Abstract

Telah dilakukan penelitian tentang kultur tunas Scoparia dulcis, Lindernia anagalis, Lindernia ciliata untuk mengetahui kemampuan penyerapan logam berat Pb, Cr dan Cd sebagai proses bioremediasi terhadap polusi logam berat.Tunas-tunas tersebut ditumbuhkan pada media Murashige-Skoog (MS) padat yang ditambah kinetin 2 ppm. Subkultur dilakukan terhadap semua jenis tunas pada umur 21 hari  kemudian dikultivasi pada media MS padat ditambah logam berat 0, 2 dan 5 ppm selama 30 hari. Semua tunas mampu menyerap Cr tetapi tidak mampu menyerap Cd, tunas Lindernia anagalis dan Lindernia ciliata mampu menyerap Pb.

Abstract

Penelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak wasbenzen dan metanolik empat jenis tumbuhan, yaitu Garcinia dulcis (Roxb.)Kurz, Blumea mollis (D.Don) Merr., Siegesbeckia orientalis L.,dan Salvia riparia H.B.K hasil koleksi dari TNGM berdasarkan metode penangkapan radikal bebas dengan DPPH dan mengetahui profil kromatografi lapis tipisnya. Ekstrak dibuat dengan proses maserasi bertingkat dengan penyari wasbenzen dan metanol, uji aktivitas antioksidan dengan metode DPPH serta analisis kualitatif dengan kromatografi lapis tipis (KLT) untuk tumbuhan dengan aktivitas antioksidan tertinggi. Hasil penelitian menunjukkan bahwa diantara keempat ekstrak metanolik, ekstrak Garcinia dulcis (Roxb.) Kurz mempunyai aktivitas antioksidan tertinggi dengan nilai IC50 38,613µg/mL. Untuk ekstrak wasbenzen, ekstrak Garcinia dulcis (Roxb.) Kurz juga mempunyai aktivitas tertinggi dengan nilai IC50 139,381µg/mL. Analisis kualitatif ekstrak metanolik Garcinia dulcis (Roxb.) Kurz dengan kromatografi lapis tipis (KLT) menunjukkan bahwa ekstrak metanolik Garcinia dulcis (Roxb.) Kurz mengandung senyawa golongan fenol dan flavonoid.

Abstract

Taman Nasional Gunung Merapi menyimpan berbagai keanekaragaman hayati yang belum dimanfaatkan. Penelitian ini bertujuan untuk menguji aktivitas antioksidan empat tumbuhan anggota suku Asteraceae, Rubiaceae dan Polygalaceae. Uji aktivitas antioksidan menggunakan metode penangkapan radikal DPPH (2,2-difenil-1-pikrilhidrazil) dengan alat spektrofotometer. Sampel diekstraksi secara bertingkat dengan menggunakan wasbenzen dan metanol. Uji penangkapan radikal DPPH menunjukkan bahwa ekstrak wazbenzen Cynara scolimus L. memiliki IC50 sebesar 496,16g/mL; Artemisia china L. sebesar 407,86g/mL; Borreria repens DC. sebesar 984,04 g/mL dan Polygala paniculata L. sebesar 633,34 g/mL. Ekstrak metanol Cynara scolimus L. memiliki nilai IC50 sebesar 334,94 g/mL; Artemisia china L. sebesar 199,99g/mL; Borreria repens DC. sebesar 402,28 g/mL dan Polygala paniculata L. sebesar 77,00 g/mL . Analisa mengunakan KLT menunjukkan bahwa ekstrak metanol Polygala paniculata L. mengandung flavonoid.

Abstract

This study was conducted to determine antioxidant activity of methanolic extract and wasbenzen extract of four plants species: Garcinia dulcis (Roxb.) Kurz, Blumea mollis (D.Don) Merr., Siegesbeckia orientalis L. and Salvia riparia H.B.K. These four plants species were collected from TNGM, based on DPPH free radicals capturing and the profile of its thin layer chromatography. Sample was extracted by gradual maceration process with wasbenzen and methanol. Antioxidant activity was analyzed by DPPH and qualitative analysis was done by thin layer chromatography (TLC) to determine the highest antioxidant activity. Antioxidant activity was showed by IC50 values and qualitative analysis of the data is presented in the chromatogram. The results shows that the highest antioxidant activity was the extract of Garcinia dulcis (Roxb.) Kurz  for both methanol and wasbenzene extract with IC50 value 38.613µg/mL and 139.381µg/mL respectively. Qualitative analysis of methanolic extract of Garcinia dulcis (Roxb.) Kurz by thin layer chromatography (TLC) showed that the extract contain of phenols and flavonoids.

