Abstract

Diabetes mellitus type 2 was induced by high fat diet and fructose. The insulin resistance and hyperinsulinemia compensatory can be measured by index homeostatic model assessment – insulin resistance (HOMA-IR). Andrographis paniculata (Burm. f.) Nees is a traditional plant can be used to treat diabetes mellitus and the main active compound of this plant is Andrographolide. The objective of this study is to evaluate the effect of purified extract of Andrographis paniculata (Burm. f.) Nees and andrographolide on HOMA-IR index of High fat diet and fructose induced Wistar Rats. This study is quasi-experimental and data analysis was using Kruskal-Wallis test. The result showed that purified extract of Andrographis paniculata and andrographolide decreased HOMA-IR index. Purified extract of Andrographis paniculata 1303,8 mg/kg decreased HOMA-IR index by 82,05 %.

Abstract

Objectives To evaluate the effect of Curcuma xanthorrhyza Roxb extract, ginger, soybean and shrimp shell in combination compared to diclofenac sodium to liver and kidney function of patients with osteoarthritis. Methods twenty one subjects randomly allocated to different groups (Trial and control = diclofenac group). The trial group were treated by the combination of 50 mg temulawak extract, 100 mg ginger, 50 mg soybean and 100 mg shrimp shell three times daily while the control groupwere treated by 25 mg diclofenac sodium three times daily for a couple of week. Blood sample was obtained before and after two weeks therapy to determine the liver enzymes SGOT and SGPT and also serum ureum and creatinin in order to monitor the kidney function. The study was a prospective randomized open end blinded evaluation (PROBE). Result both trial and diclofenac group show significantly increase of SGOT levels (p<0.05) but the difference change among these groups are not significant (p=0.312). In the trial group there was decreasing of SGPT level (p=0.871) whereas there was an increase in level of SGPT in diclofenac group (p=1.000) although the difference between these two was not statistically significant (p=0.912). The change of serum ureum and creatinin level were not significantly different in trial group compared to diclofenac (p>0.05). In the trial group there was decreasing of serum ureum level although not significant (p=0.214). Both trial and diclofenac group showed no statistically significant increasing of creatinin serum levels (p>0.05). Conclusion there is no statistically difference effect between using the combination of temulawak extract, ginger, soybean and shrimp shell compared to diclofenac sodium toward liver and kidney function of osteoarthritis patients during this two weeks treatment.

Abstract

Osteoporosis is a disease characterized by low bone mass and structural deterioration of bone tissue. Estrogen deficiency causes loss of bone mineral density which causes osteoporosis. Phytoestrogen is a potential alternative of estrogen that can be used as Hormone Replacement Therapy on osteoporosis with minimum side effects. Many edible plants contain phytoestrogens that are believed to promote bone health. Abelmoschus manihot (L.) Medik has been known as a plant that is empirically used in the traditional medicine and has potency to prevent osteoporosis. The aim of this research is to determine whether the Abelmoschus manihot (L.) Medik leaves have the potential to increase bone formation in an in vitro assays using preosteoblast cell line MC3T3-E1. The results showed an increasing activity of Alkaline phosphatase using confocal laser scanning microscopy technique.

Abstract

Kandungan karotenoid yang terdapat dalam buah-buahan dan sayuran, berperan penting dalam mencegah penyakit manusia, termasuk penyakit kardiovaskuler, kanker dan penyakit kronis lainnya. Namun, banyak metode yang digunakan dalam penentuan komposisi kimia khususnya untuk bahan makanan membutuhkan waktu yang lama serta biaya cukup mahal. Saat ini sejumlah teknik instrumentasi telah dikembangkan untuk menentukan kandungan kimia dengan cepat. Salah satu contoh adalah teknik pengukuran dengan menggunakan spektroskopi NIR. Dalam penelitian ini, spektroskopi NIR digunakan untuk analisis pendugaan karotenoid ekstrak kasar buah pisang tongkat langit (Musa troglodytarum) yang panjang dan pendek. Hasil yang diperoleh menunjukkan pisang tongkat langit mengandung β-karoten, sebab pada area bilangan gelombang 5000 hingga 5500 cm-1 terlihat adanya kemiripan antara pola spektrum β-karoten marker dengan pola spektrum pigmen karotenoid pisang tongkat langit.

Abstract

The present of chlorophyll is not expected in the isolation process of plant active constituent. Electrocoagulation is a potential dechlorophyllation method. This research aims to know the effectivity of electrocoagulation in the dechlorophyllation process of Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract as well as the effect to the total phenolic content. Electrocoagulation are performed using copper, silver, aluminum and iron plates as the electrode. Dechlorophyllation by extraction using n-hexane is performed as reference. The % absorbance of chlorophyll and % of total phenolic content of dechlorophyllated samples are measured spectrophotometrically. Electrocoagulation process reduces % absorbance of chlorophyll in Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract. The more the duration of electrocoagulaton process the lower the % absorbance of chlorophyll obtained. % absorbance of chlorophyll of the electrocoagulated extract according to paired t-test (P=0.95) are significantly different with the previous. One way ANOVA continued with LSD (P=0.95) shows that the % absorbance of chlorophyll from the electrocoagulated extract are significantly different with those extracted with n-hexane. Electrocoagulation process reduces total phenolic content along with duration of electrocoagulation.

