Abstract

The aim of the study was the determination of glycaemic index (GI) of local tubers such as Uwi (Dioscorea alata), Gadung (Dioscorea hispida), dan Talas (Colocasia esculenta). Tubers were prepared as raw flour which were administered orally to the rats. Blood were taken at 0; 0,5;  1 and 2 hour after tubers administration. The dose was 2,5 kg/kg body weight (BW) whereas glucose was administered as a standart treatment. Area Under Curve (AUC) was calculated after plotting the graph of time versus glucose levels. Glucose levels were determined using GOD-PAP kit. GI values were calculated as a ratio of AUC (0-2hr) of tubers to glucose. Porang was moderate while the other tubers were low GI.

Abstract

This study aims to determine the bioactivity of L.domesticum as cosmetic ingredient. The extraction was prepared by maceration with ethanol 96 % and ethyl acetate to obtain ethanolic extract of fruit flesh (BLE), ethanolic extract of fruit peel (KLE), ethyl acetate extract of fruit flesh (KLE) and fruit peel (KLEA). Then, determination of compounds was conducted by thin layer chromatography (TLC) and analysed of total phenolic and flavonoid content with colorimetric method. The antioxidant capacity was measured using radical scavenging assay of 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching assay (BCB) and ferric reducing antioxidant power assay (FRAP). The inhibitory effect of skin degradation enzymes (anti-aging) was carried out using elastase and collagenase assay. The tyrosinase inhibitory effect was determined using mushroom tyrosinase. Based on TLC result, extract of L.domesticum containing phenolic, flavonoids, terpenoids and steroids compounds. The largest of total flavonoid content was obtained in KLE (4,76±0,26 %b/b EQ) and phenolic total content in BLEA (6,4±0,15 %b/b EG). Sequence of antioxidant activity measured using DPPH method was BLEA> KLE> BLE> KLEA; while using BCB method and FRAP method were respectively KLEA>BLEA> KLE> BLE; and BLEA> KLE> BLE> KLEA. KLE and KLEA extracts showed the most potent to remaining activity of the enzyme elastase and collagenase. In addition, BLE extract has the higest tyrosinase inhibition than others. Based on these results it can be concluded that the extracts of the fruit flesh and fruit peel of L. domesticum can be used as a cosmetic active ingredient for anti-aging and anti-tyrosinase.

Abstract

Hydroalcoholic extract of turmeric is recommended as supplement to prevent atherosclerosis, but the common dosage form in traditional practices is less practical and unacceptable. It is therefore recommended to be mad into new dosage form, lozenges. This research aims to determine the effect of filler agent manitol-amylum manihot variations to the physical characteristics of granule, tablet, and the flavor of the tablet. The optimum formula was evaluated with simplex lattice design method. The hydro extract of turmeric was made using aquadest 80OC while the alcoholic extract using ethanol 96%. Hydro alcoholic extract was made by mixing both extracts, wet granulated and formulated into five formulas with combination of manitol-amylum manihot ratio. The result showed that the combination of manitol-amylum manihot had affected the tablet hardness, solubility time and flavor acceptance. The formula with the combination manitol-amylum manihot 80% : 20% was found to be the optimum formula.

Abstract

Iron deficiency anemia is occurred due to lack of iron within the body, so that the amount of iron for erythropoiesis does not meet the requirement. Petai (Parkia speciosa Hassk) which is widespread in Indonesian archipelago, contain high existence of iron is supposed to be able to increase hemoglobin level in white mouse blood (Rattus norvegicus). The research was carried out by using petai from Wonosari whose made to dry extract by using freezedryer. Petai seed extract (Parkia speciosa Hassk.) injected to Wistar rat (Rattus norvegicus). Data analysis was using One Sample t-Test and ANOVA test in order to find out the effect of the doses towards pharmacological effect. The result showed that petai seed extract (Parkia speciosa Hassk) gave effect towards the enhancement of hemoglobin level (Hb) within white mouse blood (Rattus norvegicus). However, the optimum dosage was not able to be determined yet.

Abstract

Sponges is one of the invertebrate Porifera phyla that produce active compounds with various structures and one of the biological activity as antimicrobial. The purpose of this research was to find out the sponge extracts that can inhibit microbial growth and potential of antimicrobial active compounds from sponge at Barrang Lompo Island. The microbes used were Escherichia coli, Salmonella typhii, Staphylococcus aureus, and Candida albicans. The sponge extracts were obtained by maceration 14 sponge samples with methanol, followed by partition using chloroform and methanol, then TLC-bioautography toward the active extract. The results showed that methanol extracts of sponge with code BRLP-009 and 010 have the most potential effect as antimicrobial agent.

Abstract

Siam citrus peel (Citrus nobilis L. var. Microcarpa) is a plant derived from Sambas Regency, West Kalimantan Province. Bioavailability of herbal active compounds can be enhanced by formulating extract into nanoparticle. The polymer used was chitosan with crosslinker Na-TPP. Stirring speed and stirring time play an important role to produce small particle size in forming nanoparticle using ionic gelation method. Enhancement of stirring speed and stirring time could reduce particle size. Nanoparticles were prepared using ionic gelation method by mixing Na-TPP, extract and chitosan (1:1:6) with varying the stirring speed 500 rpm, 1000 rpm, 1500 rpm and stirring time 1 hrs, 2 hrs, 3 hrs. The particle size of nanoparticle was found to be 85.3 nm at 1000 rpm of stirring speed and 3 hrs of stirring times, with polidispersity index 0.287, zeta potential +32.37 mV and entrapment efficiency 87.12 %.