Abstract

The research about shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata has beeen done. The aim of this research is to know ability of shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata to adsorb heavy-metal Pb, Cr and Cd as bioremediation process for heavy metal pollutant. Scoparia dulcis, Lindernia anagalis, and Lindernia ciliata shoots are inoculated in Murashige-Skoog (MS) semi-solid medium with added 2 ppm of kinetin as growth factor. Shoots are subcultured at 21 days then they are cultivated in MS semi-solid medium with added heavy-metal Pb, Cr and Cd during 30 days. Scoparia dulcis, Lindernia anagalis, Lindernia ciliata able to adsorb Cr but they can not adsorb Cd. Scoparia dulcis is not able to adsorb Pb.

Abstract

A morphological diversity analysis of Scoparia dulcis by macroscopy and microscopy has been conducted. The aim of this research is to determine the morphological and anatomical character of S. dulcis which grow in Opak watershed, Special Region of Yogyakarta. Samples were taken from Sleman (Cangkringan, Ngemplak Kalasan, and Prambanan) and Bantul (Imogiri and Pundong) region. Selection of sampling location is based on altitude difference. The samples are the third leaf order from the tip of the stem. They were measured in length and width. Microscopy analysis was performed by observing the number of glandular trichomes, palisade cells in the epidermis and stomata in the upper and lower epidermis. The data obtained were analyzed using principal component analysis (PCA). The results show that based on morphological and anatomical assessment, there were found 3 clusters of S. dulcis, i.e. Cangkringan-Ngemplak-Prambanan, Kalasan, and Imogiri-Pundong cluster.

Abstract

Sidaguri (Sida rhombifolia) is one of the herbal components used in traditional medicine. The application of chemometrics in the standardization of herbal medicine is common. The objective of this study was to classify Sidaguri from different regions based on FTIR spectra with chemometrics of principal component analysis (PCA) and to correlate the antioxidant activities with FTIR spectra using the multivariate calibration of partial least square regression (PLSR). The extraction of Sidaguri powder was performed using ultrasound-assisted extraction (UAE) at optimum conditions. The obtained extracts were subjected to antiradical scavenging activities using DPPH (2,2’-diphenyl-1-picrylhydrazyl) and ABTS (2,2′-azinobis-3-ethylbenzothiazoline-6-sulfonic acid) radicals. The PCA result shows that Sidaguri from different regions could be separated using 14 wavenumbers of FTIR spectra based on the PCA's loading plot. PLSR regression using the second derivative FTIR spectra at wavenumbers of 3662–659 cm–1 could predict radical scavenging activities (RSA) of Sidaguri with R2 values of 0.9636 and 0.9024 for calibration and validation models, with RMSEC and RMSEP values of 1.45% and 2.65%, respectively. It can be concluded that FTIR spectra treated by PCA were reliable for classifying Sidaguri from different regions. At the same time, PLSR was accurate and precise enough to predict the RSA of Sidaguri.

Abstract

Sidaguri atau Sida rhombifolia L. telah digunakan oleh masyarakat sebagai obat tradisional, yaitu: penghilang rasa nyeri, radang, asam urat, penyakit kuning, muntah darah, dan sakit gigi. Faktor yang mempengaruhi produksi kandungan metabolit sekunder adalah kondisi lingkungan seperti tempat tumbuh, iklim, interaksi intra dan inter-spesifik serta waktu panen. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan serta kadar flavonoid dan fenolik total ekstrak sidaguri berdasarkan tempat tumbuh. Sampel diekstraksi dengan ultrasonic-assisted extraction (UAE) dan diuji penangkapan radikal bebas menggunakan metode 2,2’-difenil-1-pikrilhidrazil (DPPH) dan 2,2’-azino-bis(3-diethyl4benzothiazoline-6-sulfonic acid) (ABTS), serta penetapan kadar flavonoid total dan kandungan fenolik total. Hasil uji aktivitas antioksidan sidaguri dari Cangkringan dan Ngemplak memiliki aktivitas antioksidan tertinggi baik pada metode penangkapan radikal DPPH dengan nilai IC50 masing-masing sebesar 0,400±0,004 dan 0,403±0,004 mg/mL, maupun metode ABTS dengan nilai TEAC masing-masing sebesar 40,733±0,240 dan 35,598±0,153 mg/g serbuk sidaguri, dengan kadar flavonoid total masing-masing sebesar 10,095±0,068 dan 12,066±0,025 mg/g serbuk sidaguri dan kadar fenolik total masing-masing sebesar 56,45±0,068 dan 31,502±0,025 mg/g serbuk sidaguri.
