Abstract

The form of liposome formulations is one of strategies to overcome the limitations of anthocyanin extract stability of roselle calyx (Hibiscus sabdariffa L.). In this study, antioxidant comparative analysis of antioxidant activity and total anthocyanin content from rosella extract and its liposomes form was conducted. Preparation of extract made by maceration using the best solvent consisted of methanol : formic acid (97:3) (v/v). From this procedure, the obtained extract was about 26.40% and antioxidant activity was 19.99%. Liposome formulations of methanol extract from roselle calyx were prepared by reverse phase method using rosella extract as aqueous phase and egg lecithin as the lipid phase. The stability of exctract and liposome form of rosella calyx was compared. The stability study showed that there was a reduction of antioxidant activity (DPPH test) 11.25 % for the roselle extract, while the liposome form was 0.93% after 30 days storage in low temperature (0-50C) and saturated nitrogen. In addition, total anthocyanin content (pH differential method test) decreased approximately 39.12 % for  the roselle extract and 0.72% for the liposome form. More over, the liposome had lower level of irritation than the form of extract. Therefore, it can be concluded that anthocyanin encapsulated by liposome was more stable, safe, and effective than the anthocyanin of the rosella calyx extract.

Abstract

Oxidative stress has been known to contribute to Alzheimer’s disease. Acetylcholinesterase (AChE) enzyme may lead to Alzheimer’s disease as a neurotransmitter. Antioxidants may have protective activities against oxidative damage and Alzheimer’s disease. Acetylcholinesterase inhibitors also can be used in the treatment of various neurological disorders for management of Alzheimer’s disease. This study aimed to determine antioxidant and anticholinesterase effects of Garcinia picrorrhiza Miq. fruit extract (GpKar) and its compounds, xanthone. Antioxidant activity was measured by H2O2 scavenging inhibitory activity, while anticholinesterase activity was measured using modified Ellman method. GpKar has higher H2O2 scavenging inhibitory activity (IC50= 967.28 µg/ml) compared to xanthone (IC50= 1198.95 µg/ml). In the anticholinesterase inhibitory activity, GpKar has lower activity (IC50= 70.25 µg/ml) compared to xanthone (11.80 µg/ml). In summary, GpKar has higher antioxidant activity but lower anticholinesterase activity compared to its compounds, xanthone. However, GpKar has potency as antioxidant agent to prevent Alzheimer’s disease.

Abstract

Ascorbic acid is an antioxidant which is found in rosella flower. Its production through conventional method needs long time and wide field, so an alternative method through development of callus invitro is conducted. Sucrose is a main carbon source in MS medium and this sugar plays role as precursor for the formation of ascorbic acid. This study was conducted to determine the effect of sucrose concentration on callus formation and production of ascorbic acid. Sucrose was added to the media at the concentration  of 20 g/L, 30 g/L, 40 g/L and 50 g/L. The culture was incubated for 42 days with twice subculture every 10 days.  Analysis of ascorbic acid was perfomed using iodometry. The results showed that the callus was grow better in sucrose of 20 g/L while the ascorbic acid production was better in sucrose 50 g/L.

Abstract

Disorders of carbohydrate metabolism can lead to diabetes mellitus. Carbohydrates  are metabolized in the gastrointestinal tract  into simple glucose and absorbed into the bloodstream and affected blood glucose levels. The absorption process is catalyzed by α- 1 ,4 - glycoside breaking bond  enzyme , namely α - amylase and α -1 ,6 - glycoside breaking bond  enzyme, namely α – glucosidase. They are found in the intestinal cells. Research had been conducted in an effort to develop an alternative treatment of diabetes mellitus by testing the ability of isolates of andrographolide in inhibiting α-amylase activity and α-glucosidase in vitro. Andrographolide isolates showed fairly good activity in inhibiting α-amylase ( IC50 = 1,.49 mg/mL) and weak in inhibiting α-glucosidase (IC50 = 38,86 mg/mL). Inhibition of α-amylase activity is evidence of one mechanism of andrographolide in reducing carbohydrate metabolism that can affect blood glucose levels and indicates that andrographolide is  a potential alternative medicine in addressing diabetes mellitus .

Abstract

Batang brotowali (Tinospora crispa (L.) Miers) diketahui mengandung beberapa senyawa fenolik yang mempunyai aktivitas sebagai antioksidan. Penelitian ini bertujuan menguji aktivitas penangkapan radikal 2,2-difenil-1-pikrilhidrazin (DPPH) oleh ekstrak etanolik batang brotowali dan fraksi-fraksinya. Ekstraksi batang brotowali dilakukan dengan alat Soxhlet dan pembuatan fraksi-fraksinya secara partisi cair-cair menggunakan alat Liquid-Liquid Continuous Extraction (LLCE). Pelarut-pelarutnya adalah n-heksana dan etil asetat. Adanya golongan senyawa antiradikal dideteksi  dengan metode Kromatografi Lapis Tipis (KLT) dan aktivitas penangkapan radikal DPPH. Ekstrak etanolik, fraksi etil asetat, dan fraksi air memiliki aktivitas antiradikal dan mengandung senyawa golongan kumarin dan flavonoid. Fraksi etil asetat (200 µg/ml) mampu menangkap 53,57 % radikal DPPH yang merupakan aktivitas antiradikal tertinggi dan diikuti oleh fraksi air, ekstrak etanolik, dan fraksi n-heksana.