Abstract

Premature aging skin condition characterized by dry, scaly, rough and accompanied by the appearance of wrinkles and dark spots or blemishes has become to be a women fearing thing, especially among women in reproductive age. Factors causing premature aging are internal factors (health, stamina, stress and hormonal changes) and external factors (free radicals, sunlight and pollutants). Free radicals can be prevented by antioxidant compounds either synthetic or natural materials. Tomato is one of the natural antioxidants. The most antioxidant content in tomato is lycopene. Tomato is used in the form of tomato juice made by extraction method. Tomato in their use directly is less effective so that tomato is made in the form of cream. Cream compositions were made by variation concentration of tomato juice (5%, 10%, 15%, and 20%). The antioxidant activity was tested by DPPH method. Physical stability parameters tested in this research were homogenity, viscosity, dispersive power, adhesion, cream separation ratio and pH. Data were analyzed by Kolmogorov-Smirnov test, one way ANOVA test and Tukey test. Organoleptic test data, the physical properties of the cream, and the type of emulsion were analyzed descriptively .The results showed that tomato juice has antioxidant activity with IC50 value 2,69%. The increase in the concentration of tomato juice gave significantly different results in all formulas except the formulas III and IV that were not significantly different. The IC50 values cream of tomato juice were 9,12%; 708%; 3,61%; and 2,85% respectively for the cream with tomato extract concentration of 5%, 10%, 15%, 20%. This showed that the higher the concentration of tomato juice, the higher the antioxidant activity as indicated by the lower of IC50 value. The increase in the concentration of tomato juice as active ingredients provided significantly different results for difference antioxidant activity, color, and smell, but did not cause significant differences in the consistency, viscosity, dispersive power, time and pH.

Abstract

Estrogen deficiency causes various health problems in postmenopausal women, including osteoporosis. Phytoestrogen emerged as a potential alternative of estrogen with minimum side effects. Green clover (Marsilea crenata Presl.) is a typical plant in East Java which suspected contains estrogen-like substances. The aim of this research was to report the phytochemical properties of M. crenata using GC-MS as a preliminary study. M. crenata leaves were dried and extracted with n-hexane, then separated using vacuum column chromatography to get four fractions, after that the n-hexane extract and four fractions were identified with GC-MS. The results of GC-MS analysis showed some compounds contained in M. crenata leaves like monoterpenoid, diterpenoid, fatty acid compounds, and other unknown compounds. The results obtained in this research indicated a promising potential of M. crenata as medicinal plants, especially as antiosteoporotic agent.

Abstract

Studies have been conducted to analyze flavonoids and tannins using microscopy-microchemical methods. Materials used for the analysis of flavonoid were orange peel, sonchi leaves, and orthosiphon leaves, with N. exaltata as negative control. While guajava leaves, guazama leaves, and areca seed were used for the analysis of tannins, with ginger rhizome as negative control. The study was conducted in a way of material sliced crosswise, subsequently given the identification reagent for flavonoids (ammonia, NaOH, AlCl3, sitroborat). On the other hand, using FeCl3, fast blue salt, and prusian blue for tannins. Having examined under a microscope, the area which gave a positive reaction to the reagent used, indicate the test materials contained compounds that were analyze. The results showed that this microscopy-microchemical method can be used to analyze the presence of flavonoids / tannins in the test material. Presence of flavonoid compounds will give yellow color with ammonia, NaOH, AlCl3, sitroborat reagent, while blue to black for the tannins with FeCl3, red with fast blue salt, and blue with prusian blue.

Abstract

Tuberculosis (TB), in which caused by pathogenic bacteria, Mycobacterium tuberculosis, has become the major causes of death among all of infectious diseases. The increasing incidence of multidrug-resistant tuberculosis (MDR-TB) and extensively drug-resistant tuberculosis (XDR-TB) has created a need to discover a new antituberculosis drug candidate. The aim of this study was to screen extract and fractions of Tinospora crispa for activity against Mycobacterium tuberculosis H37Rv using the Microplate Alamar Blue Assay (MABA) method. T. crispa extract was prepared by maceration in ethanol (96%) and antituberculosis activity was carried out using MABA method. The result of this study showed that ethanolic extract of T. crispa exhibit antituberculosis activity with minimum inhibition concentration of 12.5 mg/ml.

Abstract

Murraya paniculata [L.] Jack. (Kemuning) is a plant that grows widely in some areas of Indonesia. Studies related to this plant have been widely explored especially isolation of its active compounds. The plant contents several active compounds such as flanovoids. In the study, three flavonoid isolated from M. paniculata were evaluated for their effect on histamine release from mast cells (RBL-2H3 cells). These compounds were 3,3’,4’,5,5’,6,7,8-octamethoxyflavone; 3,3’,4’,5,5’,6,7-heptamethoxyflavone and 3, 3’, 4’, 5, 5’, 7–hexamethoxyflavone. The histamine inducers used in the study were DNP24-BSA dan thapsigargin, inducing the histamine release immunologically and non-immunologically, respectively. In the study, heptamethoxyflavone and hexamethoxyflavone did not influence the histamine release from mast cells significantly. However, octamethoxyflavone could increase the histamine release from RBL-2H3 cells in absence and presence the histamine inducers. The flavanoid could increase the release of histamine up to 50 %. Based on the results, polymethoxy moieties at the structure of flavonoid have a significant role to emerge the histamine-release stimulating effect from mast cells.
