Abstract

The leaves of Eupatorium odoratum L. have been used traditionally to cure abscess due to infectious diseases, leech bite and wound healing.  It was suspected that the leaves contain antibacterial compounds. The dried powder of E. odoratum leaves is extracted by Soxhlet using washbenzene and followed by methanol. Both extracts  are tested to determine their antibacterial activity by using diffusion test.  The active extract is fractionated by vacuum liquid chromatography. The active compound (2.3) obtained from the active fraction is purified by using preparative thin layer chromatography.  The UV-Vis spectrum of (2.3) (chloroform) showed λmax at 291nm. The IR spectrum of the active compound (2.3) (KBr) .0displayed signals (-OH), (-CHalifatik), (-CHalifatik), (-C=O), (C=C), (CH2), (CH3), (epoksi), and (C-O). 1H-NMR spectrum (CDCl3) showed signals at δ 1.26 ( 3H,s); 3.65 (3H,s); 3.82 (3H,s); 3.93 (3H,s); 4.53 (1H,d); 5.0 (1H,d); 5.7 (1H,d); 6.06 (1H,d); 6.14 (1H,d); 6.93 (1H,d); 7.05 (1H,dd); dan 7.16 (1H,d) ppm. This spectrum is suspected to be an epoxide chalcone type of compound. The active  compound (2.3) at minimum loading of 250 µg showed bacterial growth inhibition on S. aureus and E. coli at diameter 9.5 and 7.2 mm respectively.

Abstract

Hibiscus flower (Hibiscus rosa-sinensis L.) is efficacious in cough remedies. This flower has many variations , one of which is a red flower with a crown subject.  Ethanolic extract of the hibiscus flower has been studied and shown to have mucolytic activity. Fractionation will be closer in the process of finding active compounds. This study aimed to determine the effect of fractions containing alkaloids of red hibiscus flower varieties subject as mucolytics in vitro and determine the range of concentrations of the fractions equivalent to acetylcysteine effect of 0,1 %. This study included maceration, fractionation by VLC, the identification of fractions containing alkaloids, and mucolytic activity test fractions containing alkaloids. Mucolytic activity assay performed in vitro to decrease the viscosity of mucus cow. Test solutions were made with concentrations of fractions 0,4;0,6; and 0,8 %. Acetylcysteine 0,1 % was used as a positive control. Viscosity values were analyzed statistically using one -way ANOVA test , followed by the LSD test level of 95 % to determine differences between treatment groups. The results showed that the fraction containing alkaloids of hibiscus flower with a concentration of 0,4 % has not shown mucolytic activity, whereas at a concentration of 0,6 % and 0,8 % has been demonstrated in vitro mucolytic activity. Fractions containing alkaloids of hibiscus flower with a concentration of 0,6 % and 0,8 % have mucolytic activity equivalent to 0,1 % acetylcysteine in vitro.

Abstract

Green tea extract cream contains epigallocatechin gallate (EGCG) as the active ingredient for anti-inflammatory. Epigallocatechin gallate is easyly oxidized and able to reduce its effectivity as an anti-inflammatory. Therefore, an addition of antioxidants in order to increase its stability is required. The purpose of this study was to determine the effect of adding the antioxidant Vitamin C on the effectivity of green tea extract as an anti-inflammatory. This study uses 6 groups of male mice strain BALB/C which were given treatment as follows: normal control, negative control, base cream, green tea extract (0.2%), Vitamin C cream (1%) and green tea extract cream with addition of Vitamin C. The anti-inflammatory activity was evaluated based on the expression of COX-2, inflammatory cells and the thickness of the epidermis in the skin tissue of mice after given crotton oil (0.1%) on the back for the induction of inflammation. After treatment cream for 3 days, mice were sacrificed for histopathological tissue preparations made with hematoxylin eosin staining and immunohistochemistry COX-2. Data were analyzed statistically with one way Anova followed by t-test to determine differences between groups at a significance level of 0.05. The test results indicate that cream of green tea extract is higher in decreasing inflammatory parameters in comparison with cream of Vitamin C, except in the thickness of epidermal parameter. Green tea extract cream with the addition of Vitamin C is higher in reducing inflammatory parameters than cream of green tea extract or cream of Vitamin C. The decline percentage of cells that express COX-2, inflammatory cells and the thickness of the epidermis in the each of groups were cream of green tea extract:57.95%;53.75%;34.83%, cream of Vitamin C:48.76%;34.96%;34.27%, cream of green tea extract and Vitamin C:61,89%;65,54%;46.30%, respectively. Based on the results of this study, it can be concluded that anti-inflammatory activity of green tea extract cream increased due to the addition of 1% vitamin C as an antioxidant.

Abstract

Taman Nasional Gunung Merapi (TNGM) has various biodiversities which need to be explored. This study aims to analyze antioxidant activity four plants from Asteraceae, Rubiaceae and Polygalaceae which were collected from TNGM. Antioxidant activity is analyzed by DPPH radical capturing method using spectrophotometer. Sampel is extracted gradually by wasbenzen and methanol. DPPH radical capturing analysis shows that IC50 score of Cynara scolimus L. wasbenzen extract is 496,16g/mL; Artemisia china L. 407,86g/mL; Borreria repens DC. 984,04 g/mL and Polygala paniculata L. 633,34 g/mL. The IC50 score of methanol extract Cynara scolimus L. is 334,94g/mL; Artemisia china L. 199,99g/mL; Borreria repens DC. 402,28 g/mL and Polygala paniculata L. 77,00 g/mL . Thin layer chromatography analysis shows that methanol extract of Polygala paniculata L. contains flavonoid.