Abstract

Costus speciosus (Koen.) J.E. Smith (Pacing) is a plant widely use as traditional contraception. The objective of this study is to evaluate the effect of pacing leaves infusion on Balb/C mice spermatogenesis and its reversibility. This study was conducted on male mice. Mice was divided into 6 groups, 6 mice each group. Pacing leaves infusion was given orally at dose 275, 550, and 1100 mg/kg body weight for 14 days. The control groups received distilled water and another group received corn oil and andriol 5.2 mg/kg body weight. At day 15, 3 mice from all of groups was euthanized and the rest of mice in all of groups was euthanized in day 28 to evaluate the reversibility of spermatogenesis. The observation of spermatogenesis was conducted on quantity and quality of spermatozoa (motility, viability, and morphology). The result showed that pacing leaves infusion at dose 275, 550, and 1100 mg/kg body weight reduced the number of spermatozoa by 16-38 % (p<0.05), but it did not change the viability and morphology of the spermatozoa. At dose 275 and 550 mg/kg body weight, pacing leaves infusion reduced the motility of spermatozoa by 36-39 % (p<0.05). The ability of pacing leaves infusion in reducing the number and motility of spermatozoa is reversible.

Abstract

On Timor island, Nusa Tenggara Timur, faloak barks (Sterculia quadrifida R.Br.) has been used empirically to restore stamina. Faloak bark ethanolic extract proved to have immunomodulatory activity in vitro, which can increase macrophage phagocytosis activity. This research aimed: (i) to determine the immunomodulatory active fraction of faloak bark ethanolic extract, (ii) to determine the total flavonoid contents of faloak extract and fractions, and (iii) to evaluate the correlation of the total flavonoid contents of those extract and fractions with their macrophage phagocytosis activity. The simplisia powder is macerated with 96% ethanol. The extract was dissolved in methanol:water (9:1v/v) was then subsequently partitioned with n-hexane, ethyl acetate, and water to obtain n-hexane fraction, ethyl acetate fraction, water fraction, and insoluble fraction. Faloak extract and fractions at concentration 62,5; 125; 250; 500μg/mL were tested for their effect on the peritoneal macrophage phagocytosis of Balb/c mice in vitro by the latex beads method. Phagocytosis capacity and phagocytosis index were analyzed using one-way anova and post hoc Tukey HSD test with 95% confidence level. The results showed that ethyl acetate fraction had the highest macrophage phagocytosis capacity and the highest total flavonoid content compared to other fractions. The highest macrophage phagocytosis capacity of ethyl acetate fraction at concentration of 250 μg/mL was 51,94±4,67%, this value was significantly different from cell control (7,50±1,29%), negative controls of 0,0625% dimethylsulphoxide (6,25±0,36%), as well as positive control of 200 μg/mL echinaceae extract syrup® (9,97±0,33%). The total flavonoid content of ethyl acetate fraction determined by aluminum chloride method was 4,290±0.029 mg of quercetin equivalent/g fraction. There was a positive and strong correlation between the total flavonoid content of these extract and fractions with their macrophage phagocytosis capacity (Pearson correlation coefficient of 0,781) and showing linear relationship y=4,721x+19,663; R2=0,61.

Abstract

Cancer is a group of diseases that arise when a cell or group of cells that regulate out of control growth. Green algae (Spirogyra sp.) is one of the medicinal plants used in traditional medicine for the treatment of cancer. Green algae (Spirogyra sp.) has active substances such as melatonin. Melatonin which is a compound that has been examined by researchers world as anticancer drugs and antioxidants. This study aims to determine the effect of ethanol extract of green algae (Spirogyra sp.) on the expression of COX - 2 in T47D cells. Green algae was extracted using a Soxhlet apparatus with 96 % ethanol. This study used three groups: control cells , ethanol extract of green algae concentrations 247.668 ug / mL and 123.834 mg / mL. To ensure the expression of caspase-3 and Bcl-2, test was conducted using indirect immunocytochemistry. Observations were carried out using a light microscope to see and count the percent expression of COX - 2. The results obtained showed significant difference, decreased expression of COX - 2 in each group. It can be concluded that the ethanol extract of green algae (Spirogyra sp.) decreased COX - 2 expression in T47D cells.

Abstract

Hypercholesterolemia is a disease associated with high levels of cholesterol and LDL levels in the blood. Utilization of the commercial drugs can be given; however apart from the expensive price, adverse side effects might occur. It makes people choose alternative medication with herbal medicine through the use of natural materials. This study aimed to determine the effect of the combination of mangosteen peel extract-extract of roselle calyx and mangosteen peel extract-extract the ant nest plant as lowering cholesterol and triglyceride levels in male rats. The method used in this study was a laboratory experimental method using device posttest only control group design (simple experimental design). This study used 25 male rats of Wistar strain, divided into 5 groups; Group I: group without treatment, group II: control group solvent (NaCMC 1%), group III: positive control group (Simvastatin), Group IV: combination group mangosteen peel extract (200 mg / kg) - extract of roselle calyx (250 mg / kg), group V: group combination of mangosteen peel extract 200 mg / kg) - extract anthill (270 mg / kg). Induction of cholesterol in rats using quail egg yolk (10 ml / kg). The results showed that there was no significant difference in cholesterol and triglycerides between the combination of both extracts of mangosteen peel with a positive control (p<0,05).
