyl acetate extracts exhibited anti-TB activity in 1000 µg/ml of both extracts. The active compound group was detected by thin layer chromatography (TLC) and the separation of compounds was shown by retardation factor (Rf) and the color of the spots. Based on TLC chromatograms, it is known that there are types of compounds, such as ortho-dihydroxy compounds, phenolic compounds, and compound leads to terpenoids for both extracts.