Abstract

The snakehead (Channa striata) contained fatty acids omega-3 and omega-6 are useful as nutrients in accelerating the wound healing process. This study aims to determine the effectiveness of the oil phase extract ointment snakehead in the healing of acute wounds open stage II. Testing the effectiveness of wound healing using 6 groups: normal, gel bioplacenton (positive control), ointment base (negative control), ointments snakehead extract oil phase concentration of 10; 20 and 40% of the 4 rats with acute wounds open stage II and observation until day 16 injury. The area of the wound was measured with the image J program Macbiophotonic, calculated% healing power, and AUC values. A statistical test to the total AUC values per rat with SPSS for Windows 22.0 program using One Way ANOVA and Post-hoc LSD test. The results showed a significant difference in the negative group with ointment concentration of 20% (p <0.05). The treatment group phase ointment fish oil extract is effective for wound healing is best to have a concentration of 20% for percentage of wound healing power of 97.157% and the average AUC value of 865.683% of the day.

Abstract

Angiogenesis is a neovascularization process which its function is to supply nutrient and oxygen for cell survival. Pasak bumi is one of the Indonesian original plants having anti cancer activity. Extract of methanol, buthanol, kloroform, and water akar pasak bumi was proved to have cytotoxic effect on cancer cell culture. This research aim to know activity of antiangiogenic extract of metanol akar pasak bumi at chorioallantoic membrane of chicken embryo. Antiangiogenic activity test conducted by divide 8-9 days old egg embrio into 8 groups. Group I as paper disc control, group II as bFGF control, group III as bFGF control + DMSO 0.8% solvent, group IV, V, VI, VII, and VIII as  groups of treatment. Each groups of treatment were gave by bFGF 1ng/µL extract of metanol akar pasak bumi respectively with concentration 20, 40, 60, 80 and 100 µg/mL concentration. After 3 days incubation, then the chorioallantoic membrane macroscopic and microcospically analized. The antiangiogenic activity extract of metanol akar pasak bumi at chorioallantoic membrane bFGF induce at used concentration, that is start from 40 µg/mL concentration. The increases of the concentration also increase the prohibition activity to angiogenesis.

Abstract

Aedes aegypti is a vector for Dengue. The use of synthetic repellent cause several health issue. Zingiber officinale Roxb. “Cochin Ginger” essential oil (A) and Zingiber officinale Roxb. var rubrum essential oil (B) was used as a repellent for Aedes aegypti. Increasing of repellent activity was strived by formulating A and B with sesame oil and coconut oil as a base. The aim of this study was to compare repellent activity between A and B, to know the influence of oily base to repellent activity of A and B, to know the effective concentration of essential oil in oily base as a repellent.Isolation of essential oil was done by water and steam destilation. Quantitative and qualitative assays by GS-MS. Repellent activity was tested by three steps: initiation, effective consentration assay, and effective concentration of repellent activity. Essential oil on certain concentration (100%, 50% in the base, the effective concentration of SLD) and Lemon grass essential oil was applied on the surface of palm hand. The hand was put into 20x20x20 cm3 cage containing 25 female Aedes aegypti. The number of Aedes aegypti lied on the hand was noted to count protective capacity. First bite was counted as protective time. Analyse was done by One-way ANOVA and Post Hoc Test Tukey HSD (Homogeneous subsets) or Kruskal-Wallis and Mann-Whitney Test.  The result showed that A and B contain camphene, mirsene, 1,8-sineol, l-linalool, l-borneol, neral, geraniol, sitral, and α-kurkumen (only A) that quantitatively different. Statistic result showed that B has a greater repellent activity than A. Sesame oil base give a better effect on repellent activity of sesame oil compare than coconut oil. The effective concentration of Zingiber officinale Roxb. “Cochin Ginger” essential oil and Zingiber officinale Roxb. var rubrum essential oil in sesame oil and coconut oil was 75, 85, 85 and 90%, respectively.

Abstract

Antioxidant is an interesting topic due to their capability to inhibit free radical and prevent damage because of oxidative processes. Endophyt fungi is one of antioxidant compound resources in nature. The low yield to gain antioxidant compound from fungi challenges to look for the composition of media and optimalization of growth conditions. This research aimed to know the effect of medium condition in different carbon and nitrogen sources as well as initial pH towards antioxidant activity of endophyt fungi Bo.Ci.Cl.A3. Shaker fermentation was used on 120 rpm at room temperature for 14 days. The carbon sources were glucose, sucrose, and starch and nitrogen sources were NaNO3, NH4NO3, and yeast extract with initial pH at 5, 7, and 9. Ethyl acetate was used as extractor. The results showed that endophyt fungi can produce secondary metabolite as antioxidant at all variation of fermented media. The nitrogen source of yeast extract could increase antioxidant activity of endophyt fungi Bo.Ci.Cl.A3, while other sources such as nitrogen source, carbon sources, and different initial pH on the basal medium that were used did not give increasing antioxidant activity. The conclusion of this research was the substitution of nitrogen source with yeast extract (3 g/L) on the basal medium Czapek Dox’s Broth could increase antioxidant activity of endophyt fungi Bo.Ci.Cl.A3.

Abstract

This research aims to determine the effect of avocado seed starch as a binder to the physical characteristics of the granule that is flow rate, angle of repose and moisture content as well as to the physical characteristics of tablets that is uniformity of weight, hardness, friability and disintegration time. Root of Imperata cylindrical (Linn.) extract was obtained by remaceration process usingethanol 70 % as a solvent. Tablets were made by wet granulation method with increase the avocado seed starch concentration as binder tablet (FI: 10 %, FII: 20 %, and FIII: 30 %) then the physical characteristics of granule and tablets were tested. Data were analyzed theoretically by comparing toliterature and statisticaly using one way ANOVA with a level of confidence up to  95 %,  followed by Scheffe test. The results showed that the granule’s flow rate andangle of repose decrease byincreasing of avocado seed starch concentrations as a binder, while will increase the moist content of granules. Increasing of avocado seed starch concentration as a binder will increase the hardness and disintegration time of tablets as well, while friability of tablets are decrease and weight of tablets are more uniform.