Abstract

Banana is one of Indonesian food which has many pharmacological activities. Pisang kapas (Musa paradisiaca L.), one of Indonesian banana, is well known empirically as laxative, so it can be used in the treatment of constipation. It has been reported that both of chloroformic and ethanolic extract of pisang kapas have antidiabetic activities. However, fractination of those are not yet performed and report about their antidiabetic activities are not yet available. Here we investigated the effect of water-dissolved fraction of ethanolic extract of cotton banana to reduce the blood glucose level in rat model and identified its active ingredient. The oral glucose tolerance method with 60 min glucose loading time was used to induce hyperglicemic condition in rats, either in control group or treatment group. Glibenclamid (positive control), sodium carboxymethyl cellulose (negative control) and the fraction were orally given to the rats in a single dose. To measure blood glucose level photometrically, enzimatic reaction by using diagnostic reagent Glucose GOD FS is applied. Paper chromatography was utilized to identify the ingredient of the fraction qualitatively.  The fraction in dose of 0.25 g/kg reduced blood glucose level by 22.28 ± 0.76 %. The fraction contained reductants in addition to glucose, amino acids, and the acids in plants.

Abstract

The presence of free radicals produced inside the cells is thought to cause a range of diseases such as degenerative diseases. Antioxidant properties of carrot root (Daucus carota L.) have been reported elsewhere to inhibit the formation of these free radicals, but the compounds responsible for antioxidant activity inside its leaves have not yet determined. In Indonesia, Daucus carota L. leaves are regarded as waste product after harvesting the root. This study was conducted to isolate and identify the compound having antioxidant activity from the leaves of D.carota. n-Hexane, ethyl acetate, and methanol extracts were examined for free radical scavenger activity by using DPPH method. The n-hexane extract showing free radical scavenger activity was solid-solvent partitioned, fractionated using vacuum liquid chromatography and tested for free radical scavenger activities. The active compound was isolated by using preparative thin layer chromatography and its purity was determined by TLC with three mobile phase systems and two dimensional TLC. Based on spectroscopy UV-Vis, IR as well as 1H-NMR (CDC13) data, the active compound was suggested to be xanthophyll derivate, lutein. The compound at concentration of 0.616, 1.025, and 2.05 ppm had antioxidant activities of 0.94 ± 0.05; 18.53 ± 0.15; and 49.07 ± 0.86, respectively.

Abstract

Costus speciosus has been reported to have biological activity among antidiabetic, antihyperlipidemia, antioxidants. One of the chemical constituents that is responsible for some of the biological activity is diosgenin. This study aimed to determine diosgenin content of Costus speciosus water extract (CS). Water extract of CS was obtained with infundation method. The extract was dried using freeze dryer. The assay of diosgenin was performed by HPLC method. HPLC system used: LiChrospher® 100 RP-18 endcapped (5 µm) column length 25cm, colimn id 4 mm as stationary phase, acetonitrile-water (9: 1 v/v) as mobile phase, flow rate 2mL/min and UV detection at 205 nm. HPLC method developed was linear in the range 20-200 μg mL with R = 0.9999 ; slope = 6423.7; and intercept = -4280.5; LOD = 2.14 ug/mL and LOQ = 6.50 mg/mL; and recovery 100.63 %. Diosgenin levels in CS was 0.279 %. The developed HPLC method is relatively simple, rapid, sensitive, and accurate to determine the content of diosgenin in water extracts of Costus speciosus.

Abstract

The research about shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata has beeen done. The aim of this research is to know ability of shoot culture of Scoparia dulcis, Lindernia anagalis, Lindernia ciliata to adsorb heavy-metal Pb, Cr and Cd as bioremediation process for heavy metal pollutant. Scoparia dulcis, Lindernia anagalis, and Lindernia ciliata shoots are inoculated in Murashige-Skoog (MS) semi-solid medium with added 2 ppm of kinetin as growth factor. Shoots are subcultured at 21 days then they are cultivated in MS semi-solid medium with added heavy-metal Pb, Cr and Cd during 30 days. Scoparia dulcis, Lindernia anagalis, Lindernia ciliata able to adsorb Cr but they can not adsorb Cd. Scoparia dulcis is not able to adsorb Pb.

Abstract

A morphological diversity analysis of Scoparia dulcis by macroscopy and microscopy has been conducted. The aim of this research is to determine the morphological and anatomical character of S. dulcis which grow in Opak watershed, Special Region of Yogyakarta. Samples were taken from Sleman (Cangkringan, Ngemplak Kalasan, and Prambanan) and Bantul (Imogiri and Pundong) region. Selection of sampling location is based on altitude difference. The samples are the third leaf order from the tip of the stem. They were measured in length and width. Microscopy analysis was performed by observing the number of glandular trichomes, palisade cells in the epidermis and stomata in the upper and lower epidermis. The data obtained were analyzed using principal component analysis (PCA). The results show that based on morphological and anatomical assessment, there were found 3 clusters of S. dulcis, i.e. Cangkringan-Ngemplak-Prambanan, Kalasan, and Imogiri-Pundong cluster.

Abstract

The aim of this research to investigate the hepatoprotective and nephroprotecitve effects of Avocado seeds against carbon tetrachloride in rats. The rats was treated with the infusion and decoction of avocado seeds at doses 360.7, 642.1, 1142,9 mg/kg per oral once in a day for 6 days and carbon tetrachloride (2mL/kg) was given on the 7th day. Blood sample from all groups was obtained by sinus orbitalis after 24 hours application for the serum transaminase and creatinine. The increasing of serum transaminase and creatinine in carbon tetrachloride-treated rats, were significantly decreased (p<0.05) with both pretreatment of infusion and decoction of avocado seeds. Based on the research, it can be concluded that infusion and decoction of avocado seeds have a potent protective action upon carbon tetrachloride-induced hepatic and renal damage in rats.
